[The expression of integrin alpha5beta1 and transforming growth factor-beta in pulmonary fibrosis of rat].

OBJECTIVE: To study the roles of integrin alpha(5)beta(1) played in rat pulmonary fibrosis. METHODS: The dynamic changes of fibronectin (FN), FN receptor alpha(5)beta(1), and TGF-beta were analysed in bleomycin-induced rat pulmonary fibrosis by using immunohistochemical technique. Northern blot analysis and immunocytochemistry techniques were performed in order to detect the expression of alpha(5)beta(1), FN mRNA and their relevant proteins in cultured rat pulmonary fibroblasts (PFbs) after administration of TGF-beta1. RESULTS: (1) A strong positive reaction with anti-integrin alpha(5)beta(1) was seen in the pneumocytes, endothelial cells and proliferated mesenchymal cells on the 1st to 3rd day after bleomycin. The positive signals for integrin alpha(5)beta(1) were apparent mainly in those proliferated interstitial cells with fibroblast and myofibroblast differentiation 7 days after bleomycin treatment. Changes in FN were similar to that of the integrin alpha(5)beta(1). (2) Expression of alpha(5)beta(1), FN mRNAs and their relevant proteins was increased in rat PFbs after TGF-beta1 administration. CONCLUSIONS: Integrin alpha(5)beta(1) played a key role in inducing the activation, proliferation and differentiation of PFbs, causing an increase of extra-cellular matrix synthesis during pulmonary fibrogenesis.

[Study on the antidiuretic efficacy and mechanism of indapamide in central diabetes insipidus].

OBJECTIVE: To investigate the efficacy and antidiuretic mechanism of indapamide in central diabetes insipidus. METHODS: We observed eight patients with central diabetes insipidus treated by indapamide (2.5-7.5 mg.d-1) or dihydro-chlorothiazide (50-70 mg.d-1) or carbamazepine (150-300 mg.d-1) for 8 days with randomized block design. The therapy was changed twice in sequence after 4 days without treatment. Urine output and indexes of water metabolism were also observed. RESULTS: The urine output was reduced on the 4th day and 8th day with each therapy. The serum potassium and chloride concentration was decreased and renin activity was increased on the 4th and 8th day with indapamide therapy. CONCLUSION: Indapamide has antidiuretic action on central diabetes insipidus. The action may result from the changes of hydroelectrolytes and renin-angiotensin-aldosterone system.

[The comparison of component contents and pharmacological actions between two kinds of processed products of Pinellia rhizoma prepared by ginger juice and alum].

OBJECTIVE: To compare the component contents in Pinellia Rhizoma processed with ginger separately on orthogonal technology and pharmaceutical technology stipulated in pharmacopoeia, and study the action of the two differently processed products on animals. METHOD: Using gravimetric method and acid dye colorimetry to determine the total alkaloids, et al. RESULT AND CONCLUSION: The experiment has shown that the contents of total alkaloids of product of Pinellia Rhizoma processed by orthogonal technology are higher than those processed by pharmaceutical technology, but the contents of digged substances of the latter are higher than those of the former, and the toxicity of decoction of the latter is also higher than that of the former. Compared with raw Pinellia Rhizoma, the two products of Pinellia Rhizoma prepared by different technologies all have shown promoting action on PGE2 contents and activity of gastric proteinase in rat gastric juice. The experimental results also have shown that on the above mentioned items. There are no obvious differences between the product of Pinellia Rhizoma processed by orthogonal technology and the product processed by pharmaceutical technology. However, the component contents of raw Pinellia Rhizoma are lower than those of the above mentioned two processed products and help inhibit PGE2 contents and activity of gastric proteinase in rat gastric juice.

[Relationship of nitric oxide and immunal function of the patients with acute hepatitis and hepatic cirrhosis].

The interleukin-12(IL-12), interleukin-10(IL-10), tumor necrosis factor(TNF) and nitric oxide (NO) levels in serum of 90 patients with hepatitis and cirrhosis were measured by the method of ELISA and colorimeter. The levels of IL-12, IL-10 and NO in serum of the patients with hepatitis were in close proximity to the level of healthy control, but the TNF level was significantly higher than that of healthy control. The IL-12, IL-10, TNF and NO levels in serum of the patients of hepatic cirrhosis were significantly higher than that of healthy control and patients with hepatitis. The results suggested that the NO level was not related with the hepatic damage of acute hepatitis. The dysfunction of immunology in the patients with cirrhosis was related with the increase of NO level.

[Study on the prevention of insulitis and diabetes by complete Freund's adjuvant in NOD female mice].

The degrees of insulitis, the incidence of diabetes, and the counts of alpha, beta and delta cells in islets were studied by morphological method and avidin-biotin complex immunohistochemical method in complete Freund's adjuvant(CFA) or normal saline(NS)-treated non-obese diabetic(NOD) female mice. The results showed: The incidences of in-insulitis and the scoring means were significantly lower in CFA-treated mice than those in NS-treated ones; none of 5 CFA-treated mice and 3 out of 5 NS-treated ones developed diabetes. The positive rates of alpha, and delta cells were significantly lower in CFA-treated mice than those in NS-treated ones, but the positive rate of beta cell was significantly higher in CFA-treated mice than that in NS-treated ones. The correlations were found among the scores of insulitis and the positive rates of alpha, beta, delta cells in islets (r alpha = 0.475, r beta = -0.878, r delta = 0.869). The results indicate that CFA may lessen the degrees of insulitis and the incidence of diabetes. Its effects might be related to lessen the damage of beta cell in islets.

