A case of a suprasellar epithelial cyst in a 48-year-old man is reported. The cyst was lined with two different types of stratified epithelium: one was two to three or more layers of squamous cells with overlying ciliated columnar cells and goblet cells at the surface and the other was tyical stratified squamous epithelial cells. The tumor was neither a pure Rathke's cleft cyst nor a typical craniopharyngioma but was considered essentially to be a craniopharyngioma with ciliated columnar cells and goblet cells. Thus it might be considered a mixed form of these two tumors. This case indicates the close relationship between craniopharyngiomas and Rathke's cleft cysts.
Two types of virus particles, intracisternal type A and extracellular type C with budding, were detected in the same cells of BF osteosarcoma, its cultured cell lines, and their BFO tumors in CBA mice. The type C particles were approximately 100 microns in diameter. The buoyant density of the virions was 1.16g/ml in sucrose and 1.07 g/ml in Ficoll. A 72S RNA was demonstrated by gel electrophoresis, but no DNA was detected. Reverse transcriptase activity was also demonstrated in detergent-treated virions. Thus, the particles seem to be RNA virus. Cellular transformation and focus formation were observed after rat and mouse embryo cell monolayers were infected with the virus. The same kind of osteosarcoma was produced by inoculation of cloned transformed cells (BFOSV) of CBA embryo cells into CBA mice. Thus, the virus seems to be an oncornavirus.
Islet cells of adult rat pancreas, dissociated with EDTA-Dispase, were cultivated in Microtest wells for over 2 months. In our cultures, islet cells were free-floating and cohered with each other to reorganize histotypic aggregates resembling the nondissociated islets. Morphologically, excellent preservation of islet cells in the aggregates was confirmed during the culture using both light and electron microscopy. The function of islet beta-cells, as demonstrated by the synthesis and release of insulin, also was retained throughout the culture period. Islet beta-cells cultured for long periods exhibited better response to the short-term stimulation of theophylline than to a high concentration of glucose, as observed in the islets of fetal or newborn rat pancreas.
In 42 of 43 surgical patients who received a 10% soybeam oil emulsion (Intralipid), abnormal lipoprotein was detected in their plasma 1 to 2 days after the initial Intralipid infusion. This abnormal lipoprotein was proven to appear as a result of the infusion of soybean oil emulsion regardless of the patient's original diseases, age, sex, liver function, or concomitantly administered solutions. In addition, this abnormal -ipoprotein was found to have various similarities to lipoprotein-X (LP-X) which is found in plasma from patients with obstructive jaundice or familial lecithin:cholesterol acyltransferase deficiency. Therefore, this abnormal lipoprotein was tentatively named LP-X--like substance (LP-X-LS). A comparison of the properties of LP-X and LP-X-LS was performed and the following results were obtained: (1) LP-X-LS migrated toward the cathode on Bacto-Agar gel electrophoresis similarly to LP-X; (2) the stability of LP-X and LP-X-LS against heating and freezing were almost equal under various conditions; (3) LP-X-LS could be absorbed by anti--LP-X serum; (4) LP-X-LS existed in low density fraction (d = 1.063) separated by ultracentrifugation from plasma; (5) electron microscopic study of low-density lipoprotein particles from LP-X-LS positive plasma revealed that LP-X-LS had a similar ultrastructure to LP-X. From these results it is suggested that LP-X-LS is an abnormal lipoprotein quite similar to LP-X.

A rapid and simple R.I.A. method for serum unconjugated estriol in pregnancy had been studied for its usefulness as a routine obstetrical test to assess fetal well-being. Diluted serum samples or known amounts of standard estriol were applied with 125 I-estriol on pre-packed minicolumns of Sephadex G-25, followed by appropriately diluted antiserum. During incubation, the columns were counted in an Auto Well gamma-System to read the total counts (T). After a 90 minute incubation period, the columns were eluted with a buffer solution to remove the bound fraction, and the free estriol remaining in the columns was counted (F). Two quality control samples and sera from 74 pregnant women were analysed by this new method, and the results were compared to those obtained by the conventional radioimmunoassay method. The new column method is simple, specific and accurate. Results can be calculated within 4 hours after 30 sample sera are brought into the laboratory. Within and between assay coefficients, variations are 12.6 and 13.5%, respectively. This method correlates well with the results of the conventional method (r=0.7088, p less than 0.001), which requires almost 2 full days to process the same numbers of samples. Results of clinical cases as well as advantages of measuring free-circulating estriol in pregnancy are discussed.

