Pharmacokinetics and efficacy of amoxycillin in the treatment of an experimental Streptococcus bovis infection in racing pigeons (Columba livia).

The pharmacokinetics of amoxycillin in pigeons (Columba livia) and the efficacy of amoxycillin treatment in pigeon streptococcosis, were investigated. After intravenous administration of 150 mg.kg-1 amoxycillin, the drug plasma concentration-time profile fitted an open two-compartment model. Amoxycillin was quickly cleared from the circulation with a mean half-life of 66 min and showed a distribution volume of 0.9 L.kg-1. The absolute oral bioavailability of amoxycillin was 50.1%. A 5-day drinking water treatment with amoxycillin (1500 mg.L-1, corresponding to 112.0 mg.kg-1.day-1 during day-light hours) significantly reduced the postinoculation morbidity in pigeons experimentally infected with Streptococcus bovis, compared to untreated infected controls.

Primary sinonasal mucosal melanoma: three different therapeutic approaches to inoperable local disease or recurrence and a review of the literature.

Sinonasal melanoma is an uncommon disease with a bad prognosis and a high local recurrence rate. The use of radiotherapy in such conditions remains controversial. A review of the literature is presented in an attempt to answer some of the questions regarding therapeutic options. Data on incidence, aetiology, pathology and prognosis are also provided. Case studies are reported of three patients with sinonasal melanoma referred to our department in a relatively short time period and needing radiotherapy as a single treatment modality, using a conformal three-dimensional treatment technique. One patient was also treated with temporary 1251 seeds, while another was treated using intensity modulation. Both intensity modulation and temporary 1251 seeds are feasible techniques, providing satisfactory dose distributions encompassing the tumour volume while sparing critical structures. Surgery remains the treatment of choice for sinonasal melanoma. Radiotherapy should be used postoperatively and is a good alternative in cases of inoperable disease. Adjacent critical structures limit the radiation dose to the tumour area, especially when high fraction doses are used. Therefore, beam intensity modulation and 1251 seeds can be used to increase the tumour dose without exceeding the radiation tolerance of the surrounding structures.

Administration of doxycycline hydrochloride via drinking water to turkeys under laboratory and field conditions.

A series of experiments were carried out in order to determine doxycycline hydrochloride (DoxHCl) plasma levels in 6-wk-old turkeys medicated via drinking water containing DoxHCl at a concentration of 250 mg/L under laboratory and field conditions. Maximal plasma concentration (Cmax) values of 5.7 (+/-1.0) microgram/mL and 4.9 (+/-1.4) micrograms/mL obtained after DoxHCl administration during 2 and 7 d, respectively, were not significantly different. A significant difference was found between the area under the plasma concentration-time profile, calculated between 0 and 168 h (AUC(0-168)), Cmax, and the minimal plasma concentration (Cmin) values obtained after medication with a DoxHCl solution at a concentration of 250 mg/L (431.9 +/- 96.6 micrograms.h/mL, 4.9 +/- 1.4 micrograms/mL and 0.7 +/- 0.3 microgram/mL) and after medication with a DoxHCl solution at a concentration of 750 mg/L (1,176.5 +/- 201.8 micrograms.h/mL, 12.5 +/- 2.7 micrograms/mL and 2.9 +/- 0.4 micrograms/mL), respectively. The increase in body weight was also significantly higher for turkeys medicated with a DoxHCl solution at a concentration of 750 mg/L (83.7 g/d) than for the lower concentration (35.6 g/d). The DoxHCl solution uptake significantly decreased with the increase of DoxHCl concentration. A Cmax value of 1.7 +/- 0.6 micrograms/mL and a Cmin value of 0.5 +/- 0.1 microgram/mL were observed during the field experiment. Water consumption under laboratory conditions was followed for tap water (70 +/- 50 mL/kg.d) and for a DoxHCl solution at a concentration of 250 mg/L supplemented with 1 g anhydrous citric acid/L (119 +/- 6 mL/kg.d) and revealed to be not significantly different. The variability was significantly higher for tap water than for the DoxHCl solution. The stability of the DoxHCl solution containing 1 g citric acid/L over 24 h was 99% expressed as the percentage of the initial concentration.

