[A longitudinal study on deterioration of physical function in people aged over 55 in Beijing].

The objective of this study is to explore the index of physical performance test in detecting physical functional status among elderly people. Physical performance test, recommended by Yale University in EPESE was used in the study. A total number of 1,986 people aged 55 and over in urban and rural area of Beijing was studied in a three-year longitutinal observation. Among 1,986 cases studied in 1992, 168 died between 1992-1994. The mortality rate increased along with the degree of functional disablement, lowest in non-disabled (3.4%), followed by mild ones (5.2%), moderate (10.5%) and highest in severely disabled (18.4%). It is shown that performance test can be used as a tool in observing and detecting risk groups in the progress of functional disability It's an important index describing the decline of health status.

[Study on the immunoregulatory effect of Baoyuantang ([symbol: see text]) in patients with chronic hepatitis B].

The T lymphocyte colong, formition (TL-CFU), the T lymphocyte subsets (CD3+, CD4+ and CD8+) and interieakin-2 membrane receptor (mIL-2R) secreted by peripheral mononuclear cells of 60 patients with chronic B hepatitis were studied. The results indicated that the activity of TL-CFU, mIL-2R and CD3+, CD4+, CD4+/CD8+ were decreased, and CD8+ was increased as compared with the normal controls. After treatment with Baoyuantang, the TL-CFU, mIL-2R, CD3+, CD4+ and CD4+/CD8+ increased and CD8+ decreased as well. It is suggested that the patients with the chronic B hepatitis with immune function defficiency, and the Baoyuartan treatment is presented immunoregulatory.

[Relationship between osteoporosis and metabolism of calcium and bone].

Using metabolic balance test, bone mineral density measurement, animal models of osteoporosis, bone cell culture, we investigated the relationship between osteoporosis and the metabolism of calcium and bone. We found that the calcium intake was rather low in 20 healthy adults (480 mg per day); the average intestinal net absorption rate(%) of calcium in 124 healthy adult volunteers was 28.9(%), which increased gradually in recent 20 years (from 17.1% in 1978 to 40.6% in 1977), owing to improvement of dietetic structure. We conclude that the insufficiency of sex hormone and calcium are the main causes of osteoporosis. In bone cell culture, the whole process of bone remodelling was observed for 35 days continuously. Calcitonin damaging osteoclast was also investigated. Thus, we suggest new concept of bone remodelling coupling.

[Determination of glycosylated hemoglobin in human erythrocytes by fast protein liquid chromatography system].

The chromatography method for analysis of glycosylated hemoglobin(HbA1c) in human erythocytes was established by using a fast protein liquid chromatography system(FPLC) and a Mono S column. The average intrabatch and interbatch coefficients of variations (n = 3) were 2.5% and 4.8%, respectively. There was a positive correlation between the results of FPLC and routine microcolumn method (r = 0.413, P < 0.02). Using FPLC method, the HbA1c levels in festing blood of 54 normal donors and 26 diabetics on admission were 4.6 +/- 1.19(%) and 10.2 +/- 3.45(%), respectively. A significant difference was found statistically between the results of two groups (t = 10.8, P < 0.01).

[Internal fixation with mini-titanium plate for midface fractures].

Thirty four patients with midface bone fractures were fixed by rigid internal fixation (RIF) with min-titanium plate. An optimum exposure is provided for the fracture sites of zygomatic, zygomatic arch, orbital floor and maxilla with the coronal incision and adjuvant incisions. The approach was of benefit to accurate anatomic reduction and fixation of the fracture segments. RIF with min-titanium plate provided three-dimensional stability. The fixation of zygomatic-frontal crevice and zygomatic maxillary crevice were the key for obtaining sufficient stability, and at least two fixation screws were required above zygomatic-frontal crevice.

[Domperidone and hyperprolactinemia].

Nine patients with hyperprolactinemia and one patient with pituitary microadenona were observed for 6 years. They were induced by administration of domperidone. The clinical characteristic of the 9 patients was summarized and analysed. The results suggest that prescribe domperidone for fertile women should pay more attention to its adverse effect on inducing hyperprolactinemia and even pituitary microadenona.

[Effects of selenium polysaccharide and sodium selenite on blood selenium concentration and liver cytochrome P450 monooxygenase system in rat].

The effects of selenium polysaccharide and sodium selenite administered by single or repetitive intraperitoneal injection (i.p.) on blood selenium concentration, the activities of liver cytochrome P450, b5 as well as NAD(P)H cytochrome C reductase, glutathione S-transferase and glutathione were studied in rats. The biological effects of selenium polysaccharide and sodium selenite were also compared. The results indicated that the blood selenium concentration was increased rapidly and reached the peak in 2 hours followed by gradual decline after selenium polysaccharide and sodium selenite were i.p. injected at a dose of Se 0.6 mg/kg. The absorption and eliminating rates of Se from sodium selenite were faster than that from selenium polysaccharide. Administration of selenium polysaccharide and sodium selenite at a dose of 0.2 mg/kg by i.p. increased the blood selenium concentration to 2.6 and 2.1 times of those of control group, respectively, and the blood selenium concentration of selenium polysaccharide group was significantly higher than that of sodium selenite group (P < 0.05). The activities of liver cytochrome P450, b5 and GST were inhibited by selenium polysaccharide and sodium selenium in vivo and in vitro experiments. Those proteins were decreased to 57%, 70% and 62% of the control, respectively, by selenium polysaccharide which has particularly stronger effects on cytochrome P-450 monooxygenase system (P < 0.05). The two selenium compounds did not appear to affect the activity of NAD(P)H cytochrome C reductase. Both of the selenium polysaccharide and sodium selenite could enhance the activity of glutathion peroxidase significantly (P < 0.05).

