Biological characterization of a mesothelioma line in the nude mice. II. Some characteristics of cells cultivated in vitro prior to and after transplantation in nude mice.

Ascitic tumor cells from a female patient with a peritoneal carcinomatosis have been cultivated in vitro. The 25th in vitro passage of these cells were characterized by gowth curves. DNA-distribution patterns, chromosome patterns and drug sensitivity. Cells of the 25th in vitro passage were i.p. transplanted in nude mice. The 14th and 18th passage of ascitic cells and the 3rd passage of the solid tumor material grown in nude mice were compared with cells of the 25th in vitro passage. Changes in DNA-distribution patterns, chromosome patterns and drug sensitivity were observed: The G1/0-peak of the ascitic tumor cells (AP 14, AP 18) revealed a shift to the left. This is in coincidence with the chromosome number reduction found. Differences in sensitivity to Methotrexate and 5-Fluorouracil after transplantation in nude mice were observed. The possible reasons of instability of these biological properties are discussed.

Isolation of a multiprotein complex containing cytochrome b and c1 from Neurospora crassa mitochondria by affinity chromatography on immobilized cytochrome c. Difference in the binding between ferricytochrome c and ferrocytochrome c to the multiprotein complex.

A multiprotein complex which contains in equimolar amounts two cytochromes b (Mr each about 27,000), one cytochrome c1 (Mr 31,000) and six subunits without known prosthetic groups (Mr 8000, 12,000, 14,000, 45,000, 45,000, and 50,000) has been isolated from the mitochondrial membranes of Neurospora crassa by affinity chromatography on immobilized cytochrome c. The chromatographic separation was based upon the specific binding of the complex to ferricytochrome c coupled to Sepharose and its specific release upon conversion of the coupled ferricytochrome c into ferrocytochrome c using ascorbate as a reductant. The chromatography was performed in the presence of the nonionic detergent Triton X-100 at low ionic strengths. A monodisperse preparation of the multiprotein complex was obtained which was used for binding studies with cytochrome c from Neurospora crassa, horse heart and Saccaromyces cerevisiae. At low ionic strength (20 mM Trisacetate) and slightly alkaline pH (pH 7 to 8), more than one molecule of ferricytochrome c were bound to the isolated multiprotein complex with dissociation constants below 1 x 10(-7) M. One of these bindings appeared different from the others, since its high affinity was preserved at an ionic strength at which the affinities of the other bindings decreased. Furthermore, the affinity of only this binding decreased upon reduction of cytochrome c. It is suggested that this binding is at or near the functionally active site(s) of the mulipprotein complex.

Delayed hypersensitivity and lymphocytic transformation in patients with Hodgkin's disease and granulomas.

Noncaseating, sarcoid-like granulomas were found in the tissues of 9 out of 31 patients with Hodgkin's disease. In vivo and in vitro cell-mediated immunity was evaluated in patients with and without granulomas and compared to a group of 20 normal controls. Hodgkin's disease patients of both groups showed a significantly reduced in vivo and in vitro response when compared to the control group. However, when patients in stages IIIB and IV were eliminated and patients in stages I, II, and IIIA examined separately, a positive skin test response to one or more antigens was elicited in 85.7% of patients with granulomas, while a markedly decreased-dose dependent response was observed in patients without granulomas. In vitro lymphocyte blastic transformation by phytohemagglutinin (PHA) was severely impaired in both groups of patients as determined by dose-responses curves. These results indicate that Hodgkin's disease patients with granulomas have a significantly better skin test response than those without granulomas.

Induction of delayed-type hypersensitivity to staphylococcal antigens in guinea pigs.

Delayed-type hypersensitivity against staphylococcal antigens was induced in guinea pigs using Freund's incomplete as well as Freund's complete adjuvant. In animals sensitized with Freund's complete adjuvant the induction time and the duration of delayed-type hypersensitivity were longer and the skin reactions were stronger compared to animals sensitized with Freund's incomplete adjuvant. The difference between animals sensitized with Freund's incomplete or complete adjuvant may reflect the difference between tuberculin- and Jones-Mote type hypersensitivity.

[Computer-supported basic documentation at the accident-surgical clinic of the Hannover Medical School].

On the example of the computer-aided basic documentation at the Department of Traumatology Hannover, the necessity of constructive cooperation between the clinic staff and the medical data processors is shown. The system of basic documentation is primarily explained in its conception as far as the clinic is concerned. The fundamental principles, daily routine, and clinical application of this system are described, as are the tasks assigned to the clinical staff within the framework of medical data processing. The necessity of clinical staff quality controls by means of a computer short card is emphasized. The system has been in use for 3 years.

Rhegmatogenous retinal detachments in blacks.

Primary rhegmatogenous retinal detachment is probably much less common in blacks than whites. We reviewed 100 consecutive black patients with retinal detachment who were seen over a 3 year period, and compared them to a randomly selected group of 100 whites seen during the same period. Trauma was the most common cause of detachment in blacks (52%), but was a relatively uncommon cause in whites (12%). Posterior vitreous detachment was the most frequent etiology of detachment in whites (47%), but was rare in blacks (15%). Aphakia was the second most common etiology in whites (30%), while it was relatively uncommon in blacks (13%). Myopia and/or lattice was a more frequent associated finding in blacks with detachment (19%), than in whites (9%). These findings are discussed and the literature reviewed.
