Studies of the human testis. XIII. Properties of nicotinamide adenine dinucleotide (reduced form)-linked 17 alpha-hydroxylation.

A NADH-linked 17 alpha-hydroxylation of progesterone was examined using the washed microsome fraction prepared from human testes. The addition of NADP revealed no enhancement of 17 alpha-hydroxylation, indicating no transhydrogenation from NADH to NADP in the microsome fraction. The results further substantiate the presence of NADH-linked activity of 17 alpha-hydroxylation in addition to NADPH-linked activity. The Km of 17 alpha-hydroxylase for NADH was calculated as 4.3 x 10(-5) M at pH 7.4 and 37 C. The optimal pH of 17 alpha-hydroxylase was 7.7 in the presence of NADH and 7.9 in the presence of NADPH. An additive increase in the amount of product 17 alpha-hydroxyprogesterone was observed by adding NADH to the incubation medium containing an excess amount of NADPH. The data suggest that there are two distinct active sites for 17 alpha-hydroxylation. Furthermore, the inhibition of NADH-linked 17 alpha-hydroxylation by carbon monoxide indicates the involvement of cytochrome P450 in the electron transport system for the NADH-linked 17 alpha-hydroxylation.

A further study of 17 beta-hydroxysteroid oxidoreductase in the human testis: mechanism of in vitro activation.

The mechanism and specificity of product activation of 17 beta-hydroxysteroid oxidoreductase for interconversion of androstenedione and testosterone were investigated using cell-free homogenates of human testes as an enzyme preparation. The reactions of oxidation and reduction catalyzed by the enzyme followed the Michaelis-Menten formula Addition of testosterone to the incubation medium resulted in increased Vmax and decreased the Km of reductase function of the enzyme. Androstenedione added to the incubation medium increased both Vmax and Km of the enzyme for the oxidase function. The apparent Km of the reductase function for [3H] androstenedione as a substrate in the presence of a fixed concentration of [14C] testosterone was found identical with the apparent Kd for the androstenedione as an effector (activator) for the oxidase function of the enzyme calculated from the simultaneous increase of [14C] androstnedione produced from the fixed concentration of [14C] testosterone. Similarly, the apparent Km of the oxidase function for [3H] testosterone as a substrate in the presence of a fixed concentration of [14C] androstenedione was identical to the apparent Kd for the testosterone as an effector (activator) for the reductase function of the enzyme calculated from the simultaneous increase of [14C] testosterone produced from the fixed concentration of [14C] androstenedione. None of the free 17-oxo and 17 beta-hydroxy steroids examined showed substrate activation or product inhibition of the enzyme except for androstenedione which showed product inhibition at high concentration. The present findings in combination with our previous data suggest the presence of two sites on the 17 beta-hydroxysteroid oxidoreductase of the human testis, one specific for androstenedione and the other specific for testosterone, each serving as an active site for the substrate and also as an activation site for the other active site.

In vitro steroidogenesis in testes of three infants, two with ambiguous external genitalia and one with true precocious puberty: Evidence for the presence of active 17 beta-hydroxysteroid oxidoreductase in immature human testes.

The present report investigated steroidogenesis in vitro in testis tissues obtained from two boys aged 8 months and 4 years with ambiguous external genitalia and male vagina, and a 4-year-old body with true precocious puberty. Histologically, testes of the former two boys are still immature and the testis of the last one contains differentiated Sertoli cells and primary spermatocytes, but no mature Leydig cells are recognized in any of them. In each testis, 17 beta-hydroxysteroid oxidoreductase is active for androstenedione in the presence of an excess amount of NADPH, while delta 5-3 beta-hydroxysteroid dehydrogenase and delta 4-steroid 5 alpha-reductase activities are limited. 17 alpha-Hydroxylase and C17--20 lyase are significantly active in each testis and are enhanced in the testis of the boy with precocious puberty. Although the testis tissue used in the present study may not be biologically normal and the number of cases investigated is still limited, the above results indicate that active 17 beta-hydroxysteroid oxidoreductase is present in immature human testes and that delta 5-3 beta-hydroxysteroid dehydrogenase may become active in the human testis at the advanced stage of the development of testicular function during the puberty.

Biological evaluation on glass ionomer cement.

Biological properties of a new dental cement of glass ionomer cement were compared with other types of conventional cement. The biological test was carried out by tissue culture method and animal examination. The culture cells showed weaker reaction to the glass ionomer cement than zinc oxide-eugenol or polycarboxylate cement. Pulp tissue reaction showed no significant difference between glass ionomer and zinc oxide-eugenol cement from in vivo experiments using monkeys.

Studies of the human testis. XI. Leydig cell clusters and levels of intratesticular testosterone and 20 alpha-dihydroprogesterone.

Testosterone concentration in testes of 23 men, ages 51 to 90, was determined as 490 +/- 172 ng/g tissue (M +/- S.D.) and 20 alpha-dihydroprogesterone concentration as 10.0 +/- 2.3 ng/g tissue (M +/- S.D.) in 18 men of the same patient group. Leydig cells were estimated by the number of Leydig cell clusters per tubule. There was a high correlation (r = 0.88, p less than .001) of the Leydig cell index with intratesticular testosterone. It is proposed that the Leydig cell cluster index provides a direct index of steroid biosynthetic activity in the human testis as reflected in intratesticular testosterone levels. Although there was no significant correlation of 20 alpha-dihydroprogesterone levels in the testis with either the Leydig cell index or intratesticular testosterone, the high level of 20 alpha-dihydroprogesterone compared to peripheral levels suggests its production by the testis.