[Effects of fat emulsion on serum lipid, apo A I, apo B100, LCAT, KBR of postoperative patients with liver disorder].

Twenty post-operative patients with liver disorder were divided randomly into two groups. Ten of them received mean 0.82 g.kg-1.d-1 fat emulsion(42% of the total calorie), and the other ten received mean 1.72 g.kg-1.d-1 fat emulsion (52% of the total calorie) via vein during 5 post-operative days. Their serum lipid, apoA I, apoB100, ketone body ratio (KBR), fat clearance, LCAT and so on were investigted. The results suggest that it is safe for liver-disordered patients with slight or even moderate abnormality of liver function to receive mean 0.82 g.kg-1.d-1 and 1.72 g.kg-1.d-1 fat emulsion after operation and the use of mean 1.72 g.kg-1.d-1 fat emulsion will do no harm but good to liver function.

[A phase II clinical study of China-made paclitaxel in the treatment of cancer patients].

OBJECTIVE: To evaluate the efficacy and toxicity of China-made paclitaxel in patients with a variety of advanced malignancies. METHODS: Paclitaxel was administered by i.v. infusion at a dose of 175 mg/m2 in 3-hour every three weeks. All patients received premedication(dexamethasone, diphenhydramine and cimetidine) to prevent allergic reactions. RESULTS: Of 116 patients, 114 patients received 2 to 3 courses of therapy and were evaluable for response. The overall response rate for ovarian cancer was 46.7%, for breast cancer 56.4%, for non-small-cell lung cancer 25.0%, for small-cell lung cancer 66.7%, and for nasopharyngeal cancer 41.7%. Responses were also achieved in patients who had failed to respond to prior chemotherapy, with the overall response rate of 42.9% for ovarian cancer, 57.6% for breast cancer, 33.3% for non-small-cell lung cancer, 25.0% for small-cell lung cancer, and 40.0% for nasopharyngeal cancer. Paclitaxel did not show cross-resistance with doxorubicin and cisplatin. The major toxicity associated with paclitaxel included neutropenia, arthralgia, myalgia, numbness of hands and feet and alopecia. No severe hypersensitivity reactions were observed. CONCLUSION: At the above-mentioned dose and schedule, paclitaxel is effective with tolerable side effects.

[Immunohistochemical study of Cath-D and CD44 in laryngeal squamous cell carcinomas].

The expression of Cath-D and CD44 was examined immunohistochemically in 96 laryngeal squamous cell carcinomas and compared with clinic pathological feature. Expression of Cath-D and CD44 was detected in 52.1% and 41.7% respectively. The correlation was observed between Cath-D expression and metastasis (including lymph node metastasis and distant metastasis), but Cath-D expression was not associated with lymph node metastasis. Expression of CD44 was not associated with lymph node metastasis also. In addition, expression of Cath-D and CD44 did not correlated with clinical stage, histologic grade, size of tumor and postoperative 3 years survival rates. Our results indicate that Cath-D may play a role in metastasis.

[Mathematical diagnosis of idiopathic-hypoparathyroidism].

The present study devotes to diagnose idiopathic-hypoparathyroidism (IHP) by means of ratio of calcium and phosphorus and product of calcium and magnesium. Among 23 cases of IHP patients the coincidence of ratio (Ca/P) and low-calcium/high-phosphorus in blood was 95.7%. Among 17 cases of IHP patients the coincidence of product (Ca x Mg) low-calcium and low magnesium was 91.4%. It indicates that ratio (Ca/P) and product (Ca x Mg) are effective mathematical diagnosis of IHP.

[Expression of class II major histocompatibility antigen and factor VIII related antigen in endothelial cells of blood vessels in malignant lymphomas].

OBJECTIVE: To determine the heterogenicity in the expression of class II major histocompatible antigen (HLA-DR), factor VIII related antigen (FVIIIRAg) and the biological behavior of vessel endothelial cells in malignant lymphoma. METHODS: Standard immunohistochemical method using antiserum of HLA-DR and FVIIIRAg were applied to detect heterogeneous expression of endothelial cells in 39 cases of malignant lymphomas using neonatal lymph nodes, normal adult lymph nodes and lymph node hyperplasia as the controls. RESULTS: Both HLA-DR and FVIIIRAg reactivity was found in blood vessels in neonate lymph nodes, normal adult lymph nodes and lymph node hyperplasia. Most vessels in Hodgkin's disease and T cell lymphoma had high endothelial post capillary venules (HEV), which expressed both FVIIIRAg and HLA-DR. In B cell lymphoma, most blood vessels are capillary-like microvessels, which expressed little or no HLA-DR, only expressing FVIIIRAg. CONCLUSIONS: The above results indicate that the endothelial cell of blood vessels in Hodgkin's disease and T cell lymphoma not only participate in immunoregulation but also in blood coagulation, whereas, those in B cell lymphoma only participate in blood coagulation.

