[Color measurements of 1,944 anterior teeth of people in southwest of China-discreption].

OBJECTIVE: To study the color distribution space of natural anterior tooth crowns of people in southwest China and effects of sex and age on tooth color. METHODS: The subjects studied consisted of 162 persons aged from 20 to 73 years and born in southwest China. A total of 1,944 healthy anterior teeth were measured in-vivo with a fiber-optical spectrophotometry system. RESULTS: Statistical analysis showed that the color distribution space of Chinese tooth crowns was wide. The value of L* varied from 42.383 to 67.428, the value of a* from -4.750 to 1.371, the value of b* from 1.88 to 20.268, the value of x from 0.298 to 0.393, the value of y from 0.332 to 0.392, and the value of Y from 19.383 to 34.025. The color difference between the upper and lower limits of 95% confidence was 31.667 CIE delta E units. The difference in colorimetry between male and female was not significant. The results showed that in three sections of teeth (incisal, middle and cervical) the values of L* and a* were negatively related to age (r < 0, P < 0.01); the value of b* was positively related to age just at the cervical and middle sites of teeth (r > 0, P < 0.01) and was not related to age at the incisal site of teeth (P > 0.05). CONCLUSION: The color distribution space of Chinese anterior tooth crowns is wide. There is no difference in the anterior tooth color between male and female subjects. The values of L* and a* and negatively related to age except the value of b*.

[Study on the immunologic function of lymphocyte in cord blood].

OBJECTIVE: To explore immunologic function of T lymphocytes in human cord blood (HUCB) and provide theoretical basis for clinical application of HUCB. METHODS: Immunophenotype and co-stimulated molecules were detected by flow-cytometry using a panel of double labelled McAb, intracellular cytokines (interferon-gamma, interleukin-4) secreted by T lymphocytes by micro-culture of whole blood, and CD25 expression of T lymphocyte by PHA stimulation. T lymphocytes from adult peripheral blood (PB) were used as control. RESULTS: There are a higher CD4+/CD8+ ratio of T subset and a lower percentage of CD8+ lymphocytes in HUCB than that in adult PB (P < 0.01), and more than 90% of HUCB T-cells co-expressed CD45RA naive antigen which was significant higher than the percentage in adult PB (P < 0.01). The percentages of IFN-gamma and IL-4-producing CD4+ and CD8+ T cells from HUCB were lower than that of the control group. T cells in HUCB were able to be activated by PHA, but the phases of activation differed from adult PB T cells. CONCLUSIONS: It suggested that the low ability to secret cytokines and high percentage of naive T cells might be responsible for the decreased incidence of acute and chronic GVHD and provided possible explanation for the clinical result in HUCB transplantation. T cells in HUCB is a potential resource of adoptive immunotherapy.

[Clinicopathological features of malignant ovarian neoplasms arising from ovarian endometriosis: a report of 26 cases].

OBJECTIVE: To clarify the features of malignant ovarian neoplasms arising from ovarian endometriosis. METHODS: A total of 26 women with malignant ovarian neoplasms arising from ovarian endometriosis were analyzed retrospectively. RESULTS: Dysmenorrhea and pelvic mass were the most common clinical features. Among 18 cases who underwent B-ultrasound or color doppler ultrasound examination, solid foci in the pelvic masses were found in 10 cases. The main histologic types of ovarian malignancy were endometrioid adenocarcinoma and clear cell carcinoma. Microscopically atypical endometriosis with the tumors were found in 15 cases. International Federation of Gynecology and Obstetrics stage: stage I 21(81%) cases, stage II 3(12%) cases, stage III 2 (8%) cases. CONCLUSIONS: Clinical diagnosis of malignant ovarian neoplasms arising from ovarian endometriosis in early stage is difficult, and B-ultrasound examination is more valuable for diagnosis. It is suggested that close serutiny of endometrial hyperplasia, cellular atypia and malignancy in ovarian endometriosis be essential to understand the origin and development of malignant neoplasms arising from ovarian endometriosis.

Effect of ipratropium bromide on airway and pulmonary muscarinic receptors in a rat model of chronic obstructive pulmonary disease.

OBJECTIVE: To observe the level of muscarinic receptors in airway and lung tissues, and the effect of inhaled ipratropium bromide on these receptors in a rat model of chronic obstructive pulmonary disease (COPD). METHODS: This model was developed by exposure of rats to 250 ppm SO2 gas, 5 h/d, 5 d/wk, for a period of 7 wk. The COPD rats inhaled 0.025% aerosolized iratropium bromide for 20 min, 2 times daily, in an airtight chamber. Muscarinic receptors in airway and lung tissues of normal rats, ipratropium bromide-treated COPD rats and the recovering COPD rats were measured by the radio-ligand binding assay. RESULTS: Airway/lung pathology and pulmonary function tests showed that chronic SO2 exposure caused pathophysiologic changes similar to those observed in human COPD. The density (0.038 +/- 0.011, pmol/mg protein) and affinity (Kd, 23 +/- 11 pmol/L) of muscarinic receptors in airway and lung tissues of COPD rats were not changed compared with those of normal control rats (0.030 +/- 0.008 and 29 +/- 19, respectively, P > 0.05). Densities of the muscarinic receptors were not changed after inhalation of ipratropium bromide for 5 days, but increased significantly after inhalation for 30 days, as compared with those of the untreated COPD rats. The muscarinic receptors returned the normal levels at day 6 after cessation of ipratropium bromide treatment. There were no differences among different groups of rats in equilibrium dissociation constants (Kd). CONCLUSION: A rat model of COPD with pathophysiologic changes similar to the human counterpart was developed using chronic SO2 exposure. There was no significant change in the number and function of muscarinic receptors in airway and lung tissues of the COPD rats, but upregulation of the muscarinic receptors was observed after long-term inhalation of ipratropium bromide.

