Actin microfilaments in paramecium: localization and role in intracellular movements.

Using heavy meromyosin (HMM) or the fragment S1 of myosin as probes for actin microfilaments, we studied their organization in Paramecium both by fluorescence and electron microscopy. In interphasic cells, HMM decorates (a) most prominently the periphery of nascent and young food vacuoles and their route during the early phase of their intracellular transit; (b) a thin meshwork radiating from the gullet throughout the cytoplasm; (c) a small area beneath the pore of contractile vacuoles and beneath the cytoproct when open to release food residues. Most of these HMM-decorated structures are in close contact with microtubular arrays. All HMM decoration disappears in dividing cells and in cytochalasin-treated cells. In vivo, the drug immediately blocks food vacuole formation but does not affect cytokinesis, cyclosis, contractile vacuole pulsation, defecation, or nuclear movements. The data show that, as in the cells of other organisms, actin microfilaments form defined arrays that undergo physiologically controlled cycles of assembly/disassembly. These arrays contribute (at least in the phagocytotic process) to diverse types of movement: constriction, membrane fusion, and migration of food vacuoles. However, aside from their massive concentration along the phagocytotic tractus, actin microfilaments are neither major structural components of Paramecium cytoplasm nor the only cytoskeletal components ensuring motility or contractility processes.

Sintering technique for the preparation of polymer matrices for the controlled release of macromolecules.

A new method for making polymeric systems for the controlled release of macromolecular drugs is described. The method consists of mixing drug and polymer (ethylene-vinyl acetate copolymer) powders below the glass transition temperature of the polymer and compressing the mixture at a temperature above the glass transition point. The macromolecule is not exposed to organic solvent during the fabrication process. Kinetic studies indicate that there is sustained release, and the bioactivity of macromolecules tested is unchanged throughout the sintering and release processes.

Factors influencing the success of in vitro fertilization for alleviating human infertility.

The program for in vitro fertilization at Bourn Hall began in October 1980. Various types of infertility have been treated during this time using the natural menstrual cycle or stimulation of follicular growth with antiestrogens and gonadotrophins. Follicular growth and maturation are assayed by urinary estrogens and LH, monitored regularly during the later follicular stage. Many patients had an endogenous LH surge; others needed an injection of HCG to induce ovulation. All oocytes were recovered by laparoscopy. Wide variations occurred in the time interval between the start of the LH surge and oocyte recovery and between oocyte recovery and insemination. Embryos taken between the one- and the eight-cell stage were replaced into their mother, no standard procedure being adopted for all patients. The results of all treatments including patient's responses during the follicular and luteal phases, oocyte recovery, fertilization, cleavage, replacement, implantation, abortion, and birth and the effect of factors such as replacing two or more embryos, maternal age, and previous obstetric history are described in detail. The incidence of implantation after embryo replacement improved from 16.5% initially to 30% currently. More than 118 babies have been born, and many pregnancies are continuing.

The application of the zona-free hamster egg test for the prognosis of human in vitro fertilization.

The zona-free hamster egg test was carried out using spermatozoa from 15 men which consistently failed to fertilize their wives' oocytes in vitro. Spermatozoa from nine of these men fertilized hamster eggs in vitro, indicating that positive results in this assay are an unreliable guide to human in vitro fertilization. Donor spermatozoa were needed to fertilize the wife's oocytes in three of these cases. Nevertheless, the proportion of hamster egg penetration was significantly lower compared with spermatozoa from 15 men who could fertilize their wives' oocytes in vitro. The hamster assay also failed to indicate the establishment of pregnancy.

A4: an antigenic marker of neural tube-derived cells.

The A4 monoclonal antibody was originally found to bind to the surface of the majority of neurons in rat CNS cultures, but not to PNS neurons or non-neural cells. It was subsequently shown to bind to immature oligodendrocytes and their precursor cells but not to the most mature oligodendrocytes. In the present study, we have used immunofluorescence assays on cell suspensions and cultures and on semi-thin, frozen tissue sections to show that protoplasmic and fibrous astrocytes and most ependymal cells are also A4+. Taken together, these results suggest that in adult rats A4 is expressed exclusively by cells of the CNS and that all cell types derived from the neural tube are A4+, at least at some time in their development. While neurons, astrocytes and ependymal cells continue to express the antigen in adults, most oligodendrocytes appear to lose it as they mature. The finding that macrophages in CNS cell suspensions and cultures are A4- suggests that microglial cells are not derived from the neural tube.

Benign cryptococcaemia.

The most familiar clinical presentation of cryptococcal disease is subacute lymphocytic meningitis, generally in immunocompromised patients. Isolation of Cryptococcus neoformans from the bloodstream has been regarded as a grave prognostic sign, but we have recently seen a patient with cryptococcaemia which had an unusually benign course.

A comparison of three therapeutic formats in the treatment of secondary orgasmic dysfunction.

