The effects of intravenous infusion of solutions with different colloid osmotic pressures on rat mesenteric lymph pressure.

The effects of intravenous infusion of solutions of varied colloid osmotic pressure on mesenteric lymph pressure were measured by a glass micropipette. The lymph pressure was 4.0 +/- 1.6 cm H2O between the second and third valve of rat mesenteric lymphatic vessels with diameter of 101 +/- 25 micron (mean +/- SD). Intravenous infusion of 1 ml/100g body weight (B.W.) of isotonic saline, 5% glucose solution, and 20% glucose solution increased the lymph pressure by 1.6 +/- 0.5 cm H2O, 1.5 +/- 0.3 cm H2O and 2.0 +/- 1.0 cm H2O, respectively. Infusion of 2 ml/100g B.W. of the same solutions increased the lymph pressure by 3.3 +/- 1.3 cm H2O, 3.3 +/- 1.6 cm H2O and 3.7 +/- 0.8 cm H2O, respectively. Infusion of hydroxyethyl starch solution, with colloid osmotic pressure of 25 mmHg, minimally altered the lymph pressure. One ml/100g B.W. infusion of low molecular weight dextran solution, with colloid osmotic pressure of 162 mmHg, decreased the lymph pressure by 1.3 +/- 0.9 cm H2O, and 2 ml/100g B.W. infusion of this solution decreased the lymph pressure by 2.0 +/- 1.0 cm H2O. The different effects on lymph pressure derive from the differences in colloid osmotic pressure of these solutions in conformity with the Starling hypothesis.

Protective effect of Nocardia rubra cell wall skeleton on experimental infection in normal and immunosuppressed mice.

The protective effect of Nocardia rubra cell wall skeleton (N-CWS) on experimental infections was investigated in normal and immunosuppressed mice. Pretreatment with N-CWS provided protection against acute systemic infections due to Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, Serratia marcescens and Listeria monocytogenes in normal mice. N-CWS administered before or after challenge also showed protective activity against Herpes simplex virus infection in normal mice. N-CWS was the most effective against extracellular parasitic Pseudomonas aeruginosa infection when administered 1 day before challenge, but displayed the most potent protective activity against infection with Listeria monocytogenes, a facultative intracellular parasite, when administered 7 to 14 days before challenge. In mice with subcutaneous Pseudomonas aeruginosa infection, N-CWS suppressed abscess formation and decreased the viable cell count in the resultant abscess foci when administered either intraperitoneally or subcutaneously to the infected site. In addition, treatment with N-CWS markedly restored host defense ability against pseudomonal infection in mice immunosuppressed with cyclophosphamide, hydrocortisone and X-ray irradiation.

In vivo activation of functional properties in mouse peritoneal macrophages by Nocardia rubra cell wall skeleton.

Exudative cells in the peritoneal cavity of mice, particularly polymorphonuclear leukocytes significantly increased 1 day after intraperitoneal injection of Nocardia rubra cell wall skeleton (N-CWS). Macrophages and lymphocytes significantly increased 4 to 7 days after injection. N-CWS also enhanced peritoneal macrophage functions such as phagocytosis of latex particles, production of superoxide anion, production and secretion of lysosomal enzymes such as beta-glucuronidase, lysozyme and acid phosphatase, phagocytosis and intracellular killing of bacteria, and in vitro chemotaxis. The phagocytic function of the reticuloendothelial system was also enhanced. These results indicate that macrophages were activated in vivo by N-CWS.

Establishment of a cell line of gallbladder carcinoma (GBK-1) producing human colony stimulating factor.

A cell line designated GBK-1 was established from a patient with anaplastic carcinoma of the gallbladder, marked neutrophilia and fever, and has been propagated for the past 18 months. The cells grew as a monolayer sheet with a doubling time of 43 hr. The GBK-1 cells were of a pleomorphic polygonal epitheloid shape and they were transplantable into nude mice. Mice bearing the tumor developed marked granulocytosis. In the culture supernatant of GBK-1 cells, high colony stimulating factor (CSF) activity was evident with both human bone marrow cells and C57BL mouse bone marrow cells, and granulocytic colonies, macrophage colonies or granulocyte-macrophage mixed colonies were produced. The CSF activity was distributed in the molecular weight range of 25,000 to 60,000 with two distinct peaks at molecular weights of approximately 50,000 and 30,000. CSF activity was inactivated by heat treatment at 70 degrees for 30 min. GBK-1 is a new human cell line that produces heat-labile human GM-CSF.

Effect of Nocardia rubra cell wall skeleton on humoral and cellular factors related to immune response in mice and guinea pigs.

