Analysis of expressed sequence tags from two starvation, time-of-day-specific libraries of Neurospora crassa reveals novel clock-controlled genes.

In an effort to determine genes that are expressed in mycelial cultures of Neurospora crassa over the course of the circadian day, we have sequenced 13,000 cDNA clones from two time-of-day-specific libraries (morning and evening library) generating approximately 20,000 sequences. Contig analysis allowed the identification of 445 unique expressed sequence tags (ESTs) and 986 ESTs present in multiple cDNA clones. For approximately 50% of the sequences (710 of 1431), significant matches to sequences in the National Center for Biotechnology Information database (of known or unknown function) were detected. About 50% of the ESTs (721 of 1431) showed no similarity to previously identified genes. We hybridized Northern blots with probes derived from 26 clones chosen from contigs identified by multiple cDNA clones and EST sequences. Using these sequences, the representation of genes among the morning and evening sequences, respectively, in most cases does not reflect their expression patterns over the course of the day. Nevertheless, we were able to identify four new clock-controlled genes. On the basis of these data we predict that a significant proportion of the expressed Neurospora genes may be regulated by the circadian clock. The mRNA levels of all four genes peak in the subjective morning as is the case with previously identified ccgs.

Streptococcus australis sp. nov., a novel oral streptococcus.

Strains of streptococci were isolated from the mouths of children attending the United Dental Hospital, Sydney, Australia. These strains were analysed biochemically using the Rapid ID32 Strep microsystem, were subjected to DNA-DNA hybridization with other members of the oral streptococci and had their 165 rRNA analysed. On the basis of DNA-DNA hybridization, their nearest relative was Streptococcus parasanguinis, whereas, on the basis of 16S rRNA analysis, it was Streptococcus infantis. The name Streptococcus australis sp. nov. is proposed for the new species. The type strain is AI-1T (= ATCC 700641T = NCTC 13166T).

Force-frequency coefficient of symmetrical incomplete circular quartz crystal resonator.

The changes in the resonance frequencies of the thickness-shear vibration of symmetrical incomplete circular AT-cut quartz crystal resonators (QXRs), which are used as sensing elements in digital force sensors or pressure transducers, by the application of diametrical forces are discussed by considering the piezoelectric effect and the anistropic characteristics of crystal plates. Two-dimensional motion equations for predicting the frequency changes are derived from three-dimensional piezoelectricity equations, and equations deduced by Janiaud for solving the stress distribution in crystal plates are used to calculate the force-frequency coefficients of incomplete circular resonators. The results show that the piezoelectric effect of crystal plates decreases the force-frequency coefficient of the azimuth angle 0 degrees by 7% as compared with no piezoelectricity assumption. The incomplete circular shape can be applied to a larger load than the circular disc because the flat regions of the incomplete shape provide a distributed load application without the problems of stress concentration of a point force inherent to a circular disc. The incomplete shape gives an increased force sensitivity when compared with the complete disc near azimuth angle 0 degrees. The theoretical curves agree well with Ratajski's experimental results.

An engineered leucine zipper a position mutant with an unusual three-state unfolding pathway.

The leucine zipper is a dimeric coiled-coil structural motif consisting of four to six heptad repeats, designated (abcdefg)(n). In the GCN4 leucine zipper, a position 16 in the third heptad is occupied by an Asn residue whereas the other a positions are Val residues. Recently, we have constructed variants of the GCN4 leucine zipper in which the a position Val residues were replaced by Ile. The folding and unfolding of the wild-type GCN4 leucine zipper and the Val to Ile variant both adhere to a simple two-state mechanism. In this study, another variant of the GCN4 leucine zipper was constructed by moving the single Asn residue from a position 16 to a position 9. This switch causes the thermal unfolding of the GCN4 leucine zipper to become three state. The unfolding pathway of this variant was determined by thermal denaturation, limited proteinase K digestion, and sedimentation equilibrium analysis. Our data are consistent with a model in which the variant first unfolds from its N terminus and changes the oligomerization specificity from a native dimer to a partially unfolded intermediate containing a mixture of dimers and trimers and then completely unfolds to unstructured monomers.

Human cytomegalovirus with IE-2 (UL122) deleted fails to express early lytic genes.

