Ethanol and diet interactions in male rhesus monkeys.

Development of alcohol-related liver and other diseases appears to be modified by host and environmental factors that include diet and nutritional status, exposure to other drugs and toxins, and infection. The relative importance of alcohol toxicity and malnutrition in the induction of fatty liver and cirrhosis, the subject of this report, has been debated. Male rhesus (M mulatta) monkeys were fed purified liquid diets, adequate or marginally deficient in lipotropes (choline, methionine, and folate), containing ethanol to supply 40-50% of calories for 1.5-4.5 years. Controls, fed the diets with sucrose and fat isocalorically substituted for ethanol, grew well and were clinically normal. Ethanol-fed monkeys in both diet groups failed to gain weight and were slightly anemic, with mild derangements of serum electrolytes and small amounts of fat in their livers. None had fibrosis or cirrhosis until the severity of the lipotrope-deficiency was increased; then two of four deficient animals developed cirrhosis and one developed fibrosis. (The severe deficiency induced weight loss and fatty liver, but not fibrosis, in one of two controls.) We conclude that alcohol does not induce hepatic fibrosis or cirrhosis in rhesus monkeys fed a nutritionally complete diet, a result supported by studies in rats and another monkey, M radiata. Alcohol does induce cirrhosis when fed in combination with a lipotrope-deficient diet that is not, by itself, cirrhogenic.

Myxoma of bone in a nonhuman primate.

A myxoma of the mandibles in an adult female Macaca mulatta is described. The left mandible was markedly enlarged by the tumor, which extended from the symphysis to the temporomandibular joint. The tumor had extended a short distance across the symphysis into the right mandible. It was composed of a glistening mucinous material. Radiographs of the tumor showed a large expansile radiolucent lesion of the left mandible without soft tissue involvement. Light microscopy demonstrated capillaries, stellate shaped cells with processes up to 15 mu long, very few other cell types, and an amorphous ground substance. Electron microscopic examination revealed similarity to the myxoma of bone of man. The oxytalan fibers discovered in this myxoma have not been found in myxomas before and suggest the tumor is odontogenic in origin. The incidence of spontaneous neoplasia in general and that of spontaneous oral tumors in particular in nonhuman primates are reviewed.

Cutaneous irritation in the topical application of 30 antineoplastic agents to New Zealand white rabbits.

Of 30 antineoplastic agents studied for their primary irritation potential in rabbits, 9 showed some potential for irritation. Five of these 9 agents produced a significant dermal irritation. None of the irritation observed was considered to be irreversible skin damage. The study further showed a strong correlation between irritation observed by the Draize method and acute inflammation evaluated histopathologically. There was a tendency toward increased epidermal thickness of irritated skin sites. None of the agents produced gross or microscopically visible lesions in the internal organs observed.

Ulcerative dermatitis in rats with over fifteen generations of protein malnutrition.

1. Male and female rats with histories of up to twenty generations of protein malnutrition were found to be at a higher risk for the development of ulcerative dermatitis than rats maintained on a low-protein diet for one generation or in controls on an adequate-protein intake. 2. In all groups, female rats were more likely to have dermatitis than male rats. 3. Bacteriologic examination was performed in the intergenerationally malnourished and control animals; Staphylococcus aureus was isolated from the skins of animals in both groups, whether or not any lesion was present. In these two groups of animals, experimental inoculation with S. aureus produced dermatitis only in the malnourished animals.

Systemic fungal infections in cats.

Pneumonia and ileitis due to Aspergillus spp was diagnosed at necropsy in an adult female cat. In a 2nd cat, Aspergillus spp caused acute focal necrotizing pneumonia. A 3rd cat had severe necrotizing colitis caused by a phycomycete. Two of the cats had clinical signs and pathologic lesion compatible with feline panleukopenia, which probably increase their susceptiblity to fungal infection. The ability of Aspergillus spp and phycomycetes to invade tissue was probably enhance by prolonged treatment of the cats with antibiotics.

3-Aryl- and 3-hydroxy-3-aryloctahydropyrido[2,1-c][1,4]oxazines. Synthesis, stereochemistry, and central nervous system pharmacological actions.

A series of substituted 3-aryl- and hydroxy-3-aryloctahydropyrido[2,1-c][1,4]oxazines has been synthesized for purposes of investigating potentially useful CNS pharmacological actions of this novel heterocyclic system. The preferred conformation of the bicyclic system of the parent compounds, 1 and 2, has been shown to be trans with equatorial orientation of the 3-phenyl substituent in each compound. The various substituted aryl analogues of this series are depressants of motor activity in mice, and certain analogues exhibit significant anticonvulsant and appetite suppressant activity in test animals.

Hematologic and serum protein reference values of the Octodon degus.

