[Tuberculosis of the contralateral adrenal gland: a case report].

Unilateral adrenal tuberculosis is a very rare disease. A 66-year-old woman presented with epigastric discomfort and general fatigue. Abdominal CT scan revealed a homogeneous mass shadow in the right adrenal region. Findings of physical examination were normal except that the patient was obese. Hormonal data were in normal range. Adrenal scintiscanning demonstrated no RI uptake in the right adrenal gland. Right adrenalectomy was performed under the diagnosis of nonfunctioning tumor of the right adrenal gland. Histopathological examination, however, revealed typical tuberculosis with Langhans' type of giant cells and infiltrated lymphocytes. Of 322, 148 autopsies performed during the twelve years between 1970 and 1981 in Japan, 228 cases of adrenal tuberculosis were recognized. Furthermore, only 18 cases had tuberculous regions in the adrenal gland alone.

[The metastatic patterns of gastric cancer from its histopathological characteristics in the primary lesion--particularly in relation to peritoneal dissemination and liver metastases].

The purpose of this study was to catagorize the metastatic patterns of advanced gastric cancer into liver and peritoneal metastases from the stromal behavior of the primary lesion in 278 patients. According to our data, peritoneal disseminations were encountered 5.0% of the medullary type, 30.0% of the intermediate type and 38.0% of the scirrhous type, while the corresponding figures for liver metastases were 28.8%, 6.0% and 1.9%, respectively. The differences among the three types were significant with both patterns of metastasis. From the present data, we assume that the histological pattern in the stroma of primary gastric cancer shows the biological behavior of cancer growth and metastatic patterns, particularly in close relation to peritoneal dissemination and liver metastases.

Adrenaline involvement in the presynaptic beta-adrenoceptor-mediated mechanism of dopamine release from slices of the rat hypothalamus.

Slices of rat hypothalamus were superfused and endogenous release of dopamine (DA) was measured by high performance liquid chromatography combined with electrochemical detection. The K+ (20 mM)-evoked release in the presence of tetrodotoxin was Ca2+-dependent. The evoked release was facilitated by a beta-agonist, isoproterenol and this effect was completely abolished by a beta-antagonist, 1-propranolol. Isoproterenol also concentration-dependently facilitated the electrically (at 5Hz) evoked release of DA. The pretreatment with 1-propranolol, beta 1-antagonist, atenolol and beta 2-antagonist, butoxamine shifted the concentration-effect curve of isoproterenol to the right. On the other hand, beta 1-agonist, tazolol, beta 2-agonist, salbutamol and low concentration (10(-9) M) of adrenaline also facilitated the release. 1-Propranolol alone reduced the electrically (at 2 Hz) evoked release, and this effect was completely abolished when the adrenaline content in the brain was drastically reduced by use of a potent PNMT inhibitor, DCMB. These findings suggest that in the rat hypothalamus adrenaline released from adrenaline-containing nerve terminals probably modulates DA release via presynaptic beta 1- and beta 2-adrenoceptors on DA nerve terminals.

The protein A-gold technique. Use in the immunoelectron microscopic study of the teleost growth hormone.

By the application of the protein A-gold technique for immunocytochemistry of the tilapia pituitary growth hormone (GH), our electron micrographs revealed that the antigenicity of tilapia GH and the cytoplasmic structure were well preserved in the tissues fixed with periodate-lysine-paraformaldehyde followed by osmium tetroxide postfixation. Using the specific antiserum against tilapia GH, the immunoreactive gold particles of 20-40 nm in size were located on secretory granules (200-300 nm in diameter) exclusively of one cell type in the proximal pars distalis identified as GH cells on the basis of their ultrastructural characteristics. This technique is considered to be fairly efficient for the detection of intracellular antigenic sites on ultrathin sections.

Changes in serum concentrations of steroid hormones, thyroxine, and vitellogenin during spawning migration the chum salmon, Oncorhynchus keta.

Changes in serum concentrations of various steroid hormones (estradiol-17 beta, androgens, 17 alpha,20 beta-dihydroxy-4-pregnen-3-one (17 alpha,20 beta-diOHprog], thyroxine, and vitellogenin during the spawning migration (from the coastal sea to the spawning ground) of male and female chum salmon (Oncorhynchus keta) were investigated. In females, estradiol-17 beta levels were high during the early phases of the migration (sea and prespawning period), but significantly decreased in fish at the time of the spawning period (oocyte maturation and ovulation). The changes in estradiol-17 beta levels coincided with those of of serum vitellogenin levels. High levels of androgens were observed throughout the sampling period. Serum 17 alpha, 20 beta-diOHprog levels were extremely low during the early phases of spawning migration, but elevated dramatically in mature or ovulating females. In males, serum concentrations of androgens were high during the early phases of the migration, but sharply declined around the time of the spawning period (spermiation). Similar to females during final maturation, the serum levels of 17 alpha,20 beta-diOHprog in males rapidly increased during spermiation, although the magnitude of the elevation in males was smaller than in females. Serum thyroxine levels were highest both in females and males collected in the coastal sea, and the levels decreased during the spawning migration. These findings are discussed in relation to anadromous migration and sexual maturation of chum salmon.

