A new prolyl hydroxylase acting on poly-L-proline, from suspension cultured cells of Vinca rosea.

A new prolyl hydroxylase having a novel substrate specificity was isolated from the suspension-cultured cells of Vinca rosea. This enzyme was solubilized with 0.05 M Tris-HCl buffer (pH 7.4) containing 0.1% Triton X-100, 0.3 M NaCl and 0.5 mM beta-mercaptoethanol from the membrane fractions of the cells, and was partially purified by (NH4)2SO4 fractionation and DEAE-Sephadex A-50 column chromatography. The enzyme preparation was found to require O2, Fe2+, ascorbate, alpha-ketoglutarate and poly-L-proline to attain maximum activity. The plant enzyme does not hydroxylate free proline and di-, tri- and tetra-L-proline, but hydroxylates octa-L-proline and poly-L-proline (Mr greater than 2000). Model peptides of unhydroxylated collagen, (Pro-Pro-Gly)5 and (Pro-Pro-Gly)10 are poor substrates for the plant enzyme. This means that the plant enzyme has a novel substrate specificity in regard to peptidyl substrate, and this differs from vertebrate prolyl hydroxylase, proline,2-oxoglutarate dioxygenase (prolyl-glycyl-peptide, 2-oxoglutarate: oxygen oxidoreductase, EC 1.14.11.2).

"Living electrode" as a long-lived photoconverter for biophotolysis of water.

Living blue-green algae (Mastigocladus laminosus), immobilized on an SnO(2) optically transparent electrode with calcium alginate, functioned as an anodic photoelectrode on continuous illumination for periods of time adequate for use in a conventional electrochemical cell. This "living electrode" shows promise of use as a long-lived photoconverter of solar radiant energy to electric energy and as a suitable replacement for unstable chloroplast systems.

Delayed germination of Bacillus cereus T spores after treatment with trichloroacetic acid and their reactivation by heating.

Treatment of Bacillus cereus T spores with trichloroacetic acid delayed their germination. The extent of retardation depended on the concentration of trichloroacetic acid, and the temperature, pH and duration of treatment. The effect was completely reversed by subsequent heating, and this restoration of germination also depended on the temperature and duration of heat treatment. Fourteen compounds were examined for their ability to suppress germination of spores. The halogenated fatty acids tested, such as trifluoro-, tribromo-, and dichloroacetic acid, caused suppression of germination, whereas other compounds, i.e., free fatty acids and amino acids, did not. It is concluded that the charge distribution of fatty acid molecules is important for their effect in suppressing germination of spores.

Progression and regression of atherosclerosis.

Relation of the intimal change in aging of various arteries to progression of atherosclerosis and the morphology of regression of atherosclerotic lesion were discussed. The course of regression of experimental atherosclerosis in rabbits and its histological findings were described.

Metabolic and cardiovascular changes during physical restrains in rats.

When conscious rats were physically restrained, their body temperature, heart rate and heat production increased within minutes. The rate of increase in heat production was less than that in heart rate, particularly in warm environments. Reduction of oxygen pulse, consequently either stroke volume or (A-V) O2 difference, was suggested. Stroke volume cardiac output and its fractional distribution to most of the calorigenic organs measured by the reference sample method using 85Sr-labelled microspheres decreased significantly by restraint. However, fractional distribution of cardiac output increased greatly to the skeletal muscles. The restraint-hypermetabolism may be attributed to the increased metabolism of the skeletal muscles supported with an increased availability of oxygen. With a slight increase in mean aortic pressure, total peripheral resistance increased greatly as observed during Valsalva-like maneuver. The decrease in cardiac output and stroke volume during restraint may be caused by obstruction to the systemic venous return.

[Distribution, absorption and excretion of 198Au-colloid and Na251CrO4 in the chicken (author's transl)].

In order to clarify the suitability of 198Au-colloid and Na251CrO4 as a marker of transit of the gastrointestinal content of the chicken, the distribution, absorption and excretion of 198Au-colloid and Na251CrO4 in the chicken were examined. Orally administered 198Au-colloid was not absorbed by the gastrointestinal tract and excreted into feces. Intravenously injected 198Au-colloid was not excreted into the gastrointestinal tract through the gastrointestinal wall. On the other hand, orally administered Na251CrO4 was relatively well absorbed by the gastrointestinal tract and distributed over a whole body. Intravenously injected Na251CrO4 was excreted into the gastrointestinal tract through the gastrointestinal wall and bile ducts. It seemed that 198Au-colloid was not adhered to the gastrointestinal mucosa and consequently, the transport of 198Au-colloid in the gastrointestinal tract was not delayed. It is concluded that 198Au-colloid is highly suitable as a marker of gastrointestinal transit of the chicken but Na251CrO4 is unsuitable.

Difference in calcium metabolism following Billroth-I and Billroth-II procedures for gastric and duodenal ulcers.

There have been 7 patients with postoperative osteomalacia in our series of 500 patients who had a gastrectomy for gastric or duodenal ulcers. All seven of these patients had a Billroth-II type gastrojejunostomy or esophagojejunostomy which caused food to bypass the duodenum and the upper part of the jejunum, and six of these patients showed milk intolerance and had diarrhea on food other than cow's milk. From this finding, it may be surmised that patients are predisposed to osteopenia after gastrectomy due to disorder in the process of calcium absorption and they develop osteomalacia when treated with the Billroth-II procedure, showing milk intolerance and diarrhea on food other than cow's milk.

Relationship of relevant factors of atherosclerosis to menopause in Japanese women.

The relationship of menopause to associated factors of atherosclerosis was investigated during October 1973 to December 1976. The subjects were 1674 premenopausal and 428 postmenopausal women aged 35 to 54, who were living in Toda city, an urban area of Japan. Serum cholesterol and triglyceride were higher in postmenopausal women than in premenopausal women. This trend was also observed in serum uric acid and blood hemoglobin. On the other hand, systolic and diastolic blood pressure and relative body weight had no consistent relationship to menopause.

Effect of trichloroacetic acid treatment on certain properties of spores of Bacillus cereus T.

Spores of Bacillus cereus T treated with trichloroacetic acid (6.1--61.2 mM) were compared with untreated spores, and as the concentration of the chemical increased, the following alterations in spore properties were found: (1) the extent of germination decreased irrespective of the germination medium used; (2) the spores became sensitive to sodium hydroxide (1 N) and hydrochloric acid (0.27 N), but not to lysozyme (200 micrograms/ml); (3) loss of dipicolinate increased on subsequent heating; and (4) the spores became more sensitive to heat. However, trichloroacetic acid-treated spores were still viable and there was no significant change in spore components. The mechanism of action of trichloroacetic acid is discussed.

Carcinoma in heterotopic gastric pancreas.

A case of a 55-year-old Japanese female who died of adenocarcinoma originating from heterotopic gastric pancreas is presented. The autopsy revealed, extensive metastases of bones. Though careful examination was done, other primary focus except that of the stomach could not be detected. In review of literatures, 11 malignant tumors originating from heterotopic gastric pancreas could be found including our case.
