[Basic study on efficacy of cefmetazole against osseous and articular infections (author's transl)].

In order to evaluate the efficacy of cefmetazole (CMZ) against osseous and articular infections fundamentally, the sensitivity of clinically isolated strains of Staphylococcus aureus to the drug and its concentration in serum and bone marrow of tibia were determined. 1. The clinically isolated strains of S. aureus was found to be sensitive to 0.78 microgram-6.25 micrograms CMZ per milliliter when inoculated at 10(6) cells/ml and to 0.78 microgram-12.5 micrograms CMZ per milliliter when inoculated at 10(8) cells/ml. No strains were resistant to CMZ. 2. One gram of CMZ was injected intramuscularly before operation. Blood samples were collected from the bone marrow and the vein and measured for the concentration of CMZ. The mean concentration in the bone marrow of tibia were 30.5 microgram/ml, 16.8 micrograms/ml and 8.9 micrograms/ml 1, 2, and 3 hours later, respectively, and the mean transfer rate from serum to bone marrow was 104.8, 92.8 and 101.1% respectively. 3. CMZ concentration in bone marrow of tibia attained following an intramuscular injection of 1 g of CMZ were higher than the MIC values for most of the clinically isolated strains of S. aureus, suggesting the effectiveness of CMZ in clinical practice.

Gastrin protection against chemically induced gastric adenocarcinomas in Wistar rats: histopathology of the glandular stomach and incidence of gastric adenocarcinoma.

The effects of gastrin on the histopathology of the glandular stomach and on the incidence of gastric carcinoma induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) were investigated in inbred Wistar (W) rats. Prolonged administration of gastrin after treatment with MNNG significantly reduced the incidence of adenocarcinomas of the glandular stomach. In addition, atypical glandular proliferations were significantly less frequent and were smaller, and the incidence of marked mucosal atrophy was significantly reduced in both the antral and oxyntic gland mucosae. Both atypical glandular hyperplasia and mucosal atrophy are precursors of gastric cancers; prolonged administration of gastrin to rats after treatment with MNNG suppressed development of precursors of gastric cancer and so prevented development of gastric cancers.

Significance of pelvic retroperitoneal pneumography in the assessment of extramural invasion of rectal carcinoma.

Pelvic retroperitoneal pneumography (PRP) and barium enema were performed simultaneously in 34 patients with rectal carcinoma in an attempt to assess whether there was extramural cancer invasion. In 9 patients, no free air was visualized in the retroperitoneal cavity surrounding the mass, and histological evaluation of the excised specimens showed extramural invasion in all 9. The mean maximum diameter was 4.1 cm. In 25 patients, free air was seen around the mass, and 10 had tumors limited to the rectal wall, the remaining 15 manifested extramural invasion, however, the mean diameter was small (2.0 cm). In a mass whose center was located along the anterior rectal wall, it was difficult to assess from the free air findings whether there was extramural invasion. Based on the present findings, we concluded that PRP is useful in determining preoperatively whether there is extramural invasion in cases with rectal carcinoma involving the posterior and lateral walls.

Comparative study on the nuclear DNA content of leiomyoma, leiomyoblastoma and leiomyosarcoma of the stomach and small intestine.

The nuclear DNA content of tumor cells in 5 leiomyomas, 4 leiomyoblastomas and 2 leiomyosarcomas of the stomach and 1 leiomyosarcoma of the small intestine was examined. From the viewpoint of nuclear DNA content, leiomyoblastoma was between leiomyoma and leiomyosarcoma. A definite difference in nuclear DNA content exists among leiomyoma, leiomyoblastoma and leiomyosarcoma, and percentage of nuclei with a DNA content of 3 times or 4 times over the basic chromosome content represented good indices for distinguishing between benign and malignant conditions. A diagnosis of leiomyocarcinoma may be more accurate when over 80% of the nuclei have DNA content values over 3 times of basic nuclear DNA content and when over 50% exhibit more than 4 times of basic content.

Phenotypic reversion of SV40-transformed 3T3 cells by dimethylsulfoxide.

With dimethylsulfoxide (DMSO) (0.5 to 1.5%) in the medium, SV40-transformed 3T3 cells (SV3T3) changed morphologically from a round to a flat fibroblastic shape. The saturation density of the treated SV3T3 cells decreased and the generation time increased. These cells showed an increased anchorage dependency in soft agar. Hexose uptake by SV3T3 cells was reduced to the level in the parent 3T3 cells and susceptibility of the SV3T3 cells to concanavalin A (con A) also decreased. These phenotypes of transformed cells appeared to change concomitantly from the transformed toward the normal state with the increase of DMSO concentration.

Polyps in the acid-secreting area of the stomach.

