Complementary DNA cloning and characterization of ferredoxin localized in bundle-sheath cells of maize leaves.

In maize (Zea mays L.) two leaf-specific ferredoxin (Fd) isoproteins, Fd I and Fd II, are distributed differentially in mesophyll and bundle-sheath cells. A novel cDNA encoding the precursor of Fd II (pFD2) was isolated by heterologous hybridization using a cDNA for Fd I (pFD1) as a probe. The assignment of the cDNAs to the Fds was verified by capillary liquid-chromatography/electrospray ionization-mass spectrometry. RNA-blot analysis demonstrated that transcripts for Fd I and Fd II accumulated specifically in mesophyll and bundle-sheath cells, respectively. The mature regions of pFD1 and pFD2 were expressed in Escherichia coli as functional Fds. Fd I and Fd II had similar redox potentials of -423 and -406 mV, respectively, but the Km value of Fd-NADP+ reductase for Fd II was about 3-fold larger than that for Fd I. Asparagine at position 65 of Fd II is a unique residue compared with Fd I and other Fds from various plants, which have aspartic acid or glutamic acid at the corresponding position as an electrostatic interaction site with Fd-NADP+ reductase. Substitution of asparagine-65 with aspartic acid increased the affinity of Fd II with Fd-NADP+ reductase to a level comparable to that of Fd I. These structural and functional differences of Fd I and Fd II may be related to their cell-specific expression in the leaves of a C4 plant.

Cyclooxygenase-2 overexpression enhances lymphatic invasion and metastasis in human gastric carcinoma.

OBJECTIVE: Recent epidemiological studies indicate that there is reduced risk of all digestive carcinomas in patients who regularly take nonsteroidal anti-inflammatory drugs (NSAIDs) such as aspirin. Cyclooxygenase (COX) is a target enzyme for NSAIDs. We investigated the role of two isoforms, COX-1 and COX-2, in the development and metastasis of gastric carcinoma. METHODS: Fifteen gastric carcinoma tissue specimens and accompanying adjacent mucosa specimens were obtained from surgical resections. COX-1 and COX-2 protein expression were evaluated using Western blotting analysis, and their relative band densities were semi quantified using standard densitometry scanning techniques. RESULTS: Compared with paired noncancerous specimens, COX-2 was overexpressed in 10 of 15 carcinoma tissue specimens (66.7%). Overall, COX-2 levels in carcinoma tissue were significantly higher. Two early carcinomas (confined to the mucosa and submucosa) and three of 13 advanced carcinomas (extended below the submucosa into the muscular wall) had weak or similar COX-2 expression in paired tissue specimens. COX-2 overexpression in tumors significantly correlated with tumor invasion into the lymphatic vessels in the gastric wall and metastasis to the lymph nodes. Furthermore, the stage grouping in the TNM classification significantly correlated with COX-2 overexpression. In contrast, COX-2 overexpression did not correlate with histopathological grading, surface size, and venous vessel invasion of the tumors. COX-1 levels were similar between paired tissues. CONCLUSION: COX-2 overexpression might enhance lymphatic invasion and metastasis in patients with gastric carcinoma, implicating a poor prognosis. Therefore, the use of COX-2-specific inhibitor to suppress lymphatic metastasis in humans should be investigated.

Effects of in vitro atmospheric ammonia exposure on recovery rate and luminol-dependent chemiluminescence of bovine neutrophils and bronchoalveolar macrophages.

The effects of atmospheric ammonia, a major pollutant in animal confinement facilities, on bovine neutrophils and bronchoalveolar macrophages were evaluated in vitro. Ammonia exposure at concentrations 50, 100 and 200 ppm for one hour impaired recovery rates of neutrophils dose-dependently but enhanced their chemiluminescence activity per cell at lower concentrations (50 and 100 ppm). Macrophages were resistant to the exposure. Their recovery rates and chemiluminescence remained unaffected even at 200 ppm exposure. The present results suggest that ammonia exposure is unfavorable for bovine neutrophils in vitro, and probably in vivo also, in light of causing cell damage and triggering wider inflammatory responses.

