Analgesic effect of histamine induced by intracerebral injection into mice.

Three methods were used to study the analgesic effect of intracerebral injection of histamine (Hi) on mice: the writhing test (acetic acid and phenylquinone), the electrical stimulation of the tail and the hot plate test. At doses higher than 2 micrograms, Hi inhibited the writhing syndrome significantly, and at doses of 10 micrograms or higher, Hi displayed a marked analgesic effect during both the electrical stimulation and hot plate methods. The saline injection produced only a negligible effect. Simultaneous application of Hi and 10 micrograms of diphenhydramine, pyrilamine or promethazine, apparently causing no analgesic effect from a single administration, caused a parallel shift of the dose-response curve of Hi to the right. ED50 of Hi was increased approximately 2, 2.8 and 3.8 times, respectively. However, cimetidine did not reveal any antagonistic effect on Hi-induced analgesia. Subcutaneously administered, 3 mg/kg of morphine augmented the analgesic effect of Hi. In accordance with this, pretreatment of naloxone (0.005 mg/kg) antagonized the analgesic action of Hi almost completely. When 5 mg/kg of leucine-enkephalin, less than the minimum effective dose, was given prior to Hi injection, the analgesic effect of Hi was enhanced. In addition, 10 and 20 micrograms of Hi increased the morphine analgesia markedly and parallel shifted the dose-response curve of morphine to the left.

Effect of skim milk and yogurt on serum lipids and development of sudanophilic lesions in cholesterol-fed rabbits.

Three groups of male Japanese rabbits weighing about 2.7 kg each were given experimental diets consisting of high cholesterol food and fluid skim milk, yogurt, or water, and were bled every 4 wk to measure serum lipids. After 12 wk they were killed and concentrations of total cholesterol and atheromatous areas dyed with Oil Red O were determined in the aorta to evaluate the development of atherosclerosis. At 8 and 12 wk, the skim milk group showed significantly lower levels of serum total cholesterol, low-density lipoprotein cholesterol, and triglyceride than the control group. Although no significant difference between the yogurt and control groups in the concentration of serum lipids was observed, total cholesterol concentrations in the aorta were significantly lower in both the skim milk and the yogurt groups than in the control group. The atheromatous areas of the skim milk group (38 +/- 34%) were significantly smaller than those of the control group (75 +/- 25%). Significant difference, however, was not found between the yogurt group (51 +/- 22%) and the control group. Concentrations of total cholesterol in the liver did not differ among the three groups. These results suggest that skim milk may have a preventive effect on the development of atherosclerosis.

[A chronobiological study on toluene anesthesia in mice].

An experimental study of toluene exposure was performed on mice at various times of the day using the righting reflex and blood concentration as indicators. One hundred and sixty ICR mice were housed 8 mice per cage for 3 weeks with free access to food and water under a dark-light cycle consisting of alternate 12 hr periods of darkness and light: the dark period beginning at 7:00. The exposure was begun when the mice were 8 weeks old. They were exposed to 10,000 ppm of toluene for 40 min at 4:00, 10:00, 16:00 and 22:00, and they were sacrificed at the end of the exposure, after 3 min or after 30 min. The duration of anesthesia was 200.2 +/- 15.4 (sec) at 4:00, 361.1 +/- 21.5 (sec) at 10:00, 335.1 +/- 18.5 (sec) at 16:00 and 309.2 +/- 19.8 (sec) at 22:00. The duration of anesthesia at 4:00 was shorter than that of the other times of the day statistically (p less than 0.01). With regard to the toluene concentration in blood, there was no significant difference between those mice sacrificed at the end of exposure and those sacrificed after 3 min.

Japanese adult siblings with Tangier disease and statistical analysis of reported cases.

A 28 year old Japanese female was admitted to our university hospital because of extremely low plasma cholesterol levels (50mg/dl). On physical examination, tonsillar remnants and several flat patches of yellow-orange lymphoid tissues in the pharynx were found. Her plasma HDL was as low as 2.4 mg/dl. Laparoscopic findings showed typical yellow patches on the surface of the liver, and liver biospy specimens contained cholesterol esters demonstrated by the Schultz reaction. Colonofiberscopy showed orange-brown spots present throughout the rectum. Histologically, "foam cells" were recognized in the lymphoid tissue of the pharynx and rectal mucosa. Her sister, 26 years old, also revealed yellow-orange tonsils, low plasma cholesterol levels (39mg/dl) and low plasma HDL levels (8mg/dl), and was also diagnosed as Tangier disease. From a statistical analysis of the reported cases, it is noteworthy that yellow-orange tonsils, which were said to be the hallmark of Tangier disease, were not found in 20% of the patients. However, it is important that we could find rectal mucosa studded with orange-brown spots in all of the cases examined, and it contained "foam cells" histologically. It was concluded that when we suspect Tangier disease on account of hypocholesterolemia, we must first do a proctoscopic examination.

