Evidence for the presence of a histaminergic neuron system in the rat brain: an immunohistochemical analysis.

Histamine-containing cells in rats were identified by indirect immunofluorescent histochemistry using an antibody raised against histidine decarboxylase (HDC), the enzyme forming histamine, which was purified from fetal rat liver. HDC-like immunoreactive (HDCI) structures could be detected in the brain as well as in peritoneal mast cells and basal-granulated cells in deep crypts of the gastric mucosa of rats. Numerous HDCI neurons were found in the posterior hypothalamic area and HDCI nerve fibers with a varicose appearance of fluorescence were widely distributed in various regions of the brain.

Coagulant and fibrinolytic activities in the leukemic cell lysates.

We attempted to examine procoagulant activity (PCA), X activator activity (XAA) and plasminogen activator activity (PlgAA) of various leukemic cell lysates: 17 acute myelocytic leukemias (AML), 4 acute promyelocytic leukemias (APL), 9 acute myelomonocytic leukemias (AMMoL), 7 chronic myelocytic leukemias (CML), 4 CML with blastic crisis, 7 T cell acute lymphocytic leukemias (ALL), 8 adult T cell leukemias (ATL), 8 null cell ALL, 6 B cell lymphocytic leukemias. Among those 70 cases, 4 APL, 4 AMMoL and 5 AML were associated with overt disseminated intravascular coagulation (DIC) and 5 T cell ALL, 7 ATL and 2 null cell ALL were associated with hypofibrinogenemia not adapted for DIC. The sample used was the lysate of 10(7) cells. PCA was measured by recalcification time of normal plasma with the cell lysate, XAA and PlgAA was measured by chromogenic substrate. APL and AML, especially those associated with overt DIC, had high PCA, and lymphocytic leukemia generally had low PCA in comparison with normal controls. Total PCA (PCA multiplied by cell count/microliter) was remarkably increased in DIC and mildly increased in ALL with hypofibrinogenemia. The change in XAA and total XAA (XAA multiplied by cell count/microliter) was not remarkable in any leukemia except for T cell ALL and null cell ALL with hypofibrinogenemia. PlgAA was high in lymphocytic leukemias with hypofibrinogenemia, APL and AMMoL with DIC. Total PlgAA (PlgAA multiplied by cell count/microliter) was high especially in T cell ALL and null cell ALL with hypofibrinogenemia. Thus it is probable that PCA is the most important factor causing DIC in myelogenous leukemia and that PlgAA is the most important factor causing hypofibrinogenemia in lymphocytic leukemia. The measurement of these activities in the leukemic cells is valuable in prediction and prevention of the hemostatic disorder in leukemia.

Method for enzymatic determination of imidazole acetic acid.

A method for enzymatic assay of imidazole acetic acid (ImAA) was developed, based on the strict substrate specificity of imidazole acetate monooxygenase from Pseudomonas sp. [Maki et al. (1969) J. Biol. Chem., 244., 2942-2950], which catalyzes concomitant conversion of NADH to NAD+. Thus, ImAA was determined by measuring decrease in absorbancy at 340 nm. Tissue extracts were partially purified and/or concentrated by column chromatography on Bio-Rad AG-1 before enzymatic assay. The lowest measurable level of ImAA by this method was 2 nmol.

Effect of solution of low pH on releases of beta-endorphin-like immunoreactivity and ACTH-like immunoreactivity from human gastric antral mucosa in vitro.

The presence of beta-endorphin-like and ACTH-like immunoreactivities (beta-EpLI, and ACTH-LI, respectively) in human gastric antral mucosa and their release were studied. beta-EpLI (14.1 +/- 4.8 ng/g wet weight) and ACTH-LI (11.1 +/- 1.2 ng/g wet weight) were detected in human gastric antral mucosa. The two activities gave similar curves for inhibition of human beta-Ep and ACTH on radioimmunoassay, respectively. On gel-filtration chromatography of a gastric antral extract, three components of beta-EpLI and two components of ACTH-LI were separated. When human gastric antral mucosa was incubated with a solution of pH 4.0 or pH 2.0, the release of beta-EpLI and ACTH-LI from the mucosa into the incubation medium increased 2.3-9.8-fold. However, incubation with 20 mM glucose or 20 mM amino acids did not have any effect on the release of beta-EpLI and ACTH-LI. These results indicate that beta-EpLI and ACTH-LI present in human gastric antral mucosa are released by direct action of a solution of low pH on the gastric antral mucosa.

Highly acidic proteins from human brain: purification and properties of Glu-50 protein.

