[Effects of Nilestriol on reproductive system in ovariectomized rats].

The purpose of this study was to determine the effects of Nilestriol on reproductive system. Female SD rats, aged 4 months, were randomly divided into three groups. The OVX group's rats were subjected to bilateral ovariectomy, the SHAM group's rats received sham surgery; the Nilestriol(CEE3) group's rats were subjected to bilateral ovariectomy and treated with CEE3(0.15 mg/100 g BW, once a week). A computerized image analyzer was used to evaluate the endometrial pathological changes 10 weeks later. The weight of uterus was measured. The results showed that in OVX group, atrophic uteri; and atrophied endometria were noted. The weight of uterus was increased in CEE3 group, compared with that in OVX group (642.20 +/- 283.60 mg vs. 297.00 +/- 106.90 mg, P < 0.05). The volume fraction of endometrial glands was also significantly elevated in CEE3 group, compared with that in OVX group (1.84% vs 0.58%). The endometria in CEE3 group were at an early proliferative stage. These suggest that Nilestriol has a slight stimulating effect on the uteri of ovariectomized rats, including their endometria and uterine smooth muscles. Nilestriol is suited to be used in combination with progestogen.

[The immunohistochemical and electron microscopical study of pathological classification of Melkersson-Rosenthal syndrome].

OBJECTIVE: To prove further the pathological classification of Melkersson-Rosenthal syndrome (MRS) and observe the morphological features of the granuloma cells in MRS. METHODS: We studied 20 cases of MRS, with 10 cases of granuloma type and the other 10 cases of non-granuloma type. The negative results of periodic acid-schiff (PAS), Neelren anti-acid special staining and tuberculosis (TB)-DNA polymerase chain reaction proved that all cases were not TB and fungous disease. Then all the cases were labelled with antibody of lysozyme by immunohistochemical staining and investigated with electron microscope. We studied the expression of lysozyme in multinucleated giant cells, epithelioid cells and histiocytes. RESULTS: The results of lysozyme staining were positive in the multinucleated giant cells, epithelioid cells and histiocytes of the granuloma type and were negative in those cells of the non-granuloma type. On the other hand, the small multinucleated giant cells which had 2-3 nuclei were the features of the morphological structure of the granuloma type. The small multinucleated giant cells were not seen in the non-granuloma type. CONCLUSION: In this paper, two pathological types (granuloma and non-granuloma type) in the MRS were proved further and it is found that the different structures of granuloma can be used to discriminate between MRS and sarcoidosis and tuberculosis.

[Clinical significance of plasma PGE2 levels in pre- and post-operative patients with laryngeal carcinoma].

OBJECTIVE: To study the clinical significance of PGE2 Levels in patients with laryngeal carcinoma. METHOD: Radioimmunoassay was used to evaluate plasma PGE2 levels in 35 patients with laryngeal carcinoma. RESULT: The plasma PGE2 levels in patients with laryngeal carcinoma were significant higher than that of normal subjects (P < 0.05), but there was no correlation with the stage of laryngeal carcinoma (P > 0.05). The plasma PGE2 levels of postoperative patients with laryngeal carcinoma were significant lower than that of preoperative patients (P < 0.05). After operation, the plasma PGE2 levels of recurrent patients were significant higher than that in patients with no recurrence (P < 0.01). CONCLUSION: The determination of PGE2 levels in patient plasma may help the diagnosis and prognostic judgment of laryngeal carcinoma.

[Determination of tetramethylpyrazine in animal serum and cerebrospinal fluid by high performance liquid chromatography].

A reversed-phase high performance liquid chromatographic (RP-HPLC) method for the determination of the tetramethylpyrazine(TMP) in Chuanxiong extract, the animal(mouse) serum and cerebrospinal fluid has been developed. The TMP was separated on an ODS column Zorbax SB-C18(4.6 mm i.d. x 250 mm, 5 microns) at room temperature and detected by using UV detector at 270 nm. The mobile phase was methanol-water (50:50, V/V) containing 0.2 mmol/L of NH4H2PO4 flowing at a rate of 0.8 mL/min and 20 microL samples were injected. The detection limit of TMP was 1 mg/L and the calibration curve is linear between 5 and 500 mg/L with a correlation coefficient (r) of 0.999. The recovery of TMP ranged 98%-103%. The extract of Chuanxiong and pretreated serum and cerebrospinal fluid sample are stable for a week at room temperature.

