Unitary current through sodium channel and anomalous rectifier channel estimated from transient current noise in the tunicate egg.

1. The noise included in the Na current and the anomalous rectifier current was measured in intracellularly perfused eggs of Halocynthia roretzi Drashe under voltage clamp conditions. 2. The time-dependent component of the point-by-point ratio between the ensemble mean and the variance of successive current traces was the mirror image of the time-dependent component of the mean current itself. This can be explained by assuming an open or closed conductance state for each ionic channel, and permits estimation of unitary currents and channel number. 3. The unitary Na currents showed a potential dependence similar to that of the instantaneous currents. The unitary conductances (gamma) at zero membrane potential were 7.4 and 3.3 pmho in sea water with 400 and 100 mM-Na, respectively, and 400 mM-intracellular K. With 400 mM-Na intracellular perfusate and 400 mM-Na sea water, gamma was 15.7 pmho. In spite of the large differences in the unitary conductances, the single channel permeability for Na ions remained constant (1.15 x 10(-14) cm3 sec-1). The density of Na channels was 0.60/micrometer2. 4. The unitary currents calculated for the anomalous rectifier showed a potential dependence similar to that of the zero-time extrapolated estimates of the K inward currents. The anomalous rectifier unitary conductance was 5.5 and 6.9 pmho for sea water with 50 and 100 mM-K, respectively. The density of these channels was 0-035/micrometer2.

Degradation of histamine in the presence of ascorbic acid and Cu2+ ion; involvement of hydrogen peroxide.

In the presence, but not in the absence of Cu2+, ascorbate decomposes histamine in citrate phosphate buffer (pH 6.5) at 37 degrees, but not at 0 degrees. The breakdown is completely inhibited by catalase, but only slightly by superoxide dismutase, and scavengers of OH. like benzoic acid, ethanol or potassium iodide. A1 O2 scavenger, alpha-tocopherol also did not show significant effects on the reaction. On the other hand, addition of H2O2 to the reaction mixture markedly enhances the rate of histamine breakdown induced by ascorbate or ascorbate-Cu2+ systems. However, H2O2 alone cannot breakdown histamine even in the presence of Cu2+. Histamine breakdown induced by ascorbate appears to be dependent upon the autooxidation of this vitamin. From these results and the findings reported by Chatterjee et al. that the products of its aerobic oxidation, dehydroascorbic acid and H2O2 were ineffective in reacting with histamine in the presence of Cu2+, it is concluded that the combination of H2O2 and the intermediate of ascorbate oxidation (monodehydroascorbic acid or other unstable species), both of which are produced during the autooxidation of ascorbate, plays a major role in the histamine transformation by ascorbate-Cu2+ system.

A new adrenal computer imaging technique using dual-radioisotopes.

Computer processed adrenal imaging using dual-radioisotopes, 6 beta-iodomethyl-19-nor-cholest-5(10)-en-3 beta-ol-131I and 99mTc-phytate was performed in 12 patients with primary aldosteronism and 4 with Cushing's syndrome due to adrenocortical tumor. Adreno-photoscanning and hepato-photoscanning were performed in the same position 2-4 days following intravenous administration of radiocholesterol. The scintigraphic information was stored on cassettes and scan subtraction and a digital-computer method for data smoothing were performed on an oscilloscope. The tumor site could be determined in all cases until day 4 by this computer processed image.

Extravesical malacoplakia--possibly originated from a superficial part of the renal cortex.

A 61-year-old woman was found to have a right flank mass. Radiologically, the mass was thought to be of renal origin. Surgically, however, the tumorous lesion adhered tightly not only to the cortical surface but extended to the retroperitoneum and psoas muscle. Pathological examination confirmed the lesion to be malacoplakia which was characterized by the presence of von Hansemann cells and Michaelis-Gutmann bodies. Despite the effort made to focus on the initiating site of the tumor, it was considerably difficult to decide whether it originated from either an extremely superficial part of the renal cortex or renal capsule or retroperitoneum itself. Histochemical and electron microscopical investigations on the von Hansemann cells and Michaelis-Gutmann bodies were made. The pathogenesis of malacoplakia was briefly discussed with particular emphasis on its possible occurrence in the patients under various therapeutic regimes.

Effect of streptococcal preparation (picibanil) on the postoperative rise in serum alanine aminotransferase activity in patients with urogenital cancer.

The effect of Picibanil, a streptococcal agent, on the development of liver injury after operations for urogenital cancer was studied retrospectively in the light of serum alanine aminotransferase (ALT) activity. The series comprised 32 cases receiving Picibanil and 33 controls with otherwise comparable clinical backgrounds. Picibanil reduced the incidence of postoperative ALT rise over 50 U/l within 6 weeks but increased it thereafter. The increase in ALT activity after 6 weeks was relatively small and was seen more often in patients given blood transfusions. It was interpreted as retardation and suppression of ALT rise and as being related to the induction of interferon or to immunopotentiation. Other antihepatotoxic effects of Picibanil, due to its antioxidant activity, for example, may also account for the prevention of the early postoperative rise in ALT activity.

Primary testicular plasmacytoma: a case report.

A 73-year-old man with a left testicular tumor is presented. Orchiectomy was performed and the tumor was diagnosed as a plasmacytoma. There has been no bone lesion or immunoglobulin abnormality during a follow-up period more than one year. For several reasons, the tumor was regarded as a primary plasmacytoma of the testicle. The literature concerning testicular plasmacytoma is reviewed briefly.

The effects of intravesical instillation of Thio-Tepa on the recurrence rate of bladder tumors.

Seventy-eight patients were treated with intravesical instillation of Thio-Tepa in an attempt to prevent postoperative recurrences of bladder tumors. Fifty-six patients who were given no preventive treatment against recurrences were taken as the control group. The patients in this series presented at the Okayama University Hospital between 1961 and 1976 and only the first recurrence after the primary operation was taken into consideration. There was no significant difference in the recurrence rates of the control and instillation groups.

Nucleotide sequence of the region required for maintenance of colicin E1 plasmid.

Plasmids carrying various portions of colicin E1 plasmid (ColE1) DNA have been isolated in an attempt to determine the regions of ColE1 DNA which are required for maintenance of the plasmid in bacteria. To construct the plasmids, the DNA of a ColE1 derivative that contains a gene which controls ampicillin resistance was cleaved by the restriction endonuclease HaeII. The digestion products were joined by T4 DNA ligase and then used to transform bacteria to ampicillin resistance. The plasmid derivatives obtained in this way were always composed of certain HaeII segments. These contain approximately 10% of the ColE1 genome and include the origin of replication of ColE1. We presume that the region of ColE1 which is common to all these derivatives is required for maintenance of the plasmid. After a description of these results, the nucleotide sequence of this region is presented, and possible roles of the region in plasmid replication and maintenance are discussed.