Three extremely acidic proteins were isolated from human brain and purified to apparent homogeneity. One of them, Glu-50 protein, contained much glutamic acid (about 50% of the total amino acids). Its purification involved ammonium sulfate fractionation, DEAE-Sephadex A-50 chromatography, and gel filtration on Sephadex G-100 and G-75. Its molecular weight was determined to be 11,000 by SDS polyacrylamide gel electrophoresis and 34,000-36,000 by gel filtration on Sephadex G-75, suggesting that it consists of three identical polypeptide chains. Its isoelectric point was pH 3.9. Its N-terminal amino acid sequence was NH2-Asp-Glu-Pro-Pro-Asp-Glu and its C-terminal amino acid was Lys. It contained no detectable carbohydrate.

Acidic proteins from human brain: purification and properties of Glu-20 and Glu-35 proteins, and immunochemical comparison with Glu-50 protein.

In a continuation of previous work [Nomata et al. (1983) J. Biochem. 93, 825-831], this paper reports the purification and properties of the two other acidic proteins from human brain, and the immunochemical comparison of the three proteins. The protein described previously was named Glu-50 protein, because 50% of its amino acid residues were glutamic acid. By analogy, the proteins reported here were named Glu-20 and Glu-35 proteins on the basis of their glutamic acid contents. Both proteins were purified to a homogeneous state from human brain by ammonium sulfate fractionation, DEAE-Sephadex A-50 column chromatography and gel filtration on Sephadex G-75 and G-100, like Glu-50 protein. The molecular weights of the Glu-20 and Glu-35 proteins in the native state were 61,000 and 48,000 daltons and the values in SDS medium were 60,000 and 16,000, respectively, suggesting that Glu-20 protein is a monomer, and Glu-35 a trimer. The isoelectric points of Glu-20 and Glu-35 proteins were pH 4.2 and 3.6, respectively. Glu-20 protein contained 20.5% Glu, 18.4% Asp and 10.5% Lys, and Glu-35 protein contained 33.9% Glu and 17.2% Asp. The N-terminal amino acids of both proteins were Gly, whereas the C-termini of Glu-20 and Glu-35 proteins were Leu and Lys, respectively. Glu-20 protein gave a slightly positive periodic acid-Schiff reaction, suggesting that it is a glycoprotein, but Glu-35 protein gave a negative reaction. The three acidic proteins, Glu-50, Glu-35, and Glu-20 proteins, were compared immunochemically. Antibodies could be produced in rabbits against Glu-50 and Glu-20 proteins only after immunization with the performate-oxidized proteins, but no antibody could be produced against Glu-35 by similar immunization. Anti Glu-50 antibody reacted only with Glu-50 protein, not with Glu-35 or Glu-20 protein, calmodulin, or S-100 protein. Similarly, anti Glu-20 antibody was specific for Glu-20 protein and did not cross-react with Glu-35 or Glu-50 protein, calmodulin, or S-100. Anti S-100 antibody did not react with Glu-50, Glu-35, or Glu-20 protein. The cross-reactivities of these antibodies were tested with extracts of various tissues of a goat. Anti Glu-50 antibody reacted only with an extract of the brain, not with extracts of other tissues, whereas anti Glu-20 antibody reacted with all the tissue extracts examined.

Effect of alpha-fluoromethylhistidine on the histamine content of the brain of W/Wv mice devoid of mast cells: turnover of brain histamine.

In the brains of W/Wv mutant mice that have no mast cells, the histidine decarboxylase (HDC) level is as high as in the brain of congenic normal mice (+/+), but the histamine content is 53% of that of +/+ mice. The effects of alpha-fluoromethylhistidine (alpha-FMH) on the HDC activity and histamine content of the brain of W/Wv and +/+ mice were examined. In both strains, 30 min after i.p. injection of alpha-FMH the HDC activity of the brain had decreased to 10% of that in untreated mice. The histamine content decreased more gradually, and after 6 h about half of the control level remained in +/+ mice, whereas histamine had disappeared almost completely in W/Wv mice. It is concluded that the portion of the histamine content that was depleted by HDC inhibitor in a short time is derived from non-mast cells, probably neural cells. The half-life of histamine in the brain of W/Wv mice was estimated from the time-dependent decrease in the histamine content of the brain after administration of alpha-FMH: 48 min in the forebrain, 103 min in the midbrain, and 66 min in the hindbrain.

Mechanism of hematuria in glomerular disease. An electron microscopic study in a case of diffuse membranous glomerulonephritis.

From an electron-microscopic study in a case of diffuse membranous glomerulonephritis, we would like to propose a possible mechanism of hematuria in glomerular disease. There are several factors we should consider: (1) the deforming force of the red blood cell (RBC); (2) the deformability of the RBC; (3) the size of the gaps in the glomerular basement membrane (GBM), and (4) the thickness of the GBM. The deforming force is due to increased capillary pressure in the glomerulus that alters the contour of the RBCs. If these gaps are larger than 0.25 micron, the stretching and retracting force of the internal chamber of the GBM, combined with the capillary pulse, cause the deformed RBCs to pass through the gaps and proceed to the urinary space.

