Involvement of c-myc oncogene in lymphoma cell lines with no detectable chromosome rearrangement of band 8q24.

Two lymphoma cell lines of B-cell type were established from Japanese patients with diffuse small noncleaved cell lymphoma. Cytogenetic analysis revealed a 14q+ marker chromosome in both cell lines, and a t(8;14)(q24.1;q32.3) seemed most likely to have occurred. The chromosome 8 pair, however, had no abnormalities. Molecular analysis demonstrated c-myc amplification lacking gross rearrangement in one cell line and genetic rearrangement of c-myc at the first intron as well as aberrant sizes of c-myc mRNA in the other cell line. In the latter case, it is possible that a t(8;14)(q24.1;q32.3) was buried in an unrecognized complex translocation. A combination of cytogenetic and molecular studies to determine the precise nature of the 14q32 translocation is discussed.

Involvement of bcl-2 gene in Japanese follicular lymphoma.

A t(14;18) (q32;q21) chromosome translocation is closely associated with the follicular lymphoma, which is prevalent in the United States, and the t(14;18) causes the juxtaposition of a bcl-2 gene on chromosome 18 with an immunoglobulin heavy-chain gene locus on chromosome 14. Genomic DNAs from 30 Japanese patients with follicular lymphoma were examined for the molecular features by Southern blot hybridization. Using probe b for the major breakpoint cluster region of a bcl-2 gene, the rearrangements were detected in eight patients. Six of the eight patients had breakpoints located within the major breakpoint region, while two had breakpoints outside this cluster region but within the region of the 7.5-kb SstI fragment containing the probe b sequence. In two patients, pFL-2 probe detected the bcl-2 gene rearrangements that occurred near or within the minor breakpoint cluster region. These ten patients had a rearranged JH-containing fragment that migrated with the rearranged bcl-2 fragment. In the other 20 patients, these two chromosome 18-specific DNA probes did not detect the bcl-2 rearrangements. Compared with studies performed in the United States, the statistical analysis indicates a significant difference in frequency of the bcl-2 gene rearrangements near or within the major breakpoint cluster region (P = 0.0027) and the minor breakpoint cluster region (P = 0.029). However, the distribution difference of these events was not significant.

Difficult deflation of Probe balloon due to twisting the system.

While using the balloon-on-a wire device, we experienced a case of difficult deflation of the Probe balloon system. Blockage of the balloon system was possibly caused by twisting the system to reach and pass the lesion in the branch of left circumflex coronary artery. The duration of inflation was 20 min, causing prolonged ischemia.

Multiple coronary thrombosis in a patient with thrombocytosis.

A 59-year-old woman was admitted with acute inferior myocardial infarction. Cardiac catheterization revealed total occlusion in the right coronary artery and thrombus-like filling defect in the left anterior descending coronary artery. With simultaneous intracoronary urokinase infusion to the right and left coronary arteries, the right coronary artery became patent and the thrombus in the left anterior descending coronary artery was released to the distal coronary artery. Thrombocytosis was disclosed by laboratory examination. The relationship between myocardial infarction and thrombocytosis is discussed.

Presence of angina pectoris before acute myocardial infarction and degree of residual stenosis after coronary thrombolysis.

This study tested the hypothesis that the degree of residual stenosis after coronary thrombolysis reflected that of original stenosis presented by symptom of angina before acute myocardial infarction (AMI). The relation between the presence of angina before AMI and the degree of residual stenosis after coronary thrombolysis was observed in 57 patients with successful coronary thrombolysis for AMI. Patients with significant coronary artery stenoses other than the artery responsible for AMI were excluded from this study. In 22 patients with chronic angina for 2 weeks or longer before AMI, 14 patients had a residual stenosis of 75% or more and 8 patients had a residual stenosis of less than 75%. In 35 patients with angina for less than 2 weeks or not at all before AMI, 9 patients had a residual stenosis of 75% or more, and 26 patients had a residual stenosis of less than 75% (p less than 0.01). In the course of progression of coronary artery disease, some patients had AMI without severe underlying stenosis and others with severe underlying stenosis. Patients with chronic angina before AMI might tend to have AMI with acute occlusion superimposed on the severe organic stenosis. Patients without chronic angina before AMI would be more at risk for AMI caused by acute occlusion without underlying severe stenosis.

