Nine healthy and seven mite-sensitive allergic subjects were examined by skin sensitivity and IgE antibody to fractions from Dermatophagoides farinae, including high and low allergenic fraction to human. In the intracutaneous test, all healthy subjects reacted to 200 ng and in particular, five of them to 0.2 ng of Fr void on Sephadex G-100 filtration which was a low allergenic fraction. Furthermore, end points of intracutaneous test and specific IgE antibody titers did not correlate on Fr void. Conclusively, Dermatophagoides farinae extracts contain a nonimmunologically highly skin-reactive fraction.

A procedure was developed for obtaining both cytologic and histologic specimens from patients suspected of having hepatic malignancy at the time of ultrasonically guided liver aspiration biopsy using a 22-gauge needle of 0.4 mm inner diameter. The fine needle and the syringe used were heparinized before use to avoid blood coagulation in the materials obtained. With heparinized needle and syringe it was possible to centrifuge the materials obtained and make good smears containing abundant tumor cells, and to obtain small tissue specimens. Histologic specimens obtained with the heparinized needle were fixed in 2.5% glutaraldehyde, postfixed in 1.0% osmium tetroxide, and embedded in epoxy Epon resin. Ultrathin sections were made and usually stained with Azan-Mallory stain. In 41 patients suspected of having hepatic malignancy, a correct diagnosis was made by a combination of these examinations. The sensitivity of this procedure for hepatic malignancy was 100%, and the specificity was 95.7%. Therefore, this procedure is a very reliable method for diagnosis of hepatic malignancy.
Mucosal dysplasia and carcinoma infiltrating the submucosal layer were induced in rats with dimethylhydrazine. The nuclear DNA content of the cells was measured, and the correlation between the DNA distribution pattern and histopathologic findings was evaluated. In mild and moderate dysplasias, histograms of the nuclear DNA content showed a narrow range of distribution with a sharp peak in the diploid area. In severe dysplasia, however, there was wide distribution and no peak value; this was reminiscent of carcinoma. Based on the findings of this report, the authors suggest that severe dysplasia be regarded as intramucosal carcinoma.
An immunohistochemical (IF) method utilizing polyclonal antibodies to cytoplasmic estrogen receptors (ER) and a tissue processing procedure which allows in situ precipitation of cytoplasmic ER without denaturation of antigenic sites were first described in this journal. In this report we present data on the immunohistochemical classification of 153 cases of human breast carcinomas (123 frozen sections, 28 cytological specimens) as ER-positive (IF+) or ER-negative (IF-). Results on correlation between the biochemical vs immunohistochemical assessment of the ER status and the relationship between quantity of ER and the proportion of IF+/IF- tumor cells are presented. In addition, a procedure which has been successfully applied to study in vitro translocation of cytoplasmic ER in cryostat sections of human breast carcinomas is described. A positive correlation between ER translocatability and status of progesterone receptors (PR) was obvious. All biochemical and immunohistochemical data are collectively reviewed and the question of whether the antibodies are detecting Type I ER is examined.
Sodium butyrate induced adhesion of cultured mastocytoma p-815 cells to the surface of a standard tissue culture grade petri dish. The ratio of the number of adherent cells to that of total cells (adherent plus floating cells) was dependent on the serum concentration and on the dose of sodium butyrate. During approximately the first 6 hr after the addition of sodium butyrate, no cells adhered. The optimum conditions for adhesion were provided by 2 mM sodium butyrate and 15% fetal calf serum, 44 hr after addition of this compound. Morphologically, adherent cells consisted of spindle-shaped and round cells: the latter clustered to the former. Low concentrations of actinomycin D (0.005 microgram/mL) and of cycloheximide (0.5 microgram/mL) inhibited cell adhesion. Adherent cells were easily detached by 0.25% trypsin-0.02% EDTA but not by EDTA alone. Adherent mastocytoma cells which were cultured in the presence of 2 mM sodium butyrate, re-adhered to the surface of the dish. The ratio of adhesion in the second dish, however, was very low (35% after 2 hr incubation). Radioactive iodinated surface proteins of butyrate-treated adherent cells showed two new bands (70,000 and 92,000 D) which were not detected in control cells, but there was no difference in the extent of labeling of high molecular weight protein (250,000 D) between butyrate-treated and control cells.
The number of Na+ pump units (Bmax) and the equilibrium dissociation constant (Kd) for ouabain as well as parameters of K+ binding to the Na+ pump were examined in in vitro-grown vascular smooth muscle cells ( VSMC ) derived from Sprague-Dawley rats. The technique to measure these variables utilizes analyses of [3H]ouabain displacement from its VSMC receptors by nonlabeled ouabain and K+. The mean values for Bmax and Kd in the cultured VSMCs were 1.95 X 10(5) receptor sites per single VSMC and 2.68 X 10(-6) M, respectively. The equilibrium dissociation constant for K+ (Ki) was 0.92 mM. K+ binding to the cultured VSMCs demonstrated positive cooperativity with a Hill coefficient (n) of 1.78.
The effects of a conceptus-derived substance on the activity of 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) and 20 alpha-HSD in the ovary were studied in the rat. On day 7 of pregnancy (day 1 = insemination), rats were laparotomized and the desired number of conceptuses was aspirated from the uterus; thus, rats carrying one, two, three, four, five to seven or eight to ten conceptuses were prepared. They were autopsied on day 15 and 3 beta-HSD and 20 alpha-HSD activity in the corpus luteum (CL) or non-luteal ovarian tissue ( NLO ) was determined. Conceptus number was directly related to 3 beta-HSD and inversely related to 20 alpha-HSD activity in the CL. The serum progesterone level and CL weight were also directly related to conceptus number. Neither 3 beta-HSD nor 20 alpha-HSD activity in the NLO was affected by conceptus number. These results indicated that 3 beta-HSD and 20 alpha-HSD in the CL are probably regulated by placental hormone secreted in proportion to the number of conceptuses; in the NLO these enzymes may be controlled by a different mechanism.
Thirty-nine gastric cancer patients with liver metastasis on laparotomy were histologically analyzed. Poorly differentiated adenocarcinoma of the medullary type was the most common histologic type followed by papillary adenocarcinoma. Analysis of 32 gastric cancer patients who manifested liver metastasis after the operation showed that papillary adenocarcinoma and poorly differentiated adenocarcinoma of the medullary type were frequent. We conclude that poorly differentiated adenocarcinoma of the medullary type is the most common histologic type of gastric carcinoma metastasizing to the liver.

Among 452 patients who underwent operation for early gastric cancer, 101 were late deaths. The cause of death in these patients was studied with special reference to cancer recurrence and the occurrence of metachronous cancers in other organs. Of these 101 patients, 63 died of noncancerous diseases, the other 38 of cancer. Among the latter, 15 patients died of recurrence more than 5 years after operation; macroscopically, the protruded or elevated type combined with the depressed or excavated type were found more frequently in the primary lesions. Differentiated adenocarcinoma was a characteristic histologic findings for these lesions, and hematogenic metastasis was the most frequent recurrence pattern. Ten patients with early gastric cancer who underwent operation died of metachronous cancer in other organs. Our findings suggest that in the long-term follow-up of patients with early gastric cancer who undergo operation, the occurrence of metachronous cancer in other organs should be considered in addition to recurrence of cancer.