Validation of a high-performance liquid chromatographic method for the determination of doxycycline in turkey plasma.

A high-performance liquid chromatographic method for the analysis of doxycycline in turkey plasma samples using demeclocycline hydrochloride as the internal standard was developed, optimized and validated. A one-step extraction procedure and an isocratic HPLC method with UV detection were used. No interferences with endogenous compounds or with the anticoagulant were observed. Linear calibration curves (r2 > 0.99) were obtained in water and plasma between 0 and 600 micrograms ml-1. Good recoveries for doxycycline (> 66%) and demeclocycline (> 72%) were seen both in water and in plasma. The coefficient of variation was < 9.86% for within-day reproducibility and < 7.53% for the between-day reproducibility. The deviation between the mean value found and the true value was < 14.5% (accuracy). The limit of detection was 0.1 microgram ml-1 in plasma samples. A good stability of doxycycline was observed in water and in plasma samples after storage for six months at -20 degrees C (recovery > 91%).

Pharmacokinetics and anti-trichomonal efficacy of a dimetridazole tablet and water-soluble powder in homing pigeons (Columba livia).

The anti-trichomonal efficacy and pharmacokinetics of dimetridazole were investigated in the homing pigeon (Columba livia). Dimetridazole was formulated for drinking water medication and as a prolonged-release tablet. To suppress a Trichomonas gallinae infection successfully, medicated drinking water containing dimetridazole (400 mg/L) had to be administered for at least 3 days. A two-day treatment with a dimetridazole tablet (20 mg/tablet) in fasted, as well as in fed, pigeons was shown to be ineffective. After intravenous administration of 20 mg dimetridazole, the drug plasma concentration-time profile fitted a one-compartment open model with a mean half-life of 3.9 h. The absolute bioavailability of the tablet in fasted pigeons was 83.8%. The bioavailability of the tablet administered with food was reduced by 20%. Dimetridazole was rapidly metabolised to (1-methyl-5-nitroimidazol-2-yl) methanol.

Pharmacokinetics and bioavailability of doxycycline in turkeys.

The pharmacokinetic parameters of doxycycline (DOX) were determined in 3 day, 3-, 6- and 12-week-old fasted turkeys, after a single intravenous (i.v.) dose of 25 mg doxycycline. HCl/kg body weight. Doxycycline disposition fitted an open two-compartment model. The mean (+/-SD) elimination half-life was 10.6 +/- 0.7, 10.8 +/- 1.5, 7.9 +/- 1.4 and 10.0 +/- 0.9 h in 3 day, 3-, 6- and 12-week-old turkeys, respectively. Mean (+/- SD) total body clearance was 0.19 (+/-0.01), 0.27 (+/-0.03), 0.11 (+/-0.03) and 0.06 (+/-0.01) L/h.kg in 3 day, 3-, 6- and 12-week-old turkeys, respectively. The steady-state volume of distribution was 1.77 (+/-0.2), 2.1 (+/-0.2), 0.7 (+/-0.4) and 0.5 (+/-0.2) L/kg in turkeys of the above mentioned ages, respectively. The AUC value significantly increased with the age of the turkeys. An oral doxycycline solution at a single dose of 25 mg/kg of body weight was administered to 3 day. 3-, 6- and 12-week-old turkeys. The maximal plasma concentrations in fasted turkeys were 3.8, 5.6, 7.4 and 5.7 micrograms/mL. with tmax values of 4.7, 1.5, 2.8 and 5.4 h, for the different ages, respectively. In fed turkeys the Cmax values were 2.5, 6.1, 4.8 and 3.0 micrograms/mL. with tmax values of 4.2, 5.3, 4.5 and 7.5 h, respectively. The absolute bioavailability in fasted turkeys varied between 25.0 +/- 9.0% (for 12-week-old birds) and 63.5 +/- 7.1% (for 3-week-old birds). The relative bioavailability varied between 40.0 +/- 13.0% (for 12-week-old birds) and 83.7 +/- 14.3% (for 3-week-old birds).

Evaluation of the nebulisation of sodium ceftiofur in the treatment of experimental Pasteurella haemolytica bronchopneumonia in calves.