Capsaicin-induced release of calcitonin gene-related peptide from dorsal horn slices is enhanced in adjuvant arthritic rats.

The capsaicin (3 microM)-evoked release of the immunoreactive calcitonin gene-related peptide (iCGRP) from dorsal-half slices of the rat lumbar spinal cord was substantially reduced in slices from rats which had undergone dorsal rhizotomy. This suggests that the release is elicited mainly from the nerve terminals of primary afferents. The release was significantly increased in slices from adjuvant arthritic rats compared with those from control rats. This suggests that the amount of iCGRP in a capsaicin-sensitive pool of the nerve terminals of primary afferents is elevated by adjuvant arthritis. Since adjuvant arthritis is known to cause an enhancement of biosynthesis and axonal flow of iCGRP in primary sensory neurons, the present results imply an increased release of iCGRP from the central terminals of the primary afferents in the adjuvant arthritic rat.

Effect of phospholipase A2 inhibitors on mouse T lymphocytes. I. Phospholipase A2 inhibitors exert similar immunological activities as glycosylation inhibiting factor.

Since our previous experiments suggested that glycosylation-inhibiting factor (GIF) is a phosphorylated derivative of a phospholipase inhibitory protein, we determined whether other well-known phospholipase inhibitors may have similar biological activities. The results showed that phospholipase A2 (PLA2) inhibitors, such as recombinant human lipocortin I and ONO-RS-082, could switch T cell hybridoma 12H5 cells from the formation of glycosylated IgE-binding factors (IgE-BF) to the formation of unglycosylated IgE-BF, whereas neomycin, a phospholipase C inhibitor, failed to affect the nature of IgE-BF formed by the cells. The minimum concentrations of lipocortin I and ONO-RS-082 required for switching the 12H5 cells to the formation of unglycosylated IgE-BF were comparable to or less than IC50 of the inhibitors for PLA2. The ability of partially purified GIF to switch the 12H5 cells to the formation of unglycosylated IgE-BF was markedly enhanced by treatment of the preparation with alkaline phosphatase. It was also found that lipocortin I and ONO-RS-082, but not neomycin, facilitated the generation of GIF-producing T cells. When spleen cells of ovalbumin (OVA)-primed BDF1 mice were stimulated with homologous antigen and the activated T cells were propagated by recombinant IL-2 in the presence of GIF, lipocortin I, or ONO-RS-082, T cells obtained in the cultures constitutively produced their own GIF. Antigenic stimulation of the T cells induced the formation of unglycosylated IgE-BF and GIF with an affinity for OVA.(ABSTRACT TRUNCATED AT 250 WORDS)

Production of sterile transcripts of C gamma genes in an IgM-producing human neoplastic B cell line that switches to IgG-producing cells.

A human neoplastic B cell line SSK41 that expresses IgM on its surface switches spontaneously to IgG-producing cells. The SSK41 line contains a single immunoglobulin heavy-chain locus, the constant region (C) genes of which retain the germline configuration. The IgG-producing SSK41 line was purified by sorting, and shown to have undergone S-S recombination with deletion of the C mu gene. This line produced secretory and membrane-bound forms of gamma-chain mRNA. From cDNA libraries of a mixed population of IgM+/IgG+ SSK41 cells, we have isolated cDNA clones encoding the mature membrane-bound and secretory forms of the mu and gamma 1 heavy chains, all of which share the same variable region sequence. cDNA clones containing the mature gamma 3 chain were identified as well. We also isolated cDNA clones containing C gamma 1 and C gamma 3 sterile transcripts from the SSK41 line. These sterile transcripts contained additional exon sequences designated 'I' which were localized upstream of the C gamma 1 and C gamma 3 switch regions and homologous to murine counterparts. The I sequences were precisely spliced to the 5' ends of the corresponding C gamma exon sequences. These features of germline CH transcripts, i.e. the isotype specificity to class switching, location of exons, and sequences per se, are highly conserved between man and mouse.