Studies on thermolysin. I. Effects of chemical modifications on the activity of thermolysin.

1. When thermolysin was treated with a 100-fold molar excess of 2,4,6-trinitrobenzene-1-sulfonate at pH 8.0 and 37 degrees for 7 h, all 12 amino groups in the enzyme were almost completely trinitrophenylated. The fully trinitrophenylated enzyme still retained more than 80% of its original activity. The amino groups are therefore not essential for activity. 2. When treated with a 100- to 1,000-fold molar excess of N-acetylimidazole at pH 6.5 and 23 degrees for 2 h, thermolysin lost about 54% of its activity with concomitant acetylation of 21 tyrosine residues out of the total of 28 residues. The reaction did not easily proceed any further. This partially inactivated enzyme regained almost full activity upon treatment with hydroxylamine. These modified tyrosine residues are therefore not directly involved in the active site. 3. Thermolysin was not inactivated by treatment with about 100- to 150-fold molar excess of 2-hydroxy-5-nitrobenzyl bromide or dimethyl(2-hydroxy-5-nitrobenzyl)sulfonium bromide at pH 6.0 and room temperature for 1 h both in the presence and absence of 8 M urea. Thus the three tryptophan residues in the enzyme are not accessible to these reagents. When treated with a 4.3- to 43-fold molar excess of N-bromosuccinimide over tryptophan, the enzyme was inactivated to varying extents, depending on the reaction conditions used. In this case, the tyrosine residues appeared to be the most rapidly modified, but tryptophan and histidine residues were also modified with extensive inactivation at higher concentrations of the reagent. The presence of 8 M urea retarded the inactivation.

Studies of the human testis. VIII. Product activation of 17beta-hydroxysteroid oxidoreductase for testosterone.

Cell-free homogenates prepared from human testis tissue were incubated with either 1.3 x 10(-5)M[4-14C]testosterone and 2 x 10(-4)M NADP or 1.3 x 10(-5) [4-14C]nadrostenedione and 2 x 10(-4)M NADPH. Addition of non-radioactive androstenedione and testosterone to the incubation medium increased the formation of [14C]androstenedione from [4-14C]testosterone and [14C]testosterone from [4-14C]androstenedione, respectively, while addition of product nucleotide NADPH or NADP, respectively, decreased the conversions. The addition of androstenedione to the incubation medium changed the apparent optimal pH of 17beta-hydroxysteroid oxidoreductase for testosterone from 8.6 to 8.0. It appears likely that in a cell-free system human testicular 17beta-hydroxysteroid oxidoreductase not solubilized and still attached to membrane is activated by the product of the reaction catalyzed by the enzyme.

[Agenesis of internal carotid artery associated with aneurysm of anterior communicating artery (author's transl)].

A case of agenesis of the left internal carotid artery with an aneurysm of the anterior communicating artery was reported in detail. The patient was a 48-year-old man who had an episode of subarachnoid hemorrhage about 10 years ago. This time left heminumbness and motor weakness of the left lower extremity occured and he was brought our hospital. Right common carotid angiography, left retrograde brachial serial angiography and aortography demonstrated an agenesis of the left internal carotid artery, an aneurysm of the anterior communicating artery, tortuous megadolic hobasilar artery and anomalies on the circle of Willis such as absence of supra optic portion of the right anterior cerebral artery, and large left posterior communicating artery. We confirmed latter findings by operation. After successful neck clipping of the aneurysm, his clinical findings were remarkably improved.

The structure and function of ribonuclease T1. XX. Specific inactivation of ribonuclease T1 by reaction with tosylglycolate.

1. Ribonuclease T1 [EC 3.1.4.8] was inactivated by reaction with tosylglycolate (carboxymethyl rho-toluenesulfonate). At pH 5.5 and 8.0, alkylation of the gamma-carboxyl group of glutamic acid-58 appeared to be the predominant reaction and the major cause of inactivation by tosylglycolate, as in the case of the iodoacetate reaction, although the rate of inactivation was slower than that by iodoacetate. At pH 8.0, histidine residues were also alkylated to some extent. 2. The maximal rate of inactivation was observed at around pH 5.5 and the pH dependence of the rate of inactivation suggested the implication of two groups in the reaction, with apparent pKa values of about 3-4 (possibly histidine residue(s)). 3. In the presence of substrate analogs, ribonuclease T1 was markedly protected from inactivation by tosylglycolate at pH 5.5. The extent of protection corresponded to the binding strength of the substrate analog, except for guanosine. Ribonuclease T1 was much less protected from inactivation by guanosine than by 3'-AMP or 3'-CMP, which has a lower binding strength toward ribonuclease T1. This may indicate that glutamic acid-58 is situated in the catalytic site, at which the phosphate moiety of these nucleotides directly interacts. 4. Enzyme which had been extensively inactivated with tosylglycolate at pH 5.5 scarcely reacted with iodoacetate at pH 5.5, suggesting that these reagents react at the same site, i.e. glutamic acid-58. On the other hand, enzyme which had been inactivated almost completely with tosylglycolate at pH 8.0 still reacted with iodoacetate to some extent at pH 8.0, and the modes of reaction of tosylglycolate and iodoacetate toward ribonuclease T1 appeared to be somewhat different.
