[Significance of postoperative substitution of plasma proteins].

In the course of a pilot study 30 patients, who had to undergo extensive urological-abdominal surgery, were randomized and divided into two groups. One group received 500 ml of the stored serum Biseko on the day of operation as well as on the first postoperative day, the other group was given a placebo. The two groups were compared with regard to their complications rates (wound healing disturbance, respiratory disturbance on account of septicemia). It turned out, that there occurred clearly less complications in the group treated with Biseko than in the control group.

[In vivo study of the effects of i.v. immunoglobulin therapy. Tissue distribution of native and enzyme-treated human immunoglobulin fractions given i.v. in man].

The tissue distribution of intravenously injected native and enzyme-treated human gammaglobulin has been investigated in human rectum mucosa. FITC-labelled antibodies to human gammaglobulin were used to locate the injected antibody preparation. Three hours after application of these different gammaglobulin-preparations no enrichment in the mucosa was found. Neither the native gammaglobulin nor the enzyme-treated gammaglobulin showed any concentration in the lamina propria of the large bowel mucosa or an intracellular enrichment in the epithel. This study shows that the quicker elimination of enzyme-treated gamma-globulin from the circulation cannot be explained by an intracellular increase.

PAL-test for malignant disease.

Peripheral blood lymphocytes from a total of 300 subjects including normal controls and patients with malignant and non-malignant disorders were investigated to determine their ability to agglutinate with Poly-l-lysine 3400 (PAL-test). A high level of association between clinically evident cancer and a positive reaction was confirmed. Lymphocytes from 72 of 90 patients with malignant diseases as well as 33 of 140 patients with non-malignant processes showed positive reactions. All healthy controls were negative. The positive evidence of the PAL-test amounted to over 80%. The results and possible mechanisms of the PAL-test are discussed.

[Immunologic potency of pregnancy associated alpha 2-glycoprotein (alpha 2-PAG)].

The suggestion that the pregnancy-associated alpha2-glycoprotein could depress the immune response was investigated using the modified electrophoretic mobility test. The presence of the glycoprotein, at physiological levels, significantly reduced the inhibition of the migration velocity of indicator cells in the electric field caused by the MSF. In the contrary alpha2-PAG-free serum had no effect in the test system. Therefore, it can be supposed that alpha2-PAG may have an immunoregulatory function within the immunological mechanism which protects the conceptus and malignant tumor.

[Pregnancy associated alpha2-glycoprotein (alpha2-PAG) in the serum of patients with gynecologic carcinomas].

alpha2-PAG is a high-molecular, carbohydrate-containing glycoprotein. It occurs in traces in normal human serum. The variations in serum levels of this protein were studied during treatment of 32 gynecological cancer patients in the course of 2 years. The periodic alpha2-PAG determinations were done with an electro-immuno-assay. A correspondence between alpha2-PAG serum levels and the clinical course could be found in 60%. In 5 of 13 patients who did not respond to therapy alpha2-PAG levels rose, additional 4 had unchanged high levels, whereas 13 of 17 patients had significantly decreased levels on successful treatment. The relevance of periodic alpha2-PAG determinations is discussed.

[Diagnosis of malignant processes of the colon and rectum using a new method for the demonstration of sensitized lymphocytes (PAL-test)].

Peripheral blood lymphocytes of 45 patients with colon-rectum disease and 30 healthy controls were tested in a simple rapid microagglutination test (PAL test) for cancer diagnosis. In 30 cancer patients 28 positive reactions were obtained. All healthy controls were negative. The 15 patients with benign or premalignant lesions showed 5 positive and 10 negative reactions. Results of the PAL test were about 90% accurate.

Detection of sensitized human blood lymphocytes by agglutination with basic peptides: a possible test for malignant disease.

A simple rapid micro-agglutination test for detecting sensitized lymphocytes from human peripheral blood is described: the poly-L-lysine (PLL) 3400 agglutination test of lymphocytes (PAL test). The incubation of lymphocytes from 30 cancer patients, 30 patients with non-malignant disease and 40 healthy controls with PLL (mol. wt 3400) is evaluated. The test was positive in 83 percent of malignant and 20 percent of non-malignant cases. All healthy controls were negative. Other peptides tested showed no significant difference in reaction between malignant and non-malignant diseases. The mechanism of detection of sensitized lymphocytes from patients with malignant disease through agglutination induced by PLL 3400 is discussed.

[Correlation of alpha-2 pregnancy associated glycoprotein with the clinical course of bronchial carcinoma (author's transl)].

Alpha-2 pregnancy associated glycoprotein is a high molecular, carbohydrate containing plasma protein with unknown biological function. During clinical treatment of 25 patients with bronchial carcinoma alpha-2 PAG was measured by the electroimmuno-diffusion method according to Laurell. A good correlation was observed between alpha-2 PAG serum levels and the course of the disease in 21 patients. Patients whose course of disease could not be influenced therapeutically showed continously increasing levels of alpha-2 PAG; patients who responded to therapy, showed a significant decrease in alpha-2 PAG serum levels. These findings indicate that periodic determinations of serum alpha-2 PAG may be helpful for controlling the course of bronchial carcinoma.

Carcinoembryonic antigen, alpha 1-fetoprotein, ferritin, and alpha 2-pregnancy associated glycoprotein in the serum of lung cancer patients and its demonstration in lung tumor tissues.

Levels of carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP), ferritin and alpha 2-pregency associated glycoprotein (alpha-2-PAG) were determined in patients with confirmed lung cancer at the time of diagnosis and in serial determinations during and after radio- or chemotherapy. Whereas AFP levels were not elevated in patients with lung cancer, increased levels of CEA, ferritin and alpha-2-PAG were found in more than 50% of the patients. The results suggest that determination of CEA, ferritin and alpha-2-PAG in the serum of patients with lung cancer may be useful to detect metastases or recurrences and to monitor the results of treatment. Furthermore, in this study CEA and ferritin could be demonstrated in extracts of lung tumor tissues by specific antisera.

[Serologic breast cancer follow-up monitoring using pregnancy-associated alpha-2-glycoproteins].

The variations in serum level of the alpha 2-PAG (pregnancy associated glycoprotein) were studied during the treatment of 25 patients with breast cancer. A good correlation was found between alpha 2-PAG concentration and the course of the disease. Serum alpha 2-Pag levels rose prior to the clinical recognition of metastatic disease and decreased significantly on successful treatment. alpha 2-PAG appears to have potential as an indicator of the growth of micrometastases.
