[Changes of lipoprotein lipase and hepatic lipase and their significance in gallstone formation in rabbit model].

This experiment was made to investigate the changes of lipoprotein lipase(LPL) and hepatic lipase (HL) activity and their effects on gallstone formation in rabbit model in which the stones were induced by high cholesterol diet. Activities of plasma LPL and HL were determined; other data including concentration of plasma lipoprotein cholesterol, concentration of bile cholesterol and bile acids were also obtained. The results showed that with the rabbits continuously fed on high cholesterol diet, LPL activity heightened markedly (P < 0.05), and HL activity increased gradually (3 and 4 weeks groups vs control group, P < 0.05). The changes of concentration of plasma VLDL-C and LDL-C were the same as that of LPL activity, but the concentration of plasma HDL-C, HDL2-C, HDL3-C and bile acids showed no significant changes (P > 0.05). These results suggest that the heightened activities of LPL and HL might make the liver take up more cholesterol, secrete it into the bile duct and hence accelerate gallstone formation.

[Polymorphism of ACE gene and its relationship with serum ACE activity in the Hans in Chengdu region].

This study was aimed at the angiotensin converting enzyme (ACE) gene insertion/deletion(I/D) polymorphism distribution in Chinese and its relationship with serum ACE activity(SACE). SACE activity and ACE gene polymorphism in 90 healthy persons in Chengdu region were investigated using a spectrophotometric assay and the PCR technique, respectively. The results showed that, in the Hans, DD genotype of ACE gene was 18%, DI genotype 44%, and II genotype 38%. Allele frequencies were 0.40 for the D allele and 0.60 for the I allele. There were significant relationships between ACE I/D polymorphism and SACE activity. The results suggest that ACE genotype distribution be different between races, that II genotype and I allele be more frequent in Chinese, and that SACE activity be under the influence of a genetic polymorphism of ACE in Chinese.

[Clinical study of three-dimensional reconstruction of computed tomography in diagnosis of condylar fractures].

OBJECTIVE: To explore a method of getting good three-dimensional (3D) reconstruction images of temporomandibular joint (TMJ) by computed tomography (CT), and evaluate diagnosis value of 3DCT in condylar fractures. METHODS: Of the 14 patients studied, 12 were male and 2 were female with age ranging from 4 to 37 years old. 4 were old fractures and the other 10 were fresh fractures. All patients' orthopanotomograms were taken first, and then were scanned with CT. The scanning conditions were 300-345 mAS and 120kV, and the scanning methods included cross-sectional (11/14), coronal-sectional (2/14) and spiral (1/14) (pitch: 1, thickness of layers: 2.5 mm) scanning. Scanning scopes: The cross-sectional and spiral scanning were from 1 cm above the Frankfort horizontal plane to the middle of the mandibular ramus or Menton; The coronal-sectional scanning was from the mastoid process to the anterior point of the mandibular ramus or Pogonion. The thickness of the scanning layers was 2.5 mm (12/14) or 5 mm (2/14). Finally, 3D reconstructed images were obtained by shaded surface display (SSD). The cross-sectional images were obtained by being removed the cervical vertebra, the styloid process, the occipital bone and part of the mastoid process with Subtract Manual Irroi before reconstruction to avoid interference with the observation of TMJ and skull basis. RESULTS: 1. The best 3D CT reconstructed images were obtained by 2.5 mm-thin-layer cross-sectional scanning by being removed parts of the adjacent structures, and the bone lines were clear and smooth without adjacent bone structures shading TMJ. The quality of images taken spirally was similar to those taken cross-sectionally. While the coronal scanning neglected some important anatomic symbols which might be valuable to diagnose condylar displacement. 2. Of the 14 patients, 7 were unilateral condylar fractures and 7 were bilateral (21 sides altogether), among which 18 sides were high fractures of condyles and 3 were fractures of condylar neck. High oblique line fractures and comminuted fractures had major condylar rotation displacement which moved forward, downward and inside, whereas, vertical fractures only had minor displacement. Inward rotation displacement occurred in medial bent fractures. 10 of 14 patients (15 sides) had been conducted operation, and the same situations as seen from 3DCT reconstructed images were found. CONCLUSION: 3DCT images can display condylar fractures accurately and directly, and are very useful for surgeons to select treating methods of condylar fractures.

[A study of the mechanism of effects of solution containing trace elements on remineralization layer formation of enamel carious lesions].