[A role of nitric oxide and endothelin in bronchial hyperreactivity in patients with mild and middle asthma].

OBJECTIVES: To probe into a role of nitric oxide(NO) and endothelin(ET) in bronchial hyperreactivity(BHR) in patients with mild and middle asthma. METHODS: MchPC20 was used as an index to evaluate BHR in the asthmatic patients. Endothelin and NO level in bronchial alveolar lavage fluid(BALF) obtained from 15 patients with attacking asthma(Group I), 8 patients with stable asthma (Group II) and 7 healthy control subjects (Group III) were examined by radioimmunoassay and copper coated cadmium reduction method, respectively. RESULTS: 1. MchPC20 and FEV1% in Group III were slightly higher than those in Group I (P < 0.05), and lower than those in Group III (P < 0.05), respectively; 2. NO and ET levels in BALF from Group I were higher than those from Group II and Group III (P < 0.05), respectively; 3. ET/NO ratio in Group I was higher than that in Group III (P < 0.05) and lower than that in Group II (P > 0.05); 4. NO and ET level in BALF from Group I were positively and negatively correlated with MchPC20, respectively(r = 0.83, P < 0.01; r = 0.75, P < 0.01); the correlation of NO and ET level in BALF from Group I was negative (r = 0.70, P < 0.01); 5. NO and ET levels in BALF and ET/NO ratio from Group II were slightly higher than those from Group III (P < 0.05), respectively. CONCLUSION: NO may improve and ET exacerbate BHR in patients with mild and middle asthma. Bronchial hyperreactivity of the patients with stable asthma displays lightly abnormal, which may be due to mild abnormality of NO, ET levels and ET/NO ratio in BALF.

[Study of effects of six caries-preventive agents on enamel decalcification].

OBJECTIVE: To compare the influence of 6 caries-preventive agents containing fluoride and trace element on dental enamel decalcification and observe the relationship between enamel decalcification and time of early carious lesions. METHODS: 84 bovine teeth were selected and divided into 7 groups. Experimental groups were treated with SrF, SnF2, (NH4)2 MoO2F4, ZnF, LaF and NaF with the same [F-]concentration and deionised water respectively, then all the specimens formed artificial carious lesion in lactic acid gel. RESULTS: The [Ca2+] concentration of lactic acid gel showed a variety of enamel decalcification among specimens treated with fluoride agents after 48 hours, on the other hand, all the specimens presented a platform of enamel decalcification after 48 hours. SnF2 demonstrated the strongest inhibition on enamel decalcification, meanwhile, NaF has the minimum influence on it. CONCLUSION: The time of forming artificial carious lesion with acid etch is about 48 hours, moreover, caries-preventive agents containing fluoride and trace element are effectively able to inhibit enamel decalcification, and the potential mechanism is the interaction of fluoride and trace element.

[Application of high performance liquid chromatography to the study on the production of maltooligosaccharide by synergistic action of two-enzymes].

This paper describes the application of high performance liquid chromatograpy to the study on the production of maltooligosaccharide by synergistic action of two-enzymes. Seven sugars in products were separated on a column of Nucleosil C18 with water as the mobile phase and determined refractometrically. Meanwhile, for the preparation of maltooligosaccharide, the effect of supplementing rate of the two enzymes on the content of maltooligosaccharide in products is examined in this paper. The optimun technological conditions were obtained by using the orthogonal design and the amount of the two enzymes added was determined. Thus more than 70% of M3-M6 maltooligosaccharide in product was obtained.

[Studies on treacheostomal formation and voice prosthesis after total laryngectomy].

OBJECTIVE: To establish a tracheostomy model with no stricture and "bowl-shaped" collapse. METHODS: Twenty-five cases were reconstructed with Blom-Singer technique after total laryngectomy for tracheoesophageal voice in our department since 1996. Tracheostoma was done with 5 kinds of methods. In 6 cases, anterior cervical U-shaped incision + division of sternal attachments of bilateral sternomastoid muscles + Griffith tracheostoma enlargement (combination method) were used in the primary operation, and the third generation of indwelling prosthesis and tracheostomal valve were installed. RESULTS: In these 25 cases, 23 could speak freely. The successful rate was 92%. In tracheostoma operation, there were 7 cases with their skin sutured directly to tracheal rings, 3 cases with Myers method, 6 cases with Griffith method, 3 cases with [symbol: see text] method. All of them needed cannulation for more than 6 months and had stricture of tracheostoma and "bowl-shaped" collapse of various degrees. But those underwent "combination method" needed no cannulation and no stricture and bowlshaped collapse was found during 6 mouths' follow-up. CONCLUSION: The combination method of tracheostomy makes the installation, postoperative care and replacement of prosthesis and tracheostomal valve easy and the indwelling prosthesis has the best tone quality. The use of tracheostomal valve effectively makes the patient's hand free during speaking.

[Forecasting octane numbers of gasoline by NIR spectroscopy and the manufacture of octane number analyzer].

The predictive model of octane numbers, of gasoline was developed by using MLR and PLS method of chemometrics. It is applicable to forecast the MON, RON and PON of gasoline of catalytic cracking, catalytic reforming, alkylating as well as blended products. A near infrared optical multichannel analyzer (NIR-OMA) and a software package of octane number were developed. Combining both mentioned above a NIR-OM octane number analyzer was developed. With this NIR-OMONA one can obtain the MON, RON and PON (antiknock index) data in less than 1/2 minute with an accuracy of 0.6 ON unit and a precision of 0.1 ON unit by using 3-15 mL gasoline sample.
