[The effect of the AcD-AG stabilizer on the survival time and surface properties of fluid preserved platelet concentrates compared to concentrates from fresh blood].

In the present investigations the storage effect the AcD-AG stabilizer on thrombocytes is examined. The thrombocytokinetic parameters of 9 fresh blood concentrates and 15 concentrates of AcD-AG plasma containing platelets were determined. Storage time amounted to three days. The results show that storage with AcD-AG is only possible to a limited degree. On an average the survival time of the platelets was reduced to 2.7 +/- 1.1 days compared with 9.0 +/- 1.0 days of fresh blood concentrates. The recovery of the stored platelets amounted to 25.3 +/- 16.1%, that of the fresh blood concentrates to 63.3 +/- 23.6%. The spleen-heart quotients and those of the liver-heart or the surplus impulses over the spleen and liver respectively indicate that there is a predominant destruction in the spleen for those thrombocytes stored for three days. The liver is scarcely involved in this sequestration process. With 36.1% platelet yield was very low in concentrates gained from AcD-AG plasma containing platelets and having been stored for 3 days. In cases of emergency a clinical application of concentrates prepared in this way should not be given up. If being used, the greater requirement has to be taken into account. If the substitution therapy is continued, however, fresh blood concentrates have to be used as soon as possible.

[Drug-induced].

The rare clinical picture of the thrombocytopenia caused by drugs is described using own histories of diseases and data from literature. The questions of diagnostics and therapy are discussed. Despite the as a rule benign course it is referred to the fact to withold the noxa once diagnosed consequently from the patient for prevention of possible complications (cerebral haemorrhage).

[Quantitative determination of blood loss by means of 51 Cr-labeled erythrocytes in occult hemorrhages of the gastrointestinal tract].

By means of 51 Cr-labelled erythrocytes the stools of 17 patients were examined for occult admixtures of blood. Hereby a method well suited for routine work was developed, the manipulations with stool were reduced to a minimum measure. As measuring device an animal total body counter was used. The average excretion of blood in persons without diseases of the gastro-intestinal tract was 0.49+/- 0.38 ml/d. In three patients a haemorrhage could be proved. The quantitative determination of blood losses through the intestine is diagnostically valuable. It is useful particularly in unclear iron deficiency anaemis. A combination of the method with the determination of the survival time of erythrocytes is possible.

[Myeloproliferative syndrome].

In the present paper a description of the notion of the myeloproliferative syndrome and a survey of the clinical pictures belonging to this is given. Here the author enters into the most important diseases, such as polycythaemia rubra vera, osteomyelofibrosis, osteomyelosclerosis and megakaryocytic leukaemia, concerning diagnostics and therapy. On the basis of literature a comphrensive discussion of the problem is give, whereby it is referred to the fact that a differential diagnosis as eact as possible is necessary for the performance of an aimed therapy.

[Behavior of the heparinocytes after gold therapy].

A reduction of the total leukocytes as well as a significant decrease of heparinocytes and BAI (basophilic age index) can be observed in rheumatoid arthritis in the course of a gold therapy. As some coagulation parameters simultaneously speak in favour of an enhancement of the intravasal coagulation, a partial blocking of the endogenous heparin caused by gold may be supposed. A combined heparin or heparinoid therapy in a low dosage is being recommended for risk patients of the vascular and coagulation system.

[Thrombocyte kinetics before and after gold therapy in rheumatoid arthritis].

Before and after performing a basic therapy with sanocrysin, thromboytic kinetic examinations were carried out in 12 patients suffering from rheumatoid arthritis. The thrombocyte survival time, maximal recovery and thrombocyte turnover were determined. The surface activity values were given in the form of spleen-heart quotients and liver-heart quotients as well as by surplus impulses over these organs. The findings evaluated statistically reveal that even the untreated rheumatoid arthritis will have thrombocytic kinetics deviating from the norm. By influencing RES the gold treatment will lead to an reduction of the enhanced platelet decomposition. The behaviour of single parameters before and after gold therapy are discussed. The examination of thrombocyte kinetic could not identify a damage of thrombopoiesis in the bone-marrow and in the periphery caused by sanocrysin.

[Fibrinolysis before and after gold therapy in rheumatoid arthritis].

In 12 patients with chronic rheumatoid arthritis the immunoglobulins IgG, IgA and IgM were determined according to Mancini and the fibrinolytic split products according to Nilehn before and after a gold treatment of 3 to 6 months. The IgG values were always increased and even after the treatment there was only little regression. The fibrinolytic split products found in 5 cases speak in favour of an enhanced intravasal coagulation, which increased still further after the gold therapy. In spite of clinical improvement the immunological processes and latent actions of coagulation seem to pass almost undiminished.

