Estradiol binding capacity of peripheral blood mononuclear cells in asymptomatic HBV carriers.

Peripheral blood mononuclear cells from asymptomatic HBV carriers and normal individuals were incubated with 3H-estradiol and cold estradiol at 37 degrees C. The radioactivity bound to these cells was then counted, and the estradiol binding capacity estimated. The estradiol binding capacity of the mononuclear cells was lower in asymptomatic HBV carriers than in normal individuals. This suggested that the unresponsiveness of the mononuclear cells to estrogen in asymptomatic HBV carriers, which we have reported previously, may be due to a deficiency in the estradiol binding capacity.

Effects of spontaneous respiration on right and left ventricular function: evaluation by respiratory and ECG gated radionuclide ventriculography.

To clarify the mechanism of inspiratory reduction of left ventricular (LV) stroke volume (SV) during spontaneous respiration, we measured right and left ventricular volume changes from expiration to inspiration using radionuclide ventriculography with respiratory gating technique. In this method, scintigraphic data were acquired in a list mode with ECG R wave triggers and respiratory volume curve derived from respiratory flowmeter. Cardiac cycles occurring during the second halves of inspiratory and expiratory phases were separately selected and used to produce multigated images for the respective phases. Twelve patients with normal LV ejection fraction (EF) (greater than 50%) and right ventricular (RV) EF (greater than 40%) and without pulmonary diseases were studied. LV end-diastolic volume (EDV) decreased during inspiration in all subjects (by 11 +/- 5%), whereas LV end-systolic volume (ESV) was insignificantly changed. Accordingly, LVSV decreased during inspiration in all subjects (by 17 +/- 7%). LVEF decreased from 64 +/- 6% during expiration to 60 +/- 6% during inspiration (p less than 0.001). In contrast to the left ventricle, RVEDV and RVSV increased during inspiration by 13 +/- 11% and 22 +/- 18%, respectively. RVESV did not change significantly. RVEF increased from 48 +/- 6% during expiration to 52 +/- 5% during inspiration (p less than 0.05). These results indicate that inspiratory reduction of LVSV during spontaneous respiration is due to a decrease in LVEDV which may be derived from an increase in RVEDV during inspiration through the mechanism of ventricular interdependence.

Phenylcyclohexylamine: effect of a metabolite of phencyclidine on the efflux of dopamine in the rat.

The effect of phenylcyclohexylamine (PCA) on the efflux of dopamine (DA) in the neostriatum was examined using in vivo electrochemical techniques. Phenylcyclohexylamine produced a long-lasting dose-dependent biphasic effect on the efflux of DA in the rat. This response, to one of the major metabolites of phencyclidine, was similar in duration to but less potent than that seen with phencyclidine.

Analysis of prostatic bud induction by brief androgen treatment in the fetal rat urogenital sinus.

The androgen dependency of prostatic bud formation in fetal rat urogenital sinuses was studied using brief treatments with androgen, and the incorporation of androgens by the sinus mesenchyme was followed by steroid autoradiography. Urogenital sinuses from 16.5-day fetuses of both sexes were grown in organ culture and treated with androgens for periods ranging from 4 to 72 h and then transferred to control medium. A minimum treatment of 24 h was required to induce prostatic buds in male sinuses and of 36 h in all female sinuses. This difference in response disappeared after more prolonged treatment. In both sexes the number of prostatic buds increased with the time of exposure to androgens. Prostatic bud formation continued for 24-36 h after transfer to control medium. Steroid autoradiographic analysis showed that the labelled androgen was concentrated in the mesenchymal nuclei. The rate of incorporation rose steeply during the first 12 h and then more slowly. After transfer to control medium the amount of labelled androgen decreased rapidly to half within 12 h and then decreased more slowly. In the competition experiments a 200-fold excess of unlabelled testosterone or dihydrotestosterone in the labelling medium greatly reduced the nuclear labelling with [3H]testosterone.

A kindred of familial combined hyperlipidemia (FCHL) with proband showing type V hyperlipoproteinemia.

