Specificities of anti-rabbit VH antibodies in mammalian and fowl antisera.

Antibodies against rabbit VH-determinants were produced by immunizing mammals and birds with rabbit IgG, IgA or VH. Most of the anti-VH produced in mammals was specific for allotype VH-determinants, but small amounts of antibody against non-allotype VH-determinants were also produced. By contrast the immunization of chickens led to the production of predominantly non-allotype VH antibody. Further analysis of the antibody specificity revealed the production of antibody against VH-determinants available in the intact immunoglobulin (H- + L-chain) as well as antibody reacting with determinants only accessible in the absence of L-chain. Antibody of the latter specificity prevailed after immunization with VH. The purified VH was not deficient in antigen determinants when compared with VH in the intact immunoglobulin (IgA). Unexpectedly, none of our chicken anti-rabbit VH antisera showed crossreactivity with VH from other mammalian species.

Cytomegalovirus antibodies in epileptics receiving diphenylhydantoin.

Cytomegalovirus (CMV) antibodies were determined by indirect haemagglutination in 53 epileptics receiving long-term diphenylhydantoin (DPH) therapy and in 53 matched controls. Absorption of serum IgG, IgA and IgM performed in 12 sera showed that the antibodies were of the IgG class. A decreased incidence of high CMV antibody titres (greater than or equal to 320) was found in epileptics (11.3%) compared with controls (34.0%) (0.02 greater than P greater than 0.01), whereas antibody titres greater than or equal to 40 were found in 37.7% of epileptics and in 45.3% of controls (n.s.). The CMV antibody incidence and titre range were similar in patients with symptomatic (37.9%) and idiopathic (37.5%) epilepsy, suggesting that the DPH treatment was responsible for the decreased antibody occurrence in patients. No correlation between CMV antibody titres and the serum immunoglobulin levels or the DPH concentration or clearance could be established in the epileptics.

Reaction against liver cells by human antiactin antibodies purified by affinity chromatography.

Human antiactin antibodies were isolated from human smooth muscle antibodies by affinity chromatography using actin extracted from rabbit striated muscle and coupled to activated CH-Sepharose 4B. The eluate contained IgG and IgM smooth muscle antibodies reacting against muscularis mucosae of the stomach and blood vessels. Further the eluate contained IgM antibodies acting against the surface of isolated hepatocytes and the periphery of hepatocytes of sections of liver cells. These results indicate that antiactin antibodies possibly react with the hepatocyte membrane as has been described with regard to other antibodies.

Acute and subacute symptoms among workers in the printing industry.

The study population comprised 52 male printers and 52 controls. Each person was interviewed about job history, general health, and work-related symptoms. Symptoms from eyes and airways, neurological symptoms, and general symptoms were recorded. A lung function test and a measurement of the sense of smell were also carried out. The printers had significantly more eye, airway, and neurological symptoms than the controls; the main complaints being irritation of eyes, nose, throat, and a reduced sense of taste. The neurological symptoms were disorders of vision, vertigo, feeling of intoxication, and headache. Furthermore, abdominal pain and flatulence occurred more often among the printers. The symptoms showed no relation to age or job seniority, but neurological and general symptoms were related to shift work. No difference in lung function was found between the two groups. The printers had a slightly lower threshold of smell than the controls. Although the total load due to organic solvents and dust in the air was far below legal limits, the number of magnitude of symptoms experienced by the printers exceeded what is supposed when norms for workroom exposure are set. It is suggested that either the irritative effects of solvents are underestimated or the assumption of additive effects when great numbers of solvents are found does not hold true. A reduction of the number of solvents by eliminating the most toxic solvents or by using dyes without solvents is suggested.

Nasal histology and nickel concentration in plasma and urine after improvements in the work environment at a nickel refinery in Norway.

Nasal histology and nickel concentration in the plasma and urine of nickel workers were reexamined after 4 a for an evaluation of the effect of improvements in the refining process and work conditions. Ninety-eight (31%) nickel workers who were part of a previous study were reexamined. In addition, 33 of the original 38 workers with nasal epithelial dysplasia were studied for possible regression of the histological changes. Reduction in the atmospheric content of nickel and improved hygienic conditions lowered the concentration of nickel in plasma and urine by approximately 50%. The mean histological score and frequency of dysplasia was virtually the same in the two investigations, 12 and 13%, respectively. Nasal biopsies are of limited diagnostic value in the examination of individual cases. However, the method seems to be of value in identifying and monitoring groups of persons with increased risk of developing nasal carcinoma.

Reduction of toxic indoor emissions.