Sequence polymorphisms in the 5'-upstream region of the fibroin H-chain gene in the silkworm, Bombyx mori.

DNA fragments containing the fibroin H-chain gene from two different strains of Bombyx mori, J-139 and Nd (2) were cloned into phage lambda Charon 4A. Comparison of the restriction sites in these cloned DNAs revealed that in addition to the known polymorphism in the region coding for the repetitive amino acid sequence of the fibroin H-chain [Manning and Gage, J. Biol. Chem. 255 (1980) 9451-9457], at least two other types of polymorphism were present, one around the 5' end of the structural gene, and the other in the far upstream region of the gene. Restriction sites around the 5' end of the gene were well conserved between these strains, but some heterogeneity, suggesting the presence of small insertions, deletions or base changes, was noted. In contrast, DNA sequences of the region 2-4 kb upstream from the 5' end of the gene were markedly different between these two strains, indicating that either a deletion or an insertion of a DNA sequence longer than 2 kb had occurred in this region. Comparison with several other strains suggested that the observed changes in the far-upstream region were unique to the Nd(2) strain.

Histamine disposition in endothelial specific granules of the toad aorta.

High-performance liquid chromatography revealed that toad aorta delivered appreciable concentrations of histamine into the perfusate when perfused by Ringer's solution containing the histamine liberator. Electron micrographs of this vessel after the perfusion showed an expulsion of the endothelial specific granules into the vascular lumen. These results support to our previous hypothesis that the granules are a reservoir site of histamine and might take an important role in the regulation of vascular tone.

Hydrolytic deactivation of kyotorphin by the rodent brain homogenates and sera.

A simple analytical assay method for kyotorphin was developed by the use of amino acid analyzer and the deactivation of kyotorphin in biological media was studied with this method. Kyotorphin was hydrolyzed rapidly by enzymes in serum and brain of rat and mouse, but L-tyrosyl-D-arginine was not. The hydrolysis activity was much higher in brain than in serum. The apparent Michaelis' constants, Km, were of the order of 10(-5) for brain and 10(-4) M for serum in both species. The hydrolysis of kyotorphin was strongly inhibited by bestatin (Ki = 0.1 microM). Thus the hydrolysis might be attributed to aminopeptidase(s). A dose of 50 micrograms of bestatin administered intracisternally potentiated the analgesic activity of kyotorphin by 4.8 times or so. Thus kyotorphin seems to be deactivated by bestatin-sensitive aminopeptidase(s) in brain.

[Bilateral primary germ cell tumors of the testis: report of two cases and review of the literature].

Bilateral germ cell tumors of the testis are not common. The contralateral tumor may develop simultaneously or sequentially. In both cases, bilateral involvement is postulated to represent two primary tumors. Case 1: A 47-year-old male was seen with a painless nodular mass in the right testis. Physical examination revealed hard swelling of both testes besides a nodule in the right testis. He underwent bilateral radical orchiectomy; and, microscopical both tumors proved to be seminoma. Although bipedal lymphangiography was negative for nodal metastases, postoperative irradiation to the pelvic and para-aortic lymph nodes was given to a total dose of 5,800 rad. The patient was alive 15 years after treatment for bilateral testicular tumors. Case 2: A 53-year-old male presented with painless left scrotal swelling. Physical examination showed not only an enlarged hard testis in the left scrotum but also a palpable hard mass in the right testis. Bilateral radical orchiectomy was performed and pathological examination revealed pure seminoma in both testes; a 1 cm mass in the right testis and almost completely replaced tumor in the left testis. Evaluation including retroperitoneal lymph node dissection revealed no metastatic disease. Postoperatively, the patient received 1,900 rad of irradiation to the retroperitoneal space. He is well one year after surgery and has no evidence of recurrence or metastasis. Previous reports of bilateral germ cell testicular tumors are reviewed and the incidence, age, predisposing factors, interval between the first and second tumors, histology, pathogenesis, prognosis, and management are discussed.

[AFP producing stomach cancer--a study of transplanted tumor in nude mice].

AFP producing cancer of the stomach was divided into 2 groups; Group A demonstrated a marked elevation of serum AFP levels before gastrectomy and was positive for AFP by PAP staining; group B showed delayed elevation of AFP levels at recurrence or in the terminal stage. Experimentally, we could establish three cell lines from group A and two lines from group B from transplanted tumor in nude mice. Tumor growth, changes of serum AFP levels and PAP staining investigated in each group. Group A and B were not only thought to exhibit different biological behavior, but also different mechanisms in AFP production. Group A cells may have the ability to secrete high levels of AFP; this may be a property of these tumor cells. On the other hand, group B tumor cells initially have little ability to secrete AFP. However, they may transform into AFP producing cells as a phenomenon of cell differentiation.
