Augmentation of human immunodeficiency virus (HIV) replication in cells infected with HTLVs has been demonstrated by cellular biological studies. Here, evidence is presented that this effect could be ascribed, at least in part, to the level of HIV gene expression.

Statistical studies were made on 1,795 outpatients, 528 inpatients and 518 operative procedures at our department in 1985. The most frequent diseases among the outpatients were urogenital infections followed by anomalies, stones and tumors. The major diseases among the inpatients were hypospadias, vesicoureteral reflux, congenital urethral stenosis, bladder tumor and benign prostatic hypertrophy. A total of 518 operations were performed. The five major operations were optic internal urethrotomy, hypospadias repair, ureterocystoneostomy, and transurethral resection of prostate and of bladder tumors.

The surgical operations including 5 cholecystectomies, gastrectomy, and graft implantation for abdominal aortic aneurysm were performed on 7 patients 2 to 98 months after valve replacement. Sodium warfarin was routinely used in an amount to reduce the prothrombin activity in a range of 20-30% normal by thrombotest. In all cases, anticoagulants were reduced in doses gradually in the period of 4 to 15 days proceeding to the operation. Heparin therapy was instituted for the prevention of thromboembolism, when prothrombin activity recovered to 50% normal by thrombotest, in 5 of 7 cases. Heparin therapy was interrupted just prior to operations and it was reinstituted soon after operations in 6 cases. Heparin therapy was maintained by continuous intravenous infusion to produce ACT of 130 to 150 seconds. Heparin therapy was stopped when prothrombin activity reached a therapeutic level with sodium warfarin. There were two episodes of hemorrhage required blood transfusion, observed in patients receiving heparin therapy, but there was no thromboembolism. We believe that our method which minimizes the nonanticoagulated time appears safe and effective in patients with prosthetic heart valves who require noncardiac operations.