The relation of gastric polyps to acid-secreting mucosa was examined in 120 patients with gastric polyps. Studies were made by the endoscopic Congo red test developed in this clinic. In 23 patients (19.1%) gastric polyps were located in acid-secreting mucosa and may have arisen from normal oxyntic mucosa. These polyps were of two histological types: oxyntic gland polyps containing many parietal cells, and nonoxyntic gland polyps without parietal cells. Only the former type appeared to secrete acid, judging endoscopically from the change of Congo red from red to blue-black on the surface of the polyps after administration of gastrin.

Purification of a new protein stimulating RNA polymerase II from Ehrlich ascites tumor cells: comparison with proteins purified before.

A protein factor named S-I(b) that stimulates the activity of RNA polymerase II was purified from Ehrlich ascites tumors cells. The final preparation gave a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. This protein has a molecular weight of 24,000 and is significantly smaller than S-II and S-II', stimulatory proteins of RNA polymerase II purified previously. However, S-I(b) cross-reacted with antibody against S-II and its peptide map obtained after radio-iodination was identical with that of S-II' (a phosphorylated form of S-II), indicating that S-I(b) and S-II share a common primary structure. S-I(b) is suggested not to be a degradation product of S-II produced during the purification process, and the structural relations between S-II and S-II' are discussed.

Inhibition of gastrin secretion by pirenzepine (LS 519) in treatment of gastric ulcer.

The effects of pirenzepine on the serum gastrin levels in the basal state and in response to a liquid test meal were investigated in nine patients with gastric ulcer. Single-dose administration of the drug caused a significant decrease in the basal gastrin level and significant inhibition of the serum gastrin response. When measured 12 h after the end of drug treatment for 7 days, the serum gastrin levels in the basal state and in response to a test meal tended to be high, but the differences were not statistically significant.

Antiserum to SV40 T antigens from a rabbit inoculated with autochthonous cells transformed in vitro by SV40.

A morphologically transformed cell line was established after infection with SV40 of the primary culture derived from a kidney removed surgically from a rabbit. The transformed cells were free of detectable infectious SV40 particles but produced them upon fusion with permissive monkey cells, and produced SV40-specific large and small T antigens. Repeated subcutaneous inoculations of the transformed cells into the animal from which the parental kidney cells had been derived failed to result in tumor formation but raised a substantial level of antibodies to the SV40-specific large and small T antigens as well as to the 58,000 dalton molecular weight cellular protein (middle T antigen).

[Use of CO2 euthanasia cabinet for experimental animals (author's transl)].

The process of CO2 euthanasia on mouse, rat, guinea pig and rabbit was observed using a CO2 euthanasia cabinet. The cabinet was filled with CO2 gas and the caged animal was placed into the cabinet. These animals quietly collapsed and their respiratory movement ceased within 25--225 seconds. A-V block was the first arrhythmia recognized on ECG in all cases. The arrhythmia recorded on ECG included complete A-V block, A-V dissociation, ventricular escape rhythm, atrial fibrillation, ventricular flutter and ventricular fibrillation. It could be concluded that euthanasia was satisfactorily performed on these species by means of the CO2 euthanasia cabinet.

Carcinoembryonic antigen in gastric juice as an aid in diagnosis of early gastric cancer.

The levels of carcinoembryonic antigen (CEA) in the serum and gastric juice of 58 patients with gastric cancer were determined by radioimmunoassay. The level of serum CEA was not a satisfactory indicator of gastric cancer: it was elevated in only 19.4% of the cases of advanced and unresectable cancer and 4.5% of cases of early cancer examined. However, the CEA level in the gastric juice was significantly higher in patients with gastric cancer than in normal subjects or patients with benign gastric diseases: increased CEA concentrations and outputs in the gastric juice were observed in 54.5% and 40.1%, respectively, of the patients with early gastric cancer, and in 52.9% and 50.0%, respectively, of the patients with advanced gastric cancer. The gross type of gastric cancer had no influence on the gastric CEA level, but the CEA level in the gastric juice tended to be high when the tumors were large, or when they had little or no glandular structure or high mucus-secreting activity.

Gastric hamartomatous polyps in the absence of familial polyposis coli.

Hamartomatous polyps in the stomach have been described as gastric lesions of familial polyposis coli. Four cases of multiple gastric hamartomatous polyps not associated with polyposis coli were encountered. Histologically, all these polyps consisted of normal oxyntic glands with numerous cystic dilatations of the glandular lumens of various sizes. Endoscopically, the polyps were all located in the acid secreting area and had an acid secreting function, which was revealed by the endoscopic Congo red test.

Measurement of a stimulatory protein of RNA polymerase II in various mouse organs by the complement fixation test.

The amount of S-II, a protein specifically stimulating RNA polymerase II, was measured in various mouse organs by a micro complement fixation assay. The amount was almost the same in brain, liver, kidney, spleen, and Ehrlich ascites tumor cells on the basis of DNA, being about 3.8 micrograms/mg DNA, which corresponds to 3.6 X 10(5) molecules/cell. However, the amount of S-II decreased greatly during erythro-differentiation of Friend leukemia cells and no S-II was detected matured erythrocytes.
