[The clinical significance of deficiency coagulopathy in relationship to consumption coagulopathy (authors' transl)].

A deficiency coagulopathy may lead to a coagulation defect in the presence of massive blood loss which is characterized by a dilution of the coagulation factors especially a dilution of the plasma fibrinogen and the platelets and a dilution of their function. Hemorrhages which are triggered by a coagulation defect (consumption coagulopathy) may be aggravated by the additional loss of coagulation factors. The combination of a coagulation coagulopathy with a deficiency coagulopathy results in a bad prognosis. A case of deficiency coagulopathy and a case of consumption coagulopathy with additional deficiency coagulopathy are reported, the clinical importance and the management of these conditions are discussed.

Synthesis, turnover and compartment analysis of the free fatty acids in the placenta of rats.

The in vivo tracer method and in vitro acetate incorporation experiments were used to investigate the metabolism of placental free fatty acids. Analysis of data revealed that free fatty acids pass into the fetus from maternal plasma through a small placental compartment, which accounts for only 5 percent of all of the placental free fatty acids. The turnover time of this compartment is 0.57 min. The rate of placental fatty acid synthesis evaluated by both methods is very small and amounts to 0.003 mumols fatty acids/min/all placentas of one litter.

[Effect of run-training and run-stress on glucose assimilation and insulin release in rats of different age (author's transl)].

82 male rats of different age have been investigated after a definitive run-training, after a run-training with subsequent run-stress and after a run-stress without run-training. An intravenous glucose tolerance test has been done measuring glucose and insulin serum levels before and after i.v. glucose application. Data obtained in this study demonstrate that: 1. adult rats have a better glucose assimilation and higher insulin serum levels after i.v. glucose than older ones. 2. run-stress leads to a deterioration of glucose assimilation and diminution of insulin release in rats of any age. 3. run-training improves glucose tolerance and saves insulin at the same time. 4. in old rats, which have never done any training before, run-training improves glucose assimilation, too. 5. run-stress after run-training has a different effect on adult and old rats: In adult rats there is nearly no effect on glucose assimilation and insulin release in comparison to control animals, while old rats show a significant deterioration of glucose assimilation in comparison to control rats of the same age. Obviously the effect of run-training is less distinct in old rats despite of similar running work.

[Investigations on LH and FSH serum levels before and after application of synthetic LH-releasing-hormone in healthy men of different age].

50 male healthy volunteers of different age had measurements of plasma luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Basal levels of LH and FSH as well as maximal release of both hormones after i.v. application of 25 mug of synthetical LH-releasing hormone have been done. Results demonstrate an increasing reserve capacity of LH and FSH with age. FSH/LH ratio is altered in favor of FSH with duration of life. Sensitivity of gonadotropin producing cells of adenohypophysis to LH-releasing hormone is diminished in old age that can be recognized from a diminution of the relative rise of serum LH and FSH after LH-RH application. Correlations and partial correlations of absolute and relative body weight with age and basal LH and FSH serum levels demonstrate that age and not body weight plays a main part for increasing gonadotropin serum values. Causative responsibility for the increased LH serum concentrations is a decrease of testosterone production with age. Data obtained in this study and reports presented in literature give no convincing explanation for the increase of FSH serum levels with age.

[Age-dependent changes of hypophysis-thyroid - regulation (author's transl)].

In health men and women different age total thyroxine, free thyroxine-binding-capacity, total triiodothyronine as well as TSH in serum before and after intravenous application of 500 mcg synthetic thyrothropine-releasing-hormone (TRH) were controlled. The investigations have shown: 1. With increasing age total thyroxine level in serum does not change. 2. With increasing age free thyroxine-binding-capacity in serum does not change. 3. With increasing age in men and women a significant decrease of total triiodothyronine in serum is to be measured. 4. Basal TSH serum levels do not change with increasing age. On the other hand in men and women with increasing age significant decreased TSH increments after TRH application were measured. The investigations have shown, that with increasing age the hypothalamus-hypophysial-thyroid-axis is shifting to a lower regulation level.

Maternal plasma triglycerides as a source of fetal fatty acids.

The transfer of plasma triglyceride fatty acids from mother to fetus was studied in rats. Following i.v. injection of labelled chylomicron and very low density lipoprotein (VLDL) triglycerides into the mother, the time courses of the plasma triglycerides, free fatty acids, and fetal radioactivity were determined. The data were analysed using a mathematical model. From the results the following conclusions were drawn: To cover the need of fetal fatty acids, the placenta utilizes only VLDL triglycerides but not chylomicron triglycerides. Comparison of the amount of VLDL triglyceride fatty acids (0.04 micromoles/min/litter) and of maternal plasma free fatty acids (0.08 micronmoles/min/litter) transferred into the fetus indicates that the maternal plasma triglycerides are a source of fetal fatty acids, that cannot be neglected.

