The extreme susceptibility of the mouse kidney to infection with Staphylococcus aureus was confirmed by direct intrarenal (i.r.) inoculation with this organism. By applying Poisson's distribution formula to the results from infection of mice with small inocula (as few as one coccus or less per kidney), it was estimated that even three organisms could multiply in situ to produce abscess lesions, if the organisms were inoculated directly into the kidneys. This susceptibility of the mouse kidney for staphylococcal infection was not uniformly manifested with every strain, but correlated well with the virulence found through intravenous infection of the strains tested. The i.r. inoculation method by which infection of mice was easily established, using a small inoculum of S. aureus, similar to the amount suspected to occur in natural infections of man, is applicable for the analysis of the mechanisms of staphylococcal infections and any resulting immunity in man. The present paper describes the details and some experimental results obtained by our method.

Rat liver perfused with 0.5% Triton X-100 for 15 to 90 min followed by 1% sodium dodecyl sulfate for 30 min were studied by electron microscopy and polyacrylamide gel electrophoresis. After 15 min of perfusion, a rich network of intermediate filaments and microtubules was visualized in the cytoplasm of hepatocytes. Using stereopairs, branching was visualized. Connections of filaments were noted with nuclei, centrioles, microtubules, vesicles, and rough endoplasmic reticulum. The existence of connections supported the concept that intermediate filaments may function to integrate mechanically the cytoplasmic space as postulated by Lazarides.
In order to demonstrate the effect to the acute administration of ethanol and chlorpromazine (CPZ) on bile flow and transhepatic transport of horseradish peroxidase (HRP) into bile, male rats were administered either 5 gm per kg ethanol intragastrically (E-rats) or 3 mg per kg CPZ intraperitoneally (CPZ rats). Control rats (C-rats) received saline. Two hours after ethanol feeding or 90 min after CPZ injection HRP was injected into the portal vein, and bile samples were collected at 10-min intervals for 2 hr. Tissue samples were removed at 1, 10, 60, and 120 min to study HRP transport using electron microscopic cytochemical localization. Bile flow was reduced (p less than 0.001) both in E- and CPZ-rats compared to C-rats. In E-rats HRP secretion was significantly decreased at 30 and 40 min post-HRP injection (p less than 0.05) and the peak rate of HRP secretion was delayed by 10 min compared to C-rats. Uptake and transhepatic transport of HRP were similar to controls. These results suggest that bile secretion and flow were impaired by ethanol. CPZ inhibited secretion of HRP significantly (p less than 0.001) during the first hr after HRP injection and by 25% after 2 hr. In CPZ rats studied cytochemically HRP reaction product decreased in the cytoplasm of hepatocytes 10 min after HRP injection (p less than 0.01). These findings suggest that acute CPZ administration caused an inhibition of the uptake of HRP as well as secretion and bile flow.
To evaluate brain dysfunction of childhood-onset dentatorubral-pallidoluysian atrophy (DRPLA), three children with progressive myoclonus epilepsy, who were diagnosed as having DRPLA by DNA analysis, for the first time, underwent a study of proton magnetic resonance spectroscopy (1H-MRS). 1H-MRS obtained from both the parietal and basal ganglia regions disclosed markedly reduced ratios of N-acetylaspartate to both choline and creatine. Especially regarding the basal ganglia region, the latter (ratio of N-acetylaspartate to creatine) closely correlated to severity of genetic abnormality, i.e. number of expanded CAG repeats, suggesting that the degree of neuronal loss in the region strongly depends on genetic factors. 1H-MRS must be a valuable tool to clarify the pathophysiology of DRPLA.
To determine the characteristics of multimodal evoked potentials (MEPs) in childhood-onset dentatorubral-palli-doluysian atrophy (DRPLA) we studied three DRPLA patients with progressive myoclonus epilepsy. Brainstem auditory evoked potentials showed reduced or absent brainstem components as well as delayed latencies. In addition, short latency somatosensory evoked potentials (S-SEPs) had prolonged central conduction time and reduced amplitude of cortical components. Two patients with symptom onset in the first decade of life had extremely enlarged flash visual evoked potentials with shortened latency even in the absence of giant SEPs. Therefore, children with progressive myoclonus epilepsy and the above MEP findings are likely candidates for childhood-onset DRPLA and should undergo DNA analysis for DRPLA.
