beta(1 leads to 3) Glucan-mediated augmentation of alloreactive murine cytotoxic T-lymphocytes in vivo.

Five different beta(1 leads to 3) glucans were tested for immune adjuvant activity on the in vivo induction of alloreactive murine cytotoxic T-lymphocytes (CTL). The beta(1 leads to 3) glucans, lentinan, pachyman, pachymaran, and two differently substitute hydroxyethylated pachymans strongly enhanced the in vivo generation of alloreactive CTL. The augmenting effect of i.p.-administered beta(1 leads to 3) glucans exhibited a clear dose-response relationship and was strictly dependent on the injection schedule used. Injection of high doses of beta(1 leads to 3) glucans as well as the injection during the late phase of the immune response markedly suppressed the magnitude of the lytic CTL activity induced. When the optimal conditions for enhanced CTL responses were chosen, the augmented CTL activity within spleen cells and mesenteric lymph node cells persisted for more than 25 days. Since beta(1 leads to 3) glucans are chemically defined substances without obvious toxic side effects, they may be of potential use to augment in vivo antigen-specific T-cell responses.

[Correlations between age, risk factors for arteriosclerosis and endocrine functions (author's transl)].

In 377 males and 528 female factory workers basal serum levels of glucose, neutralfat, cholesterol, Na, K, Ca and LH, FSH, TSH, Prolactin, GH, Insulin, total corticoids and testosteron were determined. Mean values and standard deviations as well as correlations between mean basal hormone levels and other serum parameters, blood pressure and relative body weight (Broca Index) and partial correlations excluding age and relative body weight were calculated. Basal serum levels of LH, FSH, GH and prolactin show age and sex dependent secretion patterns. In men serum insulin-, GH- and total corticoid levels correlate positive significant with glucose and cholesterol levels, relative body weight and blood pressure, which possibly shows, that these hormones may play a role in the development of degenerative heart and blood vessel diseases. Testosteron levels and parameters of fat metabolism, relative body weight and diastolic blood pressure correlate negative significant. Obviously testosteron has a protective effect against arterioslcerotic lesions. In women insulin and GH do not have the same importance as in men. Prolactin serum and cholesterol level correlate negative significant. In postmenopausal women LH and FSH serum levels correlate positive significant with glucose and cholesterol levels, blood pressure and relative body weight, the so called risk factors for arteriosclerosis.

Effect of niridazole in cellular immunity in vivo and in vitro.

The influence of niridazole, an anti-helminthic drug, on cell-mediated immune responses was investigated. Allograft rejection in mice as well as the in vitro induction of cytotoxic T lymphocytes (CTL) against murine alloantigen were used as the test system. Repeated daily oral treatment of host mice with niridazole (100 mg/kg) prior to and during allotransplantation resulte in the postponement of graft rejection, inducing a transitory functional state of allograft tolerance. The time interval between the termination of niridazole administration and onset of graft rejection was estimated to be 5-7 days. In order to test the effect of niridazole or its derivatives on the in vitro induction of alloreactive CTL, the serum or urine of mice which were treated with niridazole were added to the cultures, instead of adding niridazole directly to the cultures. Such serum and urine were found to be inhibitory for in vitro induction of CTL. The serum and urine had no effect on the effector phase of CTL.

[Multivariate variability investigations of acoustically evoked slow potentials (AEP) (author's transl)].

By means of the multivariate variance analysis the ratio of intra- and interindividual variability of the AEP--time function was investigated. 90 AEP from 9 normal hearing persons (10 AEP from every person, averaging sessions spread over 10 weeks) were analyzed. The 10 AEP of every person were treated as a random sample. A multivariate pair comparison (single comparison between samples) on the basis of 12 variables (time points) showed only for 1 out of 36 sample pairs no significant differences between the sample averages. By applying the discriminant analysis false classifications resulted only for 7 out of the 90 AEP (approximately 8%). These facts mean, that even throughout the time of 10 weeks the intraindividual differences of the AEP--time behavior are significantly smaller than the interindividual ones.

[Hair cell noise damage after improved hypoxia tolerance (author's transl)].