OBJECTIVE: To study the effects of several remineralization solutions with different composition on the artificial demineralization lesions of bovine enamel and the mechanism of remineralization layer formation. METHODS: 36 bovine teeth were selected and artificial carious lesions were formed with lactic acid gel. Then all specimens were divided into 4 groups which contained no fluoride, fluoride, fluoride-calcium-phosphate-tin, and de-ionic water separately, and the de-ionic group was taken as control. The structure and composition of the sediments on the enamel surface were evaluated and detected by X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy (SEM). Histological change of enamel section was examined by polarized microscopy. RESULTS: Compared with the control group, all the solutions had remineralization effects, especially the solutions containing trace elements. XPS showed that there were sediments formed on the surface of carious lesion, the compositions of the sediments were CaF2, HAP and non-specifically absorbed fluoride (NSAF). Irregular, crystal deposits were also found under SEM on the enamel surface. The polarized microscopy results showed that the observed demineralization areas decreased after exposure to the solutions. CONCLUSION: Solutions containing fluoride and trace elements were effective on remineralization. The possible mechanism is that CaF2 and NSAF, instead of FA formed in the remineralization layer. Moreover, the remineralization effects of the fluoride solutions containing trace elements are more outstanding than that without trace elements. The potential mechanism is the synergistic action of fluoride and trace elements.

[Changes of cultured human vein endothelial cells(HVEC) in the presence of therapeutic agents of cavernous hemangioma].

OBJECTIVE: To observe the damages of cultured human vein endothelial cells treated by various drugs. METHODS: With the identification of EC, damages of which treated by drugs with different concentration were measured with MTT assay respectively, dramatic changes in the morphology of test cells observed under light and electron microscope. RESULTS: 1. According to the different inhibition on proliferation of HVEC, sodium morrhuate exhibited most cytostatic effect for HVEC, and combination of pinyangmycin and hormone produced more significant arrests of growth of HVEC than pinyangmycin used singly. 2. Positive correlation were found between cell contraction and concentration of varied drugs with the absence of structure of junctional integrity. 3. Corresponding pathological changes occurred on HVEC with MTT assay. 4. As the damages of HVEC occurred, therapeutic agents may directly injured substratum of cavernous hemangiomas through enlarged gaps between cells subsequently. CONCLUSION: Evaluation and screening of sclerosing agents for cavernous hemangioma may be feasibly demonstrated with affects on cultured HVEC by different agents.

[The changes of serum angiotensin converting enzyme activity in Type 2 diabetes and its vascular complications].

This investigation was made with reference to the changes of serum angiotensin converting enzyme (SACE) activity in type 2 diabetes and its vascular complications. SACE activity was studied in 127 type 2 diabetic patients and 90 healthy persons by using a spectrophotometric assay. The results showed SACE activity was obviously higher in diabetic patients (459.51 +/- 175.85 U) than in healthy persons (321.14 +/- 121.27 U); SACE activity was significantly higher in type 2 diabetic patients with diabetic nephropathy (548.27 +/- 166.60 U) than in patients without diabetic nephropathy (383.2 +/- 139.00 U), but there was no difference between patients with microalbuminuria and macroalbuminuria; no statistical difference was detected in SACE activity between diabetic patients with diabetic retinopathy (465.64 +/- 178.93 U) and without retinopathy (449.07 +/- 170.04 U); SACE activity was not associated with the course of diabetes, blood pressure, blood lipid and blood glucose. These data suggest that raised SACE activity might only play a role in the initiation of type 2 diabetes and diabetic nephropathy, but not relate to the progress of diabetic nephropathy, the onset of diabetic retinopathy and hypertension.

[The enlarged translabyrinthine removal of large acoustic neuromas: 18 cases report].

OBJECTIVE: To investigate the surgical methods and results of enlarged translabyrinthine approach in the removal of large acoustic neuromas. METHOD: A large mastoidectomy involved complete exposure of sigmoid sinus, the dura behind the sinus for at least 1 cm, the superior petrosal sinus and the middle fossa dura. The jugular bulb was exposed and pressed downwards if necessary. The internal auditory meatus was skeletonized and uncovered for at least 270 degrees. The debulking of the tumor began inside the anterior and inferior poles in order to find the brainstem and the facial nerve root as early as possible, then the dissection of the nerve was performed medially to laterally. Intraoperative facial nerve monitoring and postoperative CT and MRI were performed in all cases. RESULT: Total removal was achieved in all 18 patients with tumors larger than 3 cm (mean size: 4.2 cm). There were no death as well as other complications such as intracranial infection and persistent cerebrospinal fluid leakage. There were no obvious cerebral sequela. The facial nerve was preserved both anatomically and functionally in 14 cases, with Grade I or II in 8 cases, Grade III or IV in 6 cases. Nerve interruption occurred in 4 cases who all had severe facial paralysis or nerve interruption before operation. 16 patients returned to work within 1-3 months. CONCLUSION: Total removal of large acoustic neuroma could be accomplished via the translabyrinthine approach, with good result of facial nerve function and minimum incidence of morbidity.

[The semiconductor diode laser application in tympanoplasty].