[Effects of intraseptal microinjection of 6-hydroxydopamine upon the time discrimination behavior in rats].

An intraseptal 6-hydroxydopamine (6-OHDA) injection technique was used in order to examine the specific functions of the catecholamine (CA) neurons in the septal area upon the time discrimination behavior. After some preliminary CRF trainings of 5 days, 21 rats of Wistar strain were divided into 3 homogeneous groups according to their performance on the last session. 6-OHDA group (n = 6) was stereotaxically injected into the medial septal area with 8 micrograms of 6-OHDA in 0.9% saline solution containing 0.1% ascorbic acid. Ascorbic acid group (n = 5) was injected with only ascorbic acid solution and the remaining group (n = 10) received no treatments. Following the 6 recovery days and the next CRF trainings of 5 days, all subjects were exposed to a DRLL 20/LH 36 training for 30 min per day and consecutively for 30 days. As the results of these trainings, 6-OHDA group showed significantly better performance and less acceleration of operant responding just after incorrect (non-rewarded) responses as compared with other control groups in the early 10 sessions of the trainings. These findings suggested that the acquisition of time discrimination behavior was facilitated in the 6-OHDA group which was hardly affected by the emotional disturbance just after non-rewards. Therefore the authors consider that the emotional disturbance caused by non-rewards impairs the inhibition of operant responding by mediating the CA neurons in the septal area.

Light and electron microscopic studies of malignant melanoma of conjunctiva and palpebra.

The authors experienced one case of malignant melanoma of the superior palpebral conjunctiva and the inferior palpebral skin, originating from benign acquired melanosis of the bulbar conjunctiva which developed in a 48-year-old man. Orbital exenteration including superior and inferior palpebrae was performed, and the removed tissue was examined by light and electron microscopy. In the temporal bulbar conjunctiva, melanocytes containing abundant fine melanin granules proliferated over almost the whole area of the epithelial layer. Cellular atypia and karyomitosis were not observed. The basement membrane remained intact with no proliferation of melanocytes in the substantia propria. However, in the perivascular area, in addition to the moderately prominent infiltration by lymphocytes and plasma cells, infiltration by melanophages was observed relatively frequently. Based on these findings, it was confirmed that the primary lesion of the bulbar conjunctiva was an acquired melanosis of Stage IB as classified by Zimmerman. The black nodular tumors of the superior palpebral conjunctiva and inferior palpebral skin consisted of melanoma cells showing markedly prominent cellular and nuclear atypia. The nuclei showed indentations in the membrane with electron-lucent karyoplasm, each having one nucleolus. The cytoplasm contained many round or elongated melanosomes in various developmental stages, with sizes of about 0.7 micron. Scattered among melanoma cells were a number of melanophages which had phagocytized many melanosomes at various stages of maturity.(ABSTRACT TRUNCATED AT 250 WORDS)

[Intraoperative dual-mode independent lung ventilation for open-chest surgery: conventional volume-set ventilation in healthy lung and high frequency jet ventilation in diseased lung].

An effort by the anesthesiologist to maintain adequate ventilation during thoracic surgery is sometimes disturbing for the operative procedures of the surgeon. Unilateral ventilation with a large tidal volume, leaving the operative site unventilated, may provide an adequate and quiet operative field, but is opposed by the problem of disturbance in pulmonary gas exchange. The application of high frequency jet ventilation has recently been introduced to solve these problems. However, the disadvantage inherent to this technique is the tendency to produce carbon dioxide retention although it provides adequate oxygenation. In the present study, using a double lumen endobronchial tube, the large-tidal volume ventilation of the non-operative site and the high frequency jet ventilation with small tidal volume of the operative site were performed simultaneously. This technique provided the satisfactory condition of operative field, i.e., "quiet lung". Also, the serial gas analysis of the arterial and mixed venous blood samples indicated the satisfactory condition of pulmonary gas exchange.

Precursor of mitochondrial glutamic oxaloacetic transaminase isozyme exists as a dimer.

Mitochondrial glutamic oxaloacetic transaminase is synthesized in the cytoplasm as a larger precursor and then imported into mitochondria in association with its processing to mature form. The precursor corresponded to about 0.05-0.1% of the total proteins synthesized on membrane-free polysomes. Gel permeation chromatography showed that the molecular weight of the precursor was about 100,000 daltons, which was slightly larger than that of mature enzyme (homodimer: 45,000 x 2). On the other hand, sodium dodecylsulfate treated precursor had the molecular size (about 50,000 daltons) slightly larger than that of the subunit of mature one by the gel permeation chromatography. These results suggest that the precursor of mitochondrial glutamic oxaloacetic transaminase exists as a dimer.
