Purification and immunofluorescence localization of the mutant gene product of a Tetrahymena cdaA1 mutant affecting cell division.

A division arrest mutant, cdaA, of Tetrahymena thermophila is known to have a ts-defect in the formation of the fission zone which determines the position of the fission plane. A protein (Mr = 85,000; pI = 4.7, designated as p85) has recently been identified in our laboratory as a possible gene product of the cdaA locus by two-dimensional gel electrophoresis and genetic experiments (Ohba et al., submitted). In the present research, we have isolated p85, prepared its antibody, and demonstrated that in wild-type cells or in cdaA cells at permissive temperature, immunofluorescence for p85 appears on the equatorial basal bodies at the predicted fission zone just before formation of the zone. In such a case, the fission zone appears to be formed just anterior to the fluorescence-associated basal bodies, and then constriction of the division furrow occurs at the zone. However, in cdaA cells at the restrictive temperature, the equatorial p85 deposit and subsequent fission zone formation and furrowing do not occur at all. Thus, we conclude that p85 plays a key role in the formation of the fission zone and in the positioning of the equatorial fission line.

Dental care for severely handicapped children.

Dental care for the physically and mentally handicapped ought to be a normal part of the responsibilities of the pedodontist. In the case of severe handicap, however, it may be difficult to achieve an acceptable standard of care under general practice conditions. An account is given of the regimen followed in the Tokyo Metropolitan Rehabilitation Institute for the Severely Handicapped where a team approach to dental care has been in existence for the past 10 years. Data are presented which demonstrate the incidence of dental disease in various categories of handicap and examples are given of the improvement in the quality of life of the institutionalized handicapped as a result of regular, high quality dental care.

Effect of the methyl methacrylate-tributylborane sealant in preventing occlusal caries.

A newly developed pit and fissure sealant, named Enamite, was studied technologically and clinically. Major ingredients of the sealant are a solution of 3% 2-hydroxy-3-beta-naphthoxypropyl methacrylate in methyl methacrylate and poly (methyl methacrylate) powder, using tributylborane as an initiator. In comparison with other commercially available sealants, Enamite showed much greater adhesiveness to enamel, which was confirmed by scanning electron micrographic observations. The sealant was applied to 161 first molars in 99 children, aged 6-8 years, to determine the caries preventive as well as the caries retardation effect. Among the treated teeth, 98 were intact and the remaining had incipient caries. The right side teeth of each subject was assigned to the test side and the other side served as a control. Each child was evaluated every 6 months by clinical exploration and bite-wing X-ray survey. At the final examination, the electric conductivity test was also used for the caries detection. After 2 years the sealant was effective in preventing 57.1% of the occlusal caries on treated compared with control teeth. When evaluation was made by including the caries retardation effect--teeth initially carious but remaining sealed after 2 years were counted as effective--, the sealant was effective in preventing or retarding 96.3% of the occlusal caries.

Anodontia with hypohidrotic ectodermal dysplasia in a young female: a case report.

A five-year-old Japanese girl was referred to our clinic for non-eruption of the teeth. Panoramic radiographs revealed absence of all teeth of both primary and permanent dentitions. She showed hypotricosis, hypohidrosis, and anodontia. In this article, the reported cases of anodontia were reviewed and the fabricating procedure of full dentures for a young child was described.
