[Clinicopathological study of 9 cases presenting axillary masses only].

A clinicopathological study of 9 cases presenting axillary mass alone was reported. Histological diagnosis revealed 3 occult breast cancers, 2 apocrine gland adenocarcinomas of the skin, a carcinoma and a fibroadenoma in the axillary accessory breast, a neurogenic tumor in the axilla, and a benign lymphadenopathy. There were no clinical characteristics helpful in preoperative differential diagnosis. As the axilla includes the main lymphatic routes from the breast, the axillary tail of the breast and accessory breast, breast-associated tumors should be kept in mind in the management of patients with axillary mass as a sole clinical feature.

Characterization of kininogen deficiency of Brown Norway rat mutant Katholiek strain.

A deficiency in the plasma kallikrein-kinin system of Brown Norway rat mutant Katholiek strain (B/N-Ka), first reported by Damas et al. was further characterized. The prolonged activated partial thromboplastin time (APTT) of B/N-Ka rat plasma was corrected by an addition of rat HMW-kininogen, indicating B/N-Ka rat is deficient in HMW kininogen. The study of kinin-release of plasmas of the three strains of rat (B/N-Ka, B/N Ki and SD) by several kininogenases expressed that B/N-Ka rat is deficient in LMW kininogen, in addition to HMW kininogen deficiency. The plasmas of the three strains of rat were gel-filtered through Sephacryl S-200 gel and profiles of kinin-release of the fractions were examined by several kininogenases. The result demonstrated that normal rat plasma contains three kinds of kininogen (HMW and LMW kininogens, and T-kininogen), and B/N-Ka plasma contains only T-kininogen. B/N-Ka rat plasma demonstrated T-kininogen antigen but no HMW kininogen by the study of immunodiffusion using their antisera raised in rabbits.

Evidence for a role of the plasma kallikrein-kinin system in acute inflammation: reduced exudation during carrageenin- and kaolin-pleurisies in kininogen-deficient rats.

Experimental pleurisy was induced by intrapleural injection of carrageenin or kaolin in three strains of rat: Brown Norway-Katholiek (B/N-Ka), Brown Norway-Kitasato (B/N-Ki) and Sprague-Dawley (SD). B/N-Ka rats (kininogen-deficient) showed significantly less pleural fluid accumulation and exudation rate than SD rats or than B/N-Ki rats (normal). The result indicates the involvement of the plasma kallikrein-kinin system in these pleurisies and a role of high-molecular-weight kininogen is suggested.

[Immunohistochemical study of oncogene-related products in human gastrointestinal malignancies--expression of ras p 21, fes p 85 and EGF receptor].

An immunohistochemical examination by the avidin-biotin-peroxidase complex method was carried out to assess the expression of oncogene-related products, i.e., ras p 21 protein, fes p 85 protein and epidermal-growth-factor (EGF) receptors, in human gastrointestinal malignancies. The presence of ras p 21, fes p 85 and EGF receptors was detected in 48%, 62%, and 62% of 29 colorectal carcinomas and in 65%, 65% and 40% of 20 gastric cancers, respectively. More than one oncogene protein was demonstrated in 18 of 29 colorectal carcinomas and in 10 of 20 gastric cancers. These results suggest that multiple oncogenes are important in the occurrence and progress of gastrointestinal malignancies.

Inhibition of DNA synthesis in rat hepatocytes by platelet-derived type beta transforming growth factor.

Platelet-derived type beta transforming growth factor (TGF beta) is a potent inhibitor of DNA synthesis in primary monolayer cultures of adult rat hepatocytes. TGF beta induced a 50% inhibition of epidermal growth factor (EGF)-mediated DNA synthesis at approximately 5 X 10(12) M. This inhibition did not appear to be due to a delay in the peak of DNA synthesis or a toxic action, nor could it be overcome by increasing concentrations of the mitogens EGF, insulin, or glucagon. Inhibition was observed either when TGF beta and EGF were continuously present together in the culture medium or when TGF beta was added to the hepatocyte cultures after removal of the EGF stimulant. This observation together with a lack of an inhibitory effect of TGF beta on the binding of 125I-labeled EGF to hepatocytes in culture, suggests that the inhibitory action of TGF beta was not caused by a direct competition with EGF at the cell surface. TGF beta could not inhibit DNA synthesis once it had begun; however, the inhibitory action of TGF beta could be partially overcome by increasing amounts of conditioned medium produced by normal hepatocytes. Specific saturable receptors for TGF beta were found on the normal rat hepatocytes, but specific binding could not be detected on hepatocytes from regenerating liver. TGF beta is thus a potent inhibitor of EGF-induced DNA synthesis in adult rat hepatocytes. Its significance for growth control in vivo has yet to be assessed.

Exercise-induced renal dysfunction studied by 99mTc-DTPA in hypertensives and normotensive controls.

With the purpose of studying the effect of exercise on renal function in hypertensive patients, renal dynamic studies were performed with the aid of 99mTc-diethylenetriamine pentaacetate (DTPA). Serial renal images were taken at rest and during bicycle ergometric stress in 14 hypertensive patients (H) and 14 normotensive controls (N). Renograms were constructed, and glomerular filtration rate was determined by the method of Gates [2]. Exercise resulted in a prolongation of peak time, an increase in radioisotope retention rate at 10 minutes (10-min counts/peak counts) on renograms, and a decrease of GFR in both H and N. Plasma renin activity, aldosterone, and catecholamines were all elevated during exercise in both H and N. Our data indicate that the exercise-induced renal dysfunction demonstrated by 99mTc-DTPA renograms is not specific to H but can also be observed in N, which may have resulted from changes common to H and N, as indicated by the change of GFR and humoral factors. These findings suggest that the renal response to exercise may be a more complex pathophysiologic process than that measured by the DTPA renogram, at least in its simplified form, and should encourage further work in this area in order to elucidate the underlying process of hypertension.