In vitro renaturation of recombinant human pro-urokinase expressed in Escherichia coli.

OBJECTIVE: Recombinant human pro-urokinase forms insoluble inclusion body when overexpressed in Escherichia coli. It must be denatured and renatured in vitro so that it can acquire activity. This study aimed at increasing the renaturation yield of denaturant pro-urokinase. METHODS: We evaluated the basic renaturation conditions of pro-urokinase through qualitative and quantitative analysis of pH, temperature, denatured concentration, protein concentration, and the ratio of reduced and oxidized thiol reagents. We also compared the effects of nonspecific additives, step-wise dilution and urea gradient dialysis. RESULTS: We defined the optimal conditions of pro-urokinase renaturation with a yield of about 20%-30%. CONCLUSION: Different recombinant denatured proteins have different renaturation conditions due to their different molecular sizes, molecular constructions, disulfide bond numbers, and hydrophobicity. The renaturation yield can be increased by optimizing the renaturation conditions of a specific protein.

MR and cine-MR imaging in the diagnosis of infantile congenital heart disease.

OBJECTIVE: To assess the effectiveness of magnetic resonance imaging (MRI) and cine magnetic resonance imaging (cine-MRI) in the diagnosis of infantile congenital heart disease. METHODS: A total of 34 cases were studied with MRI and cine-MRI. The data were analyzed and compared with those of two-dimensional echocardiography (2DE), cardioangiography (CAG) and surgery. RESULTS: The size of the defect or its caliber obtained from MRI in 6 patients with left to right shunt congenital heart disease was compatible with that observed in surgery (P = 0.924). Comparison of cine-MRI and CAG in 28 patients with complicated congenital heart disease showed that the diagnosis of 27 cases by cine-MRI was the same as that by CAG. CONCLUSION: Both MRI and cine-MRI play an important role in diagnosing infantile congenital heart disease.

[Computing ECG based on action potential of single cardiac cell].

This paper introduces an ECG computing algorithm that computes ECG based on the action potential of single cardiac cell. Taking solid angle analysis as tool, this algorithm analyzes the field potential of a cardiac cell that connects and communicates with neighboring cells. This algorithm has been implemented in our electrophysiological model CardioAuto, which was built with extended Cellular 3.0-a cellular automata system. Algorithm and simulation results reveal that it is the transmembrane potential slope among cell group that contributes to ECG waveform generation and determines the waveform deflection. To a specific lead location, whenever cell group has a transmembrane potential slope that proximal end is higher than distal end, a downward ECG deflection is generated. Whenever cell group has a reverse transmembrane potential slope, an upward ECG deflection is generated. Whenever there is no transmembrane potential slope, ECG keeps on baseline. According to our algorithm, the significance of normal and many abnormal ECG waveforms can be analyzed and interpreted at cellular level.

[Study on the muscarinic receptor and its subtypes in patients with chronic obstructive pulmonary disease].

OBJECTIVE: To investigate the M receptor and its subtypes in the lung tissue of patients with chronic obstructive pulmonary disease (COPD). METHODS: Muscarinic cholinergic receptors have been identified and characterized by radioligand binding assay in the lung tissue specimens of patients with COPD. Competitive binding experiments with pirenzepine and methoctramine were used to characterize muscarinic subtypes. RESULTS: The contents of M-receptor were (64 +/- 10), (42 +/- 18) fmol/mg. protein in normal group and COPD patients respectively. No significant difference was observed in antagonist affinity (K(D)) among normal group and COPD patients. The ratio of subtype M(1) was higher in COPD patients (67.2 +/- 2.7)% than in normal group (74.2 +/- 4.8)%, M(2) was lower [(29.3 +/- 1.7)% vs (16.8 +/- 4.4)%] and M(3) was higher [(3.6 +/- 2.9) % vs (9.3 +/- 4.1)%] respectively. CONCLUSION: The number of muscarinic cholinergic receptors is decreased in COPD patients, but the ratios of subtype M(1) and M(3) are increased and the subtype M(2) is decreased. The changes of the distribution of the subtypes of M-receptors is an important pathophysiologic change of COPD.

