[Clinical studies on varicocele. (1). Findings on semen and testis].

The semen and testicular histology of 39 patients with varicocele on the left side, and who had high ligation of the internal spermatic vein at our Department between 1975 and 1981 was examined. The grade of varicocele was neither related to density nor motility of the sperm, nor to the score count of the left testis. In 40% of the patients, both density and motility of the sperm improved after the surgical treatment. In 80% of the patients, the left testis weighed less than the right testis. In most of the patients, the score count of both testes was lower than normal, and was found to decrease remarkably in testes weighing less than 10 g. In 30% of the patients, the score count of the left testis was lower than that of the right testis. A positive correlation was recognized between score count and testicular weight, and no correlation was found between the score count of the right testis and sperm density.

Inhibition of DNA synthesis and cell cycle in Chinese hamster cells by sterol-binding polyene amphotericin B.

Amphotericin B, a sterol-binding polyene antibiotic, was found to inhibit DNA synthesis more than protein or RNA synthesis of asynchronous cultures of Chinese hamster V79 cells. DNA synthesis in the asynchronous V79 cells was inhibited to 40--60% of the control activity in the presence of 50 micrograms/ml amphotericin B. However, addition of 50 micrograms/ml of polyene immediately after to onset of DNA synthesis (early S phase) caused a drastic reduction of DNA synthesis (below 10--20% of the control in synchronized V79 cells, whereas the inhibition was much lessened when the polyene was added 1 h later (middle S phase). In contrast, there was no inhibition of DNA synthesis by amphotericin B in an amphotericin-B-resistant (AMBR) clone that was derived from V79. Flow microfluorometry analysis confirmed that a large number of asynchronous V79 cells were arrested in the G1 phase of the cell cycle when treated with lower dose of amphotericin B. A higher dose of the polyene antibiotic also accumulated cells at the G2 (or at both S and G2) phase as well as the G1 phase. Morphological studies by scanning electron microscope showed an increased number of V79 cells with decreased microvilli in V79 cells treated with amphotericin B.

Human erythroid cell lines derived from a patient with acute erythremia.

Three continuous human cell lines, designated KMOE, derived from a patient with acute erythremia (Di Guglielmo's disease) are reported. The cell lines are the cultures of (1) bone marrow cells, (2) peripheral blood cells, and (3) cells from a tumor developed into an athymic nude mouse after transplantation of the cultured bone marrow cells. Cells of all three lines show morphology of immature erythroblast and have i(17q) marker chromosome. They are negative for both Philadelphia chromosome and Epstein-Barr virus nuclear antigen. Although all KMOE cells in suspension culture are benzidine-negative, benzidine-positive cells are found within colonies formed in semi-solid culture media. The relative number of colonies with benzidine-positive cells is increased when sodium butyrate is added to the culture.

Development of circadian rhythms in serum hormone levels in the immature female rat.

Diurnal variations in concentrations of luteinizing hormone (LH), follicle-stimulating hormone (FSH), thyroid-stimulating hormone (TSH), corticosterone, estradiol-17 beta and estrone in serum were investigated in 27- to 35-day-old female rats maintained on a 14-hour light and 10-hour dark schedule. Groups of 6-8 rats were decapitated at 3-hour intervals during a 24-hour period. Hormones were measured by radioimmunoassay. LH levels were highly variable over a 24-hour period in rats younger than 32-33 days of age, while in 34- to 35-day-old animals the levels were less variable and a moderate LH peak was seen at 24.00 h. A distinct FSH diurnal rhythm with the peak at 15.00 h was demonstrated in rats older than 32 days of age. The 27- to 28-day-old rat had a diurnal TSH rhythm with the peak at 12.00 h, but older rats had a rhythm with two peaks at 24.00 and 9.00 h. The diurnal corticosterone rhythm analogous to the adult's was established at 32 days of age, although a significant variation having a peak during the dark period appeared earlier. Similarly, a distinct estradiol rhythm appeared at 29 days of age with the peak at 12.00 h, followed by a change in the peak phase afterwards. Serum estrone levels showed a manifest variation in the 27- to 28-day-old rat, but no variation was observed in the older rats. These results not only confirm the previous reports on the existence of pulsatile LH secretion, and corticosterone and estradiol diurnal rhythms, but also demonstrate the existence of diurnal rhythms in FSH, TSH and estrone levels in the immature female rat. Furthermore, it is possible that the rhythm once developed may change in phase, or even disappear as puberty approaches.
