Identification and characterization of the genes encoding three structural proteins of the Thermoproteus tenax virus TTV1.

Three structural proteins, TP1, TP2 and TP3, of the virus TTV1 of the thermophilic archaebacterium Thermoproteus tenax strain Kra1 were mapped within the viral genome by locating the amino-terminal amino acid sequences of these proteins in the TTV1 DNA sequence. The derived amino acid sequences comprise 113, 139 and 160 amino acid residues, respectively. All three proteins are hydrophobic. The three genes are not linked, but transcribed in the same direction. No Shine-Dalgarno sequences are found in the vicinity of the initiation codons of these three genes. By Northern analysis, four mRNAs of 0.5 kb, 0.8 kb, 1.1 kb and 1.8 kb in size were found to be encoded in the region and in the vicinity of the genes, the shortest one (t1) encoding TP3 and the longest one (t4) encoding TP2. No transcript from the region encoding TP1 has been found so far. A transcriptional start site was mapped for the transcript t1. Its upstream sequence was similar to the putative consensus sequence for archaebacterial promoters.

Amino Acid Sequence

A physiological comparison of the short-leg walking cast and an ankle-foot orthosis walker following 6 weeks of immobilization.

Iatrogenically induced weakness, stiffness, and functional impairment associated with classical methods of short-leg cast immobilization have long been recognized. This study compares the effects of "controlled" immobilization with an ankle-foot orthosis (AFO) and short-leg cast (SLC) immobilization for 6 weeks. A significant reduction in loss of range of motion, muscular strength, and muscular atrophy, ie, functional impairment, at the time of cessation of immobilization was noted when an AFO was used. The additional advantages of an AFO are the elimination of multiple cast changes including economic benefits, ease in the assessment of functional improvement during treatment, and convenient adjustment for swelling and atrophy.

[Changes in the age of illness onset in cancer of the hypopharynx and larynx since 1960. The epidemiologic context in Austria].

Between 1960 and 1987, 1567 laryngeal and hypopharyngeal carcinomas were diagnosed at the First ENT Clinic of Vienna University. Patients under 50 years of age increased steadily in number and proportion. Three subgroups of carcinomas were considered: glottic (including subglottic), supraglottic and hypopharyngeal. A total of 1442 cases could be studied in detail. In the supraglottic and hypopharyngeal groups the proportion under 50 years of age rose significantly, whereas in the glottic cases no significant trend is discernible. The subgroups were standardized in 10-year age groups to the population of eastern Austria. The incidence was constant in females. In males two opposite trends were found: in glottic tumours the proportion of middle-aged patients shows a slight decrease, whereas older age groups show a significant decrease. In hypopharyngeal carcinoma, younger age groups show a highly significant increase and the older age groups a slight decrease. Supraglottic carcinoma was intermediate: in younger age groups, a slight increase was found compared with a marked decrease in older ages. The age-specific regional mortality showed a pattern similar to that of the supraglottic and hypopharyngeal tumours seen at our clinic. The alcohol and tobacco consumption of all male patients between 1977 and 1983 was reviewed. Heavy smoking was about equally frequent for all tumour sites. High alcohol intake was more frequent in supraglottic and hypopharyngeal than in glottic tumours, and more frequent in younger than in older patients. Tobacco smoke acts as a carcinogen in the long run, and this effect can be enhanced by heavy drinking.(ABSTRACT TRUNCATED AT 250 WORDS)

[Incidence and age at onset of cancers of the mouth and oropharynx since 1960. The epidemiologic context in East Austria].

Between 1960 and 1987, a total of 709 carcinomas of the floor of the mouth, tongue and oropharynx (BOPC) were diagnosed at the first ENT Clinic of the University of Vienna (555 men, 154 women). Patients under 50 years of age increased steadily in number of proportion. Age-specific rates in men based on the actual population of eastern Austria showed a highly significant increase in younger patients and a slight decrease in older patients. Crude regional mortality increased considerably in men during the whole period, and in women since 1982. The evolution of age-specific mortality in men was similar to that of the incidence at our clinic. In women the increase affected all ages. The rate of tobacco and alcohol consumption of male patients between 1977 and 1983 was compared with that of patients with chronic otitis and hypopharyngeal carcinoma. Smoking and drinking rates were substantially higher in the BOPC group than in the otitis group, and their distribution differed from that of the hypopharyngeal cancer group: heavy consumption of both tobacco and alcohol was equally frequent, but smoking rates were lower and heavy drinking more frequent in the BOPC group. High consumption in BOPC patients was more common in patients under 60 years of age. The increasing incidence of the disease and the shift to younger patients may be linked with the alcohol component within the field of influence of combined tobacco and alcohol abuse.