OBJECTIVE: To investigate the feasibility of detecting SYT-SSX fusion gene in paraffin-embedded tissues and its diagnostic significance for synovial sarcoma (SS). METHODS: Formalin-fixed, paraffin-embedded samples of 38 cases of SS and 40 cases of control tumors (including spindle cell sarcoma and metastatic adenocarcinoma) were retrived from archival materials. SYT-SSX fusion transcripts were detected in all samples by RT-PCR. House-keeping gene Porphobilinogen Deaminase (PBGD) was used to detect the quality of mRNA. RESULTS: PBGD mRNA was detected in 64 of the 78 tumor cases (82.1%). SYT-SSX fusion transcripts was detected in 33 of the 38 synovial sarcoma specimens. No SYT-SSX mRNA expression was found in control tumors. SYT-SSX mRNA expression rate reached 89.2% (33/37) in synovial sarcomas after exclusion of 1 case which was negative for both SYT-SSX and PBGD. In 33 SYT-SSX positive synovial sarcomas, 22 had SYT-SSX1 and 6 had SYT-SSX2 fusion transcripts. In 5 cases, the fusion type could not be distinguished. There was an association between SYT-SSX fusion type and histologic subtype. All 10 biphasic synovial sarcoma cases had the SYT-SSX1 fusion, whereas 12 of 18 monophasic synovial sarcoma had the SYT-SSX1 and 6 had the SYT-SSX2 fusion gene (P < 0.05). CONCLUSIONS: (1) Detection of SYT-SSX fusion gene in paraffin-embedded tissues was found to be a sensitive and specific method for the diagnosis and differential diagnosis of synovial sarcoma. (2) There was an association between SYT-SSX fusion type and histologic subtype.SYT-SSX2 fusion transcript could only be found in monophasic synovial sarcomas.

[Effects of low energy laser on tooth movement and remodeling of alveolar bone in rabbits].

OBJECTIVE: The aim of this study was to investigate effects of low energy laser on the experimental tooth movement and the remodeling of alveolar bone in rabbits. METHODS: A total of 42 white rabbits were chosen and randomized into one control group and six experimental groups, with 6 rabbits in each group. After anesthesia with 2% pentobaritone sodium, orthodontic appliances consisting of a coil spring connected bilaterally the upper first molar with the upper incisor by using a ligature wire. The force exerted at the time of insertion was approximately 80 g. The left side served as the control side, and the right side was the experimental side treated by receiving irradiation of low energy laser. The treatment periods of different groups lasted separately for 1, 3, 5, 7, 14, 21 days respectively. The displacement extent of teeth was measured by employing the computer image analyzing system. The results were analyzed statistically. Through HE staining, the histomorphological character of tissue around first molar was also investigated, and numbers of osteoclasts were counted. RESULTS: The displacement extent of teeth on the experimental side, which was irradiated by low energy laser, was more obvious than that of the normal control side. The difference was statistically significant 1, 3, 14, 21 days after the beginning of the treatment. Through histological observation under a light microscope, the osteoclasts and osteoblasts on the experimental side remained more active than those of the control side. There is significant difference in amount of osteoclasts between the experimental and the control sides, 3, 5 or 7 days after the treatment (P < 0.01, P < 0.05). CONCLUSION: The irradiation of low energy laser promote tooth movement and remodeling of alveolar bone.

[Detection of telomerase activity in laryngeal carcinoma and the association between telomerase activity and lymphatic metastasis].

OBJECTIVE: To investigate the telomerase activity in human laryngeal squamous cell carcinomas (LSCC) and the association between telomerase activity and lymphatic metastasis. METHOD: Telomerase activity was determined in 47 specimens of LSCC and 10 specimens of laryngeal inflammatory polyps using PCR-ELISA. RESULT: Telomerase activity was detected in 39 of 47 (83%) LSCC, but was not detected in all 10 laryngeal inflammatory polyps. There were significant differences between them (P < 0.01). Telomerase activation in LSCC with positive cervical lymph node involvement appeared to be higher (13 out of 13 cases, 100%) than in LSCC with negative cervical lymph node involvement (26 out of 34, 76.47%), (P < 0.05). CONCLUSION: Telomerase is activated frequently in LSCC and may become a biomarker of LSCC. The detection of telomerase activity in LSCC may have some value for predicting metastasis potential and prognosis and can guide the clinical practices applying selective neck dissection.

[Study on the genotoxicity of dibromoacetic acid in drinking water].

Dibroacetic acid (DBA) is a byproduct of drinking water disinfected by halogenation and it belongs to halogenated acetic acids. It also can be found in ozonated drinking water. Four short-term assays were performed to detect the genotoxic activity of DBA. In the Ames test, DBA showed mutagenic activity in Salmonella typhimurium strain TA100 with or without rat liver-derived metabolic activation system (S9). In UDS test, DBA was found to induce DNA damage in rat primary hepatocyte. Mouse bone marrow micronucleus test indicated that DBA induced the damage of chromosome. In NIH3T3cell micronucleus test, DBA increased the number of micronuclei in cells. Our results suggest that DBA induce DNA damage and has genotoxic activity.
