Retroviral gp70 immune complexes in NZB x NZW F2 mice with murine lupus nephritis.

NZB x NZW (NZB x W) F1 hybrid mice spontaneously develop a disease most prominently characterized by immune complex glomerulonephritis (GN), which seems to be associated with both antibodies to DNA and to the serum retroviral envelope glycoprotein, gp70. To evaluate the contribution of each of these autoimmune responses to the pathogenesis of the GN, we studied NZB x W F2 mice in which the two responses appeared to segregate relatively independently. Use of this model permitted analysis of possible correlations between each response and the G.N. The presence of circulating anti-gp 70-complexed gp70 correlated significantly with the development of fatal GN and one could predict the course of renal disease by computing the rising serum levels of gp70 complexed with antibodies. In contrast, the presence of free antibodies to either double-stranded or single-stranded DNA was not significantly associated with the development of fatal GN. This association of anti-gp70 antibody production with these animals' early death from GN strongly suggests that the gene(s) governing production of antibodies to serum retroviral gp70 may be one of the major genes responsible for spontaneous renal disease segregated in NZB x W F2 generations.

High-performance aqueous gel permeation chromatography of serum lipoproteins: selective detection of cholesterol by enzymatic reaction.

A rapid method for the quantitation of cholesterol in each lipoprotein fraction has been developed which utilizes high-performance aqueous gel permeation chromatography followed by enzymatic reaction using reaction-type high-performance chromatography. Cholesterol in serum lipoproteins eluted from the column could be sensitively and selectively detected by the absorbance at 550 nm following the enzymatic reaction. The sensitivity of the detection for cholesterol measured by A550 was compared with that for protein measured by A280 using the standard lipoprotein fractions: low-density lipoprotein (LDL) and high-density lipoproteins (HDL2 and HDL3). The effects of changing the flow-rate and lengthening the column on the resolution of LDL and HDL were examined. Analyses of serum protein and cholesterol were performed with this method for human and animal subjects.

Induction of high serum levels of retroviral env gene products (gp70) in mice by bacterial lipopolysaccharide.

In the present study, mice each given a single intraperitoneal injection of Escherichia coli lipopolysaccharide (LPS) responded with increased serum levels of the major envelope glycoprotein, gp70, of endogenous retrovirus. Concentrations of gp70 in their sera began to increase 4 hr after LPS injection, reached maximal 5- to 15-fold increases after 12--24 hr, and returned to the preinjection levels within 3 days. This response occurred only in the strains characterized by high base line levels of serum gp70 (greater than 10 micrograms/ml) such as NZB, NZB X NZW F1, BXSB, MRL, NZW, DBA/2, LG, 129(GIX+), and C57BL/6(GIX+). However, strains such as DBA/1, C3H/St, BALB/c, C57BL/6(GIX-), and 129(GIX-) with lower base line levels of serum gp70 (less than 5 micrograms/ml) made little or no response. This serum gp70 induced by LPS was structurally similar to the gp70 of NZB xenotropic virus that is dominantly expressed in sera from virtually all strains of mice. However, (i) the induced gp70 was virion-free; (ii) xenotropic virus was not isolatable from BXSB, MRL/1, or 129(GIX+) mice injected with LPS; and (iii) amounts of the major structural viral protein, p30, did not increase correspondingly in sera. All of these findings indicate that the increased expression of serum xenotropic viral gp70 in response to LPS did not result from activation of replication-competent xenotropic virus. In addition, the serum gp70 response to LPS was abolished by simultaneous inoculation of an inhibitor of protein synthesis, D-galactosamine. These results strongly suggest that LPS selectively stimulates synthesis of the env gene product, gp70, of NZB xenotropic virus but other viral gene products.

Rapid method for the quantitation of cholesterol in human serum lipoproteins by high performance liquid chromatography.

