Experimental model for peritoneal dialysis in small laboratory animals.

A simple method of performing peritoneal dialysis in rats and guinea-pigs for 48 hours is described. The method appears to be suitable for the experimental investigation of clinical problems although the long term problem of protein loss should not be forgotten when assessing the results of these short term experiments.

Metabolism and mode of action of androgens in target tissues of male rats. V. Uptake and metabolism of cyproterone acetate and its influence on the uptake and metabolism of testosterone and 5alpha-dihydrotestosterone in target organs and peripheral tissues.

In order to get more information on the mode of action of anti-androgens, two series with low but biologically active doses of cyproterone acetate were started. In the first experiments 12 mug of [3H]cyproterone acetate was injected intravenously into adult rats castrated 3 days before treatment. Thirty min after injection the radioactivity uptake in the target organs and other tissues was measured. The metabolites were separated by thin layer chromatography. A large pool of radioactivity could be shown in the liver. Thin layer chromatography revealed that in this pool cyproterone acetate had been converted by more than 80% to one metabolite. In blood plasma, too, the metabolite accounted for the major part of radioactivity. When compared to skeletal muscle, the prostate, seminal vesicles, and m. bulbocavernosus and m. levator ani accumulated more radioactivity. Within 30 min unchanged cyproterone acetate was retained selectively thus showing its relative high affinity to target organs. In a second experimental series, adult castrated male rats were given 10 mug of cyproterone acetate intravenously 30 min before the injection of [3H]testosterone or [3H]5alpha-dihydrotestosterone. Under this condition androgen uptake in target tissues was reduced to about 70% of the control values. The data parallel the results of in vivo studies on cytosol receptor displacement of androgens by cyproterone acetate. In agreement with previous investigators no significant influence of the anti-androgen on androgen metabolism was observed. The importance of the findings concerning the mode of anti-androgen action is discussed.

Myocardial hypertrophy, myocarditis, and myocardial fibrosis in a long term experiment.

In a long term experiment, a systematic analysis was made of the cellular reaction of the myocardium and the lymphatic system to application of antigenic substances. Several groups of mice received single or repeated injections of human serum, serum from rabbits with antiheart antibodies, mouse spleen cells immunized to myocardium, and human or mouse cardiac homogenate with and without incomplete or complete Freund's adjuvant. After injection of human and mouse heart homogenate, hypertrophy of myocardial cells was the first alteration seen; then followed focal myocarditis and after 10 months, interstitial fibrosis. Simultaneously, in the lymphorecticular tissue, there was marked proliferation of reticulum cells, lymphocytes, and plasma cells, as well as an enlargement of secondary follicles. All the animals produced antibodies to the injected antigens. Following injection of human serum or serum from rabbits with cardiac specific antibodies or spleen cells, no morphological changes were seen in the hearts.

[The effect of lumbar peridural anesthesia with catheter on the maternal and fetal acid-base status and the 1 minute apgar score (author's transl)].

Comparison of 650 deliveries with P.A. and of 928 deliveries without P.A. during the same period. PH from the umbilical artery and 1 minute Apgar score were studied in three groups of patients: 1.) All deliveries, 2.) Spontaneous vaginal deliveries without maternal or fetal risk, 3.) Operative vaginal deliveries. The only significant differences were found among the operative vaginal deliveries: The infants of the peridural group showed a higher incidence of pH-values above 7,2 than those of the non peridural group. Analysis of the maternal acid-base status showed less respiratory alcalosis and less metabolic acidosis in the peridural group. The neonates of this group showed a lower post partum metabolic acidosis than those in the non peridural group.

[Sialadenitis in a boy with anorexia nervosa].

Isolated sialadenosis of both sumandibular glands and anorexia nervosa are clinical pictures found extremely seldom in boys. This paper describes a case with symptomless enlarged submandibular glands, the bioptic findings which were suggesting the diagnosis of sialadenosis, the verification of the underlying disorder by child psychiatry, and the recuperation of the boy during puberty.

[Comparison of conventional methods for the determination of albumin, transferrin and ceruloplasmin in serum with immunological reference methods (author's transl)].

Radial immunodiffusion, which is a presently accepted reference method, was compared with the mechanized immunoprecipitation. Each of these specific methods was compared with two normal clinical-chemical methods for the determination of albumin and transferrin, and an enzymic method for the determination of ceruloplasmin. The correlation coefficients, the mean slope for regression, and the percentage variation of the average values from those of the average values of the reference methods are presented. In the determination of albumin and ceruloplasmin, radial immunodiffusion and mechanized immunoprecipitation give the same results. Surprisingly, these two specific methods give different results in the determination of transferrin. In as far as there is no doubt concerning the accuracy of the mechanized immunoprecipitation, this method is recommended, since it also has the advantage of being less demanding in cost and time.

[The method of demonstrating phenoloxidase].

It is reported on the evidence of phenoloxidase containing cells (=POZ) under various experimental conditions in as well spleen and small gut of white rats as small gut and skin of rabbit. A modified technic is described which gives a shortened incubation time and a regulary pigmentation of the reactive cells. POZ are demonstrable between pH = 6.2 and 8.0.

[Malignant melanoma--a PO-cell tumor? (author's transl)].

In the connective tissue, diffusely scattered phenoloxidase containing cells (PO-cells) can be found by histochemical methods. The same cells were demonstrated in a recurrent malignant melanoma of the skin and in its lymph node metastases. The theory is proposed that PO-cells are the stem cells of malignant melanoma in contrary to the common view that melanoma derives from the melanocytes of the stratum cyclindricum of the epidermis. The occurrence and distribution of PO-cells in the rat give a good explanation for the appearance of metastasis within different organs and tissues.