A case of familial hyperlipidemia incidentally found through a 16 year-old high school girl with type V hyperlipoproteinemia and abdominal bouts consistent with this type of hyperlipemia is reported for the first time in Japan. The laboratory findings of the plasma of her father revealed typical hyperlipoproteinemia of type IIa. Nineteen of her 26 kindred were investigated. Type V was seen only in the proband, type IIa in father, paternal grandmother, two paternal aunts, and two paternal cousins, type IV in three paternal cousins. The serum apolipoprotein (apo A-I, A-II, B, C-II, C-III, and E) concentrations were determined by the single radial immunodiffusion technique. The apolipoprotein concentrations were not different from those of normolipidemic control subjects except for apo B, which was higher in the hyperlipidemic members, and apo C-II, C-III, and E, which were higher in the proband.

Ultrastructural study of a clear cell chondrosarcoma.

A lesion in the left humeral head of a 15-year-old Japanese boy was light-microscopically diagnosed as clear cell chondrosarcoma. Fresh tissues from the lesion were studied ultrastructurally. In most tumor cells, that is, clear cells, paucity of organelles, sparse free ribosomes, and low protein content in cytosol were most striking. Tumor cells had many microvilli characteristic of cartilage cells. Among scanty organelles, vesicles or cisternae of dilated rough endoplasmic reticulum were rather conspicuous, and well-developed Golgi complexes were sparse. Intercellular matrix was scanty. These results suggest that intercellular matrix substance may be synthesized substantially by rough endoplasmic reticulum, whereas the synthesized substance perhaps cannot be transported smoothly into extracellular spaces because of scanty well-developed Golgi complexes.

[Androgen receptors in the epithelium during prostatic gland induction from the urogenital sinus epithelium in Tfm-mutant mice].

Epithelio-mesenchymal recombination experiments revealed that urogenital-sinus mesenchyme of normal rat fetuses failed to induce androgen-receptors in sinus epithelium of androgen insensitive Tfm (testicular ferminization) mouse fetuses. Nevertheless, the mesenchyme can induce the morphogenesis of prostate-like glands in the androgen-receptor-deficient epithelium.

[Hepatic disorder during long-term intravenous hyperalimentation in adults].

Hepatic disorder during long-term intravenous hyperalimentation (IVH) in adults was studied. Fifty-one patients (30 males and 21 females) in whom IVH was performed for more than 60 days at Tenri Hospital from 1975 to 1984, were divided into 3 groups by change of indices which showed hepatic cellular injury (CII) and bile stasis (BSI). Namely, type I patients (33 cases) showed transient abnormality of indices, type II patients (14 cases) has continuous abnormal indices during IVH and type III patients (4 cases) were lethal due to hepatic failure. There were no differences in clinical feature among 3 groups. Incidence of metabolic complications during IVH in type II was significantly higher than in type I (p less than 0.05). All patients of type III were complicated by severe infectious diseases. Chief histological findings from 13 specimens (6 biopsies and 7 autopsies) were fatty degeneration of hepatocytes and fibrosis in the portal area with proliferation of bile ducts. Change of the portal area had closer relationship to the clinical severity of hepatic disorder than change of hepatocytes. BSI was more important than CII in inspection of clinical course and estimation of prognosis on hepatic disorder during IVH.

[Clinical study of serum enzyme and metabolic changes after extracorporeal circulation--comparison with the surgery of aortic aneurysm].

The changes of serum enzyme and metabolism were studied by comparing the serial change among three groups. The first group consisted of twenty seven patients treated by surgery under extracorporeal circulation. The second group of ten patients of thoracic aortic aneurysm treated under temporary bypass and the third group of nineteen patients of abdominal aortic aneurysm under aortic cross clamp. The following results were obtained: The enzyme of serum GOT, LDH and CPK increased postoperatively in the all groups, the changes after surgery being greater in the group 1 than the other groups. The metabolism of lactic acid and glycolipid was depressed in the group 1 higher than the other groups. The tissue injury of total body after surgery with extracorporeal circulation was greater than the other groups without extracorporeal circulation. And these results suggest the necessary of improvement of circuit and perfusion technique of extracorporeal circulation.