[Effects of flutropium bromide, a new antiasthma drug, with repeated administration on bronchomotor response and hepatic drug metabolizing enzymes].

Effects of flutropium bromide, a new antiasthma drug possessing the quarternary ammonium structure of atropine derivatives, were investigated on the bronchodilatory activity with repeated inhalations in guinea pigs and on the hepatic drug metabolizing enzyme activities with repeated i.v. administration in rats. Single inhalation of flutropium bromide (0.03%) inhibited the ACh-induced bronchoconstriction without changing the fall in blood pressure induced by ACh. Flutropium bromide (0.03%) was inhaled for 5 min a day by placing the animal in an inhalation box, successively during periods of 14 and 28 days. The bronchodilatory effect of flutropium bromide after 14- or 28-day repeated inhalations was almost the same potency as that after single inhalation. Twenty-eight-day repeated inhalations did not change the body weight curve in guinea pigs; and in addition, 14-day repeated i.v. administration did not change the hepatic drug metabolizing enzyme activities in rats. From the above results, it is indicated that flutropium bromide causes neither reduction in response nor cumulative effect after repeated inhalations, and that the agent maintains the bronchodilatory effect without change in the hepatic drug metabolizing enzyme activities.

Inhibitory effects of ONO-3307 on various proteases and tissue thromboplastin in vitro and on experimental thrombosis in vivo.

The effect of ONO-3307 (4-sulfamoyl phenyl-4-guanidinobenzoate methanesulfonate), a new protease inhibitor, was studied on various proteases in vitro and in an experimental thrombosis model in vivo. ONO-3307 competitively inhibited trypsin, thrombin, plasma kallikrein, plasmin, pancreatic kallikrein and chymotrypsin; and their Ki values were 0.048 microM, 0.18 microM, 0.29 microM, 0.31 microM, 3.6 microM and 47 microM, respectively. In addition, ONO-3307 inhibited both elastase release from N-formyl-Met-Leu-Phe (fMLP)-stimulated leukocytes and tissue thromboplastin release from endotoxin-stimulated leukocytes. To examine the effects of ONO-3307 on disseminated intravascular coagulation (DIC), we developed an experimental thrombosis model. ONO-3307 (10 mg/kg/hr) completely inhibited the deposition of radioactive fibrin in kidney and lung. Gabexate mesilate (50 mg/kg/hr) was also effective in this model, but the effect of nafamostat mesilate was unclear. These results indicate that ONO-3307 exhibits a wide range of inhibitory effects on various proteases, and ONO-3307 may be useful for the treatment of protease-mediated diseases such as thrombosis and DIC.

Effects of exercise, cold, and immobilization stresses on gamma-glutamyltransferase activity in rat kidney.

Effects of acute and chronic stresses (exercise, cold, and immobilization) on gamma-glutamyltransferase (gamma GT) activity in the rat kidney were investigated. In the extramicrosomal fraction there were significant decreases in acute-exercised rats but significant increases in cold-adapted and immobilized rats. In the microsomal fraction, on the other hand, the gamma GT activity of acute-exercised rats increased definitely. The results suggest that different types of stresses have different effects on gamma GT activity in the rat kidney.

The adhesion mechanism of dental adhesive resin to the alloy. Adhesive ability of dental adhesive resin to the cleaned metal surface obtained by hydrogen gas reduction method.

