The effect of cervical dilatation by laminaria on the plasma levels of 15-keto-13,14-dihydro-PGF2 alpha.

Laminaria tents were inserted to induce cervical dilatation prior to suction abortion in 42 primigravidae. The plasma level of 15-keto-13,14-dihydro-PGF2 alpha, the principal metabolite of prostaglandin F 2 alpha, increased during the dilatation period. The gentle dilatation by laminaria tents probably induces an endogenous synthesis of prostaglandins causing a softening of the cervix.

Blood levels of 15-keto-13, 14-dihydroprostaglandin F(2alpha) during the postpartum period in primiparous cows.

Plasma levels of the PGF(2alpha) metabolite 15-keto-13, 14-dihydroprostaglandin F(2alpha) were determined postpartum in 51 primiparous Black and White Lowland cows. The highest geometric mean was 1702 pmol/l on day 3 and lowest 190 pmol/l on day 21. It should be noted that variation between animals in the concentration of the metabolite is high. For instance, on days 4, 10, 16 and 22, concentration of metabolite ranged from 775-2500, 209-2450, 45-851 and 30-398 pmol/l, respectively. The duration of the massive postpartum release of PGF(2alpha) could be determined in only 29 cows. Significant correlations were found between the duration of elevated PGF(2alpha) metabolite levels and the time required for completion of uterine involution (r = -0.41, P < 0.05) and between the duration of increased PGF(2alpha) metabolite levels and the interval from parturition to occurrence of the first ovulation followed by a normal luteal phase length (r = -0.37, P < 0.05). The occurrence of the first ovulation followed by a normal luteal phase length in the 29 cows was positively correlated with the time needed for completion of uterine involution (r = 0.54, P < 0.01).

Blood levels of progesterone and 15-Keto-13, 14-dihydro prostaglandin F(2alpha) during the estrous cycle of oxytocin-treated cows.

Six non-lactating Holstein cows were injected with 230 iu oxytocin subcutaneously twice daily from days 2 through 6 of the cycle. Controls (n=6) were given saline injections using the same schedule. Blood samples were collected at frequent intervals before and after each saline or oxytocin injection. Progesterone and 15-Keto-13, 14-dihydro prostaglandin F(2alpha) (PGFM), the major metabolite of prostaglandin F(2alpha), were analysed by radio-immunoassay. Oxytocin injections significantly increased plasma prostaglandin concentrations on days 2 and 3 when compared with the controls. In two oxytocin-treated cows, the cycle was shortened to 10 and 12 days. Estrus was preceded by a PGF(2alpha) release very similar to that preceding spontaneous estrus. Two of the oxytocin-treated cows showed estrus on day 21 and 22 preceded by luteolytic release of PGF(2alpha). Two oxytocin-treated cows developed cystic corpora lutea and had not shown heat when the ovaries were removed four weeks later. All oxytocin-treated cows showed a slower progesterone increase through day 8 than the controls. The study shows that endocrine events preceding cycle alterations in oxytocin-treated cows involve release of PGF(2alpha) and lowered levels of progesterone.

Renal excretion of prostaglandin metabolites, arginine vasopressin, and sodium during endotoxin and endogenous pyrogen induced fever in the goat.

Responses to intravenous injections of an endotoxin (E. coli-lipopolysaccharide, 1 microgram/kg b.wt.) and endogenous pyrogen were studied in euhydrated and hyperhydrated goats. The biphasic febrile response to the endotoxin was associated with a pronounced increase in the renal excretion of measured prostaglandin (PG) metabolites (11-ketotetranor PGF metabolites). This increase was time-correlated with the elevation of the rectal temperature, and (in hyperhydrated animals) with an inhibition of the water diuresis and an increase in renal excretion of arginine vasopressin (AVP). Other effects of the endotoxin were an immediate depression of renal Na and K excretion followed by the development of pronounced natriuresis, and a reduction of plasma Fe and Zn concentrations. The appearance of the febrile reactions (peripheral vasoconstriction and shivering) was accompanied by miosis. The maximum elevation of the rectal temperature was significantly greater during euhydration than during hyperhydration. Also endogenous pyrogen elicited miosis concomitant with febrile reactions, and an elevation of the renal excretion of PG metabolites which was closely correlated in time with the monophasic febrile response, and (during hyperhydration) with temporary inhibition of the water diuresis and an increase in the renal AVP excretion. However, the responses were much weaker than the corresponding endotoxin effects. No appreciable changes in renal excretion of Na and K were observed in response to the endogenous pyrogen. It is concluded that the observed effects on renal cation excretion were manifestations of direct endotoxin influences on kidney function.(ABSTRACT TRUNCATED AT 250 WORDS)

Concentrations of 15-keto-13, 14-dihydro-prostaglandin F2 alpha in the mare during spontaneous and oxytocin induced foaling.

Changes in plasma 15-keto-13, 14-dihydro-prostaglandin F2 alpha were monitored at frequent intervals before, during and after spontaneous deliveries (three mares) and foalings induced by oxytocin (eight mares). No evidence of increased concentrations of the prostaglandin metabolite was observed in the final 10 days of gestation. In spontaneously delivering mares, there was a marked increase from 3 ng/ml at -125 mins to 18 ng/ml at -65 mins to the highest observed value of 182 ng/ml at 20 mins pre-partum. Following delivery, concentrations declined rapidly to around 0.2 ng/ml. Further release of prostaglandins was seen on Days 1 and 3 post partum. In oxytocin induced mares, maximal concentrations of about 100 ng/ml were observed to occur very close to the time of delivery. Large increases were observed as early as 2 mins following oxytocin injection. The significance of these findings is discussed in relation to parturient and post parturient events and changes in levels of the hormone relaxin during the same period.