[Derivative gas chromatographic analysis of fructooligosaccharide in fermented sucrose].

As a new sweetener, fructooligosaccharide is paid more and more attention for its health improvement property. It includes trisaccharide, tetrasaccharide and pentasaccharide, and can be produced from sucrose by the fermentation of microorganism. In order to analyze the content of fructooligosaccharide in fermented sugar by gas chromatography, fructooligosaccharide was transformed into trimethylsilyl derivatives. Based on the modified gas chromatograph SP2308, and under the chosen chromatographic conditions with 0.53 mm capillary column of OV-101, the contents of fructose, glucose, sucrose and fructooligosaccharide were determined by programmed temperature chromatography. The recovery of fructooligosaccharide was satisfactory.

[A computer-aided analysis system for counting the synaptic numerical density based on disector technique].

The change in the numbers of synapse is of importance in various kinds of physiological and pathological processes. The numerical density is the structural parameter measuring up the numbers of the particles in the space. A new stereological technique, the disector technique, has been described to count the number of particles in a given volume of tissue. It is a technique for counting the number of arbitrary particles in 3-dimensional space using adjacent parallel sections at known distance apart. In this article also discussed is the software for computer assisted count of the synaptic numerical density based on the physical director technique with Visual Basic 4.0.

[Image and quantity analysis of blood-gas in rabbit's artery and Na(+)-K(+)-ATPase in their lungs during PE-SWD treated by HFJV].

This study was made to gain an insight into the mechanism of high-frequency jet ventilation(HFJV) treatment for the pulmonary edema after seawater drowning(PE-SWD). PaO2, PaCO2 and oxygen saturation (SaO2) of the rabbit and Na(+)-K(+)-ATPase in the rabbit's lungs in three groups--PE-SWD group(PE-SWD-G), HFJV group (HFJV-G) and control group(CG), were measured and analysed by the blood-gas analyser and computer image system. The results showed that, after 100 minutes' HFJV, the PaO2, SaO2 and the activity of Na(+)-K(+)-ATPase in the lung capillary endothelial cells in HFJV-G were significantly higher than those in PE-SWD-G(P < 0.01 or P < 0.05). Three parameters of Na(+)-K(+)-TAPase(G1, D1 and D2) in HFJV-G almost returned to their values in CG. The authors suggest that the increase of PaO2 and SaO2 in the rabbit's artery blood in HFJV-G is closely related to the rehabilitation of Na(+)-K(+)-ATPase activity in the lungs after HFJV. The mechanism of successful HFJV treatment for PE-SWD is that HFJV can better correct hypoxemia and improve the rehabilitation of Na(+)-K(+)-ATPase activity in the rabbit's lungs.

[Study on the mechanism of pulmonary edema after seawater drowning in rabbit].

To study the mechanism of pulmonary edema after seawater drowning (PE-SWD), the indexes of blood-gas and acid-base in rabbits artery blood were measured by the blood-gas analyser. The activity of Na(+)-K(+)-ATPase, cytochrome oxidase(CYTO) and alkaline phospharase(ALP) in the lungs were measured and analysed by computer image system. C-fos mRNA and Fos protein in the lungs were respectively determined by situ hybrioization and immunohisto chemical techniques. The distribution of phospholipid and Ca2+ of rabbits lungs was quantitatively analysed by ultrastructural location method. The results showed that, five parameters of PaO2, oxygen saturation(SaO2), pH, actual bicarbonite(AB) and base excess(BE) and the activity of Na(+)-K(+)-ATPase and CYTO decreased remarkably in PE-SWD. Both c-fos mRNA and Fos protein expression in pulmonary epithelial cells in PE-SWD were significantly elevated compared with the normal controls(P < 0.01). The phospholipids products in the pulmonary alveolar type II epithelial cells were decreased, however, the Ca2+ precipitate pellets inside the lung capillary endothelial cells and the pulmonary alveolar type I and II epithelial cells increased obviously. The arthors suggest that the injuny action of the seawater, hypoxia and metabolic acidosis may be the mian three mechanisms of the pulmonary edema induced seawater drowning. The lowering activity of Na(+)-K(+)-ATPase and CYTO in the lungs and calcium overload in the cells are not only evil consequence resulted from above three factors, but also the importent causes leading to the worse of PE-SWD. The transmiting line of Ca(2+)-fos may be also a key link to bring about the worse of PE-SWD.

