Tumor-induced platelet aggregation and growth promoting factors as determinants for successful tumor metastasis.
Two clones isolated from a metastatic variant of mouse colon adenocarcinoma 26; the high metastatic NL-17 and the low metastatic NL-44, induced similar platelet aggregation in vitro. Heterotypic aggregates of tumor cells and platelets were injected i.v. into mice. The lung colonization potential of the NL-17 was dependent on the extent of tumor cell platelet aggregation. This clearly indicates that the interaction of tumor cells with platelets can lead to enhanced tumor metastasis possibly through more efficient intravascular arrest of the heterotypic aggregates. The interaction of tumor cells with platelets could thus be an important determinant for successful metastasis. NL-44, however, did not form pulmonary metastasis even after the tumor cells formed heterotypic aggregates with platelets, suggesting that tumor metastasis, is dependent on the intrinsic nature of tumor cells. Lung extract enhanced the growth of NL-17 more effectively than that of NL-44. These results suggest that, in addition to the interaction of metastatic cells and platelets, host factors including growth-promoting factors might also have an important role in tumor metastasis.
Multifocal electroretinogram in multiple evanescent white dot syndrome.
Electrophysiologic findings including multifocal electroretinogram and visual evoked cortical potentials were studied in a patient with multiple evanescent white dot syndrome. A 19-year-old woman was diagnosed as having multiple evanescent white dot syndrome because of decreased visual acuity, white dots on fundus examination and hyperfluorescence of the white dots in the right eye. The amplitude of the flicker electroretinogram was reduced, but the single-flash electroretinogram was within the normal range. The P100 latency of the pattern visual evoked cortical potentials was slightly prolonged. The amplitudes of multifocal electroretinogram were markedly reduced in the area corresponding to the scotoma and moderately reduced in other regions of the central field in the affected eye. The results suggest that the retinal dysfunction area is wider than that detected by subjective methods or conventional electrophysiological examinations. The delayed recovery of the visual evoked cortical potential latency suggests the involvement of the optic nerve in multiple evanescent white dot syndrome.
Assessment of the presence and severity of esophagogastric varices by splenic index in patients with liver cirrhosis.
PURPOSE: To determine whether spleen size is related to the severity of esophageal varices or associated gastric varices and liver functions in patients with cirrhosis. METHOD: The authors retrospectively studied spleen size on CT (splenic index [SI] = length x width x height of the spleen), liver functions, and the results of esophagogastric endoscopy in 110 patients with cirrhosis. They also analyzed SI in 112 controls. RESULTS: In controls, body weight, height, and age affected the SI. The SI in patients with uncompensated cirrhosis was greater compared with the SI in those with well-compensated disease (p = 0.0363). The SI in patients with esophageal varices was greater than in patients without esophageal varices (p<0.0001), but patients with and without gastric varices had similar SI values. The SI in patients with the red color signs (red wale marking, cherry red spot, and hematocystic spot) on esophageal varices or with risky varices (enlarged tortuous varices with beady, nodular, or tumor shape associated with red color signs) was greater than in patients without these signs (p = 0.0029 and p = 0.0030, respectively). CONCLUSION: The SI is a good indicator of the severity of esophageal varices and hepatic functional reserve in patients with cirrhosis.
Terminal limb congenital malformations: analysis of 523 cases.
A total of 523 Japanese patients with terminal limb congenital malformations were reviewed for the characteristic features, and especially for type correlation between hand and foot anomalies. There were 281 cases of hand involvement alone, 149 cases of foot involvement alone, and 93 cases of combined hand and foot involvement. Polydactyly was commonly preaxial in the hand and postaxial in the foot, in contrast to the well-known postaxial dominance in the hand among whites. Hand anomalies were likely to show the same Swanson type of anomaly as the foot, with the affected digital ray likely to be identical in both hand and foot.
Preparation of heat-labile antigens on air-dried cytologic specimens for immunocytochemical analysis.
OBJECTIVE: To demonstrate the efficacy of Zamboni's solution and storage medium for immunocytochemistry of Ki-67, p53 tumor suppressor gene product and CD3 on long-preserved, air-dried cytologic specimens. STUDY DESIGN: Air-dried imprint cytologic specimens obtained from 13 lymphatic tissues and 13 colorectal cancers were fixed with Zamboni's solution and stored for various periods in storage medium including 50% glycerol at -10 to -20 degree C. Mouse monoclonal antibody PC, PAb 1801 and anti-Leu-4 were used for immunostaining. RESULTS: Immunocytochemical identification of Ki-67, p53 and CD3 was possible on specimens stored for 36, 32 and 16 weeks, respectively. CONCLUSION: Our method, using Zamboni's solution and storage medium, is feasible for immunocytochemistry on long-preserved, air-dried cytologic specimens using the three antibodies were employed.