[Possible availability of rapid urease tests for diagnosis of Helicobacter pylori eradication: comparative study with culture and histology].

Rapid urease tests (RUTs) are simple and economical methods to detect Helicobacter pylori infection. However, they have been thought to be unreliable to confirm successful eradication of the infection following treatment. We reviewed the efficiency of RUTs and investigated their possible indication for use after eradication therapy. The sensitivity of highly sensitive RUTs seemed to be comparable to that of culture regardless of the timing for testing, but remarkably lower than that of histology within a few months after treatment. Highly sensitive RUTs, used several months after therapy, produced excellent accuracy, suggesting that they might be available to confirm successful treatment, taking cost and time factors into consideration.

In vitro and in vivo comparison of binding of 99m-Tc-labeled anti-CEA MAb F33-104 with 99m-Tc-labeled anti-CEA MAb BW431/26.

AIM: The purpose of this study was to assess the potential for radio-immunodetection (RAID) of murine anti-carcinoembryonic antigen (CEA) monoclonal antibody (MAb) F33-104 labeled with technetium-99m (99m-Tc) by a reduction-mediated labeling method. METHODS: The binding capacity of 99m-Tc-labeled anti-CEA MAb F33-104 with CEA by means of in vitro procedures such as immunoradiometric assay and cell binding assay and the biodistribution of 99m-Tc-labeled anti-CEA MAb F33-104 in normal nude mice and nude mice bearing human colon adenocarcinoma LS180 tumor were investigated and compared with 99m-Tc-labeled anti-CEA MAb BW431/26. RESULTS: The in vitro binding rate of 99m-Tc-labeled anti-CEA MAb F33-104 with CEA in solution and attached to the cell membrane was significantly higher than 99m-Tc-labeled anti-CEA MAb BW431/261 (31.4 +/- 0.95% vs. 11.9 +/- 0.55% at 100 ng/mL of soluble CEA, 83.5 +/- 2.84% vs. 54.0 +/- 2.54% at 10(7) of LS 180 cells). In vivo, accumulation of 99m-Tc-labeled anti-CEA MAb F33-104 was higher at 18 h postinjection than 99m-Tc-labeled anti-CEA MAb BW431/26 (20.1 +/- 3.50% ID/g vs. 14.4 +/- 3.30% ID/g). 99m-Tc-activity in the kidneys of nude mice bearing tumor was higher at 18 h postinjection than at 3 h (12.8 +/- 2.10% ID/g vs. 8.01 +/- 2.40% ID/g of 99m-Tc-labeled anti-CEA MAb F33-104, 10.7 +/- 1.70% ID/g vs. 8.10 +/- 1.75% ID/g of 99m-Tc-labeled anti-CEA MAb BW431/26). CONCLUSION: 99m-Tc-labeled anti-CEA MAb F33-104 is a potential novel agent for RAID of recurrent colorectal cancer.

Usefulness of cytofluorometric DNA ploidy analysis in distinguishing benign cartilaginous tumors from chondrosarcomas.

In this study, we undertook to prove the usefulness of cytofluorometric DNA ploidy analysis in distinguishing benign cartilaginous tumors from chondrosarcomas. We analyzed the DNA ploidy of 47 cartilaginous tumors using DNA cytofluorometry, which is more sensitive than flow cytometry. All of these tumors were classified into six groups on the basis of clinical, radiologic, and histologic criteria. The 25 tumors in the No. 1 group showed no histologic signs of malignancy regardless of their clinical signs. The four tumors in the No. 2 group showed histologic signs of malignancy, but had benign clinical signs like small bone origin or Ollier's disease. The No. 3 group (13 tumors), No. 4 group (four tumors), and No. 5 group (three tumors) were conventional grade I, II, and III chondrosarcomas, respectively, and the No. 6 group included three dedifferentiated chondrosarcomas. Tumor cells isolated from fresh tumor materials treated with papain and collagenase were smeared on a glass slide and their nuclear DNA was stained with propidium iodide. The DNA content of each cell was measured by a cytofluorometer as fluorescence intensity. The results of this study showed that all of the tumors in the No. 1 group had a diploid pattern with a significantly lower (P<.001) cell proliferative activity than the grade I chondrosarcomas in the No. 3 group, all of which had a diploid pattern. Cytofluorometric analysis also indicated that grade II and III chondrosarcomas in the No. 4 and 5 groups had a higher frequency of hyperdiploid cells (%HDC), including aneuploid and polyploid cells than grade I chondrosarcomas. Importantly, all of the grade I chondrosarcomas showed a %HDC >8%, whereas all of the tumors in the No. 1 and 2 groups showed a %HDC <8%. Therefore, we believe that a %HDC value of 8% is borderline between biologically benign and malignant states in cartilaginous tumors. Four of five patients with aneuploid chondrosarcoma had tumor recurrence and two of these patients died of metastatic disease, although all of the patients except for one with diploid chondrosarcoma were continuously disease free after surgery. Based on these results, we concluded that the data of DNA ploidy analysis, especially cell proliferative activity expressed as %HDC, is more reliable and clinically more useful than the histologic and clinical signs of malignancy in distinguishing benign cartilaginous tumors from chondrosarcomas and even from low grade chondrosarcomas.