With the cleaned metal surface without passive film or an oxide layer, chromium, cobalt, and iron show excellent bonding strength, indicating values similar to 18-8 stainless steel. Therefore the existence of a passive film and oxide layer at the interface metal/resin is not essential for metal-resin bonding. The adhesive ability decreases in the order: Cr congruent to Co congruent to Fe greater than Cu greater than Ag greater than Au congruent to Ni greater than Pt greater than Pd.

Adhesion mechanism of polyelectrolyte cements to tooth structure--polyelectrolyte behavior of the cement polymers obtained by potentiometric titration in the presence of calcium ion.

Potentiomeric titration of aqueous solutions of polyacrylic acid and commercial polyelectrolyte cement polymers with sodium hydroxide solution was carried out in the presence of different concentrations of Ca2+. Polyelectrolytes all behave as weak acids without the coexisting Ca2+. However, in the presence of Ca2+, in amounts over one-half of the equivalent amount to the carboxyl group in the polymer, they have a strong acid-like behavior. This means that the carboxyl groups in the polymer chain tend to react strongly with coexisting Ca2+ as they are partially neutralized by the alkaline solution. This is also strong evidence supporting polyelectrolyte cement as an adhesion mechanism to tooth structure during cementation.

Corrosion and tarnish of dental silver-based alloys in 0.1% Na2S and Ringer's solutions. (Part 1) Electrochemical study.

The corrosion behavior of three silver-based alloys, Ag-Sn-Zn, Ag-In, and Ag-Pd-Cu, were investigated by potentiodynamic anodic polarization analysis and the polarization resistance method in 0.1% Na2S and Ringer's solutions. The corrosion activity of the Ag-Sn-Zn alloy was higher in Ringer's solution than in the 0.1% Na2S solution. In contrast, the corrosion rate of the Ag-Pd-Cu alloy was approximately 500 times higher in the 0.1% Na2S solution than in Ringer's solution. The results show that the generally accepted concept that tarnish is merely a surface discoloration due to the deposition of insoluble products is inadequate. The alloy discolors while being severely attacked in the presence of sulfides. For the Ag-In alloy, the corrosion activity in the 0.1% Na2S solution was as high as in Ringer's solution. These silver-based alloys exhibit different electrochemical activities in different solutions. The test solutions for corrosion tests must be carefully chosen for each alloy system through a screening test to replicate the predominant corrosion reaction proceeding in the oral environment.

Calphostins (UCN-1028), novel and specific inhibitors of protein kinase C. I. Fermentation, isolation, physico-chemical properties and biological activities.

A novel complex of calphostin (UCN-1028), which specifically inhibits protein kinase C (PKC) has been isolated from the culture broth of a fungi Cladosporium cladosporioides. Purification of individual components was carried out by silica gel, non-porus resin Diaion HP-20SS and Sephadex LH-20 chromatography, leading to isolation of five closely related components. A, B, C, D and I. Calphostins showed cytotoxic activities against various tumor cells, and these cytotoxicities were proportional to their inhibitory activities against PKC.

In vitro study on the corrosion behavior of three commercial Ag-Pd-Cu-Au alloys in Ringer's and 0.1%Na2S solutions.

The corrosion resistance of three commercial Ag-Pd-Cu-Au alloys was estimated in Ringer's and 0.1% Na2S solutions by electrochemical techniques and surface analyses. In Ringer's solution, the three alloys showed high corrosion resistance and there was no significant difference in the anodic polarization characteristics of the three alloys. In the 0.1% Na2S solution, the Alloy A which had the lowest noble metal content (Au + Pd) exhibited the highest anodic reactivity with the largest amount of corrosion product on the alloy surface. It was determined that the Ag-rich phase of Ag-Pd-Cu-Au alloy was preferentially attacked to form Ag2S corrosion product. The polarization resistance data showed that the corrosion rate for Alloy A in 0.1% Na2S solution was determined to be 500 times higher than that in Ringer's solution. The corrosion rate of the alloy in the freely corroded condition can be estimated quantitatively and precisely by measuring the polarization resistance.