Induction of anti-DNA antibodies by immunization with activated lymphocytes and active chromatin.

OBJECTIVE: To investigate the primary autoantigens which contribute to the production of anti-DNA antibodies. These antibodies are serological hallmark and pathogenic factor of systemic lupus erythematosus (SLE). METHODS: Nonautoimmune predisposed BALB/c mice were immunized with concanavalin A (Con A) activated, lipopolysaccharide (LPS) activated and nonactivated syngeneic spleen cells. Nuclei and chromatin from activated/nonactivated lymphocytes were isolated and syngeneic mice were immunized. Sera were taken after the third immunization. IgG anti-dsDNA antibody was determined by ELISA (calf thymus DNA treated with S1 nuclease was used as the coated antigen). The glomerular IgG deposition was observed by immunofluorescence one month after the third immunization. RESULTS: Con A activated T cells and LPS activated B cells induced anti-double stranded (ds) DNA antibody in syngeneic nonautoimmune BALB/c mice and formed the glomerular IgG deposition. Further studies showed that active chromatin isolated from activated lymphocytes induced anti-ds DNA antibody, but not resting chromatin isolated from nonactivated lymphocytes. CONCLUSIONS: Activated lymphocytes and their active chromatin could be the autoimmunogen(s) driving the anti-dsDNA antibodies. The change of chromatin's antigenicity by environmental factors and genetic background may be the common pathway to SLE pathogenesis.

[Determination of vitamin D3 in health-care foods fortified with calcium by reversed-phase high performance liquid chromatography].

"Bu Gai Bao" and "Gaoxiao Man Gai" are health-care foods fortified with calcium for people of all ages. This paper describes a method of reversed-phase high performance liquid chromatography for quantitating vitamin D3 in these products. The method is simple, sensitive, and suitable for routine analysis of vitamin D3 in a variety of health-care foods fortified with calcium. Shimadzu LC-4A high performance liquid chromatograph with SPD-2AS UV detector and C-R2AX data processor were used. The vitamin D3 in products was extracted with chloroform and separated with mu-Bondapak C18 column(300 mm x 3.9 mm i.d.) and methanol water(87:13, V/V) mobile phase with detection at UV-264 nm. Regression coefficient of correlation of calibration curves in the range of 10-50 mg/L was 0.9998. Average recoveries of vitamin D3 added to the products was 95.63%. The coefficients of variation of vitamin D3 in both products were 0.24%-0.49%.

[Isolation, purification and determination of the constituents of a toxic protein from plant amenone].

A toxic protein constituent named AME from the stems of plant amenone, have been isolated and purified. Through CM-SFF column chromatography and gel filtration on Sephacryl S-200 column with phosphate saline buffer as mobile phase. All of the operations were performed at 4 degrees C. The pulverized plant amenone material was soaked in phosphate saline buffer, homogenized, left standing overnight and then squeezed through coarse cloth by wringing. The supernatant was applied on the S-SFF column. Then, the column was eluted with the phosphate buffer containing 1 mol/L NaCl. The eluate was collected and dialyzed against water and phosphate buffer. The chromatography of the crude toxin dialyzed was carried out on the CM-SFF column with gradient elution of phosphate buffer containing NaCl. The fourth peak was collected and then applied on a gel filtration Sephacryl S-200 column using neutral phosphate buffer as mobile phase. The protein was further separated on two connected Protein 125 columns with mobile phase of 0.2 mol/L phosphate buffer (pH 6.5), the eluate was monitored at 280 nm on photodiode array detector. The protein presents typical the absorption spectrum of protein in ultraviolet region with the strong absorption at 280 nm and the weak absorption at 260 nm. The purity of peak of the protein was judged from the spectrum. The molecular weight of AME measured by two connected protein columns was approximately 35,000 D. The composition of amino acids was determined with OPA post-column derivatization/fluorescence detection.

[Effect of ginger-processing on the contents of guanosine in Pinelliae rhizoma (banxia)].

OBJECTIVE: To compare the guanosine contents in various products of Pinellia Rhizoma processed by different amounts of ginger and alum. METHOD: Using the technology of HPLC. RESULT: The guanosine contents in the two products of Pinellia Rhizoma processed in line with the pharmacopoeial method and orthogonal method respectively are very close to each other and change every little as compared with raw Pinellia Rhizoma. But the contents in the products of Pinelliae Rhizoma processed in other ways (digged in alum solution or in ginger Juice decocted with ginger juice, etc.) are much less than those of raw Pinellia Rhizoma. CONCLUSION: There are no obvious differences in guanosine contents between Pinellia Rhizoma processed by orthogonal technology and Pinellia Rhizoma processed by pharmacopoeial method. Processing technology has considerable influence on the water soluble components of Pinellia Phizoma.