Relation between cellular doxorubicin binding ability to nuclear DNA and histologic response to preoperative chemotherapy in patients with osteosarcoma.

BACKGROUND: Although chemosensitivity to antiosteosarcoma agents is the most important prognostic factor in human osteosarcoma, none of the many chemosensitivity tests reported previously are reliable and clinically useful. In this study, the authors investigated the reliability and clinical availability of doxorubicin (Adriamycin; Adria Laboratories, Columbus, OH) binding assay (ABA) as a new chemosensitivity test for osteosarcoma. METHODS: Doxorubicin (adriamycin [ADM]) binding ability (%AB) to nuclear DNA in isolated osteosarcoma cells was assessed by ABA in 14 patients with primary osteosarcoma who were treated with preoperative chemotherapy containing ADM and 6 patients with relapsed osteosarcoma after intensive chemotherapy. Histologic responses to preoperative chemotherapy were evaluated by percentage of tumor necrosis (%necrosis). RESULTS: Four of the 14 patients with primary osteosarcoma had %AB > 80% (97.3+/-3.7%) and demonstrated good histologic responses (>90% of %necrosis) to preoperative chemotherapy, whereas the remaining 10 patients had %AB < 80% (38.9+/-21.0%) and demonstrated poor responses. Patients with recurrent osteosarcoma that was clinically evaluated to be resistant to previous chemotherapy also had low %AB (34.2+/-28.3%). CONCLUSIONS: Because the results of the current study revealed that ABA is useful for predicting chemosensitivity to chemotherapy with ADM as well as chemotherapy without ADM for patients with osteosarcoma, and because ABA technically is simple and results can be assessed rapidly, the authors conclude that ABA is a clinically useful chemosensitivity test for patients with osteosarcoma.

Alteration of acetohexamide reductase activities in kidney microsomes and cytosol of cadmium-treated rats.

We examined the alteration of acetohexamide reductase activities in kidney microsomes and cytosol of cadmium (Cd)-treated rats. Acetohexamide reductase activity in kidney microsomes of male rats was markedly decreased by treatment with Cd at a dose of 1.23 mg/kg body weight. However, the decreased enzyme activity was completely restored by repeated treatment with testosterone propionate. Therefore, it is reasonable to assume that the treatment with Cd indirectly affect the androgen-dependent acetohexamide reductase activity in kidney microsomes of male rats, possibly by depressing androgen production. In the case of female rats, unlike male rats, the microsomal enzyme activity was little detectable, and was unaffected by the treatment with Cd. Furthermore, Cd treatment had no significant effect on acetohexamide reductase activity in kidney cytosol of male or female rats.

Antitumor spectra of anthracyclines against gastric cancer tissues obtained from surgical specimens with reference to P-glycoprotein expression.