A simple and rapid method for the quantitation of cholesterol in human serum lipoproteins (VLDL, LDL, HDL2, and HDL3) was developed (1). The content of cholesterol in each lipoprotein fraction was determined by means of a commercial enzymatic reaction kit after separation by high performance liquid chromatography with gel permeation columns. The quantitation of cholesterol could be performed with only 10-20 mu l of serum in less than 50 min by measuring A550 after passing the mixed eluate and enzyme solution through an on-line reactor system of a high-speed chemical derivatization liquid chromatograph. The precision, reproducibility and sensitivity of the quantitation of cholesterol with this method were acceptable, and the values of HDL-cholesterol determined by this method correlated well with those found by the heparin-manganese chloride precipitation method (r=0.958, n=93).

Fractionation of human serum lipoproteins by high performance liquid chromatography. I.

Immunologically pure lipoprotein fractions of human serum were subjected to high performance liquid chromatography (HPLC) on a combination system of aqueous gel permeation columns. Each lipoprotein fraction appeared at a distinct elution volume as a single homogeneous peak. Elution was carried out with 0.1 M Tris-HCl buffer solution, pH 7.4, or 0.15 M NaCl solution and no significant differences in elution volume and elution pattern were found. The lipoprotein mixtures (total lipoprotein fraction d less than 1.210) of healthy and pathological subjects were subjected to HPLC and showed characteristic elution patterns which could be of diagnostic significance.

Follow-up study on treatment in 27 patients with Cushing's disease: adrenalectomy, transsphenoidal adenomectomy and medical treatment.

27 patients with Cushing's disease were treated over a period of 18 years at the Departments of Medicine and Surgery, Nagoya University School of Medicine and the following results were obtained. 1) Adrenalectomy. 21 of 27 patients with Cushing's disease underwent adrenalectomy. 19 patients had total bilateral adrenalectomy and 2 patients unilateral adrenalectomy. 4 patients died, the cause of death not being related directly to adrenalectomy. 9 of 15 bilaterally adrenalectomized patients had hyperpigmentation even though they had been given substitution therapy with cortisol 20-30 mg daily. They had elevated plasma ACTH levels, which were not completely suppressed by 2 mg of dexamethasone or 2.5 mg of bromocriptine per day. 2) Adenomectomy, 5 patients had adenomectomy via the transsphenoidal approach. 3 patients were cured but one of them has required postoperative substitution therapy with cortisol for hypopituitarism for one year until today. 2 of 5 adenomectomized patients had a recurrence of Cushing's syndrome after remission for 6-8 months. One of these recurrent cases has been subsequently treated successfully with bromocriptine, a dopaminergic drug. 3) Medical treatment. 2.5 mg per day of bromocriptine has been effective in 2 patients without a pituitary adenoma and ineffective in the other 4 patients with a pituitary adenoma. 24 mg per day of cyproheptadine, an antiserotoninergic drug was not effective in any of the 4 patients with a pituitary adenoma.

Induction of leucocyte migration inhibitory factor (LIF) by stimulation with free hapten and water-insoluble epoxy resin.

The indirect leucocyte migration inhibition test was applied to the contact sensitivity to a hapten (dinitrofluorobenzene, DNFB) and a water-insoluble epoxy resin (Araldite). Sensitized guinea-pig leucocytes were pulsed with DNFB, which was not conjugated to a carrier protein, or with Araldite which was made particulate by ultrasonication. When these pulsed leucocytes were incubated for 24 hr, they released significant amounts of leucocyte migration inhibitory factor (LIF). The optimal dose for pulse-stimulation was 10 microgram/ml. A correlation was observed between the degree of contact sensitivity and the production of LIF.

High-performance aqueous gel permeation chromatography of human serum lipoproteins.

A new application of high-performance aqueous gel permeation chromatography was developed for the analysis of human serum lipoproteins. A good combination of columns (TSK GEL, type PW and type SW) was found for the separation of serum lipoproteins: very low-density lipoprotein, low-density lipoprotein and high-density lipoproteins. Analyses of serum lipoproteins from individual normal subjects and pathological subjects were performed by this combination of columns. The effects of pH and salt concentration fo the eluent on the separation of lipoproteins were also investigated.