The goal of the present study was to compare the effectiveness of three therapeutic formats: Standard Couple Therapy, Group Therapy, and Minimal Contact Bibliotherapy (self-help) in the treatment of 23 couples in which the wife was suffering from secondary orgasmic dysfunction. The results indicate that a cognitive-behavioral sex therapy program is clearly effective in changing a wide range of subjective satisfaction and behavioral measures, with concurrence of the husbands' and wives' data providing further strength to these findings. Differences in outcome which were elicited in the three experimental treatment conditions were mainly in favor of the Standard Couple condition. Since such differences were neither frequent nor great, practical issues related to increasing the effectiveness of less time-consuming treatment formats are discussed. In addition, the theoretical implications of using global versus specific therapy outcome criteria are explored.

Survey of human rotavirus propagation as studied by electrophoresis of genomic RNA.

Rotavirus RNA isolated from three different environmental sources during the winter of 1981-1982 was analyzed by polyacrylamide gel electrophoresis. Our results demonstrate the presence of novel RNA patterns, the coexistence of multiple electrophoretic patterns in rotavirus isolated from samples from a given ward, and that spread of a given electropherotype is limited.

Observations on the measurement and evaluation of endotoxemia by a quantitative limulus lysate microassay.

A quantitative microassay for bacterial endotoxin in whole blood, based on Limulus amebocyte lysate gelation and involving the use of a chromogenic substrate, was studied. The lower limit of detection was 10 pg/ml, and the mean level of recovery from plasma spiked with 1 ng of endotoxin/ml was 0.93 ng/ml. At clinically significant levels endotoxin was more easily recovered from plasma than from serum. Platelet binding had no adverse effect on the assay. Despite the use of conventional methods for the removal of nonspecific activators and inhibitors, simultaneous assay of six specimens spiked with the same amount of endotoxin resulted in variation in recovery by greater than 100%. These experiments may explain in part the conflicting results of studies of endotoxemia in humans.

Plasma concentrations of free 5 alpha-androstane-3 alpha, 17 beta-diol and related gonadal steroids during spontaneous and induced sexual maturation in the female rat.

Plasma concentrations of free 5 alpha-androstane-3 alpha, 17 beta-diol (3 alpha-diol), 5 alpha-androstane-3 beta, 17 beta-diol (3 beta-diol), 5 alpha-dihydrotestosterone (DHT) and of testosterone (T) were determined in female rats undergoing spontaneous sexual maturation or pregnant mare's serum gonadotropin (PMSG)-induced precocious ovulation. In the prepubertal period, 3 alpha-diol levels showed an inverse relationship with uterine weights and a significant drop between Days 21 and 26 of life. Postpubertally, 3 alpha-diol concentrations tended to rise again, to a lesser degree, in parallel with increments in plasma T. Concentrations of 3 alpha-diol were increased tenfold 48 h after the administration of PMSG, i.e., at a time when T and estradiol-17 beta also peaked. Concentrations of 3 beta-diol and DHT remained low throughout both experiments. In earlier studies, it was found that exogenous 3 alpha-diol suppresses reproductive function in the immature rat, but the high concentrations of endogenous 3 alpha-diol reported here for the PMSG model clearly do not interfere with ovulation. The observed constellations of plasma steroids are compatible with the postulate of a peripubertal shift in ovarian progesterone metabolism from 5 alpha-reduced C21 steroids to delta 4-3-ketosteroids.

Comparison of the sensitivity of three methods for the rapid identification of Cryptococcus neoformans.

The sensitivity of three methods for the rapid identification of Cryptococcus neoformans was compared. These were: direct microscopy of india ink preparations, acridine orange staining followed by fluorescence microscopy and detection of cryptococcal capsular polysaccharide antigen by latex agglutination. The overall limit of detection was 3.5 +/- 5.4 X 10(3) CFU/ml (mean +/- SD, n = 27). When different strains were studied, no single method was consistently superior. False positive results were rare (two of 162 observations, 1.2%) but there were eight false negatives (4.9%), five of which were with acridine orange. Tests such as these are an invaluable aid in the rapid diagnosis of cryptococcal meningitis but they cannot be relied upon to detect low grade infections.

Sympathetic skin response--a method of assessing unmyelinated axon dysfunction in peripheral neuropathies.

The sympathetic skin response (SSR) was measured in 33 patients with peripheral neuropathies and in 30 normal control subjects. Abnormalities of the response were correlated with clinical, pathologic, and EMG observations. The response was usually absent in axonal neuropathies, but present in demyelinating disorders. Abnormalities of the sympathetic skin response did not correlate well with clinical evidence of dysautonomia, but were a reliable indicator of disorders affecting unmyelinated axons.

Pharyngocutaneous fistula after laryngeal surgery: the role of the barium swallow.

Pharyngocutaneous fistula is one of the major complications following laryngeal surgery. Prior to 1979, patients undergoing laryngectomy at the University of Minnesota were not fed until 9 or 10 days postoperatively if unirradiated and 12 to 14 days if irradiated. Most fistulas were apparent by 14 days postoperatively, but occasionally a patient would develop a fistula as late as a month postoperatively. Starting in 1979, in an attempt to decrease the length of hospitalization as well as prevent fistula formation, routine barium-swallow videoesophagograms were obtained 7 days postoperatively in all laryngectomy patients. If the results were considered normal, the nasogastric tube was removed and oral feedings were started. Of a total of 45 patients undergoing wide-field laryngectomy, there were three clinical fistulas and three radiologic fistulas. Following this protocol, we shortened the average postoperative hospitalization by 2 days without an increase in the rate of fistulization.