[Bacteriological analysis of chronic sinusitis in school-age children (36 cases report)].

OBJECTIVE: To research the bacteriological characters of chronic sinusitis in school-age children and to provide basis for clinical treatment. METHOD: The purrent discharges taken from middle meatus of 36 patients were cultured for bacteria and the antimicrobial suscepility was determined. RESULT: The positive rate of bacteria culture was 83.3%, the compound infection rate was 36.1%. Aerobic bacteria were present in 76.7%, anaerobic bacteria were present in 55.6%. The result of antimicrobial susceptibility test was scattered. CONCLUSION: Infection rate of anaerobic bacteria was high in chronic sinusitis in school-age children and result of antimicrobial susceptibility was scattered. It was important to have anti-anaerobic treatment in chronic sinusitis in school-age children and to take an antimicrobial susceptibility test before medical treatment.

[Affection of exogenous gibberellic acid (GA3) on endogenus hormones of Panax quinquefolium seed during its morphological after ripening period].

According to the determination of endogenus hormones variation of Panax quinquefolium seed during its morphological afterripenging period, and the affection of exo-GA3 on the endogenus hormones dynamics, it suggested that exo-GA3 couldn't be used for accelerate the growth of Panax quinquefolium embryo, but was helpful in relieving seed dormancy during physiological afterripening period.

[Biological effects of supplements on soil properties and the growth of Panax quinquefolium].

This paper suggests that the physical and chemical characteristics of soil in American ginseng field were greatly improved after utilization of supplement on soil. Compared with the contrast, the soil gravity ratio decreased from 0.812 g/cm3 to 0.715 g/cm3, while the soil porosity increased from 69.36% to 73.03%. These improvements are benefit for the growth of Panax quinquefolium. Biological observations show that the fresh weight of ginseng root enhanced 53.8%, and the content of total saponins in root enhanced 0.5%-1% (get to 8.28%).

The preliminary study of interferon-gamma gene transfection to human Tenon's capsule fibroblasts in vitro.

PURPOSE: To investigate the results of the interferon-gamma (IFN-gamma) gene transfer and transient expression in human Tenon's capsule fibroblast in vitro in order to find a way to gene therapy in vivo. METHOD: Using LipofectAMINE, IFN-gamma gene was transferred in human Tenon's capsule fibroblasts with plasmid pcDNA3 IFN-gamma. Its mRNA transcription and protein expression were determined by RT-PCR and flow cytometry assay respectively. RESULT: The human Tenon's capsule fibroblasts transferred the IFN-gamma gene can express the IFN-gamma in transcription and protein level transiently. CONCLUSION: IFN-gamma gene can be transferred successfully and expressed expressed efficiently in human tenon's capsule fibroblast in vitro.

Preliminary study on in vitro induced differentiation of embryonic stem cells into neurons.