OBJECTIVE: To investigate the method, indications and initial results of semiconductor diode laser application in tympanoplasty. METHOD: The diode laser was used in 14 tympanoplasty procedures. The laser manipulation was mainly used in cases of malleus and incus bony fixation with the tympanum, the fibrous scar tissue around the ossicular chain and the malleus head resection. RESULT: There was no postoperative vertigo and facial paralysis. The average hearing improvement was 24 dB. Seven cases (50%) reached an air-bone gap within 20 dB. CONCLUSION: The application of diode laser in tympanoplasty could effectively reduce the manipulation trauma of ossicular chain and control the bleeding of operative field, especially in managing the bony adhesion of the ossicular chain.

[Investigation on Schistosoma japonicum infection in rodents in Yunnan Province].

OBJECTIVE: To investigate the role of rodents in the transmission of schistosomiasis in endemic areas in Yunnan Province. METHODS: Kato-Katz method was used to study the infection rate of rodents, humans and domestic animals. RESULTS: The infection rate was 0.9% (32/3,411) for rodents, 15.6% (461/2,964) for humans, and 9.6% (239/2,482), for domestic animals. The EPG(mean) was 5.8 for rodents, 1.8 for humans, 0.1 for cattle, 0.1 for horses and 0.02 for pigs. CONCLUSION: The rodents played a minor role in the transmission of schistosomiasis in the plateau area of Yunnan province.

[Studies on the supercritical CO2 fluid extraction and separation of psoralen, isopsoralen and fatty oils from Psoralea corylifolia].

Opimum extraction conditions were studied by supercritical carbon dioxide fluid extraction technology and orthogonal tests. Crystal of psoralen can be achieved directly from the extractives. Magnified experiments were carried out. The chemical constituents of fatty oils were analyzed by GC-MS. The result shows that the optimum conditions are: extraction pressure 27 MPa, extraction temperature 70 degrees C; seperation pressure 9 MPa, seperation temperature 60 degrees C. Six components such as oleic acid, linoleic acid etc. were identified in the fatty oils.

[Clinical application of the multichannel auditory brainstem implant].

OBJECTIVE: To investigate the surgical techniques and speech performance of multichannel auditory brainstem implant (ABI) in patients with bilateral acoustic neuromas (neurofibromatosis type 2). METHODS: The nucleus 21 channel auditory brainstem implant was implanted into the lateral recess of the fourth ventricle through the translabyrinthine approach in 7 patients after removal of the tumor. The accurate placement of electrode array was ensured by the electromyogram monitoring of the 7th and 9th nerves and the electrically evoked auditory brainstem responses (EABR). Initial switch-on occurred six weeks postoperatively. Speech evaluation was performed every 3 months for the first year and annually thereafter. RESULTS: During the surgery, the lateral recess could be found and the typical EABR could be recorded in 6 cases. They later reported a significant benefit from the device. Two of the cases have achieved functional open-set speech understanding. In contrast, one patient with no EABR because of difficulty of the anatomic location during the surgery had no sensations postoperatively. CONCLUSION: The multichannel ABI could effectively restore auditory sensations in patients deafened by bilateral acoustic neuromas. The accurate location of the cochlear nucleus complex during surgery was the key factor for the success of the operation.

[Survival motor neuron gene and neuronal apoptosis inhibitory protein gene deletion in patients with spinal muscular atrophy].

OBJECTIVE: To investigate the frequencies of gene deletion survival motor neuron telomere (SMNTel) exon 7 and neuronal apoptosis inhibitory protein gene (NAIP) exon 5 in 55 Chinese spinal muscular atrophy (SMA) patients, and compare the relationship between these two candidate genes and the disease. METHODS: PCR-SSCP method was used to detect the deletion of SMNTel exon 7, direct visualization of PCR products by agarose electrophoresis was used to detect the deletion of NAIP exon 5 in 55 SMA patients with type I and type II. 40 normal individuals were involved in the study as controls. RESULTS: Homozygous deletion of the SMNTel exon 7 was identified in 92% (23/25) of SMA type I patients and 90% (27/30) of SMA type II patients. The same deletion was found in two mothers and one father of SMA patients. There was no homozygous deletion found in normal controls. None of the homozygous deletion of NAIP exon 5 was found in 55 SMA patients and normal controls. Only two patients were found to have the heterozygous deletion. CONCLUSIONS: The frequency of homozygous deletion of SMNTel exon 7 was 90.1%. Our data support that SMN gene is strongly associated with SMA.

Anti-human platelet tetraspanin (CD9) monoclonal antibodies induce platelet integrin alphaIIbbeta3 activation in a Fc receptor-independent fashion.