Much evidence suggests that the major immediate-early (IE) transactivator of human cytomegalovirus (HCMV), IE-2, is likely to be critical for efficient viral replication; however, the lack of an IE-2 mutant HCMV has precluded an experimental test of this hypothesis. As an initial step toward characterizing an IE-2 mutant, we first cloned the HCMV Towne genome as a bacterial artificial chromosome (BAC) and analyzed the ability of transfected Towne-BAC DNA (T-BACwt) to produce plaques following introduction into permissive human fibroblasts. Like Towne viral DNA, transfected T-BACwt DNA was infectious in permissive cells, and the resulting virus stocks were indistinguishable from Towne virus. We then used homologous recombination in Escherichia coli to delete the majority of UL122, the open reading frame encoding the unique portion of IE-2, from T-BACwt. From this deleted BAC, a third BAC clone in which the deletion was repaired with wild-type UL122 was created. In numerous transfections of permissive human foreskin fibroblast cells with these three BAC DNA clones, the rescued BAC and T-BACwt consistently yielded plaques, while the UL122 mutant BAC never generated plaques, even after 4 weeks. Protein and mRNA of other IE genes were readily detected from transfected UL122 mutant BAC DNA; however, reverse transcription-PCR failed to detect mRNA expression from any of five early genes examined. The generalized failure of this mutant to express early genes is consistent with expectations from in vitro assays which have demonstrated that IE-2 transactivates most HCMV promoters. These experiments provide the first direct demonstration that IE-2 is required for successful HCMV infection and indicate that virus lacking IE-2 arrests early in the replication cycle.

The development and in-vitro evaluation of novel mixed metal hydroxy-carbonate compounds as phosphate binders.

The currently available phosphate binders are relatively inefficient and suffer from clinical side-effects of increased absorption of calcium and aluminium and the diarrhoea-inducing effects of magnesium. A new class of compounds based on mixed metal hydroxides has been developed and evaluated for their potential as phosphate binders. The mixed metal hydroxides were prepared using a standard procedure for hydrotalcite (Al2Mg6(OH)16.CO3.4H2O) by substituting Fe3+ for Al3+, with Mg2+ or Ca2+ as the divalent metal ion. Phosphate precipitation (binding) was examined at different pH values in aqueous solution and in various food mixtures in comparison with hydrotalcite, Al(OH)3, CaCO3 and Mg(OH)2 on the same weight-to-weight basis. A series of compounds with differing ratios of metal ions (Fe:Mg/Ca 1:2 or 1:3) gave analytically similar ratios to those predicted from the initial amounts added. CTFeCa bound > 90% phosphate in aqueous solution compared with 65% binding with CTFeMg, 85% binding with Mg(OH)2, and less than 30% binding for CaCO3 and Al(OH)3. The mixed metal compounds also bound up to 80% phosphate in various food matrices, which was relatively independent of changes in pH, compared with Mg(OH)2, where binding decreased from 85% at pH 3.0 to 25% at pH 8.0. Al(OH)3 and CaCO3 were relatively ineffective phosphate binders under all the conditions tested. The mixed metal hydroxides compounds show considerable promise as phosphate binders over those currently available and warrant further patient-based in-vivo testing.

Multipulse feed strategy for glycerol fed-batch fermentation: a steady-state nonlinear optimization approach.

During glycerol fed-batch fermentation, the process could be divided into multiple equal subintervals, and the feed operation was performed in pulse form at the start of each subinterval. Based on the macrokinetic models, the multipulse feed strategy for both glucose and corn steep slurry was determined by a general nonlinear optimization approach to maximize the final glycerol productivity and still control the residual glucose at a low concentration. The experimental results in a 600-mL Airlift Loop Reactor showed that the tested data with this strategy agreed well with the corresponding model prediction, and that the feed mode with nonlinear optimization could improve the glycerol productivity significantly compared with those determined just by limited experimental optimization in previous studies.

Electrolyte effects on electrochemical properties of osmium complex polymer modified electrodes.