PURPOSE: To study preliminarily in vitro induced differentiation of embryonic stem cells into neurons for further investigation of an alternative for the treatment of glaumatous neuropathy. MATERIALS AND METHODS: Supernatant of cultured Buffalo rat liver cells (buffalorat liver cell-conditioned medium, BRL-CM) was used for culturing embryonic stem cells (ES-D3 cell line). Morphological features of undifferentiated ES cells were studied by HE staining and electron microscopy. Based on the methods used by Bain et al, we modified the methods and used retinoic acid (RA) as an inducer to differentiate ES-D3 cells and cytosine arabinoside (Ara-C) as inhibitor of proliferative cells. The growth of the cells was observed under phase contrast microscope. RESULTS: ES-D3 cells cultured by BRL-CM grew in aggregates and remained undifferentiated. Electromicroscopy showed large nucleus and a large amount of mitochondria in undifferentiated ES cells and many processes on the surfaces. In the first day after the adding of retinoic acid, some neuron-like cells with one, two or more processes were present. In the second day after adding RA and the first day after the plus of 10 microns Ara-C, a large amount of neuron-like cells appeared, with the formation of neuron-like networks. CONCLUSIONS: Combined use of RA and Ara-C can induce ES cells to differentiate into neuron-like cells. Our present preliminary study might provide insights into an alternative for the treatment of glaucomatous neuropathy by the transplantation of embryonic stem cells.

Bifunctional effect of human IFN-gamma on cultured human fibroblasts from Tenon's capsule.

PURPOSE: To study the effect of human IFN-gamma on in vitro cultured human fibroblasts from Tenon's capsule. MATERIALS AND METHODS: The effect of different concentrations of human IFN-gamma and mitomycin-C (MMC), 5-fluorouracil (5-Fu) on cultured human Tenon's capsule fibroblasts (HTCF) was measured using a MTT [3-(4, 5-dimethylthiazo-2-yl)]-2, 5-diphenyltetrazolium bromide; Thiazolyl blue) colorimetric assay. The results were analyzed using ANOVA of the statistical package for social sciences (SPSS) 9.0 version. The difference was considered to be significant if P < 0.05. RESULTS: The effects of MMC and 5-Fu on the growth of HTCF were negative, while the effects of IFN-gamma on the growth of HTCF were both negative (10(2)-10(4) units/ml in two experiments) and positive (10(6), 10(5), 10 units/ml in two experiments). The inhibition rate of MMC ranged from 5.73% to 46.9%, which was similar to the inhibition rate of 5-Fu ranged from 12.49% to 38.92% (P = 0.351). The inhibition rate of IFN-gamma in two experiments was smaller than MMC and 5-Fu (P < 0.05). CONCLUSION: IFN-gamma has bifunctional effect (both enhancement and inhibition) on proliferation of cultured HTCF. The antiproliferative effect of IFN-gamma was weaker than MMC and 5-Fu. Further study has to be carried out to document the inhibition of scar formation of filtration bleb by IFN-gamma and the molecular mechanisms of its bifunctional effect on HTCF proliferation.

The analysis of pedigree GZ (Guangzhou). 1 with primary open angle glaucoma.

PURPOSE: To analyze the hereditary modality of primary open-angle glaucoma in China. METHODS: The genetic form of Pedigree GZ. 1 was analyzed using Mandalian hereditary rules. RESULTS: Pedigree GZ. 1 had following characteristics: 1) The pedigree had four generations, and there existed POAG patients in each generation; 2) Each patient had a parent with POAG. If the parents didn't suffer from the disease, their children would not. 3) The incidence of POAG in the relatives of the patients was 1/2. In addition, The age of onset, intraocular pressure, fundus and prognosis was different from each other in the patients. CONCLUSIONS: 1) Pedigree GZ. 1 is inherited as an autosomal dominant trait. 2) There exists individual differences of clinical manifestations in POAG patients.

[The clinical application of stereography in laryngology with "the three-dimensional reconstruction system of medical faultage photograph"].

