Fulminant tropical splenomegaly syndrome.

Eight patients with the tropical splenomegaly syndrome who presented with an acute febrile illness and severe anaemia due to haemolysis are described. None was pregnant. All were treated with anti-malarial drugs, four received steroids, and another two steroids and other cytotoxic drugs. Two patients died from causes which were, in part, attributed to this complication of the tropical splenomegaly syndrome. This mode of presentation is seldom documented in this condition. The management and differential diagnosis are discussed.

Electrocardiographic changes in African trypanosomiasis caused by Trypanosoma brucei rhodesiense.

The electrocardiographic findings in 40 patients with Trypanosoma brucei rhodesiense infection are reported. Using rigid diagnostic criteria 7 out of 18 patients (39%) had abnormal electrocardiograms before any form of therapy and 22 of the 40 patients (55%) had abnormal electrocardiograms at some stage of the disease or its treatment. The electrocardiographic abnormalities are described and discussed and the literature is reviewed.

The incidence of hepatitis-B-antigen and anti-hepatitis-B among blood donors in Ndola, Zambia.

Hepatitis-B antigen (HBsAG) was detected in the sera of 29 or 250 (11.6%) blood donors in Ndola, Zambia, most of whom were prisoners or students. The incidence among prisoners was 15.1%, among students only 1.6%. Thirteen sera were subtyped, nine carried the y and four the d antigenic determinant. Although there was a marked difference in the incidence of HBs antigen between prisoners and students, when donors were tested for antibody to HBsAg (anti-HBs) this positive. Overall, anti-HBs was detected in 132 of 250 blood donors (52.8%). The sensitivity of radioimmunoassay, counter-immunoelectrophoresis and a newly developed passive haemagglutination (PHA) test, employing tanned formalinised antibody coated turkey erythrocytes, was compared. Although radioimmunoassay provided the most sensitive method, the PHA test compared extremely favourably and was considered to be particularly suitable for use in the tropics.

Ribonuclease-sensitivity of covalently closed rat liver mitochondrial deoxyribonucleic acid.

Preparations of covalently closed mitochondrial DNA of rat liver contain 10-30% of molecules that are converted into relaxed circular molecules after treatment with ribonuclease. Control experiments, with covalently closed bacteriophage PM2 DNA, indicate that ribonuclease-sensitivity cannot be induced either by depurination or by incubation with reducing agents.

Isolation and characterization of mitochondrial deoxyribonucleic acid of Acanthamoeba castellanii.

DNA was prepared from isolated mitochondria of Acanthamoeba castellanii and was shown to behave as a single component in density gradients, on ;melting' and on renaturation. From measurements of renaturation kinetics, sedimentation coefficient and electron micrographs the genome size of the mitochondrial DNA was calculated to be about 3.4x10(7) daltons. A small proportion of the preparations could be isolated as relaxed circular molecules of mean contour length 16.2mum.

Purification and properties of rabbit muscle L-glycerol 3-phosphate dehydrogenase.

A modified procedure has been developed for the purification of rabbit muscle l-glycerol 3-phosphate dehydrogenase. The product of the preparation satisfies all criteria of homogeneity. Some physical properties of the enzyme have been re-investigated. The results suggest that previous preparations may have been contaminated with significant amounts of heavy-molecular-weight protein.

Mitochondrial deoxyribonucleic acid from Tetrahymena pyriformis and its kinetic complexity.

1. Mitochondrial DNA from Tetrahymena pyriformis strain T has a buoyant density (rho) of 1.685 compared with rho1.688 for whole cell DNA. Mitochondrial preparations from T. pyriformis strain W show an enrichment of a light satellite (rho1.686), although this is not obtained free from nuclear DNA (rho1.692). 2. T. pyriformis mitochondrial DNA renatures rapidly and the kinetics of this process indicate a complexity of approx. 3x10(7) daltons. 3. The base-pairing in the renaturation product is of a precise nature, since the ;melting' temperature (80.5 degrees C) is indistinguishable from that of the native DNA (80.5 degrees C). 4. Centrifugation of mitochondrial DNA in an alkaline caesium chloride density gradient gives two bands, implying the separation of the complementary strands.
