Modification of hepatotoxic effects of aflatoxin B1 in rabbits by immunization.

Reduction of acute toxic effect of aflatoxin B1 was achieved by immunizing the rabbits with small amounts of bovine serum albumin-aflatoxin B1 conjugate. Rabbits after immunization showed lower mortality, near normal serum isocitric dehydrogenase activity, no abnormality in livers when challenged with a single dose of aflatoxin B1. The results suggest that immunization might be used prophylactically against aflatoxicosis.

Production of antibody against aflatoxin B1.

Antibody against aflatoxin B1 was obtained after one multiple-site injection of bovine serum albumin-aflatoxin B1 conjugate into rabbits. The antibody has greatest binding efficiency for aflatoxin B1, less efficiency for B2, G1, and Q1, and least for aflatoxicol, G2, and M1. Sterigmatocystin, coumarin, and 4-hydroxycoumarin did not give a cross-reaction with the antibody. The sensitivity of the binding assay for detection of aflatoxin B1 is in the range of 0.2 to 2.0 ng per 0.5-ml sample. Detailed methods for the preparation of the conjugate, production of immune serum, and methods for antibody titer determination are described.

Sex and age differences in luteoskyrin hepatotoxicity.

1. Luteoskyrin, a hepatotoxic and hepatocarcinogenic mycotoxin from the fungus of "yellowed rice" Penicillium islandicum Sopp, shows different toxicities depending on the age and sex in mice. The lethal toxicity was higher in young or male mice than in adult or female mice. 2. 3H-luteoskyrin was accumulated in the livers, especially in their mitochondria, of suckling or male mice in larger amounts than in those of adult and female mice. 3. Intravenously injected 3H-luteoskyrin was excreted into the feces more rapidly in female than in male mice. 4. Luteoskyrin added to a diet was accumulated in the liver at a much higher rate in male than in female mice. 5. The susceptibility of mice to luteoskyrin hepatotoxicity slowly correlates to the rate of its accumulation in their livers, which in turn reflects the rate of its biliary excretion.

Pharmacokinetic studies on the hepatotoxicity of luteoskyrin. (III) quantitative determination of luteoskyrin in the liver.

A method for the quantitative determination of luteoskyrin in mouse liver is described. Luteoskyrin was extracted with ether from the liver homogenate after acidification with with HCl, purified by thin-layer chromatography and determined by spectrophotometry. Five mug of luteoskyrin added to the liver homogenate which was equivalent to one gram tissue was determined with a recovery rate of 97% and 1 mug, under the same conditions, with a recovery rate of 70%. Luteoskyrin in the amount of 0.5 mug could be detected on the chromatogram, but could not be determined quantitatively. A radioactivity corresponding to 0.92 mug/g tissue of 3H-luteoskyrin in the livers from the mice injected intravenously with this compound was extracted into ether with a recovery rate of 93% and separated on the thin-layer chromatogram with a recovery rate of 81%. The lower limit of quantitative determination of luteoskyrin in one gram of liver was 1 mug. Amounts of luteoskyrin in the livers of mice poisoned with this mycotoxin were estimated. Luteoskyrin was recovered in a large quantity in livers from male mice while detection in the liver of females was nil.

[Existence of triphasic flow pattern in regional cerebral blood flow of prolonged unconscious patients (author's transl)].

Anticipating to get some new informations about the cerebral circulations of the comatous patients, two kinds of radioisotopic techniques were combined and applied to twelve unconscious patients of various etiologies. The first step was the conventional gas clearance method using xenon 133 as the indicator. Regional cerebral blood flows were measured at the six areas over each hemisphere. Immediately following this procedure, about 150 muCi of I-131 MAA (I-31 macroaggregated human serum albumin) were injected into internal carotid artery via the same catheter inserted already for the first procedure. Radioactivities of the head and lungs were measured and the relative shunt flow was calculated based on the formula already described elsewhere. Profil scanning of the whole body of the patient was also performed after the intracarotid injection of I-131 MAA. Consciousness states of the ten patients were all severely damaged and causes of the disturbed consciousness were various, including 5 of subdural hematoma, 2 of occlusive cerebrovascular disease, 2 of brain tumor and 1 of ruptured intracranial aneurysm. This investigation revealed the following results; 1) In all patients, regional cerebral blood flows were shown decreased over all areas detected. The regional cerebral blood flows could not be increased by 5% carbon dioxide inhalation, suggesting angioparalysis in these areas. 2) Graphical analysis of the clearance curves revealed triphasic flow pattern in rCBF in four cases. In three cases among the four, the intracarotid injection of I-131 MAA showed the radioisotope labelled particles were captured not only in the brain, but also in the lungs and the relative shunt flow calculated increased up to twice as much as normal controls. It indicates that some of the particles larger than the cerebral capillary size passed through the brain and were captured by the capillary net work of the lungs. From the above described data, it might be concluded that the initial rapid component of triphasic flow pattern in rCBF measurement does not represent the hyperemia of luxury perfusion of metabolic origin, but arteriovenous shunting blood flow, probably, through the precapillary thoroughfare channels which have been anatomically demonstrated by Hasegawa et al.