Severe acute bronchopneumonia was induced in 24 conventional Friesian Holstein calves by inoculating them intratracheally with Pasteurella haemolytica type A1. Twelve of the calves were treated intramuscularly with sodium ceftiofur and 12 were treated with an aerosol of sodium ceftiofur. The mortality rate in the group of calves treated with the aerosol was significantly lower, and their clinical and haematological parameters returned to normal significantly faster than in the calves treated intramuscularly.

Chondrosarcoma of the larynx. A diagnostic and therapeutic dilemma.

Chondrosarcoma of the larynx. A diagnostic and therapeutic dilemma. Chondromas and chondrosarcomas of the larynx are very rare. Differential diagnosis between benign chondromas and the different degrees of malignancy in chondrosarcomas is difficult. Behaviour of chondromatous tumors of the larynx differs from those of the overall head and neck region. A case of chondrosarcoma of the larynx is described. A literature review regarding chondro(sarco)mas of the larynx and the head and neck region is given presented.

Effect of adjuvant administration on weight gain, infectious diseases, and mortality of veal calves.

Young weaned calves are susceptible to viral and bacterial infections. In two studies 279 and 500 weaned calves were injected at random with an adjuvant, with saline or were left untreated in order to evaluate their weight gain or illness and mortality during the first 10 weeks of fattening on a fat-stock farm. However, in contrast to studies reported in the literature, no significantly higher increase in weight gain and no significant improvement of illness and mortality were found with adjuvant treatment.

Pharmacokinetics and anthelmintic efficacy of febantel in the racing pigeon (Columba livia).

This paper reports on the pharmacokinetics and the efficacy of febantel against Capillaria obsignata and Ascaridia columbae in the racing pigeon. Febantel was rapidly cleared from the circulation and highly metabolized. The efficacy of febantel against Capillaria and Ascaridia was studied on lightly and heavily infected pigeons. The efficacy against Ascaridia was 100% for the faecal egg count reduction (FECR) as well as for the worm reduction in all treated pigeons. After two treatments with febantel against Capillaria in heavily infected pigeons the FECR amounted to 96.7% and worm reduction was 95%. In order to effectively remove both parasites from the host, repeated treatments with febantel at a short interval could be the treatment of choice.

Pharmacokinetics and bioavailability of ronidazole from a prolonged release tablet in the homing pigeon (Columba livia).

The pharmacokinetics of ronidazole and the bioavailability of a prolonged release tablet were studied in the homing pigeon. After intravenous administration of 5 mg ronidazole, the drug plasma concentration profile fitted a one-compartment open model. The mean half-life of the drug was 11 h and the volume of distribution was 0.86 l/kg. Total body clearance was 0.056 l/h/kg. A sustained release matrix tablet exhibited prolonged drug release in vitro. After oral administration of the matrix tablet to pigeons drug absorption was nearly complete. When given on an empty stomach, the tablet failed as a prolonged release system. Administration to previously fed pigeons resulted in an increase in tmax and a decrease in Cpmax.

Anesthesia for foreign bodies in the tracheo-bronchial tree in children.

The authors present the anesthetic and ventilation techniques, used in 106 children, who were suspected of foreign body aspiration in the respiratory tract. In 62 children a foreign body was found. The youngest child was 8 months old and the oldest 13 years, with an age distribution peak in the 1 to 2 years age group. A predominance for the male sex (60%) was present. Foreign bodies of organic nature were found most frequently (80%), 39 of them consisting of peanuts. The bronchi were involved more often than the trachea and the foreign body was located more frequently at the right bronchus (38 pt). The children were ventilated initially with an intermittent oxygen jet injection technique, using a home made apparatus, but since 1978 with HFPPV, using the AGA Bronchovent. Induction of anesthesia was done with halothane and maintenance with etomidate infusion (10-20 micrograms/kg/min.) or thiopental increments (2 to 3 mg/kg). The technique so far used, proved to be satisfactory, specially since HFPPV is used. Few complications occurred. One child died during the bronchoscopic procedure and in an other child a tracheostomy had to be performed for extraction of the foreign body.