The purpose of this study was to determine whether the force of a finger could be measured with a new system based on an attachment to the Kin-Com dynamometer, and to define the reproducibility of the finger force measurement. Eleven male subjects (age, 27.6 +/- 6.12 years) took part. All were free of orthopedic problems in the tested hand. Mean forces of the index and little finger for concentric and eccentric motion were measured using the new system. Overall coefficients of variation (OCVs) were calculated as an index of reproducibility. The reproducibility of force curves was considered good when an OCV was less than 20%. The OCVs for mean force of the index finger and for eccentric motion of the little finger were less than the 20% standard, at 10.7 and 14.0, respectively. The OCV for concentric motion of the little finger was greater than 20%. The findings suggest that measurement of finger force is feasible with the new Kin-Com attachment.
The objectives of the present study were (1) to determine whether oxidized low-density lipoprotein (LDL) and lysophosphatidylcholine (lyso-PC), a major phospholipid component of oxidized LDL, stimulate the production of endothelin-1 (ET)-1 in cultured human coronary artery smooth muscle cells (SMCs), and (2) to examine the possible effect of an antiatherogenic agent, eicosapentaenoic acid (EPA), on oxidized-LDL- and lyso-PC-stimulated ET-1 production in these cells. Oxidized LDL (10-50 microg/ml) and lyso-PC (10(-7) to 10(-5) mol/l) stimulated ET-1 production in a concentration-dependent manner. By contrast, the effects of native LDL and phosphatidylcholine were modest or absent. Lyso-PC (10(-7) to 10(-5) mol/l) and oxidized LDL (10-50 microg/ml) significantly induced particulate protein kinase C (PKC) activation. Lyso-PC- and oxidized-LDL-stimulated ET-1 production was significantly inhibited by PKC inhibitor, PKC (19-36). EPA (80-160 micromol/l) clearly suppressed ET-1 production stimulated by oxidized LDL and lyso-PC in a concentration-dependent manner. Furthermore, EPA (160 micromol/l) significantly inhibited lyso-PC (10(-5) mol/l)- and oxidized LDL (50 microg/ml)-induced particulate PKC activation. Results suggest that oxidized LDL and lyso-PC stimulate ET-1 production by a mechanism involving activation of PKC, and that EPA suppresses ET-1 production stimulated by lyso-PC as well as oxidized LDL probably through the modulation of PKC in human coronary artery SMCs. EPA may exert an antiatherosclerotic effect, in part, through these mechanisms.
alpha-Synuclein has been isolated as a component of amyloid in addition to the major A beta peptide in Alzheimer disease (AD). However, there are conflicting reports regarding the association of alpha-synuclein gene polymorphism with AD. Using a novel and common polymorphism in intron 3, we examined the relationship between AD and alpha-synuclein and apolipoprotein E (ApoE) genes in 183 Japanese AD patients and 210 controls. Carriers of the alpha-synuclein deletion (D) allele had a 2.2-fold increased risk of developing AD than noncarriers in women. The odds ratio for the ApoE epsilon 4 and the alpha-synuclein D allele was 11.4 in women. The results showed that the alpha-synuclein gene is associated with sporadic AD in women, independent of ApoE epsilon 4 status.
We radiographically evaluated the response of primary lesions to chemotherapy in 101 patients with inoperable advanced gastric cancer. The median survival time in the 50 responders (49.5%) was 379 days, as compared with only 210 days in the 51 non-responders (50.1%) (p<0. 001). Morphologic changes of the gastric mucosa were assessed endoscopically. Among the changes observed, we focused on regenerative mucosa and its relation to response and survival. Regenerative mucosa was seen in 56 (55%) of the 101 patients. The regenerative mucosa was examined endoscopically and classified into four patterns: flat, radial, granular and cobble-stone. Biopsy specimens were taken from the regenerative mucosa and examined histopathologically. The response to chemotherapy was found to be closely associated with the incidence of regenerative mucosa. In particular, a flat or radial pattern significantly correlated with a positive response to treatment (p=0.004) and was associated with significantly longer survival than granular or cobblestone pattern or no regenerative mucosa (p<0.001). Flat or radial pattern was also associated with low tumor-positive rate of biopsy specimens. We conclude that endoscopic evaluation of regenerative mucosa in patients with inoperable advanced gastric cancer who receive chemotherapy can provide useful information in the assessment of response and prognosis.
