Insect immunity: isolation and structure of cecropins B and D from pupae of the Chinese oak silk moth, Antheraea pernyi.

The immune system in the Chinese oak silk moth, Antheraea pernyi, has been compared with that of the Cecropia moth which has been characterized earlier. Antibacterial activity against Escherichia coli was induced in diapausing pupae by injection of viable E. coli or Enterobacter cloacae. The activity reached a maximum on day 7-8 after which the response gradually declined. The pupae produced a set of immune proteins with P4 and P5 as major labelled components similar to that earlier found in Cecropia. The major antibacterial factor in A. pernyi was cecropin D. A procedure is described for the isolation of cecropin B and D, which is in principle similar to the one used for the isolation of the corresponding cecropins from Cecropia pupae. Amino acid sequence analyses of the A. pernyi cecropins show the D form to contain 36 amino acid residues and that both cecropins have blocked C-termini. The general structure of cecropins having a charged N-terminal region (residues 1-21) followed by a long hydrophobic stretch (residues 22-32) is well conserved. Cecropin B and D from A. pernyi differ from the corresponding proteins in Cecropia by four and three conservative amino acid replacements, respectively. The homology between the cecropins from the two insects suggests that they orginate from a single ancestral gene. The antibacterial activity was tested against nine different bacterial species. Evolutionary aspects of the cecropins are discussed.

The acute administration ward as a therapeutic community.

The authors evaluated six acute short-stay admission wards in a community mental health center using the sociometric Ward Atmosphere Scale. There was an attempt to run three of the six wards as therapeutic communities. No differences were found on Ward Atmosphere Scale dimensions between the therapeutic communities and other types of programs, nor did the therapeutic communities conform to an empirically derived therapeutic community program dimension profile. Possible reasons for this failure and its implications for community mental health centers are discussed.

[Course of recurrent nerve paralysis after thyroid operation].

In congenial collaboration with the otolaryngological department, Landeskrankenanstalten Salzburg, we have examined 216 patients with recurrent paresis within a postoperative period between half a year and 17 years. Thereby, we could note in 40% of the patients a complete disappearance of vocal cord paralysis. Disruption of the nerves, intubation damage and postoperative edemas have to be assumed to be causes of this temporary paresis. The result of the laryngological examination in no case coincided with the recovery of voice quality. The percentage of patients who were satisfied with their voice was much higher (90%), which is most important.

[Dose-effect relationship of glucagon on the stomach, pylorus and duodenum].

The effect of glucagon on the stomach, pylorus and duodenum was evaluated within the first 30 minutes p.i. Glucagon was administered intravenously in a dose of 0.1, 0.2, 0.4, 0.6 and 1 mg. The hypotonic effect was greater on the duodenum than on the stomach. The dose response of the pylorus was not uniform. It was possible to point out a direct relationship between the intensity and duration of the hypotonic effect and the administered dosage.

Sequence and specificity of two antibacterial proteins involved in insect immunity.

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacteria. WE recently purified and characterized P9A and P9B, which are two small, basic proteins with potent antibacterial activity against Escherichia coli and several other Gram-negative bacteria. We believe that P9A and P9B plays an important part in the humoral immune responses described previously and that the P9 proteins represent a new class of antibacterial agents for which we propose the name cecropins. We describe here the primary structures of cecropins A and B. We also show that cecropin A is specific for bacteria in contrast to melittin, the main lytic component in bee venom which lyses both bacteria and eukaryotic cells.

Pediatric and psychiatric collaboration in the management of childhood asthma.

With many asthmatic children for whom emotional factors are significant, joint pediatric and psychiatric management is essential. A three-level model of collaboration is presented, including (1) pediatric care, (2) psychiatric consultation, and (3) inpatient psychosomatic treatment. The organization, professional roles, and treatment protocol of the psychosomatic inpatient service are described. A review of the first 16 asthmatic children who received inpatient care in the program suggests that such a model may present a viable alternative to long-term residential care.

[Morphological and stereological examinations of placenta of patient with unruptured tubal pregnancy at full term (author's transl)].

Optimum conditions for foetal life were recorded from morphological and stereological findings obtained from a patient at full-term tubal pregnancy. Basiotrophic implantation, no reduction of chorion frondosum, large placental exchange surface, and extremely good capillarisation of chorionic villi proved to be the conditions for survival of the child.

[The mean blood pressure in the second trimester (map-2) as a valuable aid in the early recognition of the pregnancies with a risk of hypertension (author's transl)].

The mean blood pressure in the second trimester of pregnancy was calculated with a formula (Formula: see text) which stands for systolic pressure plus twice diastolic pressure divided by 3 equals mmHg. In 285 patients 43% had a MPA-2 value of 85 mmHg. or higher. At this threshold value 89% of all later hypertensions were recognized. An MAP-2 value of 90 mmHg. was found in 21% of the patients. In this group 63% became hypertensive. However, this group contained 10 of the 11 moderately severe and severe cases. In the outcome of the pregnancies there was a significant increase in prepathological fetal monitoring changes and a significant increase in suspicious Apgar values. The calculation of the MAP-2 value should become a part of routine prenatal care.

[Estimation of organ dose during radiological and computer tomographic examinations with calculations of the somatically significant doses (author's transl)].

The risks from radiation exposure during radiological diagnosis has usually been estimated in relation to genetic changes. Relevant information has been expressed as the genetically significant dose. In this paper we attempt to produce an analogous measure for evaluating the somatic risk in the form of a somatically significant dose index for radiological and CT examinations. It is shown that, for both types of examination, the two risk factors may be entirely different.

Insect immunity. Purification and properties of three inducible bactericidal proteins from hemolymph of immunized pupae of Hyalophora cecropia.

Three inducible bacteriolytic proteins, designated P7, P9A and P9B, from the hemolymph of immunized pupae of the giant silk moth Hyalophora cecropia have been purified using a two-step procedure with cation-exchange chromatography. Purified protein P7 has a molecular weight of 15000 and its amino acid composition shows a great similarity to that of the lysozyme from the wax moth Galleria mellonella. Moreover, heat stability, pH-rate profile and bacteriolytic specificity also indicate that protein P7 is a lysozyme. The other purified proteins, P9A and P9B, are highly potent against Escherichia coli and some other gram-negative bacteria. The amino acid compositions of proteins P9A and P9B are very similar, although the contents of glutamic acid and methionine were different. The molecular weights of these very basic proteins are around 7000. The P9 proteins are heat stable; their activities were retained after 30 min incubation at 100 degrees C. Both forms of protein P9 clearly differ from the lysozyme class of enzymes and they may represent a new type of bacteriolytic protein.

The use of two-channel telemetric systems in obstetrics: an ideal monitoring method for oral PGE2 induction of labor.

Our results demonstrate that free mobility in labor has many advantages. Because of the accelerated uterine contractility, the induction-delivery interval is shortened. No unphysiologic baseline pressure alterations were found. The maximal labor amplitudes did not differ from bedside-monitored labor. No FHR-changes occurred and the fetal outcome was excellent. Being able to move about, women are not influenced by the unfamiliar surroundings of modern labor wards. Pain relief and breathing are better than in the recumbent position. The ideal compound to support labor is oral PGE2.

[Unruptured tubal pregnancy at full term with survival of mother and child (author's transl)].

A clinically intact child was delivered from breech presentation in the 40th week of pregnancy. The amniotic sac was removed by salpingectomy. Basiotrophic implantation, the absence of chorion frondosum reduction, and the presence of amniotic fluid appeared to have been major prerequisites for unimpaired development. Diagnostic possibilities and symptoms of complete tubal pregnancies are discussed.
