Phospholipase C of an oral strain of Propionibacterium acnes purification and partial characterization.

Phospholipase C of the oral Propionibacterium acnes strain D 7 was purified from culture supernatants and partially characterized. The molecular weight was found to be 32.000 and the optimum pH was situated between 7.0 to 8.0. This nonhemolytic enzyme hydrolyzed relative intensively acidic glycerophospholipids and produced 1,2-diglyceride from phosphatidyl choline.

Protective effect of butylated hydroxytoluene against induction of gastric cancer by N-methyl-N'-nitro-N-nitrosoguanidine in Wistar rats.

The effects of butylated hydroxytoluene (BHT) on the incidence and histology of gastric cancers induced by oral administration of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) were investigated in Wistar rats. Oral administration of 1.0% BHT in regular chow pellets during treatment with MNNG for 25 weeks significantly reduced the incidence of gastric cancers in experimental week 40. BHT had no influence on the histological type of adenocarcinomas induced in MNNG-treated rats. BHT alone induced slight glandular hyperplasia, but not moderate glandular hyperplasia or gastric cancer.

Diagnosis of minute cancers by the endoscopic Congo red--methylene blue test.

The clinicopathological features of minute cancers of the stomach were investigated, and the accuracy of diagnosis of minute cancers by routine endoscopic examinations and by the endoscopic Congo red - methylene blue test were compared. From 1959 to 1979, 56 minute cancer were found in 54 patients. Minute cancers are defined as those of less than 5 mm in the longest diameter. None involved the muscularis propria, but when minute cancers, especially the depressed type, were more than 4 mm in diameter, cancer cells were frequently found to invade the submucosa. Therefore, minute cancers must be diagnosed as early as possible. A correct diagnosis of minute cancers was made in only 25.0% of the cases by routine endoscopic examinations. But with the Congo red--methylene blue test the diagnostic rate was raised significantly to 75.0%. In this test, tumors are seen as areas in which the dyes become bleached to white, in sharp contrast to the unaffected mucosa. This test is more accurate than other methods for diagnosis of minute cancers, because the target area can be biopsied with greater accuracy.

Electron microscopic observations of cell division in Mycobacterium vaccae V1.

Cell division of Mycobacterium vaccae was initiated by deposition of new wall material in the cross wall. The surface layers of the old wall remained continuous until septum formation was complete. Subsequently, rupture of the outer cell wall layers occurred circumferentially, leaving rings on the cell wall. The two daughter cells remained connected with each other at the new pole and bent to form V-shaped structures at the connecting point.

Purification and characterization of mucopolysaccharidase from an oral strain of Bacteroides sp.

A mucopolysaccharidase in the cell extract of an oral strain of Bacteroides sp. was purified to homogeneity by ammonium sulfate precipitation, DEAE-cellulose column chromatography, gel filtration on Sephadex G-200, and isoelectric focusing. Specific activity increased 110-fold and recovery was 2%. The molecular weight was determined to be 89,000 by gel filtration, and the isoelectric point was 7.0. The optimum pH for the activity was 6.5. The enzyme was inactivated by heating at 60 degrees C for 5 min. The purified mucopolysaccharidase degraded hyaluronic acid more rapidly than chondroitin and chondroitin sulfate A and C. However, it had no activity against chondroitin sulfate B, heparin, and heparan sulfate. Since unsaturated disaccharides were derived from the enzyme substrate, this enzyme was considered to be a mucopolysaccharide lyase.

Production, purification, and properties of a bacteriocin from Staphylococcus aureus isolated from saliva.

