Effects of nonsteroidal antiinflammatory drugs on the proton exchange reaction between octanol and water: its correlation with their inhibitions of inflammation and prostaglandin synthesis.

Effect of various antiinflammatory drugs on proton exchange between octanol OH and water after partition equilibrium between octanol and water at pH 7.4 was examined. These drugs accelerated the proton exchange reaction to different extents. The rate constant of the proton exchange induced by the drugs was well correlated with their biological potencies in inhibiting Carrageenan-induced edema and prostaglandin synthesis, suggesting that proton or electron movement in an apolar environment is very important for exhibition of these biological effects.

A cyanine dye tri-S-C7(5). Phosphate-dependent cationic uncoupler of oxidative phosphorylation in mitochondria.

The trinuclear cyanine dye, tri-S-C7(5), at about 10 microM stimulated State 4 respiration of rat liver mitochondria more than 6-fold and released oligomycin-inhibited respiration completely. Thus, the dye is concluded to be a very effective cationic uncoupler of oxidative phosphorylation in mitochondria. However, for exhibition of its uncoupling action, the presence of Pi (or arsenate) was necessary, and a phosphate-transport inhibitor, N-ethylmaleimide or mersalyl, inhibited its action. The stimulation of phosphate transport via the Pi carrier by the dye is suggested to be directly related to the uncoupling action.

Severe hepatitis during ketoconazole therapy.

Ketoconazole is an imidazole derivative recently developed as an antifungal agent. There have been only a few established cases of hepatotoxicity in the literature. This report describes a case of presumed ketoconazole hepatotoxicity characterized by the development of severe hepatitis with marked centrilobular necrosis. The lack of hypersensitivity reactions in clinical findings and centrilobular location of necrosis suggested a host idiosyncrasy with metabolic abnormality as the effect of ketoconazole in the present case.

Labelled-calcium release from rat mandibles exposed to prostaglandins in vitro.

Bone resorptive effects of prostaglandins (PGs) in cultured mandibular bone tissue of rat fetus prelabelled with 45Ca were examined in comparison with those in radius-ulna. PGE1 (3 X 10(-6)-10(-4) M) and PGE2 (3 X 10(-8)-10(-4) M) could significantly stimulate the release of 45Ca from mandible with a peak effect at 3 X 10(-6) M. The effects of PGEs were more marked in mandible than in radius-ulna. Salmon calcitonin (SCT) inhibited the stimulating effect of PGE2. PGF 2 alpha did not cause the significant increase of 45Ca-release. Parathyroid hormone (PTH) at concentrations of 0.01-1.0 IU/ml dose-dependently stimulated 45Ca-release and the effects were greater in mandible than in radius-ulna. Dibutyryl cyclic (DBc)-AMP stimulated the release of 45Ca in mandible with a sharp peak effect at 5 X 10(-4) M. The results indicate that mandible is more susceptible to these bone resorptive agents than radius-ulna.

Effect of tungsten absorption edge filter on diagnostic x-ray spectra, image quality and absorbed dose to the patient.

The X-ray spectra from a tungsten-target diagnostic tube were measured with a lithium-drifted silicon detector. Four characteristic X-ray peaks were clearly observed. When a 0.05-0.3 mm thick tungsten absorption edge filter was added to the tube, the number of photons in the spectra above the K-absorption edge decreased dramatically. The effect of the absorption edge filter on image quality and on the absorbed dose were investigated by both measurement and Monte Carlo calculation. The absorbed dose to the patient is reduced without image quality, being degraded.

The cyanine dye triS-C4(5) as a cationic uncoupler of oxidative phosphorylation: interaction with mitochondria detected by derivative spectrophotometry.

Derivative spectrophotometry was used to study the interaction of the cationic uncoupler triS-C4(5) with mitochondria. The uncoupling action of this dye is dependent on the presence of Pi in the incubation medium. The second derivative spectrum of the dye changed with the incubation period, becoming similar to the spectrum in chloroform; but, after a time, the spectral pattern reverted to the original spectrum. The change in the spectrum in the presence of Pi was much more rapid than in its absence. The degree of spectral change agreed with the relative amount of bound dye determined directly. Thus, the spectral change reflects the binding of dye to the mitochondria, dependent on their energy state. The greater binding without Pi does not cause uncoupling but does cause shrinkage. In contrast, the lesser binding in the presence of Pi causes uncoupling and the swelling of mitochondria. These facts indicate that the dye does not penetrate the mitochondrial membrane. This refutes the idea that uncoupling by lipophilic cations is caused by the electrophoretic transfer of the uncoupler to the mitochondrial matrix space.

Wolff-Parkinson-White syndrome with gradual transition from type A to type B.

This report documents a case which showed type A, type B and intermediate patterns of pre-excitation on different days. A vagotonic maneuver and digitalis induced a type A pattern, while exercise, atropine and isoproterenol caused a type B pattern of activity. A gradual transition from type A to B was demonstrated with vectorcardiograms. Despite the variations in the QRS morphology, the direction of the initial vector was not altered and was directed straight anteriorly. In this case, an accessory pathway may be located in the posterior paraseptal region or the lateral free wall of the left ventricle, and a variable size of pre-excited area may have caused type A and type B patterns of pre-excitation.

[Multiple meningioma].

Meningiomas are the most popular benign intracranial tumors, but they are rarely seen as a multiple growth. The incidence of multiple meningiomas, defined by Cushing and Eisenhardt, is about 1 to 2 per cent of all meningioma cases. Though many cases of the multiple meningiomas were reported previously in the world, some of them were concomitant with von Recklinghausen's disease or acoustic neurinoma. Most of the cases of the multiple meningiomas reported showed multiple lesion at the time of operation or after a few years of the initial operation. We have encountered two patients with multiple meningioma without the stigmata of von Recklinghausen's disease in our clinic. Case 1. A 58-year-old female visited the hospital complaining of headache and occasional nausea on February 6, 1980. Plain and enhanced cT confirmed a large tumor in the right parietal region and three small tumor nodules in the right occipital region. Carotid angiogram detected only two tumors of frontal falx. Apparent two tumor stains were seen on the region, and they were fed by meningeal frontal and parietal region, and they were fed by meningeal arteries through the right ophthalmic artery. A large tumor of parietal and a small tumor of frontal region on the right side of falx were removed. Three nodular tumors of right occipital convexity were extirpated at the same time. Histological examination of the tumors disclosed all extirpated tumors were fibroblastic meningioma. Case 2. A 61-year-old male developed convulsive seizure of the right upper limb and right side of the face was diagnosed as having convexity meningioma in the left parietal region.(ABSTRACT TRUNCATED AT 250 WORDS)

H NMR detection of the specific change to the uncoupled state of the mitochondrial membrane.

The spin-lattice relaxation times T1 of H2O in mitochondrial suspensions at various energy states were measured at 28 degrees C. It was found that de-energization on the addition of an uncoupler of oxidative phosphorylation affects T1: after the addition of the uncoupler, T1 first decreases and then increases to a value greater than those at other different energy states. The T1 values of the unfrozen water in mitochondrial suspensions were measured at -15 degrees C after the energy state of the mitochondria was freeze-trapped. It was found that they increase only in the presence of an uncoupler. These changes in T1 are discussed in relation to the specific change of the mitochondrial membrane in the uncoupled state.