Rapid analysis of cholesterol of high density lipoprotein and low density lipoprotein in human serum by high performance liquid chromatography.

A simple and rapid method for the determination of cholesterol in human serum lipoproteins (VLDL, LDL, HDL2, and HDL3) was described. The contents of cholesterol of lipoprotein fractions from a very small amount of serum (20 microliter) can be determined by the enzymatic assay after separation by high performance liquid chromatography in less than 50 min. The method is very useful for studying lipoprotein metabolism and related diseases.

Analysis of cholesterol in high density lipoprotein subfractions by high performance liquid chromatography.

We succeeded in the separation of human serum high density lipoprotein into their subfractions (HDL2 and HDL3) by high performance liquid chromatography with gel permeation columns (1, 2). The cholesterol contents in HDL2 and HDL3 for normal men and women (n = 42) and for the patients with atherosclerotic heart disease (n = 20) were measured by this separation method. The results in our experiment agree closely with those reported by other investigators using the ultracentrifugal method.

[Evaluation of the exposure to organic solvents by means of urinalysis for metabolites (author's transl)].

The method of biological monitoring of the exposure to organic solvents by means of urinalysis is discussed. The topics include the variety of the solvents to which the method of monitoring is applicable, choice of time for urine sampling, possible correction of the results for urine density, and establishment of the biological threshold. Recent progress is reviewed in particular, in the cases of benzene, toluene, xylenes, styrene, trichloroethylene, tetrachloroethylene and 1, 1, 1-trichloroethane.

[The correlationship between basal prolactin levels and the suppressive effect of CB-154 or L-DOPA on GH release in acromegalic patients (author's transl)].

It has been reported that the plasma growth hormone (GH) and prolactin (PRL) levels in acromegalic patients were suppressed after intensive administration of dopaminergic drugs, but we found that the plasma GH levels in some acromegalic patients were not suppressed. Plasma GH and PRL levels after a single oral administration of CB-154 (2.5mg) and L-DOPA (1g) were measured in 18 active acromegalic patients with and without galactorrhea. 1. The mean plasma GH levels after the administration were clearly suppressed in 8 patients with galactorrhea, while they were not suppressed in 10 patients without galactorrhea. These drugs were more effective in suppressing plasma GH levels in acromegalic patients with galactorrhea and hyperprolactinemia. 2. We calculated the responsiveness of the plasma GH level as follows: GH responsiveness (%) = (mean plasma GH level after the administration of CB-154 or L-DOPA)/ (basal GH level) x 100. The basal plasma PRL levels were inversely correlated with GH responsiveness (CB-154: r=-0.690, p less than 0.01. L-DOPA: r=-0.541, p less than 0.05). It was found that the effect of dopaminergic drugs on plasma GH levels was closely correlated with basal PRL levels in acromegalic patients. This implies that the chronic administration of CB-154 may be effective in acromegalic patients with galactorrhea and hyperprolactinemia.

Immunochemical properties of phosphatidyl cholesterol and its homologue.

1,2-Dipalmitoyl-rac-glycerol-3-phosphoryl-3'-cholesterol (PCH) and 1,2-dipalmitoyl-rac-glycerol-3-phosphoryl-20'-(3-hydroxy norpregn-5-ene) (PET) were combined with poly-L-lysine to form a PCH-poly-L-lysine complex and a PET-poly-L-lysine complex, respectively. These complexes were subcutaneously injected into rabbit foot pad with Freund's complete adjuvant. PCH antiserum showed specificites against the phosphatidyl group, the cholesterol moiety and the side chain of cholesterol. PET antiserum contained the specific antibodies against the phosphatidyl group, the cholesterol moiety and the OH-group at the C3 position of the cholesterol molecule.