[Effect of gold therapy in rheumatoid arthritis on leukopoiesis and granulocyte phagocytosis].

In 12 patients with rheumatoid arthritis the investigations of leukopoiesis and granulocytic phagocytosis were carried out before and after a gold therapy. A marked reduction of granulocytic phagocytosis could be observed here which decreased after the gold therapy, yet remained below the values of a normal collective. Partly contrary informations are discussed. The disturbance of the phagocytosis capacity of granulocytes may possibly be due to a rheumatic factor.

[Rapid enzymatic-colorimetric method for detecting organophosphorous insecticide residues in milk].

The authors developed a new enzymatic-colorimetric rapid method for testing milk for residues of organophosphorous insecticides. The test is based on the colour reaction of indoxyl acetate with the esterase of the OROSERAN (Dessau) horse serum which is inhibited by organophosphorous insecticides. Calibration curves were established for Trichlorphon and Dichlorphos (DDVP). The limit of detection lies at 0.02 mg/kg. The time actually needed per sample is about 15 min. The result is obtained within 90 min. Since this procedure requires no clean-up operations, it is labour-saving compared to other thin-layer or gas chromatographic methods. It may be recommended as a screening test for the detection of organophosphorous insecticides in milk samples.

[Effects of plant protective agents and pesticides on the processibility of milk with special consideration to microbiological technology].

The authors studied the effects of the active ingredients contained in 43 pesticides and insecticides on dairy cultures and the suitability of milk contaminated in vitro with selected organophosphorus insecticides. The acid-forming property of yoghurt, kefir, butter and cheese cultures, the clotting property of milk and the ripening process of Camembert were used as testing criteria. The herbicides 2-methyl-4,6-dinitrophenol (DNOC), 2,4, 5-trichlorophenol, 2-methyl-4-chlorophenoxyacetic acid (MCPA), 4-(2-methyl-4-chlorophenoxy) butyric acid (MPCB), 2,4-dichlorophenoxyacetic acid (2,4-D), 2,2-dichloropropionic acid (dalapon), the chlorinated insecticide heptachlorepoxide and the fungicide pentachlorophenol exerted inhibitory effects when applied at concentrations of more than 100 p.p.m., whereas most of the organophosphorus and chlorinated insecticides under investigation produced no detectable effects on the culture activity when used at concentrations up to 100 mg/kg. The clotting property of milk was not impaired by the pesticides and insecticides at concentrations up to 100 mg/kg; the same was true of the ripening process of Camembert.

[Patholgenesis of anemias in chronic liver diseases].

In a complex examination of clinically healthy persons, with postprandial glucosuria or affected with a hereditary diabetes mellitus differences were found in the dynamics of the insulin secretion in oral and venous glucose tolerance dependent on the character of the glucose tolerance. With the deterioration of the latter the insulinaemia increase and prolongs in oral tolerance, in venous tolerance, however, it will be lower and lower. The through analysis of the results refers to the pathogenetic role of the peripheral resistance to the insulin as well as the disturbed secretory function of the islet apparatus. The established connections allow of the derivation of pathogenetically proved criteria for the differentiation of normal, subnormal and pathologic glucose tolerance. The confronting of the insulinaemia with the adiposity of the persons examined detects the fundamental difference between persons with normal and disturbed carbohydrate tolerance. The increase of weight contributes to the manifestation of the genetic defect. From the prophylactic point of view apart from the significance of the adequate dietetic regimen the attention is also drawn to the effect of biguanides.

[Clinical aspects of osteomyelofibrosis and osteomyelosclerosis].

The clinical and radiological parameters, the laboratory findings as well as the results of the nuclear-medical examination established in 78 patients with an osteomyelofibrosis or osteomyelosclerosis are compared with the data existing in literature and the problems of diagnostics and therapy of these diseases are derived from this.

[The question of thrombocyte substitution as a prophylactic measure in splenectomy for idiopathic thrombocytopenic purpura (ITP)].

The problem of intraoperative thrombocyte transfusion in splenectomy of patients with ITP is discussed. The indication for splenectomy cannot be equalized with that for intra-operative platelet substitution. Thrombocyte kinetic examinations with a concurrent determination of the thrombocyte turnover enable those patients to be recognized by their bleeding tendency who are particularly endangered by operations. The intraoperative platelet substitution should be limited to those patients with ITP whose turnover is markedly lowered as a manifestation of reduced thrombocytopoiesis. In these cases it is advisable to shift splenectomy to a later date.