BACKGROUND AND OBJECTIVES: Although the mechanism of P-glycoprotein (Pgp)-related resistance of doxorubicin is known, it has not been clarified for other anthracycline derivatives. We have examined the chemosensitivity of gastric cancer tissues to three anthracyclines in relation to Pgp expression. METHODS: Sixty-six surgical specimens obtained from patients with gastric cancer were subjected to histoculture drug response assay using doxorubicin (DXR), epirubicin (EPI), and 4'-O-tetrahydropyranyldoxorubicin (pirarubicin; THP). The cutoff concentrations used were 15 microg/ml for DXR and EPI and 17 microg/ml for THP. RESULTS: A 50% or more inhibition index (I.I.) was regarded as sensitive, at which the correlation rates were 95.8% (23/24) and 74.2% (49/66) for DXR-EPI and DXR-THP, respectively. Twenty-six specimens were immunohistochemically stained with monoclonal antibody to Pgp, with a positive rate of 53.8% (14/26). In Pgp-positive specimens, all cases were resistant to DXR and 28.6% (4/14) of cases were sensitive to THP, while the antitumor activity of EPI was essentially identical to that of DXR. CONCLUSIONS: The expression of Pgp might affect resistance to DXR and EPI, although THP may partially impair this resistance, suggesting the clinical usefulness of THP in treatment of DXR-refractory gastric carcinoma.

[A rare case of 2 free thrombi in left atrium with mitral stenosis].

A 64-year-old female was admitted with general fatigue and orthopnea. Preoperative echocardiography showed a free ball thrombus in the left atrium, mitral stenosis and severe tricuspid regurgitation. To avoid a herniation of thrombus to the mitral orifice, an emergency operation was performed. Two free and small mural thrombi were found in the left atrium. Thrombectomy, mitral valve replacement and tricuspid annuloplasty were performed successfully. Postoperative course was uneventful, and she was discharged in good condition on the 21st postoperative day.

[A case of cardiac lipoma in the ventricular septum].

Lipoma in the ventricular septum is very rare. Our review of the English literature revealed that our case is the sixth of removal of lipoma in the ventricular septum. A 60-year-old male was admitted because a mass in the ventricular septum was found incidentally in abdominal CT taken following type B hepatitis. CT scanning of the heart showed 2 cm diameter of tumor in the ventricular septum. The tumor had very low radiodensity, so it was thought to be identical to fat tissue. The tumor was more clearly visualized by MR imaging and the signal intensity was high on the T1-weighted image. The tumor was suspected to be lipoma. The intraoperative histological diagnosis showed the tumor was lipoma. The tumor adhered strictly on the myocardium of the septum, and it was located near the left anterior descending coronary artery. It could not completely resected, in these reasons. Postoperative course was uneventful, and echocardiogram taken 9 months after the operation showed no evidence of enlargement of the residual tumor. It is necessary to follow-up rigidly for the potential of enlargement of the residual tumor.

Setting and stress relaxation behavior of resilient denture liners.

STATEMENT OF PROBLEM: Resilient denture liners are widely used for the patients who are not comfortable with correctly made conventional hard-based dentures because of thin and relatively nonresilient mucosa or severe alveolar resorption. There are several materials used for denture liners and the efficacy in their use is influenced by their viscoelastic properties. PURPOSE: This study evaluated the setting behavior and viscoelastic properties of various types of resilient denture liners and the changes in viscoelasticity with the passage of time. MATERIAL AND METHODS: Four types of resilient denture liners were used. Setting behavior of 5 autopolymerizing materials was evaluated with an oscillating rheometer. Stress relaxation tests were conducted to evaluate the viscoelastic properties of 9 materials and changes that occurred over time by means of Maxwell model analogies. RESULTS: Significant differences were found in the setting behavior of the autopolymerizing materials. The acrylic resin and fluoroethylene materials demonstrated viscoelastic properties and the silicone and polyolephin materials were found to be elastic. The acrylic resin materials exhibited the greatest changes in viscoelastic properties over time when compared with silicone, polyolephin, and fluoroethylene materials. CONCLUSIONS: The results suggest that it is important to select denture liner materials according to clinical situations because of the wide ranges of setting behavior, viscoelastic properties, and durability over time.

Rheology of tissue conditioners.

