Rosette-forming cells during immune response to Toxoplasma gondii in mice.

The rosette-forming cell (RFC) response of mice immunized with varying doses of Toxoplasma gondii was studied by immunocytoadherence (ICA). The specificity of ICA in the present system was tested by passive sensitization with hyperimmune serum in vivo and in vitro. A slight increase in RFC was observed with the latter. Prior treatment of spleen cells from immunized animals with rabbit anti-mouse immunoglobulin resulted in total inhibition of ICA. During the primary and the secondary response after 10 days, the number of RFC rose rapidly to reach the peak on the 3rd day. With secondary immunization 30 days later, the peak shifted to the 2nd day. Mice infected with a lower dose of Toxoplasma had a greater number of RFC during the secondary response after 10 days than with a larger dose.

[Demonstration of specific IgM antibody in cases of suspect congenital toxoplasmosis: interpretation of results (author's transl)].

From the aspect of laboratory Toxoplasmosis examination, a series of clinically and parasitologically confirmed cases of congenital Toxoplasmosis (CT) were selected. Serological findings (CFT, DT, IgM-IFAT) in infants and mothers were critically examined. The IgM titers in CT as a rule lay between 1:16 and 1:64, the highest recorded being 1:256. The serum IgM globulin levels showed no dependence on the high CFT titers. T recognize unspecific reactions amongst infants with positive IgM values, the serological testing of mothers was indispensable. It is also absolutely necessary with infants to repeatedly test serologically at intervals to clearly bring out the results of all the three serological reactions. The technical problems encountered for a definative diagnosis of CT and the clinical picture in the present critical examination are discussed.

Lymphokines which influence electrophoretic mobility of macrophages.

Supernatants of blood lymphocytes from immunized guinea pigs after incubation with purified protein derivative (PPD) were fractionated on Sephadex G-75. The fractions were tested for activity to change the electrophoretic mobility of macrophages. Dependent on incubation time, three different regions of activity with an average molecular weight of 13 000, 40 000 and more than 100 000 were estimated.

[Life threatening infection with bacteroides fragilis in connection with Dalkon shield (author's transl)].

A case of severe peritonitis in connection with the application of a Dalkon Shield is reported. By culturing the peritoneal exsudate a monoculture of Bacteroides fragilis was found as causing agens. The general aspects of severe infection in connection with IUD, particularly the Dalkon Shield, are discussed. By analysing the reports in the literature it cannot be ruled out, that Bacteroides fragilis may to be a major factor in cases with lethal outcome.

[The effect of clotrimazole on the pathogen of toxoplasmosis, Toxoplasma gondii].

By means of a toxoplasma infection in the mouse it was tested whether the antimycotic agent bis-phenyl-(2-chlorophenyl)-1-imidazolyl-methane (clotrimazol, Canesten¿) is also effective against the asexual stages of replication of T. gondii as they occur in man. Canesten did not influence the forms of the proliferative phase of replication of T. gondii (trophozoites, pseudocysts = tachyzoites) either in vivo in infected mice or in vitro in the tissue culture.

[Adenoma of the ciliary epithelium of unusual structure and localisation (authors transl)].

The authors describe an adenoma of the ciliary epithelium of low degree of malignacy. The tumor is clinically and histologically remarkable because of its uncommom site behind the iris and the abundance of mucoid stroma. The clinical data, the course as well as the histological findings are described and compared with the rare similar cases in the literature. Electron microscopic data of this kind of tumor are reported for the first time. The tumor are characterised by basal membranes, microvilli, desmosomes, occasional melanin granules, a well differentiated RER and specially by the massive content of intracytoplasmatic filaments.

[Statistical importance of tuberculosis and toxoplasmosis of the eye in recent years (1971-1974, Eye-Clinic, Davos) (author's transl)].

Tbc has not yet been eradicated in Western Europe. Among 311 human/bovin-tuberculin tested cases out of 500 patients with uveitis (1971-1974), 37-50% (average age 40-45 years) reacted highly positive. Chorioretinitis disseminata and periphlebitis retinae (Eales' disease): 90% positive. The criteria for a long term tuberculostatic treatment are exposed. 20% of all cases of uveitis and 90% of Jensen Retinochorioditis have been interpreted as caused by toxoplasmosis. 75% are relapses. The gigantic toxoplasmotic lesions of the eyeground is stressed. As a rule low Sabin Feldmann titers are found. Quantitative serology is valuable. In the closing discussion the immunologically well known adjuvans-effect of Koch's bacillus-extract is supposed to be of importance in highly tuberculin positive uveitis.

[Urogenital myiasis with Fannia scalaris].

A 15-year-old boy excreted a fly larva in his urine. It constituted the third larval state of the fly Fannia scalaris. Microhaematuria, mild albuminuria and leucocyturia ceased after the parasite had been passed.

Manipulation of neurotransmitters by acupuncture (?) (A preliminary communication).