Identification of T-kininogen in high and low molecular weight kininogens deficient rat (brown Norway Katholiek strain).

Kinin release in Brown Norway Katholiek (B/N-Ka) rat plasma was compared with those of Brown Norway Kitasato and Sprague-Dawley rats by treating with rat plasma kallikrein, rat urinary kallikrein, snake venom kininogenase and trypsin. B/N-Ka rat plasma yielded no detectable amount of kinin by either plasma kallikrein, urinary kallikrein or snake venom kininogenase, but yielded variable amount of kinin by trypsin. The released kinin was proved to be isoleucylseryl-bradykinin by high performance liquid chromatography and bioassay profiles. B/N-Ka rat plasma formed a precipitation line against antiserum to T-kininogen, but no line against antiserum to HMW kininogen-light chain.

Rat plasma high-molecular-weight kininogen. A simple method for purification and its characterization.

High-molecular-weight kininogen has been isolated from rat plasma in three steps in a relatively high yield. The purified preparation gave a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the absence and presence of 2-mercaptoethanol, and the apparent Mr was estimated as 100,000. On incubation with rat plasma kallikrein, rat high Mr kininogen yielded a kinin-free protein consisting of a heavy chain (Mr = 64,000) and a light chain (Mr = 46,000), liberating bradykinin. The kinin-free protein was S-alkylated, and its heavy and light chains were separated by a zinc-chelating Sepharose 6B column. The amino acid compositions of rat high Mr kininogen and its heavy and light chains were very similar to those of bovine high Mr kininogen and its heavy and fragment 1.2-light chains, respectively. A high histidine content in the light chain of rat high Mr kininogen indicated the presence of a histidine-rich region in this protein as in bovine high Mr kininogen, although this region was not cleaved by rat plasma kallikrein. Rat high Mr kininogen corrected to normal values the prolonged activated partial thromboplastin time of Brown-Norway Katholiek rat plasma known to be deficient in high Mr kininogen and of Fitzgerald trait plasma. The kinin-free protein had the same correcting activity as intact high Mr kininogen. Rat high Mr kininogen also accelerated approximately 10-fold the surface-dependent activation of rat factor XII and prekallikrein, which was mediated with kaolin, amylose sulfate, and sulfatide. These results indicate that rat high Mr kininogen is quite similar to human and bovine high Mr kininogens in terms of biochemical and functional properties.

Demonstration of arginyl-bradykinin moiety in rat HMW kininogen: direct evidence for liberation of bradykinin by rat glandular kallikreins.

The amino acid sequence around kinin moiety in rat High-Molecular-Weight (HMW) kininogen was determined by isolating a peptide containing bradykinin after cyanogen bromide treatment of the purified kininogen as follows; NH2-Thr-Ser-Val-Ile-Arg-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg-Ala-Pro-Arg- Val-Lys-Lys-. The data indicated that rat HMW kininogen contains the arginyl-bradykinin moiety, instead of lysyl-bradykinin. Kinins liberated from rat HMW kininogen by rat urinary and submaxillary kallikreins were identified to be bradykinin, not arginyl-bradykinin.

DNA synthesis in rat hepatocytes: inhibition by a platelet factor and stimulation by an endogenous factor.

Primary monolayer cultures of adult rat hepatocytes can be induced to undergo DNA synthesis in serum-free medium in the presence of insulin, glucagon, and epidermal growth factor (three factors). We have found that hepatocyte DNA synthesis is affected not only by an endogenous stimulant produced by the hepatocytes and released into the culture medium. Serum has a strong inhibitory effect on hepatocyte DNA synthesis. Partially purified human platelet extract ("platelet inhibitor") inhibits the three-factor-induced DNA synthesis in a concentration-dependent manner. Pure beta TGF at 0.5 ng/ml as well as HPLC-purified PDGF at 10 ng/ml completely inhibit the three-factor-induced DNA synthesis. Determination of the time required for the presence of the three factors and the platelet inhibitor to exert their effects indicated that the inhibition of DNA synthesis is caused not by competition of the platelet inhibitor with any of the three factors but through an independent pathway. Hepatocyte DNA synthesis is density-dependent and is greater if medium is not changed during the course of an experiment than if medium is changed daily. Hepatocyte-conditioned medium is also affective in stimulating DNA synthesis beyond the level induced by the three factors. These results suggest that an endogenous stimulant for hepatocyte DNA synthesis is produced by the hepatocytes themselves. Our studies demonstrate that hepatocyte DNA synthesis is subject to both stimulatory and inhibitory controls. Unlike the three factors, the endogenous stimulant can overcome the inhibition by the platelet inhibitor, suggesting the importance of these factors in the physiological control of hepatocyte DNA synthesis.

Tubular adenoma with focal squamous metaplasia of the ascending colon.

A case of tubular adenoma with focal squamous metaplasia of the ascending colon in a 66-year-old male is reported. The tumor was a pedunculated polyp with a size of 1.5 X 1.4 X 0.9 cm. Histologically, the tumor showed tubular adenoma with moderate to severe atypia, was accompanied by focal squamous metaplasia. It is thought that squamous epithelial polyp may be derived from colonic adenoma with squamous metaplasia, and that the malignant change of colonic adenoma with squamous metaplasia may lead to squamous cell carcinoma, adenosquamous carcinoma, and mucoepidermoid carcinoma. Only seven cases of colonic adenoma with squamous metaplasia have been reported previously in the literature, and it is yet necessary to discuss the histogenesis, significance, and natural history of colonic adenoma with squamous metaplasia.
