[Diagnostic value of B-scan in exudative age-related macular degeneration].

PURPOSE: To explore the image features of exudative age-related macular degeneration (AMD) with B-scan. METHODS: Using NIDEK US-3300 model B-scan to observe 31 patients (54 eyes) with AMD. RESULTS: The membranous arc echo was found at the posterior polar and there was a shuttle absence of echo between the echo band and the wall of eyeball in the period of retinal pigment epithelium detachment. During the exudation and hemorrhage absorbed not comletely, mixed echo was seen. A small parenchyma body with protrusion was seen with stronger echo in the late phase when the exudation and hemorrhage were replaced by the scar tissues, it was like a convex lens or a triangle and the top pointed to the globe behind, but it was flat towards the vitreous. CONCLUSION: The B-scan examination is valuable in diagnosis, treatment and follow-up for exudative AMD.

[The application of basic fibroblast growth factor to auricles of rabbits with compound injury].

OBJECTIVE: To study the role of basic fibroblast growth factor (bFGF) in the repair of irradiation- and surgery-induced auricle trauma. METHODS: The bilateral auricles of New Zealand white rabbits were received surgical trauma and beta-active irradiation(90Sr-90Y), bFGF was then applied to the wound. The efficacy of bFGF on wound healing was compared with the control group. RESULTS: On the 10th day after the trauma, the mean epithelalized area was (34.17 +/- 0.37) mm2 and the mean number of microvessels/high power field was (10.25 +/- 2.34) in the treated group, whereas both were (26.25 +/- 0.20) mm2 and (4.38 +/- 0.92) respectivety in the control group. There was a significant difference between the two groups (P < 0.05). On the 7th and 14th days after the trauma, the mean epithelialized area in the treated group was significantly different from that of the control group. The measurement by the image cytometry showed that the treated group had a mean nuclear peak area value of (38.28 +/- 7.23) micron 2 and the mean S-stage cell value of (31.28 +/- 10.35)%, and those of the control group were (28.02 +/- 8.13) micron 2 and (20.14 +/- 9.46)% respectively. There were significant differences (P < 0.05) between them. CONCLUSION: Application of bFGF can, to a certain extent, accelerate the repair of irradiated and surgically injured soft tissue.

[Modification of cochlear active micromechanism by auditory cortex].

OBJECTIVE: To investigate the modification of active micromechanism of outer hair cells by auditory cortex. METHODS: 1. Effects of selective attention on distortion product otoacoustic emissions (DPOAE) were analyzed in 9 human subjects with normal hearing. Visual tasks, two letters, O and Q, were alternatively displayed on a computer screen. Letter Q was target stimulus flashing randomly (probability was 20%), and letter O was non-target stimulus occurring regularly (probability was 80%). 2. In 13 guinea pigs, DPOAEs were measured before and after decerebration. RESULTS: 1. Visual tasks suppressed DPOAE significantly in both 1 kHz and 2 kHz (P < 0.05). 2. DPOAE reduced significantly in decerebrated guinea pigs. CONCLUSION: The study further demonstrates that the efferents from the auditory cortex do innervate the cochlea, and the efferent pathway passes from the auditory cortex, the super olive complex to the cochlea. Through this pathway the auditory cortex modulates the cochlear active micromechanism. The underlying mechanism was discussed.

Coral as a carrier for recombinant human bone morphogenetic protein-2.

By combining coral with recombinant human bone morphogenetic protein-2 (rhBMP-2), rhBMP-2/coral composite was obtained in this study. Following implantation of the composite into the muscle pouches of mice, cartilage growth was induced in the pores or on the surface of the implants at one week, woven bone at three week and lamellar bone with bone marrow at six week, and coral was absorbed partially. The induced formation of endochondral bone was time-related and rhBMP-2 dose-related. The results of this study indicate that the composite possesses a superior ability of osteogenesis, and coral acts as one of the most suitable rhBMP-2 slowrelease carriers currently available. The composite will be a new type of bone substitute to be used in orthopaedics and maxillofacial surgery.

A rapid preparation of extracts for DNA replication in vitro.