Staphylococci from samples of human saliva were isolated on staphylococcal-selective agar plates. These strains were tested for the inhibition of the growth of Staphylococcus aureus FDA209P. The frequency of inhibitory strains among all of the staphylococcal isolates was 5.2%. Strain IYS2, which formed the biggest inhibitory zone against the growth of the indicator strain, was used as the producer of bacteriocin. IYS2 was identified to be S. aureus, based on its biological properties. The bacteriocin was purified by sequential procedures, including ammonium sulfate precipitation, fractionation with ethanol, ion-exchange chromatography, and gel filtration. Its molecular weight was determined to be 5,000. The isoelectric point was 10.0. In amino acid composition, lysine, histidine, aspartic acid, valine, and phenylalanine were predominant. The bacteriocin was heat stable but inactivated by proteases or peptidase. The bacteriocin had a bactericidal effect on susceptible cells. An analysis of the inhibitory spectrum among typical oral indigenous bacteria showed that Streptococcus salivarius, Propionibacterium acnes, Corynebacterium parvulum, and Actinomyces israelii were susceptible to the bacteriocin. Streptococcus mutans, Streptococcus sanguis, Streptococcus mitis, Actinomyces viscosus, Actinomyces naeslundii, Fusobacterium nucleatum, Bacterionema matruchotii, and Bacteroides melaninogenicus were resistant. The majority of S. aureus tested were susceptible, and all Staphylococcus epidermidis strains tested were resistant.

Cardiac beat-to-beat variation as an indicator of neuropathy and the frequency of its abnormality in diabetes mellitus.

The cardiac beat-to-beat variation and the maximum constriction velocity and acceleration of the pupillary light reflex have been considered indices of parasympathetic neuropathy. The abnormality of these indices was evaluated in 191 non-diabetics and 151 diabetics. The cardiac beat-to-beat variation decreased gradually with age in non-diabetics as well as diabetics. It was small in diabetics compared to non-diabetics in corresponding age group between 10 and 59 years. In diabetics it was still smaller in the presence of peripheral neuropathy than in its absence. The overall prevalence of this abnormality was 30.1% in diabetics but only 3.0% in non-diabetics. In diabetics it rose to 40.0, 42.6 and 52.2% in the presence of motor, sensory and motor plus sensory nerve neuropathy, respectively. In 63 diabetics examined for both cardiac beat-to-beat variation and pupillary light reflex, no statistical difference was observed between the frequency of these abnormalities. These observations indicate an extremely high prevalence of abnormality in autonomic nerve function in diabetics especially in the presence of abnormality in autonomic nerve function in diabetics especially in the presence of other types of neuropathy, so that diabetics with peripheral neuropathy should be carefully examined for autonomic neuropathy and vice versa. Besides, the measurement of the cardiac beat-to-beat variation is similar in sensitivity in the detection of neuropathy to the measurement of pupillary light reflex.

Hepatocellular carcinoma: sonographic and histologic correlation.

Sonographic and histologic correlation was studied in 20 cases with a combined total of 23 resected lesions of hepatocellular carcinoma. All cases were examined by sonography within 1 week before surgery. Tumor sonograms were classified as hypoechoic, complex, or hyperechoic on the basis of the difference in echogenicity between the lesion and the surrounding liver parenchyma. Fourteen excised pathologic specimens were examined after surgery by a fixed-position sectional sonographic technique that permitted detailed comparison of the sonograms with the histologic findings. Each of the three types of lesion classified by echo pattern exhibited specific histopathologic characteristics. The hypoechoic lesion corresponded to a solid tumor without necrosis; the complex lesion corresponded to a tumor with partial necrosis; and the hyperechoic lesion was represented histopathologically by two distinct types of tumor, one with fatty metamorphosis and the other with marked sinusoidal dilatation. Fixed-position sectional sonography is a useful technique for comparing histologic and sonographic findings in a given tissue section and it contributes to the investigation of tissue characterization by sonography.

Promotion by histamine of carcinogenesis in the forestomach and protection by histamine against carcinogenesis induced by N-nitroso-N-methylnitroguanidine in the glandular stomach in W rats.

The effect of histamine on induction of tumors in the forestomach and the glandular stomach after N-nitroso-N-methylnitroguanidine (MNNG) administration was studied in male inbred W rats. Animals were given 50 micrograms MNNG solution/ml orally for 25 weeks and then 4 mg histamine dihydrochloride sc per day in depot form. Administration of histamine in depot form after MNNG significantly increased the incidence of tumors in the forestomach, but it significantly decreased the incidence of adenocarcinomas in the glandular stomach. All of the tumors induced in the forestomach were of the squamous cell type, and 50% of them were squamous cell carcinomas. Histamine alone had no apparent carcinogenicity in rats.
