Prevalence of antibody to human calicivirus in general population of northern Japan.

Serum specimens from children and adults living in Saporo, Japan, were tested for antibody against human calicivirus by immune electron microscopy (IEM), using virus-rich faecal extracts as the source of antigen. Of 83 serum specimens tested, 49 (59%) were positive for calicivirus antibody. Age-related prevalence of antibody to calicivirus was as follows: 23% (3/13) in the 0-5-month-old group, 30% (6/20) in the 6-23-month-old group, 65% (13/20) in the 2-5-year-old group, and 90% in school children (18/20) and adults (9/10). As for IEM antibody ratings scored from 0 to 4, almost all positive sera from older infants and preschool children scored 3 to 4. Antibody scores were rather more scattered in school children. The results indicated that caliciviral infection is prevalent in younger children in this part of Japan.

The 33- to 39-nm virus-like particles, tentatively designed as Sapporo agent, associated with an outbreak of acute gastroenteritis.

An outbreak of nonbacterial gastroenteritis occurred in an orphanage in December 1979. Of 54 residents, 43 were affected with acute gastroenteritis. Diarrhea was the commonest symptom. The 33- to 39-nm virus-like particles, tentatively designated as Sapporo agent, were detected in 17 (48.6%) of the patients' 35 fecal specimens. Morphologically, it was difficult to distinguish the Sapporo agent from other agents without geometrical pattern of the surface. Antigenically, the Sapporo agent related to the Otofuke agent detected in an outbreak of the institution for mentally retarded adults and also to the SRV ;76 detected in familial cases of infectious gastroenteritis. The Sapporo agent might belong to a new group of those gastroenteritis viruses which were larger in size than the Norwalk virus group.

Effects of prostaglandin on experimental bone malignancy and on scintigrams of bone and marrow.

The correlation between prostaglandin E (PgE) and scintigrams of bone (Tc-99m MDP) and bone marrow (Tc-99m SC) was investigated in normal and VX-2-bearing rabbits. PgE in plasma of normal rabbits was 486.2 +/- 185.7 pg/ml (n = 86) and the maximum-to-minimum (max/min) ratio was 1.85 +/- 0.26 at 4 wk after tumor implantation. In rabbits with VX-2 transplanted into femoral muscles, PgE was in the normal range unless the tumor invaded bone. PgE did not increase significantly in rabbits when the tumor was transplanted into the marrow cavity. When tumor invaded bone, PgE increased markedly (to 1335 +/- 584 pg/ml). Elevation of PgE did not necessarily coincide with the appearance of positive bone scans. PgE in an indomethacin-treated group was not higher than in the untreated group. There was no significant difference between the two groups regarding the time of appearance of abnormal bone scans. However, when the number of transplanted cells in the bone marrow was reduced, the treatment with indomethacin delayed the increase in tracer uptake in the affected bone and resulted in a photon-deficient area. Indomethacin may suppress the local acceleration of calcium metabolism.

Combined treatment of syngeneic murine tumors and xenotransplanted human lung cancer by immunotherapy and radiotherapy.

The synergistic effect of nonspecific immunotherapy with cell-wall skeleton of BCG on radiotherapy against two syngeneic murine tumors, a methylcholanthrene-induced tumor (MCA) and a spontaneous well-differentiated mammary adenocarcinoma (Br-1), was studied in (+/+) balb/c mice and (nu/nu) mice of BALB/c background. Single irradiation of tumors with a dose of 2000 rad induced complete shrinkage in about 18% of MCA and Br-1 tumors in (+/+) mice. Single irradiation did not induce complete shrinkage of tumors in (nu/nu) mice. When immunotherapy was combined with radiotherapy, the rates of complete shrinkage of MCA and Br-1 tumors increased to 82 and 61%, respectively. In contrast, such a strong synergistic effect was not observed in (nu/nu) mice. Moreover, human lung cancers (two squamous cell carcinomas and two small cell carcinomas) xenotransplanted to nude mice were treated with the combined therapy. The effect was stronger on squamous cell carcinomas than on small cell carcinomas.

Experimental and clinical studies on differential diagnosis of bone diseases with nucleomedical procedures.

This report is a study of the differential diagnosis of bone diseases using nucleomedical procedures. Experimental and clinical studies on the accumulation of 201Tl and 67Ga in bone inflammation and bone tumors were performed. Also, the degree of deposition of 99mTc-phosphorous compounds represented as a color difference was evaluated for a differentiation between malignant and benign diseases of the spine and shoulder. Uptake of 201Tl in inflammation of the spine and in tumor of the pelvis of rabbits was very low, while that of 67Ga was high. Uptake of 67Ga decreased gradually with lapse of time after the onset of inflammation. These experimental results agreed with clinical studies. The detectability of malignant tumors with 201Tl was inferior to that with 99mTc-label and 67Ga, although osteolytic lesions were occasionally 201Tl-positive. In terms of color range, the difference was 2 or less in spines of normal subjects and 3 or more in the metastatic bone disease and compression fracture. This difference was not statistically significant. Since 67Ga scan was occasionally negative in compression fracture, a differentiation was considered to be possible in such a case. A combined study of bone imaging with 99mTc-phosphorous compound including color difference, 67Ga imaging, bone-marrow imaging and radiography are considered to be helpful for the differentiation.

Fecal shedding of virus in relation to the days of illness in infantile gastroenteritis due to calicivirus.

Fecal shedding of virus in relation to the days of illness was studied by electron microscopic examinations of stool specimens collected during two consecutive outbreaks of gastroenteritis associated with calicivirus in an orphanage in the city of Sapporo, Japan. Of 61 stool specimens examined, 29 (48%) were found to contain typical calicivirus particles. Although caliciviruses were found in none of the seven stools obtained by chance before the onset of illness, they were found in 18 (95%) of 19 stool specimens collected within four days after the onset of illness. Seven (50%) of 14 specimens collected during the next five days were virus-positive, and the viruses were rarely detected in the stools collected thereafter. Thus correlation between viral shedding and the days of illness was clearly demonstrated. This finding should provide additional evidence for the etiologic role of calicivirus in acute infantile gastroenteritis.

Effect of radiotherapy on lymphocyte cytotoxicity against allogeneic lung cancer cells in patients with bronchogenic carcinoma.

Cytotoxicity of peripheral blood lymphocytes against allogeneic target cells of bronchogenic carcinoma was examined by a microcytotoxicity test before, during, and after radiotherapy in primary lung cancer patients. Before the treatment, cytotoxicity was depressed only slightly in patients in stage III and strikingly in those in stage IV, as compared to the values in patients at earlier stages of lung cancer such as stages I AND II. Local irradiation scarcely affected cytotoxicity at stages II and III, but augmented remarkably at stage IV. The number of peripheral blood lymphocytes decreased profoundly during and after radiotherapy in all cases of stages II, III, and IV. Although radiotherapy exhibited various effects on the cytotoxic activity of lymphocytes and the number of peripheral blood lymphocytes, only the cytotoxic activity at the end of radiotherapy correlated well with the reduction in tumor size.