A rapid preparation of cytoplasmic extracts using a small number of cells was developed for SV40 DNA replication in vitro. Compared with methods published previously, this new method has two advantages: First, cells of several human cell lines can be processed at the same time. Second, the time of preparation has been reduced from 10 h to 2 h by adding sucrose and reducing the concentration of NaCl in the dialysis buffer. Activities of extracts prepared from small numbers of cells with this new method to support SV40 DNA replication in vitro are high, reproducible and comparable to that of extracts from large numbers of cells with the methods traditionally used. These advantages will make it possible to study the regulation of DNA replication in irradiated or drug-treated human cells more efficiently. Therefore, this simple method should be a useful complement to the large-scale preparation for the general study of regulation of DNA replication in human cells.

Tobramycin-EDTA conjugate: a noninnocent affinity-cleaving reagent.

The design, synthesis, and ribozyme inhibitory activity of a novel EDTA-aminoglycoside conjugate are reported. This affinity cleaving reagent is a noninnocent RNA binder: its RNA affinity, judged by its ability to inhibit the hammerhead ribozyme HH16, is different than the parent natural product and is markedly dependent on the oxidation state of the chelated metal ion.

[The polymorphism of two microsatellites closely linked to the locus for polycystic kidney disease 1 in Chinese population].

OBJECTIVE: To evaluate the polymorphism of AC2.5 and SM7, two microsatellite markers closely linked to the locus for polycystic kidney disease 1 (PKD1), in the Han (Chinese majority) and the Zhuang(the largest minority in China) populations and make clear whether the allele frequencies of these markers exhibit significant differences between populations. METHODS: The two microsatellite markers were amplified by polymerase chain reaction(PCR), followed by 10% non-denatured polyacrylamide gel electrophoresis(PAGE) and DNA sequencing to analyze the PCR products. RESULTS: (1) Ten AC2.5 alleles and eleven SM7 alleles were observed in 104 Chinese Han people, and eight AC2.5 alleles and nine SM7 alleles were observed in 75 Chinese Zhuang people. (2) The expected heterozygosities of AC2. 5 and SM7 were 68.4% and 77.4% in the Han population and 67.1% and 57.5% in the Zhuang population respectively. The polymorphism information content(PIC) values of them were 0.68 and 0.77 in the Han population and 0.67 and 0.57 in the Zhuang population respectively. (3) Several allele frequencies of the two markers exhibited significant differences between populations(P 0.05). CONCLUSION: AC2.5 and SM7 are highly polymorphic and can be used in genetic diagnosis of both Chinese Han and Zhuang PKD1 families. The allele frequency distributions of them exhibit significant difference between populations.

Purification and characterization of novel ribosome inactivating proteins, alpha- and beta-pisavins, from seeds of the garden pea Pisum sativum.

Two ribosome inactivating proteins designated alpha- and beta-pisavins were isolated from seeds of the garden pea Pisum sativum var. arvense Poir with a procedure involving affinity chromatography on Affi-gel Blue gel, immobilized metal ion affinity chromatography on Iminodiacetic acid-agarose, cation exchange chromatography on Resource-S, and gel filtration on Superose 12. alpha- and beta-pisavins are nonglycoproteins with a molecular weight of 20.5 kDa and 18.7 kDa respectively. The sequences of the first sixty N-terminal amino acids of alpha- and beta-pisavins were identical. In isoelectric focusing these two proteins merged into one band with a pI greater than 9.3. Inhibition of protein synthesis by a rabbit reticulocyte lysate system was achieved at an IC50 of approximately 0.5 nM. Activity of the proteins toward tRNA was observed. The proteins acted on ribosomal RNA through its RNA N-glycosidase activity to release an Endo's fragment, and converted the conformation of DNA from supercoiled and circular forms into a linear form.

Ribosome inactivating protein and lectin from bitter melon (Momordica charantia) seeds: sequence comparison with related proteins.