Blood and serum from normal degus (Octodon degus) that ranged in age from 3 to 48 months were analyzed to determine reference hematologic and serum protein values. Both sexes were evaluated and were similar. The hematologic and serum protein values for males were: erythrocytes, 8.69 +/- 0.19 X 10(6) /microliter; packed cell volume, 42.1% +/- 0.59%; hemoglobin, 12.0 +/- 0.15 g/dl; leukocytes, 8.50 +/- 0.39 X 10(3)/microliter; neutrophil-to-lymphocyte ration, 40:60; and total protein, 5.70 +/- 0.20 g/dl. The hematologic and serum protein values for females were: erythrocytes, 8.94 +/- 0.16 X 10(6)/microliter; packed cell volume, 40.0% +/- 0.61%; hemoglobin, 11.7 +/- 0.17 g/dl; leukocytes, 8.20 +/- 0.36 X 10(3)/microliter; neutrophil-to-lymphocyte ratio, 40:60; and total protein, 5.62 +/- 0.18 g/dl. The hematologic and serum protein values for the degu were similar in some respects to values reported for guinea pigs and rats.

Mycotic enteritis in a chameleon and a brief review of phycomycoses of animals.

Rectal prolapse in an adult chameleon was surgically replaced. The animal was given tetracycline and dextrose orally, but became comatose 7 days postoperatively and was euthanatized. Necropsy revealed intussusception of the terminal portion of the colon. Phycomycotic hyphae accompanied by necrosis and a mixed leukocytic infiltrate were found in the area of intussusception.

Adverse effects of mouse hepatitis virus on ascites myeloma passage in the BALB/eJ mouse.

During experimental serial passage of ascites myelomas through BALB/cJ mice, unexpected illness and premature deaths occurred. Postmortem examination of affected mice revealed focal or diffuse discolored depressed areas in the liver and, in some cases, splenomegaly. Histopathologic findings consisted of focal to diffuse areas of necrosis with minimal leukocytic infiltration. Aerobic and anaerobic bacterial cultures of livers and spleens from affected mice were negative. Mouse hepatitis virus (MHV) was isolated from livers of clinically ill mice and from the ascites myeloma lines. An MHV contaminated ascites myeloma line, when passed into nude (nu/nu) mice, killed the animals in 6 days; the virus was isolated from livers of inoculated mice. Attempts to determine the source of the infection were unsuccessful. Serologic survey of newly acquired mice indicated no evidence of antibodies to MHV while mice in holding rooms had titers that ranged from 1:10 to 1:40. Two solid myeloma lines (being maintained by subcutaneous passage) were negative for MHV when tested by virus isolation techniques, and nine lines were negative to 11 murine viruses when tested by mouse antibody production assay. Attempts to demonstrate Eperythrozoon coccoides in control BALB/cJ mice were unsuccessful. Because of the outbreak, changes were made in animal handling procedures. A colony of BALB/cAn mice negative to MHV antibodies was established to provide animals for experimental passage of tumors, and animals in both the breeding and transfer room were placed under filter tops. The results were encouraging. In the four newly established tumor lines, one having been passed 46 times, no illness or unexplained deaths were observed.

Ulcerative dermatitis in the rat.

Ulcerative dermatitis observed in several shipments of rats was found to be limited to one strain (Sprague Dawley) provided by one supplier from two specific breeding areas. The incidence of lesions was higher in males than in females. Rats receiving an agar base diet also appeared to have a higher incidence of the lesions. The lesion consisted of a moist ulcer usually involving the lateral aspect of the shoulder and often the sides of the neck. Staphylococcus aureus was isolated from lesions of affected rats and from the skin of unaffected rats. Inoculation of S aureus into susceptible animals resulted in lesions in 25-40% of the rats, but usually at sites other than the site of inoculation. Serum neutralization tests for sialodacryoadenitis virus indicated that about half the rats with lesions and half the rats without lesions were positive. Toenail clipping of the hind feet of affected rats reduced the severity of the lesions. Efforts to isolate fungi and ectoparasites were unsuccessful. The selective procurement of rats from specified breeding areas, eliminated the population susceptible to or harboring the etiologic agent or agents responsible for the development of the ulcerative dermatitis described.

Conformational aspects of the antifibrillatory activity of procaine.

The effects of procaine and four semi-rigid conformational analogs (compounds 1,2,3 and 4) were tested and compared on isolated rabbit atria. Procaine and the four analogs produced positive inotropic effects at all dose levels tested. The antifibrillatory activity of procaine and its analogs arranged in decreasing order of activity was compound 4 greater than 3 greater than 2 greater than 1 greater than procaine. The antifibrillatory activity of the compounds correlated to the distance between the ring nitrogen and the ester oxygen; that is, as the N-O distance increased the concentration required to reduce the following frequency decreased. However, the compound became more toxic as the N-O distance increased. Our data do not confirm the commonly regarded direct relationship between local anesthetic activity and antifibrillatory activity of procaine. Differences in activity displayed by the isomers of procaine could reflect differences in the ability of these analogs to bind to receptors responsible for the respective actions.