A toxicological evaluation of the emission from building materials is given for irritants, allergens, odorous substances, carcinogens and teratogens and systemic toxic agents. The experience from a notification system within the National Danish Inventory of Toxic Substances and Products shows that it is possible to effectively survey building materials and products and their areas of application. As an example, data on nearly 1500 epoxy and polyurethane products (sealants, glues/adhesives and paints/lacquers) have been to estimate the percentage of those containing (a) substances known for danger or risk of irreversible effects, (b) organic solvents (greater than 5%) or (c) chemicals with irritant properties. A labelling of products is now prescribed.

Eggs: conveniently packaged antibodies. Methods for purification of yolk IgG.

Eggs from immunized chickens may provide a convenient and inexpensive source of antibodies. We describe two simple and efficient methods for purification of IgG from yolk. The antibody is rendered useful for most currently employed immunological techniques. Amounts of antibody corresponding to almost half a litre of antiserum may be recovered from a chicken in one month.

Gamma-glutamyltransferase, aspartate aminotransferase and alkaline phosphatase as markers of alcohol consumption in out-patient alcoholics.

Serum activity of gamma-glutamyltransferase, aspartate aminotransferase and alkaline phosphatase were determined in 316 patients attending an out-patients clinic for treatment of alcoholism. The activity of gamma-glutamyltransferase was raised in 34% and that of aspartate aminotransferase and alkaline phosphatase in 18% and 7%. Neither the activity of gamma-glutamyltransferase, aspartate aminotransferase nor alkaline phosphatase showed any significant (P greater than 0.05) correlation with the history of alcohol consumption. The activities of gamma-glutamyltransferase and aspartate aminotransferase were raised significantly more often in patients with recent alcohol consumption than in patients who had abstained for more than 9 days. The concentration of alkaline phosphatase was not significantly (P greater than 0.05) different in these groups. The predictive value of raised and normal activities of gamma-glutamyltransferase, in deciding whether a patient had had recent alcohol consumption or not, was not superior to the predictive value of raised and normal activities of aspartate aminotransferase.

Human response to controlled levels of combinations of sulfur dioxide and inert dust.

Under controlled conditions in an environmental chamber 16 healthy young volunteers were exposed to combinations of sulfur dioxide (SO2) (2.6 or 13 mg/m3) and inert plastic dust (2 or 10 mg/m3) or of SO2 (13 mg/m3) and dust (10 mg/m3) coated with vanadium. During periods of clean air and during exposures of 5-h duration nasal mucus flow rate, nasal airflow resistance, forced vital capacity, and subjective discomfort were measured. Reductions in nasal mucus flow rate, forced expiratory flow (FEF25-75%), and discomfort were related principally to the SO2 concentration. The combined effects of SO2 and dust were, at the most, additive, and there was no indication of potentiation effects. No effect could be attributed to the coating of the dust with vanadium.

Smooth-muscle antibodies in rheumatoid arthritis.

Smooth-muscle antibodies (SMA) detected by the indirect immunofluorescence method were found more often in rheumatoid arthritis (RA) patients (15.3%) than in normal controls (7.6%) (0.02 > p > 0.01). The increased frequency was due to IgG-antibodies which occurred in 12.2% of RA patients, in 4.1% of normal controls and in 7.3% of patients with other arthritic diseases. Eight of 9 IgG-SMA-positive RA sera reacted with F-actin, and one serum contained non-actin antibodies. In RA, ANA were found in 35.7%, glomerular antibodies in 1.0%, parietal cell antibodies in 4.1% and mitochondrial antibodies in 2.0%. SMA were associated with the occurrence of rheumatoid factors and mitochondrial antibodies (0.02% > p and 2p = 0.04, respectively). In the SMA-positive group the erythrocyte sedimentation rate was higher and elevated serum alkaline phosphatase values were found more often than in the SMA-negative group.

Variations of nickel in plasma and urine during the work period.

One of the basic problems in monitoring exposure of nickel refining workers has been the unclear correlation between plasma nickel, urine nickel, and airborne nickel during the eight-hour work period or throughout the 24-hour day. In previous studies the authors have demonstrated the value of biological sampling in an occupational health program in the nickel processing industry. Whether a single plasma or single urine sample is an adequate gauge of exposure or whether 24-hour urine specimens or a three-day average plasma level are more accurate indices remained to be determined. The purpose of this paper is to report the results of studies of diurnal variations which affect the significance of single biological samples. In addition, data gathered on the correlation of plasma and urine nickel levels with soluble and insoluble nickel compounds in the atmosphere are presented.