One of the ribosome inactivating proteins (RIPs), beta-momorcharin, and a lectin were isolated from seeds of the bitter melon Momordica charantia (Family Cucurbitaceae) in accordance with published procedures. Both beta-momorcharin and M. charantia lectin were then subjected to amino acid sequencing. beta-momorcharin exhibited considerable homology in sequence to other Cucurbitaceae RIPs, and also to the A chains of abrin and ricin which are type II (double-chained) RIPs. The resemblance between alpha-momorcharin and other RIPs is closer than that between beta-momorcharins and other RIPs. M. charantia lectin manifested a certain extent of sequence similarity to beta-momorcharin and other RIPs, and some degree of homology in sequence to lectins from Cucurbita maxima, Cucurbita argyrosperma, Sambucus nigra and Ricinus Communis.

Apoprotein B100 has a prolonged interaction with the translocon during which its lipidation and translocation change from dependence on the microsomal triglyceride transfer protein to independence.

When lipid synthesis is limited in HepG2 cells, apoprotein B100 (apoB100) is not secreted but rapidly degraded by the ubiquitin-proteasome pathway. To investigate apoB100 biosynthesis and secretion further, the physical and functional states of apoB100 destined for either degradation or lipoprotein assembly were studied under conditions in which lipid synthesis, proteasomal activity, and microsomal triglyceride transfer protein (MTP) lipid-transfer activity were varied. Cells were pretreated with a proteasomal inhibitor (which remained with the cells throughout the experiment) and radiolabeled for 15 min. During the chase period, labeled apoB100 remained associated with the microsomes. Furthermore, by crosslinking sec61beta to apoB100, we showed that apoB100 remained close to the translocon at the same time apoB100-ubiquitin conjugates could be detected. When lipid synthesis and lipoprotein assembly/secretion were stimulated by adding oleic acid (OA) to the chase medium, apoB100 was deubiquitinated, and its interaction with sec61beta was disrupted, signifying completion of translocation concomitant with the formation of lipoprotein particles. MTP participates in apoB100 translocation and lipoprotein assembly. In the presence of OA, when MTP lipid-transfer activity was inhibited at the end of pulse labeling, apoB100 secretion was abolished. In contrast, when the labeled apoB100 was allowed to accumulate in the cell for 60 min before adding OA and the inhibitor, apoB100 lipidation and secretion were no longer impaired. Overall, the data imply that during most of its association with the endoplasmic reticulum, apoB100 is close to or within the translocon and is accessible to both the ubiquitin-proteasome and lipoprotein-assembly pathways. Furthermore, MTP lipid-transfer activity seems to be necessary only for early translocation and lipidation events.

Bcl-2-mediated resistance to apoptosis is associated with glutathione-induced inhibition of AP24 activation of nuclear DNA fragmentation.

Studies on the mechanism of apoptosis in this laboratory support a model in which signal transduction involving caspase 3 leads to activation of a serine protease called Mr 24,000 apoptotic protease (AP24), which then induces internucleosomal DNA fragmentation in the nucleus. This study examined the effect of Bcl-2 overexpression on activation of AP24 and the induction of DNA fragmentation by AP24 in isolated nuclei. It was demonstrated that overexpression of Bcl-2 in either HL-60 or PW leukemia cell lines suppressed activation of AP24 induced by either tumor necrosis factor or UV light and protected cells from apoptosis. Furthermore, nuclei isolated from Bcl-2-overexpressing cells were relatively resistant to internucleosomal DNA fragmentation induced by AP24 isolated from apoptotic cells. Bcl-2-overexpressing cells that were nutritionally depleted of glutathione (GSH) became sensitive to tumor necrosis factor- or UV light-induced activation of AP24 and underwent apoptotic cell death. Moreover, nuclei isolated from Bcl-2-overexpressing cells that were depleted of GSH became sensitive to AP24-induced DNA fragmentation. The addition of exogenous GSH blocked the proteolytic activity of AP24, as well as its ability to induce DNA fragmentation in normal isolated nuclei. These results indicate that Bcl-2 can attenuate at least two events in the AP24 apoptotic pathway: activation of AP24 and induction of DNA fragmentation by activated AP24. Furthermore, agents that deplete intracellular levels of GSH may have therapeutic use in the sensitization of Bcl-2-overexpressing cancer cells to apoptotic cell death.