STATEMENT OF PROBLEM: Tissue conditioners can be used to condition abused tissues, record functional impressions, make temporary relinings, and for other clinical applications, mainly because of their specific viscoelasticity. However, little information is available on the rheology of the materials, manipulation, and suitability for various clinical applications. PURPOSE: This study evaluated the gelation times, the viscoelastic properties after gelation of tissue conditioners, and the influence of the powder/liquid (P/L) ratio. MATERIAL AND METHODS: Ten tissue conditioners were used and gelation times were obtained with an oscillating rheometer. A series of stress relaxation tests were also conducted to evaluate the viscoelastic properties after gelation and the changes with the passage of time by means of Maxwell model analogies. RESULTS: Significant differences were found in the gelation times and flow properties after gelation among the materials mixed with the P/L ratios recommended by the manufacturers. The flow properties tended to increase with time of storage. Large differences in the limits of the clinically acceptable P/L ratios and the adjustable limits of elasticity and viscosity by altering P/L ratios were found among the materials. CONCLUSIONS: The results suggested that each material should be selected according to each clinical purpose because of the wide ranges of viscoelastic properties and changes in viscoelasticity with time among the materials. Furthermore, gelation times and the viscoelastic properties after gelation can be controlled to improve handling and suit various applications by altering the P/L ratios within the acceptable limits.

Aberrations of chromosomes 1 and 17 in six human osteosarcoma cell lines using double-target fluorescence in situ hybridization.

Analysis of six human osteosarcoma cell lines was performed by using double-target fluorescence in situ hybridization (FISH). FISH was applied to interphase nuclei, not to metaphase chromosomes. In this study, numerical aberrations of chromosomes 1 and 17 or structural chromosomal aberrations of chromosome arm 1p or 17p, in which it has been suggested that there are one or more tumor suppressor genes in various malignant tumors, were examined with this technique. All six of the human osteosarcoma cell lines studied had extra copies of chromosomes 1 and 17. A high frequency of deletions (> 60%) in chromosome 1 was found in two cell lines and deletions of chromosome 17 were found in one cell line.

The effect of saliva or serum on Streptococcus mutans and Candida albicans colonization of hydroxylapatite beads.

OBJECTIVE: Several recent reports imply the possibility of cariogenicity of denture plaque containing Candida albicans. Hence the purpose of this study was to investigate the effects of salivary and serum pellicles on C. albicans and Streptococcus mutans colonization on hydroxylapatite beads. METHODS: The colonization of three isolates of C. albicans and two isolates of S. mutans was examined by the use of a bioluminescent adenosine triphosphate (ATP) assay based on the firefly luciferase-luciferin system. RESULTS: In the preliminary study, a good correlation was observed between the cell number and ATP amount of each isolate tested, and the results yielded a level of significance (P < 0.001; Student's t-test), confirming the validity of this method. When the relative ATP content of the 48 h colonization of both isolates of S. mutans were compared, a saliva pellicle was significantly more effective in promoting bacterial colonization than either uncoated or serum pellicle (ANOVA; P < 0.01). In contrast, in the case of colonization of C. albicans isolates, a serum pellicle was significantly more effective in promoting the colonization of C. albicans GDH 18 and GDH 19, than both uncoated specimens and saliva pellicle (ANOVA; P < 0.01). Similar trends were observed with C. albicans GDH 16, though significant differences were not observed (ANOVA; P > 0.05). CONCLUSION: The results suggest that the mechanism involved in fungal colonization on hydroxylapatite (HAP) should be different from that of mutans streptococci.

Viscoelastic properties of tissue conditioners--influence of molecular weight of polymer powders and powder/liquid ratio and the clinical implications.

The effect of the molecular weight of polymer powders and the polymer powder/liquid ratio on the viscoelastic properties after gelation of tissue conditioners was studied by means of a stress relaxation test. The results are summarized as follows. The lower-molecular-weight polymer powders produced the larger flow after gelation especially at long times. The use of a lower powder/liquid ratio produced a greater flow after gelation at both short times and long times. The difference in the molecular weight of polymer powders and the powder/liquid ratio was found to have no influence on changes in viscoelastic properties with the passage of time. It would be possible to control the viscoelastic properties of tissue conditioners suitable for each clinical purpose by making variations in composition and structure.
