[24,25-3H]Cholesterol: presence of tritium at additional sites in the side chain.

In order to measure the distribution of radioactivity present in the side chain of [24,25-3H]cholesterol prepared by a sequence involving catalytic tritiation of 3 alpha, 5 alpha-cyclocholest-24-en-6 beta-ol 6-methyl ether, the cholesterol was oxidized to 4-cholesten-3-one, which was then cleaved between C-24 and C-25 to afford the C24 alcohol. Oxidation to the corresponding cholenoic acid, followed by alkali equilibration and esterification completed the sequence. It was found that about 20% of the tritium in the labeled cholesterol is not lost when this tracer is physiologically converted to bile acids. Consequently, measurements of bile acid formation using this tracer must be corrected upward by this amount.

Side-chain oxidation of lipoprotein-bound [24,25-3H]cholesterol in the rat: comparison of HDL and LDL and implications for bile acid synthesis.

The purpose of the study was to test the hypothesis that high density lipoprotein (HDL) cholesterol would be more easily oxidized in vivo than low density lipoprotein (LDL) cholesterol. Homologous plasma was incubated with [24,25-3H]cholesterol and fractionated by ultracentrifugation to obtain HDL and LDL each labeled with [3H] free sterol. HDL and LDL labeled with [24,25-3H]cholesteryl esters were prepared by ultracentrifugation of plasma from donor rats injected 24 hr previously with [24,25-3H]cholesterol in propylene glycol. These four labeled lipoproteins were administered to recipient rats. It was found that more tritium oxide (3H2O) was produced after the HDL doses than after the corresponding LDL doses, from 2--3-fold more when lipoprotein free cholesterol was labeled and from 2--6-fold more when lipoprotein cholesteryl esters were labeled. More 3H2O was produced from free cholesterol-labeled lipoproteins than from cholesteryl ester-labeled lipoproteins. Since oxidation of cholesterol is a measure of bile acid formation, it is concluded that under the conditions of the study HDL-cholesterol is a better precursor of bile acids than LDL-cholesterol.

[Lipid mobilization in obesity without carbohydrate intolerance. 2. Caffeine-stimulated lipolysis].

The lipolysis induced by i.v. injection of caffeine (3 mg/kg body weight) has been investigated in 11 normal subjects and 18 obese patients with normal 50 g oral glucose tolerance test and normal thyroid-function. The pattern of blood-glucose and immunoreactive insulin (IRI) secretion were not influenced by caffeine in both groups. Both in the controls and obese group a significant response of FFA and glycerole were noted. In the obese group the caffeine-induced lipolysis was slightly higher as compared with controls. There are no differences in FFA-response between obese group and controls. The observations suggest that an impaired lipolysis is not a pathogenetic factor for development of obesity.

[Technic for the presentation and evaluation of pathognostically significant erythrocyte changes].

In this paper is described in all details a method of staining and evaluation of blood smears which was originally proposed by Clara Fonti and was revised by the authors. With the help of this method characteristic changes of the erythrocytes can be demonstrated. The changes are depending on growth processes. Without exception they appear in an increased range in the growth of malignant tumours.

[Studies on the cancer-diagnostic evidence of erythrocyte changes during the course of neoplasm treatment].

In the present communication is reported on the results of further observations of the course of the index of the change of erythrocytes of 12 patients in connection with four clinical findings and parameters. The data got in these cases continue the previously presented reports on the results. In 9 out of 12 cases there is a correspondence with the indices of the change of erythrocytes and the clinical data belonging to this; in the remaining three cases the indices of the change of erythrocytes cause to presume a continuation of the malignant processes, though the other findings do not indicate this at present.

Preparation and antigenic properties of androsterone-7-BSA conjugate.

The 7-carboxymethoximino derivative of androsterone (1) has been prepared from dehydroisoandrosterone-17-ethyleneketal by a sequence involving inversion at C-3, introduction of a carbonyl at C-7, and reduction of the double bond at C-5. The substance was condensed with BSA by the carbodiimide procedure to afford a conjugate which produced anti-androsterone antiserum in innoculated rabbits. The antiserum is sufficiently active to be useful in radioimmunoassay procedures.

[Studies on lipid mobilization in obesity without glucose intolerance. 1st communication. Noradrenaline-stimulated lipolysis].

1. In obese patients with normal glucose tolerance the norepinephrine-induced lipolysis in vivo was significantly higher as compared with a non-obese group. This was observed both during weight-related and constant norepinephrine infusion. 2. In the obese group a discordant release of free fatty acids and glycerol and a significantly lower quotient of FFA/Glycerol than in the controls was found. It is supposed that in obesity the re-esterification of FFA is increased.