The electrolyte effects on the electrochemical behaviors of osmium complex polymer modified electrodes were investigated by a comparison between two osmium complexes, [Os(bpy)2(PVI)10Cl]Cl (Os-PVI10) and [Os(bpy)2(PVP)10Cl]Cl (Os-PVP10). The electrode process at Os-PVI10 modified electrodes is reaction-controlled, while a diffusion-controlled electrode process exists at Os-PVP10 modified electrodes. Both the cation and anion in supporting electrolytes strongly affect their electrochemical behaviors, such as the redox potential, wave shape and peak current. These phenomena are attributed to a change in the film structure and polymer swelling in various supporting electrolytes. The influence of electrolyte anions on the electrochemical behaviors is related to their hydrophobicity. The electrode process of Os-PVP10 depends on the pH value of solutions, exhibiting different electron transfer mechanisms.

[Effects of taurine on brain development and learning ability in filial mice fed with high iodine].

OBJECTIVE: To observe the effects of taurine on brain development and learning ability in filial mice fed with high iodine. METHODS: Mice were divided into three groups, the first one fed with adequate dose of iodine, the second with high-dose iodine, and the third with high-dose iodine plus 0.6% taurine. All the male and female mice were mated three months after treatment. Relevant biochemical indicators in the brain tissues and their memory and learning ability were measured for their offsprings with 30 days old in the three groups. RESULTS: Brain contents of protein and DNA reduced in the high-dose iodine group, and there was no significant difference in them between groups with taurine and adequate-dose iodine 30 days after their birth. Activity of acetylcholine esterase (ACHE) increased and content of acetylcholine decreased in the hippocampus of filial mice at age of 30 days with high-dose iodine. Content of acetylcholine increased in the filial mice with high-dose iodine plus taurine than that with adequate-dose iodine, but there was no statistically significant difference in ACHE between them. CONCLUSION: Taurine could antagonize disturbance of brain development in filial mice caused by high-dose iodine.

Inhibition of steroidogenesis and induction of apoptosis in rat luteal cells by cell-permeable ceramide in vitro.

The present study was undertaken to investigate the effects of ceramide on progesterone production and apoptosis in rat luteal cells in vitro. Luteal cells were prepared from the ovaries of pregnant mare serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG) primed female Wistar rats and incubated with cell permeable C2-ceramide. The effects of ceramide on progesterone production and apoptosis in luteal cells were assessed by radio immunoassay (RIA) and flow cytometry analysis. In addition, changes in nitric oxide synthase (NOS) activity and nitric oxide (NO) production by luteal cells treated with C2-ceramide were also evaluated. Ceramide was found to reduce hCG-stimulated progesterone production in a dose-dependent manner, whereas it had little effect on basal progesterone content. Spontaneous apoptosis in luteal cells was observed after a 12-hour incubation in vitro and 5 mumol/L of ceramide significantly increased the apoptotic rate (P < 0.05). Enhanced apoptotic peak was seen in the histogram by flow cytometry. Moreover, 50 mumol/L of C2-ceramide significantly increased NOS activity (P < 0.01) and NO production (P < 0.001). It is suggested that ceramide may serve as an important signaling molecule mediating certain ovarian processes, such as luteal regression.

Ilioinguinal approach to manage benign pelvic and acetabular tumors.

Benign osteoblastic and cystic lesions involving the anterior column and wall of the pelvis and acetabulum are technically difficult areas to approach and achieve successful surgical outcomes. Tumors in this area are rare, leaving little information in the literature regarding how best to surgically approach them. Previous authors have reported small series of patients treated using anteromedial, iliofemoral, and intracapsular dissections. The ilioinguinal approach has been extensively described and used in the modern-day treatment of pelvic and acetabular fractures involving the anterior column and wall. Despite its use for acetabular trauma, application of the ilioinguinal approach in the treatment of benign pelvic and acetabular lesions has not been described. We reviewed the management of 5 patients with benign pelvic or acetabular tumors treated using the ilioinguinal approach. Diagnoses included aneurysmal bone cysts in 3 patients and osteoid osteomas in 2 patients, involving the pelvic anterior column and wall. The approach has permitted satisfactory exposure of the lesions for operative treatment. At an average follow-up of 3 years (range, 2-4 years), excellent functional outcome and no recurrences were noted. There were 2 patients with lateral femoral cutaneous nerve palsies, both of which resolved completely by 8 weeks. Encouraging results from our study demonstrate a new application for the ilioinguinal approach in the treatment of benign anterior column and wall lesions of the pelvis and acetabulum.