[Intracranial benign chondroblastoma. A case report and review of the literature].

A case of benign chondroblastoma extending from the nasal cavity to the frontal region, was reported. A 1-year and 9-month old girl was admitted to our hospital in April 1981 because of generalized convulsion. On admission, she was intact neurologically and had It. nasal obstruction and It. eye discharge. Her laboratory examinations, including serum electrolytes and A1-P, were all in normal range. Plain skull X-P showed marked calcification from midfrontal region to the nasal cavity, and destruction of the frontal base especially in It. side. Plain CT showed isodensity mass with spotty calcification in the nasal cavity and marked high density mass in the frontal region. By contrast enhancement, nasal mass was markedly enhanced and new enhanced mass appeared around the intracranial calcification. Lt. ext. CAG showed the enlarged middle meningeal artery which is the feeder of the tumor, fine tumor vessels and slight tumor stain. Lt. int. CAG only showed the mass effect. After three-stage operation of bifrontal craniotomy, sublabial transnasal approach and rhinotomy, all the tumor was removed and she was discharged with no neurological deficits. The tumor was well demarcated and consisted from solid, cystic and calcified region. The solid region was grayish and easy-hemorrhagic. Histological examination revealed characteristic features of benign chondroblastoma, such as diffuse proliferation of polygonal or round cells, foci of chondroid matrices, scattered polynuclear giant cells and some mitosis.

[Research on experimental cerebral infarction in cats with in vivo TMR (topical magnetic resonance) approach].

Metabolic changes in the ischemic brains of cats were investigated in vivo with high energy phosphate compounds as parameters by using a topical magnetic resonance (TMR) spectrometer. The experimental focal cerebral ischemia was made in four cats by modifying the method of O'Brien and Waltz. The stem of left middle cerebral artery was exposed and set the occlusive device 5-7 days before the in vivo measurement. 31P-NMR spectrum was taken under general anesthesia with Ketamine HCI and with decreased blood flow. The following points must be considered in obtaining 31P-TMR spectrum in a cat brain: Firstly, as much muscle as possible must be removed from the detective area because it contains phosphate compounds. Our experiment showed that bone and blood had little or no effect on the 31P-TMR spectrum. Secondly, although same procedure was repeated, it was difficult to obtain constant ischemic lesion; in site and size. The detective area in setting was not changed in the particular area. However there was a possibility of the detective area also including various non-ischemic regions. Thirdly, 31P-TMR spectrum had several peaks in a cat brain; which were sugar phosphate, inorganic phosphate, phosphodiesters, phosphocreatine, gamma-, alpha-, beta-ATP in the pre-occlusive conditions. These peaks did not appear in clear volume separation with one another. We drew the perpendicular line from through between two neighbour peaks to the horizontal base line and artificially divided two peaks. The area of each peak did not represent correctly the density of each component. Fourthly, unnecessary components were rationalistically excluded from the 'raw' spectrum.(ABSTRACT TRUNCATED AT 250 WORDS)

Prognosis and pathogenesis of sudden deafness with scaling out.

Recovery of hearing in sudden deafness is said to be such that the severer the hearing loss and the longer the period from the disease onset to the start of treatment, the poorer the recovery of hearing. However, the prognosis of the so-called scaling out cases has not yet been fully studied. We have therefore studied 96 patients with severest sudden deafness who consulted us within 14 days after disease onset and who had scaling out in all the frequencies. We evaluated the extent to which recovery of hearing could be expected and whether any characteristics are seen in pathogenesis, etc. The results showed that there was no patient who experienced complete recovery, even when treatment was started as early as within 4 days. Recovery of hearing was better in low frequencies than in high frequencies. The site of the lesion was the inner ear in many cases, and some had retrocochlear lesions. Concerning pathogenesis, we think the difference in degree of lesion should be considered important, rather than there may be a substantial difference, compared to that of slight deafness.

Histochemical studies on enzyme-digested protein plugs of patients with chronic pancreatitis: a preliminary report.

Sulfated acidic mucopolysaccharides have been found to be significant components of "protein plugs" in patients with chronic pancreatitis. The precise identification of the mucopolysaccharides and their distribution within the protein plugs may clarify the pathogenesis of the plugs. Pure pancreatic juice from five patients with chronic pancreatitis was obtained by endoscopic retrograde catheterization of the papilla of Vater. Enzymes for digestion of the plugs included hyaluronidase of the bovine testes and streptomyces hyalurolyticus, chondroitinase ABC and AC, and sialidase (neuraminidase). Our study indicated that: I) Sialic acid is distributed throughout the plugs and may be a major component, followed by a lesser amount of chondroitin sulfate B. 2) Chondroitin sulfate A, C, D and E and chondroitin may be minor components. 3) Hyaluronic acid is negligible in the plugs.