UNLABELLED: Guinea pigs were gradually adapted to a simulated altitude of 10,000 m. The round window microphone potential (RMP) frequency response was measured 24 hours after pure tone exposition (2.7 kHz, 130 dB, 1 hr). The electrophysiologically demonstrable sound induced damage of the organ of Corti of the adapted guinea pigs was significantly smaller than those of the nonadapted (normal) group. Altitude adapted animals: m = 18, Hb: 18.5 +/- 1.8 g%, Hc: 57.3 +/- 4.4+ RMP mean loss: 1.7 +/- 0.8 dB. CONTROLS: n = 18, Hb: 12.7 +/- 1.8 g%, Hc: 40.5 +/- 1.6%. RMP mean loss: 5.8 +/- 2.4 dB. All differences significant p less than 0.001. The evident possibility to reduce noise induced hair cell damage by means of previous adaptation to high altitude might be explained by the greater hypoxia tolerance and perhaps additional better oxygen supply to the receptor cells.

The analysis and source of 1-diphenylmethyl-4-nitrosopiperazine in 1-diphenylmethyl-4-[(6-methyl-2-pyridyl) methyleneamino]piperazine: a case history.

Analytical methods were developed to determine whether there was less than one ppm of II in IV. Purified IV contained a compound with the same GC retention time as authentic II on OV-17 and Silar 10C columns, using a FID or a nitrogen/phosphorus detector. The GC-coupled mass spectra contained major peaks at m/e 251 and 167 and the GC-coupled methane CI spectra gave the quasi-molecular ion, m/e 282. However, the CI selected ion monitor indicated three compounds at m/e 282. The nitrosamine II was separated from IV by HPLC on muBondapak C18. The GC-coupled mass spectra of the HPLC nitrosamine fraction had peaks at m/e 251 and 167 and other peaks for a compound of greater MW than II, although the GC retention times were the same. Evidence for II and a decrease in the detection limit to one ppm was obtained with a TEA interfaced with a HPLC. To determine if II was formed from IV, it was exposed to ozone at -78 degrees C in methylene chloride. The DSC, IR and NMR of the product were coincident with those of the standard of II. In other experiments, IV in methylene chloride was exposed to light and dry air in the presence and absence of methylene blue. The PMR and 13C NMR of the product formed in the presence of methylene blue were the same as that of II. It is postulated that this nitrosamine was formed by a singlet oxygen mechanism.

Quantitative evaluation of the fetal fatty acid synthesis in the rat.

The fetal fatty acid synthesis has been evaluated by 3H2O in vivo incorporation experiments. The results indicate that the fetus synthesizes 0,16 mumoles fatty acids/min/litter. From the results the conclusion can be drawn that the fetus gets fatty acids from the mother and from its own fetal fatty acid synthesis at equal amounts.

Studies on the paraplacental free fatty acid transport in rats.

Using the tracer method and a mathematical model the paraplacental transport of free fatty acids (FFA) (via uterine fluid--yolk sac--fetus) has been investigated in 21 days pregnant rats. The results show that only 0.3 nmoles FFA per min pass into the fetus via this paraplacental route. This accounts for less than 1% of FFA transported from the maternal plasma across the placenta into the fetus (0.14 mumoles FFA per min). From these results we would like to draw the conclusion that the paraplacental transport of FFA to the rat fetus is of minor importance from the quantitative point of view.

[Tracer kinetic studies on maternal-fetal amino acid transport in Wistar rats].

The experiments were designed to determine the amount of free amino acids which passes through the placenta from the maternal pool in plasma to the fetus. On day 19 of gestation we determined the growth rate of the rat fetus to be about 39 microgram amino nitrogen/minute/litter. Uniformly labelled [14 C]-L-leucine was used in our experiments. The specific radioactivity was measured in the maternal plasma amino acids and in fetal amino acids and proteins from zero to sixty minutes after pulse labelling. By means of compartmental analysis we calculated the transfer rate of amino nitrogen from the mother to the fetus to be 89 microgram/min/litter. The rate of amino nitrogen acccumulation in fetal carcass proteins was 39 microgram amino nitrogen/min/litter. The importance of our results in fetal metabolism is discussed.