Human T-cell leukemia virus type II (HTLV-II) isolated from a T-cell variant of hairy cell leukemia contains gag, pol and env genes as well as a fourth gene termed X, which can code three major open reading frames Xa, Xb and Xc. Proteins with molecular masses of 26 kDa (p26Xb) and 24 kDa (p24Xb) encoded by the Xb open reading frame were identified with antisera directed against synthetic peptides corresponding to the N-terminal and C-terminal amino acid sequences deduced from the structure of the Xb open reading frame. More than half the Xb products were found to be located in the nuclear fraction of HTLV-II-infected cells.
The effect of cholecystectomy on colon carcinogenesis induced by methylazoxymethanol (MAM) acetate was examined in four groups of Syrian golden hamsters. For the sexes combined, the incidences of total large intestinal neoplasms and adenomas in Group 1, which received cholecystectomy and a single intravenous injection of MAM acetate (20 mg/kg body weight), were significantly higher than those of hamsters in Group 2, which were given MAM acetate alone. The combined multiplicities of total large intestinal neoplasms from male and female hamsters, and the multiplicities of those in females of Group 1 were also significantly higher than those in animals in Group 2, respectively. No intestinal tumors were observed in hamsters in Group 3 (cholecystectomy alone) or Group 4 (untreated control). These results indicate an enhancing effect of cholecystectomy on MAM acetate-induced large intestinal carcinogenesis in hamsters.
The effect of dietary chlorogenic acid on methylazoxymethanol (MAM) acetate-induced carcinogenesis was examined in Syrian golden hamsters. The combined incidence of total large intestinal tumors from male and female hamsters, and the combined incidence of large intestinal adenocarcinomas or the incidence of the carcinomas of male or female animals of the group given a single intravenous injection of MAM acetate (20 mg/kg body wt) and then fed the diet containing 0.025% chlorogenic acid for 24 weeks were significantly lower than those of hamsters given MAM acetate alone. The numbers of hyperplastic liver cell foci in male and female hamsters given MAM acetate and chlorogenic acid were also significantly smaller than those of hamsters given MAM acetate alone. These results indicate an inhibitory effect of chlorogenic acid on MAM acetate-induced carcinogenesis in hamsters.
Sertoli cell cultures were monitored for müllerian inhibiting substance with a solid phase radioimmunoassay. Sertoli cells from newborn calf testes were placed in defined media free of serum in monolayer culture after treatment with trypsin-collagenase followed by gravity separation. Immunoreactive müllerian inhibiting substance was detected in the culture media of Sertoli cells but not National Institutes of Health 3T3 cells. To verify that the Sertoli cells were intact, cyclic adenosine monophosphate levels were determined after follicle-stimulating hormone stimulation. Cyclic adenosine monophosphate levels were elevated in Sertoli cells but not National Institutes of Health 3T3 cells. The newborn calf Sertoli cell culture provides a useful system in which to study factors affecting müllerian inhibiting substance production and release, and documents this substance as another reliable marker for the Sertoli cell. Müllerian inhibiting substance levels also could be measured in media beneath testicular fragments in organ culture, and were increased and prolonged in comparison to müllerian inhibiting substance release from Sertoli cell monolayer cultures. Modulation of müllerian inhibiting substance release from monolayer and organ cultures then was attempted. Neither gonadotropin nor steroid additions affected the release of müllerian inhibiting substance during 24 to 72 hours in organ or tissue culture. We are using the müllerian inhibiting substance radioimmunoassay to monitor attempts to immortalize a Sertoli cell line capable of continuous müllerian inhibiting substance production using viral deoxyribonucleic acid transfection techniques.
Serum levels of luteinizing hormone and follicle-stimulating hormone before and after luteinizing hormone-releasing hormone stimulation, and levels of testosterone before and after 3 days of treatment with human chorionic gonadotropin were determined by radioimmunoassay in 98 boys with hypospadias (2 to 8 years old). The doses of luteinizing hormone-releasing hormone and human chorionic gonadotropin were calculated for body surface. The basal and peak levels of serum luteinizing hormone, follicle-stimulating hormone and testosterone in patients with hypospadias were compared with 9 endocrinologically normal boys of the same age without hypospadias. Luteinizing hormone response to luteinizing hormone-releasing hormone stimulation was significantly impaired in boys with hypospadias and also simple hypospadias (no associated anomaly). Basal levels of luteinizing hormone in boys with simple hypospadias and levels of follicle-stimulating hormone in patients with hypospadias were significantly low. The maximum testosterone response to human chorionic gonadotropin stimulation was significantly decreased in boys with severe hypospadias in direct proportion to the degree of hypospadias.
The effect of betel nut on chemical carcinogenesis in the upper digestive tract and liver was examined in two different experimental models with ACI rats. The incidences of neoplasms and preneoplastic lesions of the tongue in animals given 5 ppm 4-nitroquinoline-1-oxide (4-NQO; CAS: 56-57-5) in the drinking water for 16 weeks and followed by 20% betel nut in the diet for 40 weeks were significantly higher than those in animals given 4-NQO alone. No enhancing effect from betel nut on the incidences of neoplastic and preneoplastic lesions in the upper digestive tract was found in animals administered 4-NQO for 12 weeks. The number of altered liver cell foci in rats given 200 ppm N-2-fluorenylacetamide (FAA; CAS: 53-96-3) in the diet for 8 weeks and followed by the betel nut diet for 16 weeks was significantly greater than that in animals fed the FAA diet alone. These results indicate enhancing effects of dietary administration of betel nut on oral carcinogenesis by 4-NQO and hepatocarcinogenesis initiated by FAA.
Fourteen patients with hypospadias were treated surgically for either an enlarged prostatic utricle (6) or vagina masculina (8). Six vaginae masculinae were removed by an extravesical abdominal approach. One enlarged prostatic utricle was removed by the perineal approach. Three enlarged utricles were removed by an abdominoperineal approach. The transtrigonal approach was recently used for three enlarged utricles and one vagina masculina in four patients. Surgical results in these 14 patients were satisfactory. One post-operative complication of transient mild bilateral vesicoureteric reflux was seen after a transtrigonal removal of an enlarged utricle. Potency was not affected in any approaches in four patients who were older than 12 years. The transtrigonal approach was found to be the best for good exposure of the lesion.