Varying reactions of the vegetative nerve system to various point combinations (for example: vomiting, dizziness, diarrhea, urge to urinate, fatigue or drowsiness, headache), especially to the needling of Tai Chong (Li 3), induced us to perform biochemical studies before and after acupuncture treatment. A group of children and a group of adults were studied. The material studied was urine and blood; from the children, urine only. The following were determined in the urine: indolacetic acid, 5-hydroxy-indol-3-acetic acid, homovanillic acid, and vanillic-mandelic acid; in the blood, tyrosine and tryptophan (free and bound). Individual points with wide influence (He Gu = LI 4; Zu San Li = St 36; Tai Chong = Li 3) and their combination with generally effective points were tested. The needling of Tai Chong especially showed a clear increase in indolamine metabolism. Isolated increases in metabolites of catecholamine metabolism could be correlated with the patient's increased physical activity after acupuncture. Noteworthy is the observation that no significant chemical reactions were evident if local reactions to the needling no longer appeared at the end of a series of acupuncture treatments.

The nutritive effect of glucose on the structure and function of jejunal self-emptying blind loops in the rat.

In an attempt to obtain further information on the influence of the intestinal contents on the development of mucosal structure and function, self-emptying blind loops of rat jejunum were constructed, and the oral end was exteriorized with a stoma to permit infusion of different solutions into the loop. Ringer solution or media containing glucose or galactose was instilled into the loops three times daily for 27 days before functional and structural examination of the loops. The body weight and food intake of the animals did not differ significantly from one group to another. Treatment with glucose, but not with galactose or Ringer solution alone, induced a significant increase in the villus height. Disaccharidase but not dipeptidase activity was concomitantly increased. Infusion of glucose or galactose both led to an increase in the transport capacity of the loop, as determined by glucose absorption in vivo. These results indicate that glucose has a nutritive effect, probably due to its intracellular metabolism, which is responsible for the structural alterations observed. On the other hand, the increase in transport capacity may be attributed to substrate-induced stimulation of the specific monosaccharide transport system in the epithelial cell.

[Comparative Chemotherapeutical studies on the effect of Bactrim and other combined preparations on the proliferative and cystforming phase of Toxoplasma gondii in NMRI mice. A contribution to experimental chemotherapy of toxoplasmosis (author's transl)].

In the present paper, the efficacy of different sulphonamides and potentiators alone and in combination (BACTRIM, FANSIDAR) in mice infected with Toxoplasma gondii is evaluated. The experimental chemotherapeutical studies were performed in mice either freshly infected with trophozoites or cystozoites, or suffering from acute disease or a chronic infection; For the components (see article) a proportion of 10 times 1 turned out to be the most effective mixing ratio. In comparison with the standard combination of sulphamethoxypyrazine times pyrimethamine sharing the best degree of activity, the therapeutic effect of a combination of sulphadoxine times pyrimethamine (components of FANSIDAR) in a proportion of 20 times 1 was either the same or less, depending on the experimental design. The duration of therapy was found to be decisive for the therapeutic result. If it is too short the parasite will return to the cyst stage; Even at low dosage, the components of FANSIDAR, sulphadoxine and pyrimethamine in combination, are capable of acting upon all forms of the proliferative multiplication phase and the cyst formation phase, inclusive of young cysts, and eliminated them completely from the host organism.

[Bacteroides and appendicitis (author's transl)].

Bacteroides fragilis is frequently recovered from cases of appendicitis with perforation and from infections developing secondary to appendicitis. In order to assess the part played by B. fragilis in the aetiology of appendicitis, quantitative aerobic and anaerobic culture studies of the contents of 49 inflammated appendices were performed. Anaerobic gram-negative non-sporing rods were cultivated from 43 appendices in the range 10(3)-10(9)/g. A total of 1,473 isolates was differentiated by biochemical methods, and 1,374 cultures were found to belong to the saccharolytic species of the genus Bacteroides (B. fragilis, B. thetaiotaomicron, B. vulgatus, B. distasonis etc.). B fragilis was detected in 31 appendices; the species predominated in 18 samples. B theraiotamicron, recovered from 27 samples, was prevalent in 4 appendices. In one sample, B. fragilis and B. thetaiotaomicron outnumbered the other appendicular bacterial. B. vulgatus was cultivated from 12 appendices, but did once constitute the prevalent group. It has been previously shown that B. vulgatus (43% of intestinal isolates) and B. thetaiomicron predominate in the normal narge bowel flora. On the other hand, approximately 80% of pyrogenic Bacteroides strains belong to B. fragilis, B. thetaiotaomicron accounting for 19% and B. vulgatus being virtually absent. From these striking differences in species distribution the conclusion was drawn that B. fragilis possesses the highest virulence for man. Species distribution within the 1,374 appendicular isolates of saccharolytic Bacteroides (percentages of 62, 19 and 4.3 for B. fragilis, B. thetaiotaomicron, and B. vulgatus, respectively) was very similar to that encountered in clinical specimens. From the results obtained it becomes evident that pyrogenic Bacteroides, in particular B. fragilis, plays an important role in nearly 50% of cases of appendicitis.