OBJECTIVE: This study characterized the activation of platelet integrin alphaIIbbeta3 induced by two anti-human platelet tetraspanin monoclonal antibodies (mAbs), HI117 and SJ9A4. METHODS: Using 125I-labeled human fibrinogen(Fg), specific Fg binding to human platelets induced by HI117 and SJ9A4 was measured as indication of activation of platelet integrin alphaIIbbeta3 by the two mAbs. RESULTS: HI117 and SJ9A4 (10 microg/ml and 20 microg/ml) induced evident specific Fg binding to human platelets, suggesting that the two mAbs evoked activation of platelet integrin alphaIIbbeta3. Further study indicated that HI117 and SJ9A4 induced integrin alphaIIbbeta3 activation independent of platelet Fc-receptors, and that HI117 and SJ9A4-induced integrin alphaIIbbeta3 activation was inhibited by sphingosing, aspirin, apyrase, and/or PGI2. CONCLUSION: The anti-platelet tetraspanin (CD9) mAbs, HI117 and SJ9A4, can induce platelet integrin alphaIIbbeta3 activation independent of Fc-receptor. Three signaling pathways, i.e. thromboxane, secreted ADP, and cAMP pathways may be involved in the process, with protein kinase C activation presumably being the common step of the three pathways.

Serum levels of vascular endothelial growth factor in patients with angina pectoris and acute myocardial infarction.

OBJECTIVE: To determine whether serum vascular endothelial growth factor (VEGF) concentrations are altered in several kinds of coronary heart disease patients. MATERIALS AND METHODS: Using a VEGF enzyme-linked immunosorbent assay (ELISA), serum VEGF concentrations were determined in antecubital venous blood of 16 patients with stable angina pectoris (SAP), 16 with unstable angina pectoris (UAP) and 16 with acute myocardial infarction (AMI) before and after thrombolytic therapy, and of 16 age- and sex-matched healthy volunteers who used as controls. RESULTS: The concentrations of serum VEGF in patients with SAP (98.60 +/- 26.99 pg/ml) and UAP (103.61 +/- 24.89 pg/ml) tended to be higher than those in control subjects (80.44 +/- 24.57 pg/ml), but the differences did not reach statistical significance (P > 0.05 for each). Before thrombolytic therapy, the concentrations of serum VEGF in patients with AMI (285.92 +/- 125.15 pg/ml) were significantly higher than those in patients with SAP, UAP or control subjects (P < 0.01, respectively), and correlated with synchronous serum creatine kinase (CK) and its MB isoenzyme (CK-MB) contents (r = 0.866, P < 0.001 and r = 0.948, P < 0.001; respectively). Three hours after thrombolysis, the concentrations of VEGF had fallen to 111.57 +/- 31.29 pg/ml (P < 0.01 vs x before thrombolytic therapy and P < 0.05 vs x control subjects). CONCLUSION: The present study shows that serum concentrations of VEGF in patients with AMI are markedly elevated and that increased serum VEGF levels may be one of the most sensitive indexes in diagnosing AMI and judging reperfusion.

[Effect of Al3+ on luminescence intensity of Tb in silica glasses].

The silica glasses doped with Tb3+ and Al3+ iones was synthesized through Sol-Gel process in this paper. The effect of Al3+ on luminescence properties of Tb3+ were studied. The results show that no obvious effect of Al3+ on the emission peak position of Tb3+, but the emission intensity of Tb3+ doped Al3+ by sol-gel process increased obviously for all samples. The emission intensity of 1% Al-99% SiO2 glass was 5 times larger than that of 100% SiO2 glass. We concluded that the relation between the energy levels of Tb3+ and the energy levels of Al3+ may be appropriate for the energy transfer, the function of Al3+ was to absorb efficiently energy and to transfer fully to Tb3+.

[Quantitative determination of methyl-sulf amino-acid by internal standard-derivative infrared spectroscopy].

With pressed kBr disk technique the methyl-sulf amino-acid--the nutritious restrictive amino-acid of poultry was determined by internal standard-derivative infrared spectroscopy. The suitable internal standard--CaSO4 was selected, and peak height, peak area, first derivative and second derivative methods were used. In the determination, we selected undisturbed peak at 553 cm-1 as standard. The correlation coefficient of standard curve equation were in the range of 0.994-0.998. The result was satisfied.

[Quantitative determination of benzene in cyclohexane by derivative infrared spectroscopy].

Trace benzene in cyclohexane was determined by derivative infrared spectroscopy. In the determination, we selected peak at 674 +/- 2 cm-1 as quantitative peak, and peak height, peak area, first derirative (maximum, minimum, region) and second derivative methods were used. The correlation coefficient of standard curve equations were in the range of 0.990-0.995. There is a linear relationship between absobance and concentration of solution in the range of 0.010-0.10 g.10 mL-1. A quick and precise method for the determination were provided.