[The function of 5'flanking region of mouse neuronal CDC2-like kinase regulatory subunit p35].

OBJECTIVE: To clarify the possible mechanisms which control p35(nck5a) neural-specific expression. METHODS: Thirteen luciferase expression vectors which contain different parts of 5'flanking sequence were constructed. Transient expression assay, DNase 1 hypersensitive site(DHSS) assay and primer extension assay were made to reveal the possible mechanisms of p35(nck5a) expression control. RESULTS: It was found that 5'flanking sequence of p35(nck5a) contained 5455bp and there were a few possible promoters in the fragment. The result of transient expression assay in primary cerebrum cortex cell of rat (Neuron) and established non-neuron cell such as HeLa showed the loss of neural-specific expression of all the luciferase expression vectors. The result of DNase 1 hypersensitive site assay revealed a DHSS located at the -400bp upstream of p35(nck5a) in brain but not in liver. The primer extension assay found the putative transcription initiation site near the position of DHSS. CONCLUSION: The results above suggest that there is no cis-element to control the neural-specific expression in the 5'flanking sequence, and the control on the chromosome level may be critical to the neural-specific expression of p35(nck5a).

[Expression of telomerase reverse transcriptase and its relationship with tumor suppressor gene p53 in HCC].

OBJECTIVE: To explore the expression of human telomerase reverse transcriptase (HTERT) and its relationship with tumor suppressor gene p53. METHODS: RNA-RNA in situ hybridization (ISH) technique was applied to localize the expression of HTERT mRNA and immunohistochemistry (IHC) was used to observe the p53 protein expression in 34 specimens of HCC and their corresponding non-cancerous tissues. Both the significance of HTERT mRNA expression in the different histopathologic characters and its relationship with p53 protein were investigated. RESULTS: HTERT was detected in tumor tissue in 30 of 34 cases (88.2%), and all the 6 cases of small cancer (diameter<3cm) were positive. The positive rate of HTERT mRNA was higher in the tumor tissues with low-differentiation, extra- or intra-hepatic metastasis, uncompleted fibrotic capsule and/or the non-cancerous tissue with chronic active lesions or positive HBsAg. Intensive signal was seen both in the HCC cells along the front of invasive area and in a few non-cancerous liver cells with dysplasia. Of all 34 HCC, p53 protein was presented in 25 cases. There was a correlation between the expression of HTERT mRNA and of p53 protein. The coincidence rate of the expression (HTERT+/p53+ and HTERT-/p53-) was 79.4%. More intensity of HTERT signal could be seen in the cases of positive p53. HTERT mRNA was not found in normal liver tissues. CONCLUSIONS: High expression of HTERT mRNA is proved in most cases of HCCs. HTERT expression may relate to the pathogenesis and the invasion of HCC. p53 may play a role in regulating the expression of HTERT.

Inhibition of amyloidosis using low-molecular-weight heparins.

BACKGROUND: Amyloid diseases are characterized by the tissue deposition of extracellular proteinaceous material, which results in organ dysfunction and death. Colocalization of heparan sulfate (HS) proteoglycans to amyloid deposits suggests that they may be an early event in amyloid formation and play an important role in fibril formation. Structural analysis has demonstrated that HS interacts with amyloidogenic proteins resulting in structural changes that allow for an increase in beta-sheet content, possibly enhancing fibrillogenesis. Recent studies have shown that small-molecule anionic sulfonates or sulfates can arrest inflammation-associated (AA) amyloid induction. MATERIALS AND METHODS: In the present study, we examine the effect of low-molecular-weight heparins (LMWHs) on the development of amyloid in the mouse model of AA amyloid. In addition, in vitro fibril formation assays were performed to determine the effect of LMWHs on fibrillogenesis. RESULTS: Injection of mice with clinically relevant doses of LMWHs (enoxaparin and dalteparin) demonstrated a reduction in AA amyloid deposition. These compounds were capable of arresting the progression of AA amyloid and eventually resulting in regression of the amyloid deposits. In vitro analysis indicated that LMWHs prevented AA and Abeta peptide fibril formation by impeding the structural changes necessary for fibril formation. CONCLUSIONS: Our findings suggest that the LMWHs may provide beneficial effects against the development of amyloidoses, including Alzheimer's disease.