OBJECTIVE: To study the value of the clinical application of stereography in laryngology. METHODS: Taken by Microtek E6 scanner, the CT faultage images were inputted in personal computer and reconstructed to a three-dimensional stereograph by "the three-dimensional reconstruction system of medical faultage photograph". The software was compiled using IDL5.2, a complete, integrated software environment for data analysis, visualization and application. RESULTS: The three-dimensional laryngeal graph could be incised in random, displayed in optional degree, and was rotatable, semitransparent and measurable. The tumor could identified be identified by a special color and revealed through translucent parenchyma and cartilage. Reconstruction stereographs of laryngeal and hypopharyngeal cancer of 26 patients were investigated. CONCLUSION: It is convenient for popularization because the three-reconstruction process is built on a personal computer without the need of any equipment for image acceleration. It is very helpful for ENT surgeon to define the range of the tumor, to fix on surgical pattern accurately and to anticipate the operational course.

[Effects of 1 alpha(OH)D3 on trabecular and cortical bone in ovariectomized rats].

OBJECTIVE: To study the pharmaceutical effects of 1 alpha(OH)D3 on trabecular and cortical bone in ovariectomized (OVX) rats. METHODS: 41 female Wistar rats of six months old were randomly divided into 5 groups: (1) Baseline control; (2) Sham control; (3) 6 weeks after OVX (OVXb); (4) 14 weeks after OVX (OVXe); (5) OVX + 0.1 microgram/(kg.d) 1 alpha(OH)D3 (O + VD), treatment started 6 weeks after OVX and lasted 8 weeks. Histomorphometry analysis of tibia, peripheral quantitative computed tomography (pQCT) scanning of femur, bone biomechanical test and serum and urinary biochemical parameters were determined. RESULTS: The levels of bone turnover indexes increased in OVX rats, OVX also resulted in reducing of trabecular, bone mass and biomechanical properties. The ratio of urinary deoxypyridinoline crosslink/creatinine was decreased by 67.0% in O + VD group compared with OVXe group 67.0% [(43.50 +/- 11.20) nmol.L-1/mmol.L-1 vs(131.80 +/- 14.90) nmol.L-1/mmol.L-1, P < 0.01]. Percent trabecular area (Cn-BV/TV) was increased by 89.8% in O + VD group compared with OVXe group (11.03 +/- 0.73 vs 5.81 +/- 1.29, P < 0.05). Trabecular bone mineral content and density were increased by 77.3% and 91.3% compared with OVXe respectively (P < 0.05). Although cancellous maximal load and stiffness increased in O + VD group, but no statistical significance. 1 alpha(OH)D3 also enhanced polar moment of inertia (PMI) and maximal load of cortical bone in femur compared with OVXe (23.70 +/- 1.63 vs 18.23 +/- 1.41, P < 0.01 and 171.69 +/- 9.92 vs 147.58 +/- 11.29, P < 0.05 respectively). CONCLUSION: 1 alpha(OH)D3 inhibited the higher bone turnover induced by OVX, increased trabecular bone in proximal tibia and bone mass in distal femur. 1 alpha(OH)D3 also improved the mechanical properties of cortical bone in femur.

[Immobilization and properties of angiotensin converting enzyme].

OBJECTIVE: To study the immobilization of angiotensin converting enzyme (ACE) for purifying ACE inhibitor from a native peptide mixture. METHODS: The experiment was carried out under the low water activity condition, using tosylate chloride activating side-chain hydroxyl group of Sepharose CL-4B agarose to form a high active group which could react with the free amino-group of ACE to link the enzyme with agarose. RESULTS: Immobilized ACE not only had a wider pH range, but also had a 0.6 unit right-move optimum pH than soluble ACE. After treated in pH9.0 and pH 6.5 conditions for 24 h respectively, 82% and 68% enzyme activities of immobilized ACE was maintained, and soluble ACE remained 64% and 39%. Immobilized and soluble ACE both appeared maximum enzyme activity at about 50 degrees C, the soluble ACE would lose almost all its activity when temperature kept rising. When kept at 40 degrees C and 50 degrees C for 2 h, the activity of immobilized ACE remained 82% and 34% respectively, while the soluble ACE remained 52% and completely inactivated. After two kinds of enzymes were stored at 20 degrees C for one month, immobilized ACE remained 61% activity, as compared with the 20% activity residual of soluble ACE. CONCLUSION: The immobilized ACE had a better stability than soluble ACE in conditioned pH and temperature.