An oat cell carcinoma occurring in the esophagus of a 62-year-old man is described. A strong argyrophilia of the oat cells together with the presence electron microscopically of neurosecretory type granules in their cytoplasm suggests a derivation of this tumor from the cells of the APUD series, and the tumor is diagnosed as apudoma. An aberrant columnar epithelium on the esophageal surface is presumed to be the source of its origin. A rapid postoperative course of the patient despite a rather circumscribed growth pattern of the primary tumor appears to characterize the clinical aspect of this case. An apparent accumulation of the cases with esophageal apudoma in Japan is discussed.

Biopsy specimens, mostly lymph nodes, removed under aseptic conditions, were emulsified and the homogenate was inoculated in various media, which were incubated aerobically or anaerobically. Only Propionibacterium acnes was isolated in high incidence and the highest concentration of bacteria was 6.5 X 10(5) per g tissue: 28 and 31 out of 40 lymph nodes were positive on solid and on liquid media, respectively. P.acnes was also isolated from control non-sarcoidosis lymph nodes: 4 and 8 out of 14 were positive on solid and on liquid media, respectively. The highest concentration of bacteria was 1 X 10(4). A bacterial component derived from P. acnes was used for the passive hemagglutination test to estimate antibodies against the bacterium and also for skin test on patients. These results showed only slight differences between patients with sarcoidosis and control patients. No other bacteria or fungi were isolated.

A comparative study was made by the double-blind technique in order to make clear the usefulness of pivmecillinam in the treatment of intractable complicated urinary-tract infections using amoxicillin as a reference drug. Pivmecillinam was given in dosage of 400 mg (potency) per day which was one-fifth the dose of amoxicillin 2,000 mg (potency) per day. In global judgement, pivmecillinam was found superior to amoxicillin. It showed a "significant" superiority over amoxicillin for the treatment of, among others, the urinary-tract infections after prostatectomy, which are intractable diseases. When evaluated by symptoms, pivmecillinam improved bacteriuria "significantly" better than amoxicillin. When seen by causative organisms, the pivmecillinam treatment was "significantly" superior to the amoxicillin treatment against E. coli infections. Pivmecillinam was active against amoxicillin-resistant E. coli. Incidence of adverse reactions was less frequent with pivmecillinam than with amoxicillin. These results indicate that pivmecillinam is a drug of high usefulness for the treatment of intractable complicated urinary-tract infections when evaluated using amoxicillin as a reference drug.

The ocular involvement of sarcoidosis is particularly important in Japan, because 1. the frequency of the eye involvement is as high as 20 to 40%, higher than in other countries- 2. the presence of this involvement proved to be an influential factor relating to the whole sarcoidosis lesions of a patient, and 3. the patients with this involvement will become blind unless adequately treated. In diagnostic routines of ocular sarcoidosis, chest X-rays will give the most useful information of the disease. If the chest X-ray findings are normal, it is not easy to make a diagnosis of a case whose ocular findings are very suspicious of sarcoidosis. This paper will give several case experiences in which we could successfully make a diagnosis, using mediastinoscopical biopsies though the X-rays had no findings.
Leukaemia cells of C12/0 line, derived from Gross virus-induced W/Fu rat thymoma, have been successfully maintained in a completely synthetic, serum-free RPMI 1640 medium. The cells propagated as good as in serum-supplemented medium, though they could not grow from a low cell density. Cells adhered to the substratum and did not survive. This problem was overcome by use of silicone-coated substratum, or of microexudate from C12/0 cells adsorbed on the substratum. The cells underwent degeneration soon after they reached their maximum density. C12/0 cells released macromolecules might possibly have both growth-promoting and toxic effect on the cell itself.