The in vitro and in vivo effects of Nocardia rubra cell wall skeleton (N-CWS) on induction of serum ceruloplasmin, hemopexin, haptoglobin, complement and interferon in mice and guinea pigs are described. After intraperitoneal administration of N-CWS to mice, serum hemopexin increased markedly and also induced an increase of haptoglobin, but not of ceruloplasmin. The drug activated complement in vitro but did not affect serum complement level in guinea pigs. Intravenous N-CWS induced serum interferon in mice, although there were strain differences. Addition of N-CWS to mouse spleen cell culture also induced interferon. Intraperitoneal N-CWS increased peritoneal cells in mice: in particular polymorphonuclear leukocytes increased soon after dosing, followed by macrophages and lymphocytes. However, the drug did not affect peripheral leukocyte counts or alter peripheral cell population.

[The effect of artificial hypotension on postischemic cerebral edema in the reperfusion of middle cerebral arterial occlusion in cats--experimental study].

The effect of hypotension on postischemic cerebral edema was studied by the occlusion-reperfusion model of the middle cerebral artery (MCA) of a cat. The middle cerebral artery was transorbitally occluded for 3 hrs and reperfused for 3 hrs. We chose cats whose CBF decreased lower than 10 ml/100 g/min by the MCA occlusion. We studied 10 cats as a control group and 6 cats as the hypotension group. The mean arterial blood pressure was lowered artificially about 50 mmHg from the previous value using ATP and dipyridamole just before and during the reperfusion. The CBF was measured by the hydrogen clearance method and extradural pressure was recorded. The water content of cerebral hemisphere was measured by the drying-weighing method after sacrifice. In the control group the preocclusive value of CBF was 65.9 +/- 20.6 ml/100g/min and the value just after reperfusion was 35.0 +/- 21.5 ml. The value 3 hours after reperfusion was 8.2 +/- 11.0 ml. In the hypotension group, these values were 53.9 +/- 14.0 ml, 40.6 +/- 19.6 ml and 18.1 +/- 12.2 ml respectively. The ICP increased from 9.1 +/- 8.6 to 90.0 +/- 22.6 mmHg in the control group and in the hypotension group from 4.5 +/- 6.5 to 37.7 +/- 10.1 mmHg. The water content of the affected hemisphere was 79.7 +/- 0.5% in the control group and 79.1 +/- 0.5% in the hypotension group. The hypotension didn't change the CBF just after reperfusion and also maintained it more than the control until 3 hrs after the reperfusion.(ABSTRACT TRUNCATED AT 250 WORDS)

[Metabolic response in the acute stage of cerebral infarction--with special reference to oxygen consumption and resting metabolic expenditure].

Oxygen consumption, carbon dioxide production, respiratory quotients, and resting metabolic expenditure were measured in 23 patients with cerebral infarction in their acute stage. Metabolic measurement were carried out with the technique of indirect calorimetry on their admission within two days from the onsets. At the same time, urine was collected twenty-four hours to measure urinary catecholamine excretion. Mean value of resting metabolic expenditure was 115.1% and this positively correlated with urinary catecholamine, especially noradrenaline excretion. Maximum value of resting metabolic expenditure was up to 187.1% of that expected for an uninjured resting person of equivalent age, sex, and body surface area. Oxygen consumption and carbon dioxide production also positively correlated to urinary catecholamine excretion. On the other hand, respiratory quotients did not have any significant correlation with oxygen consumption, carbon dioxide production, resting metabolic expenditure, or urinary catecholamine excretion. From those facts, it was implied that overactivity of sympathetic nervous system existed in their acute stage of cerebral infarction, and the overflow might directly influence oxygen consumption, carbon dioxide production, and resting metabolic expenditure, and moreover we had to take into consideration of hyper-metabolic state to manage patients with ischemic cerebrovascular diseases as malnutrition might cause weight loss and immune incompetence.

[The effect of low dose dopamine on CBF in patients with subarachnoid hemorrhage].

Cerebral blood flow was measured in 10 patients with subarachnoid hemorrhage using the 133Xe inhalation methods. Regional cerebral blood flow (r-CBF) values were calculated with initial slope index, their values were compared with the blood flow response to the administration of low dose dopamine (DA). CBF values of just after and continuous administration of DA were compared with control values. The measurement of r-CBF was performed immediately after the infusion of 5 micrograms/kg/min DA for 30 min. The focal ischemic lesions decreased, bi-hemispheric mean CBF value (7 cases without A-Com patients) increased by 10.3% significantly (paired T test), affected hemispheric mean CBF value increased by 8.7% significantly and non-affected hemispheric mean CBF value increased by 16.6% significantly. The PaCO2 value didn't change but the mean systemic blood pressure increased by 5.1% significantly. The CBF values of continuous administration of DA for 10 days to 2 weeks were measured. The CBF values increased during the administration of DA. The mean CBF values increased without the steal phenomenon and the mean arterial blood pressure increased slightly after the administration of DA. These phenomena were observed more significantly in non affected side than affected side. We suggest that the administration of DA increase the CBF values in patients with subarachnoid hemorrhage due to the stimulation of DA receptor.

