Detection of histidine decarboxylase (HDC) in paraffin sections of formalin fixed rat stomach using avidin-biotin-peroxidase-complex method.

Immunohistochemical localization of histidine decarboxylase (HDC) in rat stomach was studied. Formalin fixed, paraffin sections were prepared and immunohistochemical localization of HDC was performed by the avidin-biotin-peroxidase-complex method (ABC method). As a result, HDC was clearly detected in the basal portion of the oxyntic glands. The shape of many HDC-positive cells were flasklike. No reaction products were seen in chief cells or parietal cells. These findings are in agreement with the results reported by many workers. Therefore, ABC method of HDC is thought to be a useful technique for the detection of enterochromaffin-like cells using paraffin sections.

Demonstration of immunohistochemical and immunochemical cross-reactivity of L-histidine and L-dopa decarboxylases using antibodies against the two enzymes.

Both rat L-histidine decarboxylase (HDC) and guinea-pig L-DOPA decarboxylase (DDC) were shown immunohistochemically and immunochemically to react with anti-rat HDC antibody. No cross-reaction was observed in immunoprecipitation experiments, but both anti-rat HDC antibody and anti-rat DDC antibody immunostained neurons in the substantia nigra, raphe nucleus and locus coeruleus of guinea-pig brain. Moreover, on immunoblotting, anti-rat HDC antibody recognized not only rat HDC but also guinea-pig DDC, but not rat DDC. However, anti-rat DDC antibody showed no immunohistochemical or immunochemical cross-reactivity with rat HDC.

Spectrally selective flash early receptor potential (ERP) in dichromats.

The human spectrally selective flash early receptor potential (ERP) was studied in 12 dichromats: 6 protanopes (12 eyes) and 6 deuteranopes (12 eyes). Color filters used were Kodak Wratten filters No.23A, No.57, and No.47 for the red, green, and blue flash ERPs, respectively. The ERP amplitude was measured between the summits of R1 and R2. Mean amplitudes of the red flash ERP and green flash ERP were highly significantly decreased in the protanopes (p less than 0.001) and deuteranopes (p less than 0.01) as compared with the corresponding data in 10 normal subjects (20 eyes). The mean amplitude of the blue flash ERP was significantly lower than normal (p less than 0.001) in the deuteranopes. The mean ratio of the blue flash ERP amplitude to the red flash ERP amplitude showed a highly significant increase in the protanopes (p less than 0.001) and a highly significant decrease in the deutoranopes (p less than 0.001) compared with the mean ratio in the normal subjects, indicating a new, useful index for the objective clinical detection of congenital color defects.

Determination of the molecular size of the hepatic H1-receptor by target size analysis.

The molecular sizes of histamine H1-receptors of rat, rabbit, human, pig, guinea-pig, chicken, dog, and bovine liver were investigated by radiation inactivation and determined to be 100,000 to 160,000 daltons in all the animals examined. Statistical analysis showed that the hepatic H1-receptors have a common size of 128,000 +/- 63,000 daltons. Saturation analysis showed that the [3H]mepyramine binding constant was not changed by irradiation, while the binding capacity decreased with increase in the radiation dose.

Histaminergic axons in the neostriatum and cerebral cortex of the rat: a correlated light and electron microscopic immunocytochemical study using histidine decarboxylase as a marker.

Histaminergic nerve fibers and their axonal varicosities in the neostriatum and cerebral cortex were light and electronmicroscopically examined by means of peroxidase-antiperoxidase immunocytochemistry with histidine decarboxylase (HDC) as a marker. A majority of HDC-like immunoreactive axonal varicosities observed in serial thin sections for electron microscopy exhibited no synaptic contacts in either the neostriatum or cerebral cortex. The remaining small proportion of immunoreactive axonal varicosities formed synaptic contacts with non-immunoreactive dendritic shafts and spines.

Molecular cloning of rat mitochondrial glutamic oxaloacetic transaminase mRNA and regulation of its expression in regenerating liver.

cDNA clones for rat mitochondrial glutamic oxaloacetic transaminase (mGOT) have been isolated from a rat liver cDNA library. One of the clones, designated p501, contained a cDNA insert of 1.4 kilobase pairs in length and hybridized to a mRNA of 2.4 kilobases from rat liver. We measured mGOT mRNA content in a regenerating rat liver. In a regenerating rat liver, mGOT activity was increased and reached maximum (170% of control activity) at about 48 h following the operation. Using the cDNA of mGOT, it was revealed that the increase of mGOT in the regenerating rat liver depended on its mRNA content.

Adenosine deaminase and histidine decarboxylase coexist in certain neurons of the rat brain.

The enzymes adenosine deaminase (ADA) and histidine decarboxylase (HDC) were immunocytochemically detected in rat brain. The gross distributions of ADA- and HDC-immunoreactive neurons in the basal hypothalamus were very similar. The superficial layers of the superior colliculus showed only ADA-containing neurons. Using adjacent thin-sections of basal hypothalamus, stained alternately for ADA and HDC immunoreactivity, it was possible to show the two labels localized within the same neurons. These observations imply a relationship between two neurochemically distinct putative neurotransmitter/modulator systems, that of histamine and adenosine.

Immunohistochemical demonstration of histamine N-methyltransferase-like structures in rat kidney.

Histamine N-methyltransferase (S-adenosylmethionine: histamine N-methyltransferase, E.C. 2.1.1.8) was purified to homogeneity from rat kidney, and antibody was raised against it in guinea pigs. The antibody immunoprecipitated histamine N-methyltransferase. Immunofluorescent histochemical studies with anti-histamine N-methyltransferase antibody as the first antibody and goat anti-guinea pig IgG conjugated with fluorescein isothiocyanate as the second, showed the presence of immunoreactive structures in the proximal tubules of rat kidney. The brain showed no immunoreaction with the antibody.

Unruptured intracranial aneurysms in polycystic kidney disease.

To detect unruptured intracranial aneurysms, we performed cerebral angiography in five patients with polycystic kidney disease (PKD) who had no neurological deficits and no history of subarachnoid haemorrhage. Three of the five patients had unruptured intracranial aneurysms and two underwent surgery with no mortality or morbidity. Our review of the literature revealed that the surgical risk of unruptured intracranial aneurysms is smaller than the risk of bleeding in conservatively treated patients. We discuss the importance of an early diagnosis, and early operation for unruptured aneurysms in patients with polycystic kidney disease and stress the need for intensive care for their renal dysfunction and hypertension during and after the operation.
