Trials of an inactivated equid herpesvirus 1 vaccine: challenge with a subtype 2 virus.

Serological responses following two and three doses of an inactivated equid herpesvirus 1 ( EHV -1) vaccine containing a subtype 1 strain were examined in yearling ponies. Complement fixing antibody responses were significantly higher against the subtype 1 vaccine strain than against a subtype 2 virus. Complement fixing antibody responses declined rapidly after the second dose of vaccine and had returned to almost pre-vaccination levels eight weeks after the second dose of vaccine. Complement fixing antibody titres to the heterologous subtype 2 strain increased after each successive dose of vaccine. The neutralising antibody responses following vaccination were weak although less strain specific than the complement fixing antibody responses. When challenged with a subtype 2 strain, 15 ponies which had received three doses of vaccine, two, eight and 12 weeks previously resisted challenge infection. However, 11 out of 15 ponies which had received two doses of vaccine eight and 12 weeks previously were susceptible to infection. While two doses of vaccine did not reduce duration of virus excretion or febrile responses in ponies challenged eight weeks after the second dose, the amount of virus excreted was significantly reduced.

The detection of rotaviruses in products of wastewater treatment.

Rotaviruses present in products of wastewater treatment were assayed in MA104 cells by indirect immunofluorescence. Levels in settled sewage, activated sludge and effluent were greater than 10(3) per litre in March and April but virus was not detected during later months. This pattern correlated with the decline in laboratory reports of human rotavirus infection.

An affinity technique for the isolation of Clostridium perfringens type C from man and pigs in Papua New Guinea.

Necrotising enteritis caused by Clostridium perfringens type C (CwC) is an important cause of morbidity and death in highland regions of Papua New Guinea (PNG) and the organism is readily isolated from clinical cases. Although detected in fluorescent antibody stains of faecal cultures, CwC has previously only once been isolated from normal people or from pigs in PNG. The difficulty results from the greater numbers of the much more common Cl. perfringens type A (CwA) which is morphologically and culturally indistinguishable from CwC. Separation of CwC from the more numerous CwA has been achieved by utilising their antigenic dissimilarity. Silicate beads covered with specific antibody to CwC added to a mixed culture bound CwC cells while most of the unbound cells were washed away. When the beads were plated out type C organisms were recovered. In laboratory tests CwC was isolated from mixtures with CwA where the type A organisms predominated by one million to one. With this system type C organisms have been isolated from human and pig faeces from PNG.

Investigations of tissue folates in normal and malignant tissues.

The folates present in liver, gut and tumour tissue were examined before and after autolysis. Before autolysis 10-formylfolate tetraglutamate (10-CHOFA(glu)4), 5-methyltetrahydrofolate triglutamate (5-CH3THF(glu)3) and possibly tetrahydrofolate polyglutamate(s) (THF(glu)n) were detected. Liver contained all 3 species whereas no 5-CH3THF(glu)3 was present in the tumours; gut showed an intermediate situation. After autolysis the predominant monoglutamates formed were 5-CH3THF in the liver, 10-formylfolates in the gut and possibly tetrahydrofolate (THF) in the tumour extracts. These differences illustrate changes in tissue folates with the proliferation rate of the tissue and suggest an explanation for the methionine auxotrophy of Walker 256 carcinosarcoma cells.

Analysis of gross migration profiles in England and Wales: some developments in classification.

"In this paper [the authors] describe the development and application of two approaches to the analysis of age and sex specific interarea migration. The data relate to local authority areas in England and Wales. These approaches are developments of established migration profile-fitting and cluster analytic methods. The paper shows how these methods have been used to develop complementary classifications from which potentially valuable criteria for migration analysis can emerge. The work described is part of a wider project concerned with the development of methods to improve the official estimation of internal migration in England in the context of local area population forecasting."

Pathogenesis of neonatal necrotising enterocolitis.

Neonatal necrotising enterocolitis (NEC) mostly affects the small premature infant in a neonatal intensive care unit (NICU). It is proposed that, because of physical isolation, the cleanliness of nursing procedures, and, in some cases, antibiotic treatment, the normal bacterial colonisation of infants in NICUs may be delayed. In such babies, colonised with one or a few species, the organisms multiply in the gut, unhindered by competitors. The immature gut takes up macromolecules intact, especially in the lower ileum, and toxic products from the growing bacteria may be absorbed and cause mucosal damage, initiating NEC. Thus, NEC may be a result of the NICU environment.