[Visual disturbances in moyamoya disease].

Visual disturbances in moyamoya disease are evaluated. Forty-three among our series of 178 cases of moyamoya disease were suffered from various visual symptoms such as visual field defect, attack of blindness, decreased visual acuity, scintillating scotoma. These visual manifestations were resulted from the occlusive lesions in the posterior circulation. In comparison with the cases of adult onset, those of juvenile onset tend to develop visual disturbances. STA-MCA anastomosis with EMS exerts indirect redistribution effect upon the posterior circulation in moyamoya disease. In most cases, improvement in the visual disturbances followed these bypass operations. However, in five cases, omentum transplantation to the occipital lobe as the method of direct revascularization were needed.

Antileukemic effect of coral-prostanoids clavulones from the stolonifer Clavularia viridis on human myeloid leukemia (HL-60) cells.

To elucidate the biological activities of coral-prostanoids, clavulones, discovered from the Japanese stolonifer Clavularia viridis, we examined the effect of clavulone on the cell growth of human cancer (human promyelocytic leukemia (HL-60) cells and HeLa cells) and normal (Chang liver cells and lung fibroblasts) cells in vitro. Clavulone showed strong antiproliferative and cytotoxic activities in the human cells and it had some selectivity to leukemic (HL-60) cells over other HeLa cells or normal cells on the basis of the IC50 values and cytotoxic effect of the cells. The IC50 value of clavulone in the HL-60 cells was about 0.4 microM (0.2 micrograms/ml). Over 1.0 microM (0.5 micrograms/ml), clavulone showed a significant cytotoxic activity on the HL-60 cells. The data on DNA synthesis and flow cytometric analysis revealed that clavulone arrests the cells in the G1-phase and inhibits the cell growth of HL-60 cells by inhibiting S-phase DNA synthesis. These results suggest that clavulone has a potent antileukemic effect on HL-60 cells.

Effect of phenobarbital on the activity of RNA polymerase II during diethylnitrosamine-induced hepatocarcinogenesis in the rat.

The effects of dietary phenobarbital (PB) on RNA polymerase II (nucleoside triphosphate: RNA nucleotidyltransferase, EC 2. 7. 7. 6) in the liver of male Sprague-Dawley rats previously administered diethylnitrosamine (DENA) were studied. When a diet containing 0.05% phenobarbital was administered, the activity of DNA-dependent RNA polymerase II in the liver nuclei of rats was temporarily increased 1 and 2 weeks after the onset of feeding and expressed to be relatively higher afterwards, compared with the activity in rats fed the control diets.

Hyaline bodies in ameloblastoma: histological and ultrastructural observations.

An exceedingly rare case of ameloblastoma, plexiform type, which contained numerous hyaline bodies was reported. Hyaline bodies occurred within the parenchyma, and showed similar histochemical reactions to those of hyaline bodies in odontogenic cysts. Ultrastructurally, hyaline bodies found in the present case of ameloblastoma were composed of a peripheral electron-dense area and central electron-lucent area. Well-developed endoplasmic reticulum containing electronlucent and homogeneous substance were seen in the epithelial cells adjacent to hyaline bodies. Furthermore, electron-dense spherical granules, with or without limiting membrane, were scattered in the cytoplasm of epithelial cells. It is thought that hyaline bodies in the present case of ameloblastoma are particular products formed by odontogenic epithelial cells.

[The antiinflammatory effect of budesonide].

The systemic and topical antiinflammatory activities of budesonide (B) were studied in rats and mice and compared with those of commercially available steroids. Betamethasone 17-valerate (BV) was used as the main reference compound, and fluosinolone acetonide (FA), hydrocortisone 17-butyrate (HB) and hydrocortisone 21-acetate (HA) were also used. B given systemically had stronger antiinflammatory effect than BV on carrageenin edema, cotton pellet granuloma, adjuvant arthritis, croton oil edema, PCA reaction, Arthus reaction, contact hypersensitivity and histamine or serotonin skin reaction. The potency of antiinflammatory activity of the 5 compounds in carrageenin edema, croton oil edema and contact hypersensitivity tests was in the order of FA, B, BV, HB and HA. B given locally also produced stronger antiinflammatory effects than BV on carrageenin edema, cotton pellet granuloma, croton oil edema and contact hypersensitivity. The order of potency of the 5 compounds in carrageenin edema, croton oil edema and contact hypersensitivity tests was the same as by systemic application. In general, the ratio of the dose required to cause atrophy of the thymus and adrenals to the dose required to produce the antiinflammatory effect was the greatest with B by both systemic and local application. The results suggest that B has a stronger antiinflammatory activity with fewer systemic side effects than conventional steroid compounds.
