A study on immunodiagnosis of gnathostomiasis by ELISA and double diffusion with special reference to the antigenicity of Gnathostoma doloresi.

In order to diagnose gnathostomiasis immunologically, Gnathostoma doloresi was evaluated for the antigenicity in comparison with G. hispidum which was recently reported in Japan by using micro-ELISA. The study revealed that G. doloresi can be used as the alternate source of antigen in the test. A significant increase of specific IgG antibodies was seen in 22 (73.3%) out of 30 gnathostomiasis cases. Although double diffusion was slightly less sensitive than ELISA, it was considered more specific than the latter method.

IgE response in Strongyloides ratti-infected rats with special reference to the life cycle of the parasite.

Parasite-specific IgE antibody response was examined in Strongyloides ratti-infected rats. The results showed that the parasite-specific IgE antibody response was generated after a primary infection. However, repeated infections rather depressed the level of parasite-specific IgE antibody in the serum. Immunization limited to specific stages of the parasite revealed that stimulation of parasite-specific IgE antibody was related to the intestinal adult stage. On the other hand, depression of IgE titers was related to the tissue-migrating larval stage. The capacity of the each stage of the parasite to induce specific IgE response may be related to the variable results of the IgE responses in human strongyloidiasis.

Antigenic similarity of Onchocerca volvulus to other helminths examined by monoclonal antibodies against O. volvulus.

Antigens in Onchocerca volvulus were characterized using anti-O. vovulus monoclonal antibodies (McAb). A total of 17 McAb which were not species-specific to O. volvulus was examined for the antigenic similarity of O. volvulus with 12 parasitic helminths using indirect immunofluorescence. The range of reactivity of these McAb was wider in nematodes in contrast to species of cestoda or trematoda. The antigenic similarity within helminths examined was generally in agreement with their taxonomic relationships but was not shown in the lower taxonomic hierarchy. The helminth which holds the antigens most in common to O. volvulus was not the filarial parasites but Ascaris suum, an intestinal nematoda. The least antigenic similarity to O. volvulus was found in Schistosoma mansoni and S. japonicum, trematoda, among helminths examined. On the other hand, the existence of a common antigen shared with all the 13 helminths examined was demonstrated by a IgM McAb.

Dirofilaria infection in the breast of a woman in Japan.

Dirofilaria infection in the breast of a woman was reported from Japan. A nodule in the breast was excised and histologically examined. The sections of a female nematode were found in the center of the nodule. Based on the morphological features of this worm, it is suggested that this may represent the second human case of Dirofilaria immitis occurring in the breast.

[Basic and clinical study of liver cancer using intra-arterial infusion of an emulsion of the oily contrast medium lipiodol with 5-FU].

An emulsion of Lipiodol and the anti-cancer agent 5-FU has been made using the microtip of an ultrasonic homogenizer, and we have been performing super-selective intra-arterial infusions of the emulsion by the Seldinger method on patients with liver tumor. In our basic study, the emulsion was in a water-in-oil state in which 5-FU granules 5-45 mu in diameter were enclosed in the Lipiodol. In an in vivo experiment, the concentration of 5-FU in the tumor tissue remained as high as that existing 30 minutes after infusion of the emulsion, while almost no activity was detected 30 minutes after infusion of 5-FU only. Therefore, the use of Lipiodol as a carrier enables the anti-cancer agent to reach the affected site and makes it possible to obtain a high concentration of the drug within the tumor itself in cases of liver tumor with abundant vasculature significantly involving the hepatic artery. In addition, with this method, it is possible to select different drugs according to the drug-sensitivity of the particular tumor, and the side-effects are extremely slight. In conclusion, we believe that this method can be an effective therapy not only for unresectable primary liver tumor, but for metastatic liver tumor as well.

Development of Onchocerca volvulus larvae in Simulium pintoi in the Amazonas region of Venezuela.

The intake and development of Onchocerca volvulus in Simulium pintoi from the Parima mountain region of the Federal Territory of Amazonas in Venezuela, were studied experimentally. When wild females fed on the lower half of the legs and lower third of the back of an onchocerciasis patient harboring 23 and 264 microfilariae per skin snip, respectively, at each site, an average (median) of 14 (range, 1-77) and 245 (range, 58-495) microfilariae was ingested. However, within 24 hours of microfilarial ingestion a mortality of 47% (16/34 flies) was observed in the group of flies which fed on the back, as compared with 2% (2/101 flies) in the other group which fed on the legs. At a temperature varying between 16 degrees C and 24 degrees C, the development of O. volvulus larvae in S. pintoi was synchronous and orderly; no abnormal nor deformed larvae were observed. Third-stage larvae were first seen in the head of flies dying between 8 and 9 days after microfilarial ingestion, and 98 of 100 larvae recovered from days 10-16 were in the third stage. The proportions of females harboring third-stage larvae among flies which lived through day 8 in the two groups which fed on the legs and back, respectively, were 55% (21/38 flies) and 63% (5/8 flies). Although only two of five positive flies in the latter group contained third-stage larvae in the head (1 and 12, respectively), 71% (15/21 positive flies) of the former group had an average of 2.7 third-stage larvae in the head (range, 1-10).(ABSTRACT TRUNCATED AT 250 WORDS)

[2 cases of the independent coexistence of early cancer and leiomyosarcoma in the same stomach].

In the past 10 years, malignant diseases have been revealed in 157,836 autopsy cases (in Japan). Among them, gastric cancer was found in 17.5% and leiomyosarcoma in 0.45%. In addition, the frequency of leiomyosarcoma of the stomach was only 0.08%. In the literature, 20 cases of the coexistence of cancer and leiomyosarcoma in the same stomach (were) reported in Japan prior to (data). Eight of these were early cancer. Subsequently, three cases, including two of our own, were added. In our two patients, early cancer and leiomyosarcoma coexisted independently in the same stomach. It is suggested that the stomach with leiomyosarcoma may have a tendency to be cancerous.

Strongyloides ratti: the role of enteral antigenic stimuli by adult worms in the generation of protective immunity in rats.

Immunogenicity of adult Strongyloides ratti was studied in rats. Immunization of rats by intraduodenal implantation of adult worms could completely inhibit the egg production and hasten the expulsion of challenged worms which were developed from subcutaneously inoculated L3 or were implanted intraduodenally as adults. Enteral immunization by intraduodenal implantation of adult worms was, however, not able to affect the esophageal larval output of the challenge infection with L3. In contrast to enteral immunization with adult worms, immunization by full sequence of a primary infection or by a combination of drug-abbreviated infection and adult worm implantation could suppress the esophageal larval output of the challenge infection. The relationship between the host defense mechanism and the life cycle of S. ratti is discussed.

Karyotypes of Brugia pahangi and Brugia malayi (Nematoda: Filarioidea).

Using air-dried preparations of the testis and ovary, karyotypes were analyzed and compared to each other in two species of filarial parasites, Brugia pahangi and B. malayi. Both species had a diploid number of 10 chromosomes and were karyotypically very similar. C-banding analyses disclosed that the sex-determining mechanism of these species was of the XY-XX type, where the X chromosome was the largest, and the Y chromosome was of medium-size.