Estimation of migration profiles in England and Wales.

"Migration profiles by age have been increasingly used in the analysis of migration data, and a theoretical function has been developed by researchers at the International Institute for Applied Systems Analysis (IIASA). This paper refines the approach by applying the principles of maximum likelihood to the estimation of the coefficients for the function. This enhances the statistical basis and in particular enables [the authors] to carry out tests of similarity between different areas on the basis of the calibrated coefficients. The method is applied to 1971 Census data for the local authorities of England and Wales."

Isolation of indigenous enteroviruses from chemically treated and dewatered sludge samples.

Samples of wastewater sludge were examined for infectious enteroviruses before and after they had been chemically conditioned and dewatered. The least virus was recovered from the cake produced by filter pressing of sludge, which had a greatly increased solids content (39 to 45% [wt/vol]) relative to the untreated sludge (4.2 to 6.2% [wt/vol]) and in one plant was at pH 11 due to the lime conditioner used. Conditioning with a cationic polyelectrolyte before dewatering by centrifugation produced a watery sludge (2.7 to 5.3% [wt/vol]) from which high titers of infectious virus were recovered which were often greater than those isolated from the untreated sludge (0.6 to 1.4% [wt/vol]). This was thought to be due to saturation of virus and sludge floc adsorption sites by the polyelectrolyte, resulting in the liberation of virions from the sludge solids.

Exponential analysis of the lung pressure-volume curve as a predictor of pulmonary emphysema.

To compare the predictive value of different pulmonary function tests in the diagnosis of morphologic emphysema, we performed measurements of subdivisions of lung volume, gas exchange, maximal expiratory flow rates, and static deflation pressure-volume curves on 55 subjects prior to surgery for removal of an isolated peripheral pulmonary lesion. Emphysema was graded on the resected lung specimen and the pressure-volume data were fitted to an exponential equation (V = A - Be-KP). By chi-square analysis, K was the best predictor of emphysema in individual subjects and it was the only test that distinguished subjects with moderate emphysema from subjects with mild or without emphysema, but K did not distinguish those with mild emphysema from those without emphysema. As a group those with mild emphysema were distinguishable from predicted normal with K and elastic recoil pressures at 90 and 60% of predicted total lung capacity. We conclude that minimal emphysema may be detected by exponential analysis of the lung pressure-volume curve.

The effect of severe zinc deficiency on serum levels of albumin, transferrin, and prealbumin in man.

Concentrations of three serum transport proteins, albumin, transferrin, and prealbumin, were determined in seven patients with severe zinc deficiency. Zinc deficiency was manifested not only by depressed serum zinc concentrations, but also by skin lesions typical of zinc deficiency that corrected with zinc supplementation only. Concentrations of all three serum proteins were significantly depressed in zinc-deficient patients compared to healthy controls, and levels of all three proteins improved or corrected with a short period of zinc supplementation as the sole form of therapeutic intervention. Prealbumin levels dropped and corrected most rapidly, probably due in part to its short half-life of 2 days. This study demonstrates that zinc plays an important role in protein metabolism in man and is necessary for the maintenance of normal levels of certain transport proteins. These results support the possibility that zinc deficiency may alter tissue availability of other nutrients such as vitamin A or iron through its effect on transport proteins.

Recovery of indigenous enteroviruses from raw and digested sewage sludges.

We examined different types of raw sewage sludge treatment, including consolidation, anaerobic mesophilic digestion with subsequent consolidation, and aerobic-thermophilic digestion. Of these, the most efficient reduction in infectious virus titer was achieved by mesophilic digestion with subsequent consolidation, although a pilot-scale aerobic-thermophilic digester was extremely time effective, producing sludges with similarly low virus titers in a small fraction of the time. Although none of the treatments examined consistently produced a sludge with undetectable virus levels, mesophilic digestion alone was found to be particularly unreliable in reducing the levels of infectious virus present in the raw sludge.

Major coagulopathy and "nonpoisonous" snake bites.

Two florid examples of disseminated coagulopathy following the bite of a rear-fanged snake (normally considered "nonvenomous") are reported. The posibility of clinically significant cases of snakebite envenomation due to "harmless" snakes in the United States is raised because of the diagnostic and therapeutic implications.