[Incorporation of work disability into the prolonged life expectancy process].

The various reasons of deficiency, as e.g., mortality, invalidity and notification of illness result in a decrease of the potential of work of the labour. Because of the mutual influence of these reasons, an isolated assessment of them only can be performed in a restricted manner. The loss of working years in male workers and clerks of the GDR (SV FDGB 1982) is calculated and assessed on the basis of the various reasons of deficiency.

[The behavior of polychlorinated biphenyls (PCB) in water purification plants. I. Mathematical balancing in a model purification plant].

The behaviour of 39 PCB-isomers will be tested in a model sewage plant. Their distribution is described mathematically and the quantities of the partial streams are calculated. Because there is no loss of PCB through the ventilation of the activated sludge tank and according to the isomeric mass balance, it can be concluded that the less chlorinated PCB are decomposed microbially while all of the higher chlorinated PCB are found in the sludge and the effluent of the model plant.

[The behavior of polychlorinated biphenyls (PCB) in a water purification plant. II. A mathematical balancing of the Ruhleben purification plant in Berlin].

Studies on PCB concentration and their distribution in waste water sludge of a municipal sewage plant are made. The incoming sewage has an average concentration of 813 ng/l, the effluent still contains 108 ng/l. The biggest part of the PCB is accumulated in the sludge. A comparison of the in- and output of PCB and the average PCB-patterns at the various sampling sites of the sewage plant shows that the PCB are not transformed or decomposed microbially.

[Hyaluronidase in cytostatic therapy of ENT tumors].

In 27 patients with squamous cell carcinomas of the head and neck region hyaluronidase was added to cytostatic chemotherapy, in part of them from the beginning and in part after development of chemoresistance. We administered either ampoules containing 750 i. u. of Permease in a dosage between 7,500 i. u. and 22,500 i. u., or alternatively a preparation of hyaluronidase containing 200,000 i. u. Hyaluronidase was well tolerated; there were reversible allergic reactions in only 2 patients. We obtained CR 14/27, PR 5/27, NC 3/27. After giving hyaluronidase to chemoresistant patients CR 8/16, PR 3/16 and NC 3/16 were achieved. The course of the disease in chemoresistant cases make it probable that hyaluronidase can improve the prognosis in these patients. With regard to the kind of activity it seems noteworthy that the amount of mucopolysaccharides is increased in many malignancies; furthermore, the transport of cytostatic agents to the tumour cells can be improved.

Ceftriaxone pharmacokinetics during peritoneal dialysis.

The purpose of this study was to investigate the pharmacokinetics of intraperitoneally (IP) administered ceftriaxone (CRO) in patients maintained on chronic peritoneal dialysis. A single 2 g dose of CRO was administered IP to six adult patients who did not have peritonitis at the time of study. After a 5 hour dwell, the peritoneal fluid was exchanged with CRO-free fluid. Exchanges were carried out every 4 to 8 h, over a 24- to 28-h period. The peak total plasma CRO concentration was 104 micrograms/ml. An average of 74.1% of the IP dose of CRO was absorbed. Plasma protein binding was nonlinear; mean free fraction ranged from 12.8 to 17.9% at low and high concentrations. Dialysate concentrations at the end of subsequent exchanges ranged from means of 19.9 to 2.9 micrograms/ml. Total CRO clearance from plasma was 10.1 ml X kg-1 X h-1 and the mean terminal t 1/2 was 12.7 h. Dialytic clearance averaged 0.69 ml X kg-1 X h-1, only 6.9% of total clearance. A model which incorporates known characteristics of CRO binding and distribution in anuric patients was used to simulate plasma and peritoneal concentrations of CRO during multiple dose IP drug administration.