Studies on the hypertriglyceridemia in the pregnant rat.

The in vivo tracer method and in vitro incorporation experiments were used to obtain information on the mechanism responsible for the hypertriglyceridemia found in 21 days pregnant rats. The results indicate that the uptake of plasma free fatty acids (FFA) by liver triglycerides of pregnant rats (2.0 mumoles FFA/min/300 g) did not differ from that of non-pregnant rats (2,1 mumoles FFA/min/300 g). The rate of liver fatty acid synthesis in pregnant rats (0,4 mumoles fatty acids/min/liver) is higher than that of non-pregnant rats (0,05 mumoles fatty acids/min/liver). In non-pregnant female and pregnant rats the turnover rates of plasma chylomicron and VLDL triglycerides were almost identical (about 2.8 mumoles triglyceride fatty acids/min/300 g, and about 2.3 mumoles triglyceride fatty acids/min/300 g, respectively). These various observations, when taken together, strongly suggest that increased hepatic triglyceride production cannot be the mechanism responsible for the hypertriglyceridemia in pregnant rats.

Operative lengthening of the femur.

Femoral lengthening with the aid of the apparatus described in this article can be accomplished safely and predictably. However, it is not without considerations and problems which must be fully understood and appreciated. The healing process requires months to years; with a cooperative patient and a diligent attentive orthopedist, the equalization of leg lengths is more than justified.

Effect of saralasin on plasma renin activity and arginine-vasopressin in hypertensive man.

This study was carried out to assess the influence of saralasin (SAR), an angiotensin II-analogue, on peripheral and central angiotensin II-receptors by measurements of plasma renin activity (PRA) and arginine-vasopressin (AVP) release. Before and during i.v. infusion of 10 microgram/kg/min of SAR over a 30 minute period, blood samples were obtained from 15 recumbent hypertensive patients (7 renovascular, 8 essential) to determine hormone activities by radioimmunoassay. In 10 patients with a decrease of blood pressure following SAR, PRA increased significantly whereas AVP levels increased significantly in only 7 of these patients. In the remaining 5 patients without a fall of blood pressure, PRA and AVP remained virtually unchanged. The results indicate that an enhanced AVP release may be due to a hypotensive stimulus induced by SAR in angiotensinogenic hypertension. A direct influence of SAR on central receptors is unlikely under the conditions studied.

[Possibilities of ultrasonic diagnosis in fetal abdominal monstrosities (author's transl)].

The diagnostic possibilities of intrauterine ultrasonic assessment of the fetal abdomen by rapid B-scan are described on the basis of two cases (megalocystis/megaloureter syndrome and omphalocele with cloaca formation). Other examination methods of prenatal diagnosis to confirm the findings are also discussed, but their diagnostic significance is even more limited than that of sonography, individual cases excepted. Although early detection of neural tube anomalies is the most important application of ultrasound, pronounced monstrosities of the fetal abdominal region are also definitely recognisable via ultrasonic examination.

[Technic of surgical reduction plasty of the female breast].

The surgical principles for the reduction operation of the female breast were established in numerous publications during the last decades. Improvement of results can be obtained in our opinion only by refinement of previous described techniques. We are describing the operation for mammahyperplasia designed in this institution which contains modified steps of Stroembeck, Biesenberger and McKissock technique. The cosmetic results are acceptable and the complications of slough of the nipples is greatly reduced.

Primary in vitro sensitization of virus specific cytotoxic T lymphocytes.

CBA mouse-derived splenic lymphocytes treated with either beta-priopionlactone-inactivated or u.v. light-inactivated parainfluenza (sendai) virus stimulated in vitro unprimed syngeneic T-lymphocytes to differentiate into cytotoxic T-lymphocytes (CTL). The CTL generated were virus specific and H-2 restricted. For optimal CTL responses to be induced (i) a critical treatment of stimulator cells, (ii) an optimal ratio of responder to stimulator cells and (iii) an in vitro incubation period of 5 days was required. The in vitro system for the induction of primary virus-specific CTL responses may be useful to analyse the sensitization phase of H-2 restricted virus-specific CTL.