Dilatation of the cervix by oral PGE2 before first-trimester termination of pregnancy.

Twenty-four patients--all primigravidae with an especially narrow, rigid cervix--were given PGE2 tablets or placebo tablets in order to prime the cervix before first trimester termination of pregnancy. 1.5 mg PGE2 or placebo tablets were administered hourly for 12 hours on the day before vacuum aspiration. The diameter of the cervix was measured by means of a Hegar dilator prior to administering the tablets and also before the vacuum aspiration the following day. The plasma level of 15-keto-13,14-dihydro-PGF2 alpha was determined before the treatment and again before the vacuum aspiration. All the 12 patients treated with PGE2 tablets had an average change in cervix diameter of 2 mm, whereas in the placebo group the average change was 0.58 mm.

Vasopressin and prostaglandins in premenstrual pain and primary dysmenorrhea.

Both vasopressin and PGF2 alpha are effective uterine stimulants in the non-pregnant human uterus, especially around the onset of menstruation. In order to clarify the relationship of these hormones to menstrual pain, plasma concentrations of vasopressin and two prostaglandin metabolites (15-keto-13,14-dihydro-PGF2 alpha and 11-ketotetranor PGF metabolites) were measured in serial blood samples taken premenstrually and during menstruation. Five women with premenstrual pain gave 7-9 blood samples at intervals of 30 minutes on the day preceding the onset of menstruation. From 5 women with severe primary dysmenorrhea a corresponding series of blood samples were taken during the first day of menstruation. Two groups of 5 women with no symptoms served as controls, either premenstrually or during menstruation. In the women with premenstrual pain the vasopressin concentrations were significantly higher than in the corresponding control group. Even higher and markedly fluctuating vasopressin levels were found in the women with dysmenorrhea who, in general, had more intense pain than the women with premenstrual symptoms. In the group with dysmenorrhea there was also a significant rise in plasma concentration of the PG metabolites. No such increase was seen in the group with premenstrual pain. It is concluded that the pathophysiology of premenstrual pain could imply increased vasopressin secretion. The more severe pain in primary dysmenorrhea seems to be the result of a combined effect of vasopressin and PGF2 alpha.

Laparoscopy in post partum sows with special emphasis on the effects of uterine manipulation on the release of prostaglandin F2 alpha.

One hundred laparoscopies were performed on 30 primiparous post partum sows and complications during and after examinations recorded. Nineteen of these animals were separated into two groups and subjected to different treatments: group 1 (10 sows), with conventional laparoscopy; group 2 (nine sows) with two minutes extra manipulation of the uterus after laparoscopy. Blood was sampled before and after the operations. The plasma was analysed for its content of 15-keto-13,14-dihydroprostaglandin F2 alpha by radioimmunoassay. The results indicate that laparoscopies were successful and can be used in post partum sows during lactation and after weaning without any severe complications. In some sows laparoscopy alone or laparoscopy followed by manipulation of the uterus for two minutes caused a peak of the prostaglandin metabolite. However the height and duration of this peak were not sufficient to influence reproductive performance in pigs.

Concentrations of prostaglandin F2 alpha in the pig oviductal fluid.

Oviductal fluid was continuously collected from cycling, unrestrained gilts during late proestrus, standing heat and early luteal phase. Collection was performed via cannulae exteriorized to single vented collection tubes attached to the skin. In some cases one of the oviducts was transected at isthmic level to compare fluid production rates with those of the contralateral intact tube. A consistently elevated fluid production was observed during proestrus and standing heat, being highest at the second day of the cycle, thereafter decreasing. No significant differences were seen between daily collection rates from right or left nor between intact and isthmic transected oviducts. Levels of PGF2 alpha were determined by RIA in the collected samples. Relatively large day to day fluctuations in PGF2 alpha values were evident and a consistent relationship was found between concentration and the stage of the estrous cycle. The highest PGF2 alpha mean values (up to 50 ng/ml fluid) were also found on the second day of standing heat, concurrently periovulatory.

Involvement of prostaglandins in vasopressin stimulation of the human uterus.

The involvement of prostaglandins (PG) in the vasopressin (VP) action on the human uterus was investigated in healthy women during three menstruations. Intrauterine pressure was recorded and total pressure area measured. Repeated plasma samples were taken for estimations of arginine(A)- and lysine(L)-VP, 15-keto-13,14-dihydro-PGF2 alpha and 11-ketotetranor PGF metabolites. During the first menstruation LVP was infused in a dose of 0.08 micrograms/min. During the second menstruation the infusion of LVP was repeated with the same dose, but 70 min before infusion the women received an oral dose of 500 mg of naproxen. During the third menstruation PGF2 alpha was administered intravenously in a dose of 25 micrograms/min. LVP infusion per se caused a significant increase in uterine activity and plasma levels of LVP and PG metabolites. When the women were pretreated with naproxen practically the same uterine activity was induced and closely similar plasma levels of LVP were obtained, but the levels of PG metabolites decreased significantly in comparison with the first series of experiments. Infusion of PGF2 alpha caused an increase in uterine activity but no change in the plasma levels of AVP. The results indicate that uterine stimulation with VP is possible without an obligatory last step of PG synthesis and release. The results also support the concept that an elevated VP level in primary dysmenorrhoea may be of aetiological importance and is not just released as a 'stress'-hormone because of the dysmenorrhoeic pain.