Cell-mediated immunity in lumphocytic choriomeningitis. I. The specificity of the cytotoxic T lymphocytes.

The specificity of cytotoxic T lymphocytes (CTL) generated during murine lymphocytic choriomeningitis (LCM) has been investigated. CTL were obtained from the spleens of mice injected i.p. with LCM virus. The cytotoxic activity of the CTL was tested in an in vitro 51Cr cytotoxicity assay using infected macrophages or fibroblasts as target cells. At the peak of the cytotoxic T cell response (7-8 days after infection) the cytotoxic action was restricted to syngeneic virus-infected target cells. Using H-2 recombinant mice the target antigen of the CTL generated could be identified as products coded for by either the H-2 K or H-2 D region of the major histocompatibility complex. I region identity between CTL and infected target cells was insufficient for optimal lysis to occur. During the early phase of LCM virus infection there was a transient phase during which non-infected H-2 histocompatible targets were lysed as efficiently as virus-infected target cells. This finding may suggest, that during the early phase of LCM disease self-reactive cytotoxic T lymphocytes are temporarily present in LCM virus-infected mice.

[Activity distribution of SDH in liver of mice. A histophotometrical study (author's transl)].

The paper presents histophotometrical measurements of the histochemical SDH reaction with INT as redoxindicator in comparison with biochemical techniques of the determination of SDH activity (with K3[Fe(CN)6] and INT as indicators) and a histochemical method of elution (with INT as indicator). The measurements were performed on liver of mice. For each method absolute values of activity (10(-9) mol substrat/min-mg liver) are calculated as relative values of activity [%]. The standard deviation of the biochemical methods is approximate +/- 4%, of the histochemical methods +/- 8%. The smallest value of detection is 2.5-10(-14) mol formazan of histophotometrical measurement. The correspondence between the measurements of the relative values of activity by the described methods was secured by statistical tests. A statistically significant proportion of activities between periportal zone and central zone like 100:82 was determinated by histophotometrical measurement.

[Partusisten as a possible cause for renal colics (author's transl)].

Two patients were treated with Partusisten and Isoptin to inhibit uterine contractions. One patient was in the 36th and the other in the 32th week of pregnancy. Both developed colic like complaints related to the urogenital system, which disappeared after disruption of treatment and reoccured after restarting. The case description seemed to indicate an as yet not described side reaction of Partusisten.

Induction of cytotoxic T lymphocytes against I-region-coded determinants: in vitro evidence for a third histocompatibility locus in the mouse.

Determinants controlled by the I region of the murine H-2 complex provoked the generation of cytotoxic T lymphocytes (CTL) in both a secondary and primary mixed lymphocyte culture. The stimulating determinants appeared to be controlled by loci within the I-A subregion. The target antigens of the CTL generated were present on both lipopolysaccharide- and concanavalin-induced blast lymphocytes, but were barely detectable on phytohemagglutinin-induced blast cells. The stimulating capacity for CTL induction of a complete H-2 complex incompatibility by far exceeded the sum of H-2D/K-region and I-region incompatibility, respectively.

[Clinical investigations of the effect of tolbutamide and glibenclamide on pancreatic glucagon secretion (author's transl)].

The effects of tolbutamide, glibenclamide, arginine and arginine in combination with glibenclamide upon insulin, glucagon and glucose serum levels have been studied in healthy young men. Sulphonylurea-induced hypoglycemia is followed by a reactive hyperglucagonemia. Arginine-induced hyperglucagonemia is not suppressed by sulphonylureas. While there exists no difference between tolbutamide and glibenclamide-stimulated glucagon secretion, there is one with regard to insulin secretion.

Effects of synthetic somatostatin on endotoxin-induced changes of growth hormone, cortisol and insulin in plasma, blood sugar and blood leukocytes in man.

In normal man synthetic somatostatin inhibits the growth hormone response to intravenous administration of endotoxin (Piromen), but exerts no influence on cortisol plasma levels. Endotoxin-induced leukocytosis in blood is significantly reduced by somatostatin. Plasma levels of insulin and the blood sugar in man receiving endotoxin are firstly reduced in a significant manner and then increasing to normal values during somatostatin infusion.
